Interplay of the nasal microbiome and epigenome among adolescents.

BACKGROUND: The respiratory microbiome, including that of the nasal cavity, is involved in host defense and airway pathophysiology. Interactions of the microbiome with the host immune system may impact health and disease susceptibility through changes in the epigenome. In this study, we aimed to analyze cross-sectional associations of nasal microbiome composition and the nasal epigenome among adolescents in the Project Viva cohort (N = 372, mean age: 13.0 years). We collected nasal swabs from anterior nares, profiled the microbiome by 16 S rRNA gene sequencing, and grouped samples into 6 clust

Encapsulation of Verbascum sinaiticum Leaf Extract as a Natural Antimicrobial for Controlling Microbial Growth in Beef During Refrigerated Storage.

The efficacy of plant-derived antimicrobials in meat systems is frequently limited by interactions with proteins, lipids, and other food matrix components that reduce the bioavailability and antimicrobial activity of phytochemicals. This study evaluated the antimicrobial effectiveness of Verbascum sinaiticum (V. sinaiticum) leaf extract encapsulated using maltodextrin (MD), gum arabic (GA), and a maltodextrin-gum arabic blend (MDGA, 8:2 w/w) through freeze-drying for application in raw beef during refrigerated storage (4 °C). The encapsulation systems exhibited process yields of 42.5-54.7%, en

Gut microbiota and constipation: from causal evidence to therapeutic strategies-a state-of-the-art narrative review.

Constipation is a common functional disorder of the gastrointestinal tract with a global prevalence of approximately 10-20%, which seriously affects patients' quality of life and imposes a heavy socioeconomic burden. In recent years, the role of the gut microbiota in the pathogenesis of constipation has received increasing attention, particularly in the context of the brain-gut axis theory. In this narrative review, we critically examine the research literature on constipation and intestinal microecology published over the past decade, focusing on four aspects: (1) the characteristics of the g

Stevia-derived sweeteners in reduced-sugar foods: formulation strategies, sensory optimization, nutritional evidence, and safety perspectives.

BACKGROUND: Excessive intake of added sugars is an important contributor to obesity, type 2 diabetes mellitus (T2DM), dental caries, cardiovascular disease, and other metabolism-related disorders in humans. Stevia-derived sweeteners, particularly high-purity steviol glycosides (SGs), have been widely used in reduced-sugar and sugar-free foods because of their high sweetness intensity, low energy contribution, and plant-derived origin. However, their broader application remains limited by bitterness, lingering sweetness, licorice-like or herbal aftertaste, thin mouthfeel, and food-matrix depend

Urinary Volatilomic Profiling Reveals Candidate Metabolomic Signatures Associated with Colorectal Cancer.

Colorectal cancer (CRC) remains one of the leading causes of cancer-related mortality worldwide, largely due to delayed diagnosis. Current screening approaches are limited by invasiveness, cost, and low patient adherence, underscoring the urgent need for non-invasive biomarkers that could complement the existing strategies. Cancer-associated metabolic reprogramming, together with alterations in host-microbiota interactions, is reflected in the urinary volatilome, offering a potential source of candidate biomarkers. In this exploratory, case-control pilot study, urinary volatile organic metabol

Exploring the oral microbiome: from traditional techniques to advanced omics and databases.

The oral microbiome comprises analysis of microbes within the oral cavity. Omics, molecular, and bioinformatic methods have significant functions in examining the diversity, composition, and function of the oral microbiome. Molecular techniques, especially 16S and 18S rRNA sequencing, have transformed the study of the oral microbiome. Analysis by 16S rRNA gene sequences of the oral sample creates a broad portrait of the species composition of the oral microbiome and both existing and new species of bacteria. It is instrumental in detecting bacterial pathogens at specific levels, specifically t

Multi-omics integration reveals the link between gut microbiota dysbiosis, host metabolic dysregulation, and aortic inflammation in abdominal aortic aneurysm.

BACKGROUND: An abdominal aortic aneurysm (AAA) is a localized expansion of the abdominal aorta, in which the vessel diameter exceeds 3 cm or is increased by more than 50% compared to the normal expected size. It has been increasingly recognized as a systemic disease involving complex interactions between vascular tissues and extra-aortic factors, particularly host metabolism and inflammation. The aim for the study was to integrate multi-omics approaches to systematically investigate the molecular mechanisms underlying AAA. RESULTS: Significantly reduced α-diversity Pielou’s evenness was observ

Ecdysone signaling acts as a regulator of immunity and microbiota balance, shaping Leishmania infantum infection in Phlebotomus perniciosus.

BACKGROUND: The steroid hormone ecdysone, acting through its nuclear receptor (EcR), is a key regulator of insect development and reproduction and also contributes to immune regulation and gut microbiota homeostasis. In sand flies, both immunity and the gut microbiota are critical determinants for Leishmania establishment in the midgut. Phlebotomus perniciosus is the principal vector of Leishmania infantum in the western Mediterranean basin; however, the mechanisms affecting interactions among parasite, vector immune system, and resident microbiota remain poorly understood in this species. Artificial intelligence in food and nutrition science: a paradigm-centric review of computational frameworks and system-level integration.

Precision nutrition on a global scale necessitates an understanding of food not as static collections of so-called macronutrients but rather as dynamic and heterogeneous biochemical matrices. These complex non-linear interactions between food composition, gastrointestinal digestion and the human microbiome are difficult to capture using traditional empirical experimental methods. This review articulates a paradigm-based framework that reconceptualizes artificial intelligence (AI) in food and function science, from generic industrial applications to the computational modeling of physiological a

Life in sediments fosters 'sexual' speciation in the Shewanella baltica complex.

Understanding how intra- and interspecific differentiation arises in natural microbial populations is central to explaining the processes that drive bacterial evolution. Motivated by the co-occurrence of multiple putative genospecies closely related to Shewanella baltica in Baltic Sea sediments, we investigated the genomic structure of this species complex across fine spatial scales. We analyzed 112 genome sequences from strains collected across several sediment cores and depths (0-6 cm) at Vaxön (Stockholm archipelago, Sweden) as well as earlier isolates from this site and allopatric strains

Effects of bisphenol A on root traits and rhizosphere bacteria: exploring the link between rhizosphere bacterial and root growth.

BACKGROUND: Bisphenol A (BPA), a widespread environmental pollutant, has been extensively studied for its effects on bacteria and plant, but its impact on rhizosphere bacterial communities and plant root traits is less understood. At the same time, the role of bacteria in helping plants resist adversity is widely recognized, but the relationship between BPA-induced with rhizosphere bacterial changes and root development is still unclear. Therefore, this study investigated the effects of varying BPA concentrations (1.5, 17.2, and 50 mg/L) on soybean root traits and rhizosphere bacterial communi

The role of gut microbiota-derived metabolites in modulating poultry immunometabolism.

The poultry sector is crucial to global food security, but it faces increasing challenges from heat stress, viral diseases, and restrictions on antibiotic use. These stressors highlight immunometabolism, the junction of immune function and metabolic pathways, as a crucial factor in determining the productivity and health of poultry. There is growing evidence that the gut microbiota is a dynamic metabolic organ that produces a diverse range of bioactive metabolites in addition to its function in nutritional digestion. The immunometabolism of poultry is significantly influenced by these microbio

Synthesis and computational investigation of novel 2-mercaptoimidazolones as dual antimicrobial and anti-proliferative agents with potential multitargeting kinase inhibitory activity.

Anticancer drug therapy-induced immunosuppression increases the incidence of treatment-resistant infectious diseases in cancer patients. Combining antibacterial and anticancer capabilities into a single medication can potentially enhance the treatment outcomes. In this light, the current study focused on developing innovative 5-arylidene-3-phenyl-2-thioimidazolones 1a-e and evaluated them as antimicrobial candidates against a panel of Gram-positive and Gram-negative bacterial and fungal isolates. Compounds 1e and 2d exhibited the most potent microbial activity, with MIC values ranging from 24

A Parent-Metabolite Middle-Out PBPK Model for Genistein and Its Glucuronide Metabolite in Rats: Integrating Liver and Enteric Metabolism with Hepatobiliary and Enteroluminal Transport to Assess Glucuronide Recycling.

Background: Glucuronide recycling in the gut and liver profoundly affects the systemic and/or local exposure of drugs and their glucuronide metabolites, impacting both clinical efficacy and toxicity. This recycling also alters drug exposure in the colon, making it critical to establish local concentration for drugs targeting colon (e.g., drugs for colon cancer and inflammatory bowel disease). Methods: In this study, a parent-metabolite middle-out physiologically based pharmacokinetic (PBPK) model was built for genistein and its glucuronide metabolite to estimate the systemic and local exposure

The role of dysbiosis in shaping host immunity in endometrial cancer development.

In recent years, research into the background of carcinogenic processes has increasingly focused on the role of the tumor microenvironment (TME) in tumorigenesis. In addition to the presence of tumor cells and non-malignant components, which include immune cells, extracellular matrix elements, stroma, and endothelial cells, the microbiome is now increasingly being classified as an integral part of the TME. The establishment of the Human Microbiome Project (HMP) in 2007 along with the development of next-generation sequencing (NGS) techniques proved to be a breakthrough in terms of human microb

Immobilized inclusion bodies of recombinant cold-adaptive lipase from Antarctic Pseudomonas sp. as catalysts.

Immobilization of lipase has been receiving attention for a long time; this is because of the need for robust catalysts by industries. Numerous literature has reported improvements in the properties of immobilized lipase in terms of stability when exposed to extreme conditions of temperature, pH, and organic solvents commonly encountered in most industrial settings. However, some microbial lipases that have the potential to catalyze significant reactions do occur in the form of inclusion bodies when expressed in Escherichia coli. This research aimed to immobilize catalytically active inclusion

A Salmonella enterica Surrogate for Future Validation Studies for Nonfat Dried Milk and Low-Moisture Food Environments.

Low-moisture food persistent bacterial populations (LMF PBPs) may be responsible for pathogen harborage in LMF processing environments, resulting in outbreaks and recalls. To address this issue, industry requires a surrogate to understand the survivability of LMF PBP and perform in-plant validation studies to reduce the level of this pathogen to an acceptable level. Hygienic controls for the LMF industry must be tailored to different equipment, environments, and food types. In-plant validations with a surrogate would assist with evaluating the efficacy of sanitation protocols that eliminate LM
