Real-time multiple-particle tracking: applications to drug and gene delivery.

Complex biological environments, such as the cell cytoplasm or the mucus lining the airways of the lungs, can pose significant barriers to efficient therapeutic drug and gene delivery. Biological barriers are particularly important in controlled drug delivery applications that utilize a large carrier particle, such as a liposome or a polymer micro- or nanosphere. The dynamic transport of particulate drug and gene delivery vehicles through these barriers is poorly understood, having been primarily studied with static methods in the past. Recently, the transport of synthetic drug and gene carrie

[Visceral leishmaniasis: new drugs].

The standard treatment of visceral leishmaniasis is pentavalent antimony (meglumine antimoniate or sodium stibogluconate), but toxicity is frequent with this drug. Moreover, antimony unresponsiveness is increasing, both in immunocompetent and in immunosuppressed patients. Amphotericin B is a polyene macrolide antibiotic that binds to sterols in cell membranes. It is the most active antileishmanial agent in use. Its infusion-related and renal toxicity may be reduced by lipid-based delivery. Liposomal amphotericin B (Ambisome) seems to be less toxic than other amphotericin B lipid formulations (

[Exoftalmos and palpebral ptosis in the immunocompromised patient].

The mucormycosis is an opportunistic mycosis which has a high rate of global mortality with a bad prognosis. The present clinical case shows a patient attended in the Intensive Care Unit because of a breathing insufficiency secondary to a pneumonia. Among his antecedents there is a larynx carcinoma totally remittent after chemo- and radiotherapy. He develops an infrapalpebral injury which spreads rapidly in a bilateral way acquiring a necrotic look that it did not have initially. Having that evolution, it is implemented a treatment with liposomal amphotericin B and granulocyte colony-stimulati

In vivo clearance of glucuronoxylomannan, the major capsular polysaccharide of Cryptococcus neoformans: a critical role for tissue macrophages.

Cryptococcus neoformans produces a life-threatening meningitis in patients who are immunocompromised by AIDS. A striking feature of cryptococcosis in AIDS is high serum levels of the major capsular polysaccharide, glucuronoxylomannan (GXM). Soluble GXM has numerous biologic activities that may contribute to the pathogenesis of infection. The objective of the study was to further understand in vivo processing of GXM. Mice were injected intravenously with GXM, and the tissue distribution was determined. A macrophage suicide technique that used liposome-encapsulated dichloromethylene diphosphonat

Amelioration of the dystrophic phenotype of mdx mice using a truncated utrophin transgene.

Duchenne muscular dystrophy (DMD) is a severe, progressive muscle-wasting disease that causes cardiac or respiratory failure and results in death at about 20 years of age. Replacement of the missing protein, dystrophin, using myoblast transfer in humans or viral/liposomal delivery in the mouse DMD model is inefficient and short-lived. One alternative approach to treatment would be to upregulate the closely related protein, utrophin, which might be able to compensate for the dystrophin deficiency in all relevant muscles. As a first step to this approach, we have expressed a utrophin transgene a

Smart nanoparticle delivery systems for curcumin: a targeted strategy to enhance anticancer efficacy and bioavailability.

Curcumin, a natural polyphenol derived from Curcuma longa, exhibits potent multimodal anticancer activity by modulating critical oncogenic pathways (e.g., NF-κB, STAT3, PI3K/Akt/mTOR), inducing apoptosis, suppressing angiogenesis, and reversing multidrug resistance (MDR). However, its clinical translation is severely hindered by poor aqueous solubility, rapid metabolism, and negligible oral bioavailability (typically <1% in serum), which result in subtherapeutic concentrations at tumor sites. Smart nanoparticle delivery systems have emerged as a transformative strategy to overcome these limita

Targeted Immunoliposomal Delivery of a 5-Fluorouracil Analog in EGFR-Expressing Pancreatic Cancer Models.

IntroductionDysregulated epidermal growth factor receptor (EGFR) signaling is a key mechanism driving cancer progression and metastasis. Owing to its frequent overexpression in pancreatic cancer (PCa), EGFR has become a desirable molecular target for targeted therapies. XYZ-I-73 (N-(5-fluoro-2-oxo-1-(tetrahydrofuran-2-yl)-1,2-dihydropyrimidin-4-yl) dodecanamide), a structural analog of 5-fluorouracil (5-FU), has been previously synthesized and shown to exhibit cytotoxicity against PCa cells.MethodsXYZ-I-73 was entrapped in liposomes via thin-film hydration and subsequently conjugated to EGFR a

Targeted nanosensitizer-augmented sono-immunotherapy with STING agonist to remodel the immune microenvironment in hepatocellular carcinoma.

Hepatocellular carcinoma (HCC) is the most common primary malignant disease of the liver. Although immunotherapy offers new opportunities for treating advanced HCC, its therapeutic effect is still limited by the immunosuppressive tumor microenvironment (TME). Herein, a nanosensitizer RGD@Ce6@MSA-2@Liposome (RCM-Lip) is synthesized to specifically initiate the HCC tumor immune microenvironment through sonodynamic therapy (SDT)-triggered immunogenic cell death (ICD) and MSA-2-activated cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) pathway. RCM-Lip consists of a sonosensit

Myeloid Cell Derived IL1β Contributes to Pulmonary Hypertension in HFpEF.

BACKGROUND: Pulmonary hypertension (PH) in heart failure with preserved ejection fraction (HFpEF) is a common and highly morbid syndrome, but mechanisms driving PH-HFpEF are poorly understood. We sought to determine whether a well-accepted murine model of HFpEF also displays features of PH, and we sought to identify pathways that might drive early remodeling of the pulmonary vasculature in HFpEF. METHODS: Eight-week-old male and female C57BL/6J mice received either Nγ-nitro-L-arginine methyl ester and high-fat diet or control water and diet for 2, 5, and 12 weeks. The db/db mice were studied

Formation of hydrated PEG layers on magnetic iron oxide nanoflowers shows internal magnetisation dynamics and generates high in-vivo efficacy for MRI and magnetic hyperthermia.

Multicore magnetic iron oxide nanoparticles, nanoflowers (NFs), have potential biomedical applications as efficient mediators for AC-magnetic field hyperthermia and as contrast agents for magnetic resonance imaging due to their strong magnetic responses arising from complex internal magnetic ordering. To realise these applications amenable surface chemistry must be engineered that maintain particle dispersion. Here a catechol-derived grafting approach is described to strongly bind polyethylene glycol (PEG) to NFs and provide stable hydrogen-bonded hydrated layers that ensure good long-term col

Preparation, Physicochemical Properties, and In Vitro Toxicity towards Cancer Cells of Novel Types of Arsonoliposomes.

Arsonoliposomes (ARSL) are liposomes that incorporate arsonolipids (ARS) in their membranes. They have demonstrated significant toxicity towards cancer cells, while being less toxic towards normal cells. In this study, we sought to investigate the possibility to prepare novel types of arsonoliposomes (ARSL) by incorporating a lipidic derivative of curcumin (TREG) in their membrane, and/or by loading the vesicles with doxorubicin (DOX). The final aim of our studies is to develop novel types of ARSL with improved pharmacokinetics/targeting potential and anticancer activity. TREG was incorporated

Relative effectiveness of additive pain interventions during vaccination in infants.

BACKGROUND: Vaccine injections can cause acute pain and distress in infants, which can contribute to dissatisfaction with the vaccination experience and vaccine hesitancy. We sought to compare the effectiveness of additive pain interventions administered consistently during vaccine injections in the first year of life. METHODS: We conducted a multicentre, longitudinal, double-blind, add-on, randomized controlled trial. Healthy infants were randomly assigned to 1 of 4 levels of pain management for all vaccine injections at 2, 4, 6 and 12 months: (i) placebo control; (ii) parent-directed video

Pleural effusion as the initial clinical presentation in disseminated cryptococcosis and fungaemia: an unusual manifestation and a literature review.

BACKGROUND: Cryptococcus neoformans infection usually presents as chronic meningitis and is increasingly being recognized in immunocompromised patients. Presentation with pleural effusion is rare in cryptococcal disease; in fact, only 4 cases of pleural effusion as the initial clinical presentation in cryptococcosis have been reported in English-language literature to date. We report the first case of pleural effusion as the initial clinical presentation in a renal transplant recipient who was initially misdiagnosed with tuberculous pleuritis but who then developed fungaemia and disseminated c

Structural and Mechanistic Insights Into GSDME Pore Formation and Functional Regulation.

Gasdermin E (GSDME) is a unique member of the gasdermin family linked to deafness, cancer, and neurodegenerative diseases, and mediates chemotherapy-induced pyroptosis; however, its pore-forming mechanism remains poorly understood. Here, we present cryo-EM structures of GSDME pores with oligomeric stoichiometries ranging from 23 to 29 protomers. Using mutagenesis, liposome leakage assays, and analysis of cancer-associated mutations, we identified key residues responsible for membrane binding, shallow membrane insertion, and oligomerization through three distinct structural interfaces. Multiplexed spatial screening of lipid-based nanoparticles for extrahepatic messenger RNA targeting.

Lipid-based and other types of nanoparticles (NPs) enable target- and organ-specific, efficient extrahepatic delivery of synthetic messenger RNA (mRNA) or other cargos in vivo. However, clinical translation of targeting mRNA NP products is hampered by the lack of detailed control of targeting selectivity. Here, we introduce multimodal bioluminescence and mass spectrometry imaging (MSI) for active nanoparticle multiplex screening. It combines luminescence and fluorescence imaging on an organ and cellular level, respectively, as functional readouts of the mRNA payload with direct multiplexed ass

A Comprehensive Review on Strategies of Topical Niosomes and Their Synergistic Effect for Enhanced Therapeutic Outcomes.

The review aims to assess the potential of niosomes-nonionic surfactant-based vesicular systems-as carriers for topical and transdermal drug delivery. Niosomes enable targeted and controlled drug release while minimizing systemic toxicity. The investigation centers on their structure, stability, and capacity to entrap both hydrophilic and lipophilic drugs, as well as their use in managing various dermatological and systemic disorders. Recent studies have examined the formulation of niosomes, particularly highlighting the roles of nonionic surfactants and cholesterol in enhancing the stability

Polarized ATR-FTIR Spectroscopy Reveals Light-Induced Peptide Insertion of LAH4 Into Photoswitchable Membranes.

Photoswitchable biomimetic membranes can be controlled by light and offer the possibility to study the role of membrane dynamics for the interaction with antimicrobial peptides. The α-helical peptide LAH4, a well-studied model system, interacts with membranes in a pH-dependent manner. It binds to the membrane surface at acidic pH and inserts into the membrane at basic pH conditions. Polarized attenuated total reflection (ATR) Fourier-transform infrared (FTIR) spectroscopy was employed to probe peptide orientation and structural changes, with particular emphasis on the pH-dependent response of

Solid lipid nanoparticles for targeted drug delivery in breast cancer: Biointerface interactions, therapeutic potential, and translational challenges.

Breast cancer (BC) is a significant cancer that is responsible for a huge disease burden globally. Therapeutic effects against BC are often hampered due to poor drug bioavailability, systemic toxicity, and multidrug resistance development. Solid lipid nanoparticles (SLNs) started as advanced lipidic nanocarriers that could offer biocompatibility, physicochemical stability, and controllable interaction with the bio-interface. This review examines the design, functionalization, and therapeutic potentials of SLNs against breast cancer in-depth and critically. The cellular uptake, tumor penetratio

Engineering Lipid Nanoparticles through Integrated Compositional and Ligand Targeting Enhances β Cell-Directed RNA Delivery.

Beyond their deployment as COVID-19 vaccines, lipid nanoparticles (LNPs) have emerged as versatile vehicles for therapeutic nucleic acid delivery. However, achieving efficient and cell-targeted transfection in extrahepatic tissues, particularly pancreatic β cells, remains a major challenge. Here, we develop a dual-targeting LNP engineering strategy that integrates high-throughput compositional screening with surface conjugation of β cell-specific targeting ligands to enable selective gene delivery to pancreatic β cells. Compositional optimization identified LNP formulations that achieved over
