Metabolomics-Based Analysis Reveals Flavonoid-Mediated Defence in Alfalfa (Medicago sativa 'Gannong No. 5') Against Pea Aphid (Acyrthosiphon pisum) Infestation.

The pea aphid (Acyrthosiphon pisum) is a destructive phloem-feeding pest of alfalfa. Its overreliance on chemical insecticides necessitates the development of sustainable, plant-based resistance strategies. Plant flavonoids are important in induced defense, but their dynamic responses to aphid feeding and subsequent insecticidal effects remain unclear. Using the highly resistant alfalfa variety 'Gannong No. 5' (GN5), this study integrated behavioral assays, performance bioassays, untargeted and targeted metabolomics, and exogenous flavonoid feeding assays to investigate how pea aphid infestati

Endocrine-Disrupting Pesticides as Drivers of Human Disease: Mechanistic Toxicology and Life-Course Health Effects.

Endocrine-disrupting pesticides (EDPs) are environmental toxicants capable of perturbing hormonal homeostasis through multiple molecular and cellular mechanisms. Growing evidence indicates that these compounds contribute to a broad spectrum of adverse health outcomes extending beyond classical endocrine dysfunction. This review critically synthesizes current knowledge on the toxicological mechanisms of EDPs and evaluates epidemiological evidence linking exposure to human disease. Mechanistically, EDPs act through modulation of nuclear hormone receptors, disruption of membrane-associated signal

Integrated physiological, transcriptomic, and metabolomic analyses provide insights into zinc-doped carbon quantum dot-mediated alleviation of cadmium toxicity in lettuce.

Cadmium (Cd) accumulation in crops seriously threatens agricultural productivity and food safety, highlighting the need for effective strategies to mitigate Cd toxicity. Uniformly dispersed zinc-doped carbon quantum dots (Zn-CQDs; 2.67 ± 0.36 nm) were synthesized in this study. The synthesized dots demonstrated enzyme-mimetic activities in vitro and potent radical-scavenging capacity. Foliar application of Zn-CQDs markedly alleviated Cd-induced growth inhibition in lettuce (Lactuca sativa L.), increasing shoot fresh and dry biomass by 30.8% and 23.9%, respectively. Moreover, Zn-CQDs treatment

Multi-omics integration uncovers mechanisms of chromium tolerance in Cosmos sulphureus.

Chromium (Cr) is a highly toxic, mobile heavy metal pollutant that is easily taken up by plants, whereas the Cr tolerance and molecular mechanisms underlying Cr stress responses in C. sulphureus remain poorly elucidated. In this study, we comprehensively investigated phenotypic, physiological, transcriptomic, and metabolomic alterations to uncover the potential molecular mechanisms governing Cr tolerance in C. sulphureus. Phenotypic and photosynthetic physiology results showed that seedlings exposed to low concentrations of Cr exhibited normal leaf and root growth, accompanied by significantly

Cestode infection is linked to transcriptional shifts in neuropeptide signalling and caste-specific ageing pathways in a social insect.

Social insects are key models for phenotypic plasticity, as queens and workers show striking differences in behaviour and lifespan despite sharing the same genome. Neuropeptides are central regulators of behavioural plasticity, and caste-specific ageing is governed by the insulin/insulin-like growth factor 1 signalling-target of rapamycin-juvenile hormone network. Parasites that manipulate host behaviour and lifespan may exploit these regulators by targeting conserved neuropeptides or by altering ageing pathways. Here, we examine a host-parasite system in which the cestode Anomotaenia brevis e

FABP4 characterizes metabolic-immune interactions and serves as a prognostic biomarker across gynecologic malignancies: a multi-omics integrative analysis.

Gynecological malignancies-including breast, cervical, ovarian, endometrial cancers and uterine sarcoma-pose a serious threat to women's health owing to their high incidence, aggressiveness and frequent recurrence. Fatty acid-binding protein 4 (FABP4) is a key regulator of lipid metabolism and tumor metabolic reprogramming, yet systematic investigations of its role across gynecological malignancies remain scarce. Here, by integrating data from TCGA, GTEx and multiple public repositories, we performed a multi-omics, multi-scale analysis of five gynecological malignancies, encompassing transcrip

Elevated CO2 reprograms carbon allocation and attenuates ABA sensitivity to balance photosynthesis and structural resilience in foxtail millet (Setaria italica).

Rising atmospheric CO2 profoundly influence plant physiology, yet integrated responses from molecular regulation to carbon allocation remain poorly characterized in C4 cereals. We employed an integrated physiological, biochemical, and metabolomic approach to investigate how e[CO2] (ambient +200 μmol mol-1) modulates carbon and nitrogen metabolism in leaves and stems of foxtail millet (Setaria italica). e[CO2] significantly increased net photosynthetic rate (+27.4%) and photosynthetic pigment contents. This enhanced carbon gain was accompanied by increased cuticular wax deposition (+27.1%) and

Effects of drought stress on growth, physiology and bioactive compound accumulation in licorice (Glycyrrhiza spp.): a meta-analysis-based integrated evaluation.

INTRODUCTION: Drought stress is a major environmental factor affecting the growth, physiological processes, and secondary metabolism of licorice (Glycyrrhiza spp.). However, the integrated responses of licorice to drought stress remain unclear. METHODS: This meta-analysis synthesized evidence from multiple studies to evaluate the effects of drought stress on growth, physiology, and bioactive compounds in licorice. Random forest analysis and partial least squares path modeling (PLS-PM) were further applied to identify influential predictors and potential pathway relationships. RESULTS: Drough

From polymer to commercial product: Unravelling the ecotoxicological effects of PVA-based dishwasher pods.

Water-soluble polymers (WSPs) are emerging as a largely overlooked class of contaminants. Despite their widespread use, the lack of specific regulation contributes to the uncontrolled release, especially into aquatic ecosystems, raising concerns for environmental and human health. Among WSP-based consumer products, dishwasher pods are widely used and are typically enclosed in a water-soluble film made of polyvinyl alcohol (PVA) and formulation additives. This study provided an integrated ecotoxicological assessment of such films in zebrafish (Danio rerio), combining physicochemical characteriz

Plant-rhizosphere control of thallium mobility and detoxification in contaminated soils: Insights from multi-omics and in situ DGT.

Thallium (Tl) is an extremely toxic and strongly bioaccumulative metal increasingly detected in agricultural soils, yet its behavior at the plant-rhizosphere interface remains poorly constrained. Here, we integrated in situ diffusive gradients in thin films (DGT) with multi-omics analyses (transcriptomics, metabolomics, and 16S rRNA sequencing) to elucidate how plant-rhizosphere interactions regulate Tl mobility and detoxification in Brassica rapa. In situ DGT profiling coupled with the European Community Bureau of Reference (BCR) sequential extraction identified the root-soil interface (0-3 c

Microbiome and cancer: mechanistic insights, diagnostic potential, and therapeutic strategies.

The human microbiome is now recognized as an active and dynamic participant in cancer biology rather than a passive bystander. Increasing evidence demonstrates that microbial dysbiosis contributes to tumor initiation and progression through chronic inflammation, genotoxic toxin production, metabolic reprogramming, immune modulation, and direct reshaping of the tumor microenvironment. Specific microbial factors including colibactin, Bacteroides fragilis toxin, CagA, and Fusobacterium adhesins intersect with canonical oncogenic pathways. Linking microbial activity to genomic instability and immu

Single-cell profiling reveals senescent bone marrow mesenchymal stem cell subpopulations driving pathological ossification in ankylosing spondylitis.

OBJECTIVES: Aberrant new bone formation is a hallmark of ankylosing spondylitis (AS), yet the cellular mechanisms remain unclear. Bone marrow-derived mesenchymal stem cells (BMSCs) are crucial for skeletal homeostasis and may be pathologically reprogrammed under chronic inflammation. We aimed to characterize the cellular heterogeneity and differentiation states of BMSCs in AS, identify BMSC subpopulations associated with structural damage, and explore their molecular features and potential therapeutic targets. METHODS: Single-cell RNA sequencing was performed on BMSCs from 12 AS patients stra

Mechanistic insights into the neuroprotective effects of monoterpene-rich Eucalyptus citriodora essential oil: integrated in silico and in vitro analysis in 6-OHDA-treated N2a neuronal cells.

Parkinson's disease (PD) is characterized by progressive dopaminergic neuronal loss driven by mitochondrial dysfunction, oxidative stress, and proteostasis collapse, for which current therapies remain symptomatic. This study investigated the neuroprotective effects of Eucalyptus citriodora essential oil (ECEO) on 6-hydroxydopamine (6-OHDA)-induced toxicity in N2a neuroblastoma cells. GC-MS analysis showed that ECEO was dominated by oxygenated monoterpenes, mainly citronellal (60.7%), isopulegol (16%), and citronellol (5.7%). In silico analysis of these constituents showed favorable ADMET profi

Single-cell transcriptomic analysis reveals age-related remodeling of brain endothelial cells.

Blood-brain barrier (BBB) integrity naturally declines with age. Brain endothelial cells (ECs) and pericytes (PCs) form the BBB, and aging impairs tight junctions, likely via altered PC-to-EC signaling. However, the molecular mechanisms underlying this impairment remain unclear. Using single-cell RNA sequencing, we profiled 68,316 brain ECs expressing 15,564 genes from young and old mice. Unsupervised clustering and annotation revealed five distinct EC subtypes-Capillary EC1, Capillary EC2, Arterial EC, Venous EC1, and Venous EC2-defined by marker genes Mfsd2a, Plvap, Bmx, Nr2f2, and Vcam1, re

Single-cell transcriptomics reveals phenotypic and functional alterations of erythroid progenitor cells in tuberculosis patients.

Anemia is a common complication of tuberculosis (TB), closely associated with impaired host immune function and poor prognosis; however, its impact on the dynamic alterations of erythroid progenitor cells (EPCs) remains largely unexplored. In this study, we integrated and analyzed single-cell transcriptomic data from peripheral blood mononuclear cells (PBMC) of healthy controls, latent tuberculosis (LTB), TB, and disseminated tuberculosis (DTB) cohorts, as well as 18F-FDG-labeled TB lung tissues and adjacent uninvolved regions. At single-cell resolution, we identified ten EPCs clusters within

Type 2 diabetes Reprograms Bone Marrow Hematopoiesis and Dysregulates Immune Signaling in Response to Stroke.

Type 2 diabetes mellitus (T2DM) worsens stroke outcomes, but the underlying mechanisms linking T2DM to systemic immune dysfunction remain unclear. We investigated whether T2DM alters bone marrow (BM) hematopoiesis and dysregulate immune signaling following ischemic stroke in mice. Single-cell RNA sequencing, GeoMx digital spatial profiling (DSP), nCounter, and flow cytometry were used to analyze BM cells from control (db/+, Ctrl) and diabetic (db/db, T2DM) mice underwent experimental stroke or sham surgery. Diabetes caused marked structural remodeling of BM, with reduced cellularity and imbala

GLP-1 and the Degenerating Brain: Exploring Mechanistic Insights and Therapeutic Potential.

Neurodegenerative disorders, including Alzheimer's disease (AD), Parkinson's disease (PD), stroke, and depression, are marked by progressive neuronal dysfunction and loss, yet current treatments remain largely symptomatic with limited disease-modifying efficacy. Glucagon-like peptide-1 (GLP-1), an incretin hormone traditionally associated with metabolic regulation, has emerged as a promising neuroprotective agent. Its receptor, GLP-1R, is expressed in key brain regions implicated in cognition, emotion, and motor control, including the hippocampus, frontal cortex, and substantia nigra. GLP-1R a

Immunomodulatory effects of phthiocol: An aryl hydrocarbon receptor ligand regulating conventional dendritic cells and macrophages to alleviate sepsis and lupus severity.

Excessive inflammation in sepsis and systemic lupus erythematosus (SLE) is driven by immune dysregulation involving conventional dendritic cells (cDCs), which regulate adaptive immunity, and macrophages, which mediate inflammation and tissue repair. The aryl hydrocarbon receptor (AhR) plays a crucial role in regulating these innate immune cells, suggesting that targeting AhR is a promising strategy to mitigate hyperinflammation in sepsis and SLE. In this study, we demonstrate that phthiocol, a vitamin K analog and AhR ligand, ameliorates hyperinflammation and improves outcomes in both cecal li
