Metabolomic study of natrin-induced apoptosis in SMMC-7721 hepatocellular carcinoma cells by ultra-performance liquid chromatography-quadrupole/time-of-flight mass spectrometry.

Natrin, a new member of the cysteine-rich secretory protein (CRISP) family purified from the snake venom of Naja naja atra, has been demonstrated to have anticancer activity. However, the underlying molecular mechanisms need further elucidation. In this study, MTT was used to evaluate cell viability. Apoptotic cells were analyzed by employing a transmission electron microscope (TEM). Metabolomic study of the metabolic perturbations caused by natrin-induced apoptosis in differentiated SMMC-7721 cells was performed for the first time by using integrative ultra-performance liquid chromatography c

Metagenomic Shotgun Sequencing and Unbiased Metabolomic Profiling Identify Specific Human Gut Microbiota and Metabolites Associated with Immune Checkpoint Therapy Efficacy in Melanoma Patients.

This is the first prospective study of the effects of human gut microbiota and metabolites on immune checkpoint inhibitor (ICT) response in metastatic melanoma patients. Whereas many melanoma patients exhibit profound response to ICT, there are fewer options for patients failing ICT-particularly with BRAF-wild-type disease. In preclinical studies, specific gut microbiota promotes regression of melanoma in mice. We therefore conducted a study of the effects of pretreatment gut microbiota and metabolites on ICT Response Evaluation Criteria in Solid Tumors response in 39 metastatic melanoma patie

Biochemical studies of Piper betle L leaf extract on obese treated animal using 1H-NMR-based metabolomic approach of blood serum samples.

Piper betle L. (PB) belongs to the Piperaceae family. The presence of a fairly large quantity of diastase in the betel leaf is deemed to play an important role in starch digestion and calls for the study of weight loss activities and metabolite profile from PB leaf extracts using metabolomics approach to be performed. PB dried leaves were extracted with 70% ethanol and the extracts were subjected to five groups of rats fed with high fat (HF) and standard diet (SD). They were then fed with the extracts in two doses and compared with a negative control group given water only according to the stu

Mass spectrometry-based metabolomic profiling identifies alterations in salivary redox status and fatty acid metabolism in response to inflammation and oxidative stress in periodontal disease.

Periodontal diseases represent the most common chronic inflammatory diseases in humans and a major cause of tooth loss. Combining mass spectrometry-based ionomics and targeted lipidomics on fatty acid metabolites, we identified significant alterations in redox status and fatty acid metabolism in saliva in response to chronic inflammation and oxidative stress in periodontal disease in a cohort of nonsmoker subjects with chronic periodontitis. For the first time, ionomic profiling of around 30 ions in saliva revealed significantly decreased levels of redox-active metal ions including Mn, Cu, and

Ferric iron availability and species identity jointly shape the metabolic profiles and spoilage potentials of grass carp spoilage bacteria: A combined metabolomic and physicochemical evaluation.

Ferric iron is essential for bacterial growth and spoilage-associated phenotypes, yet its specific influences on the metabolism and spoilage potential of fish spoilage bacteria remain unclear. Therefore, this study aimed to investigate the metabolic and spoilage potential changes of grass carp spoilage bacteria under varying ferric iron accessibilities, using combined metabolomic and physicochemical detection. The results showed that high ferric iron accessibility stimulated bacterial carbohydrate metabolism (glycolysis and tricarboxylic acid cycle) and fatty acid β-oxidation. Conversely, the

The Micro-battlefield under climate change: the domino effect of freeze-thaw cycles on Tributyl phosphate accumulation in grains via nutritional, metabolomic, and microbial profiling.

Under the context of climate change, the risks posed by emerging combined pollution to food security, particularly the accumulation and toxicity of contaminants in the edible parts of crops, remain poorly understood. This study investigated the individual and combined effects of freeze-thaw cycles (FTC) and the flame retardant tributyl phosphate (TBP) on rye growth, physiology, rhizosphere microbiome, quality, and grain accumulation through short-term hydroponic and long-term soil cultivation experiments. The results showed that combined FTC and TBP stress exerted a significant synergistic tox

Integrated metabolomic-transcriptomic profiling of dwarf Camellia reticulata 'Hentiangao' reveals key gibberellin metabolism genes and multi-hormone crosstalk under GA3 treatment.

BACKGROUND: Camellia reticulata 'Hentiangao', a dwarf camellia cultivar, possesses distinctive ornamental traits and high value for urban landscaping. Dwarfism is an important agronomic and ornamental trait, and gibberellin (GA) plays a central role in regulating stem elongation and plant height. However, the molecular mechanisms underlying its dwarf phenotype and the interactions between stem development and endogenous hormone networks remain unclear, limiting the promotion and targeted breeding of this cultivar. We analyzed shoot tips of C. reticulata 'Hentiangao' treated with 800 mg·L-1 GA3

Metabolomic Insights on Obesity and Diabetes from Feeding Diets Varying in Carbohydrate-Fat Ratios in Zucker Diabetic Fatty (ZDF) and Lean Zucker (Zlean) Rats.

In population health the highly cited Atherosclerosis Risk in Communities study indicated a U-shaped association between carbohydrate intake and mortality, whilst the Prospective Urban Rural Epidemiology study linked higher fat intake to lower mortality risks. The Malaysia Lipid Study reported high-fat and high-carbohydrate dietary patterns were associated with increased cardiometabolic risks, including insulin resistance and small dense LDL particles generation. This animal model study therefore was purposely designed to evaluate metabolic outcomes of carbohydrate-fat permutations in Zucker d

Sucrose and starch metabolism pathways as candidate regulatory pathways in the salt stress response revealed by transcriptomic and metabolomic analysis during soybean (Glycine max) germination.

Soybean is a globally important grain and oil crop, and salt stress severely restricts its germination and seedling establishment, ultimately reducing yields. However, the core metabolic regulatory network underlying its salt-stress response and during seed germination remains poorly understood. In this study, the soybean cultivar 'Heihe 91' was cultivated under control (0 mmol/L NaCl) and 150 mmol/L NaCl salt treatments. Phenotypic and physiological assays combined with transcriptomic and non-targeted metabolomic analyses were performed to explore the salt response mechanism during germinatio

Integrated physiological, transcriptomic and metabolomic analyses reveal the mechanism of UV-B radiation enhancement to alleviate drought stress damage in Tibetan hulless barley (qingke).

Drought (D) and ultraviolet-B (UV-B) radiation present severe, frequently co-occurring constraints on crop productivity, particularly in high-altitude environments such as the Qinghai-Xizang Plateau. Although drought severely impairs plant growth and photosynthetic capacity, UV-B radiation can counterintuitively mitigate drought-induced damage; however, the molecular and metabolic mechanisms underpinning this stress cross-talk remain poorly understood. Here, using integrated physiological, transcriptomic, and metabolomic profiling, we examine how Tibetan hulless barley (qingke) responds to ind

Preoperative gut microbiota depletion and metabolomic signatures predict postoperative pneumonia in patients with intracranial space-occupying lesions: a multi-omics prospective cohort study.

BACKGROUND: Patients with intracranial space-occupying lesions are highly susceptible to postoperative pneumonia (POP) due to extensive surgical trauma, impaired consciousness, and prolonged immobilization, which severely compromise recovery and increase mortality. While the "gut-lung axis" highlights the role of gut microbiota in distal pulmonary immune regulation, how preoperative gut signatures predict neurosurgical infections remains poorly understood. METHODS: This prospective study enrolled 50 patients, collecting preoperative and postoperative fecal and plasma samples. 16S rDNA sequenc
