Novel CNS malformations and skeletal anomalies in a patient with Beaulieu-boycott-Innes syndrome.

THO/TREX (transcription/export) is a conserved eukaryotic complex that plays a crucial role in gene expression and prevents DNA damage during mitosis and meiosis. In mammals, TREX is essential during embryogenesis, determining stem cell fate specification by regulating posttranscriptional self-renewal and differentiation in several tissues. It is composed of a core called THO, consisting of THOC1, 2, 5, 6, 7, and additional proteins. Bi-allelic mutations in THOC6 have been associated to Beaulieu-Boycott-Innes syndrome (BBIS), a syndromic form of intellectual disability (ID). To date, nine pati

Update: Noncongenital Zika Virus Disease Cases - 50 U.S. States and the District of Columbia, 2016.

Zika virus is a flavivirus primarily transmitted to humans by Aedes aegypti mosquitoes (1). Zika virus infections also have been documented through intrauterine transmission resulting in congenital infection; intrapartum transmission from a viremic mother to her newborn; sexual transmission; blood transfusion; and laboratory exposure (1-3). Most Zika virus infections are asymptomatic or result in mild clinical illness, characterized by acute onset of fever, maculopapular rash, arthralgia, or nonpurulent conjunctivitis; Guillain-Barré syndrome, meningoencephalitis, and severe thrombocytopenia r

Insulin-like growth factor type 1 deficiency in a Moroccan patient with de novo inverted duplication 9p24p12 and developmental delay: a case report.

BACKGROUND: 9p duplication is a structural chromosome abnormality, described in more than 150 patients to date. In most cases the duplicated segment was derived from a parent being a reciprocal translocation carrier. However, about 15 cases with de novo 9p duplication have been reported previously. Clinically, this condition is characterized by mental retardation, short stature, developmental delay, facial dysmorphism, hand and toe anomalies, heart defects and/or ocular manifestations. CASE PRESENTATION: We report here the case of a 2-year-old Moroccan girl with a de novo duplication of 9p24

Renal thrombotic microangiopathy in patients with cblC defect: review of an under-recognized entity.

Methylmalonic aciduria and homocystinuria, cobalamin C (cblC) type, is the most common genetic type of functional cobalamin (vitamin B12) deficiency. This metabolic disease is characterized by marked heterogeneity of neurocognitive disease (microcephaly, seizures, developmental delay, ataxia, hypotonia) and variable extracentral nervous system involvement (failure to thrive, cardiovascular, renal, ocular) manifesting predominantly early in life, sometimes during gestation. To enhance awareness and understanding of renal disease associated with cblC defect, we studied biochemical, genetic, clin

Neuropathogenesis of Zika Virus Infection : Potential Roles of Antibody-Mediated Pathology.

Zika virus (ZIKV) is an enveloped, positive-sense, single-stranded RNA virus that belongs to the genus Flavivirus, family Flaviviridae, which includes many human and animal pathogens, such as dengue virus (DENV), West Nile virus, and Japanese encephalitis virus. In the original as well as subsequent experimental and clinical reports, ZIKV seems to have moderate neurotropism (in animal models) and neurovirulence (in human fetuses), but no neuroinvasiveness (in human adults). Intrauterine ZIKV infection (viral pathology) has been linked to an increased incidence of microcephaly, while increased

Fetal brain disruption sequence versus fetal brain arrest: A distinct autosomal recessive developmental brain malformation phenotype.

The term fetal brain disruption sequence (FBDS) was coined to describe a number of sporadic conditions caused by numerous external disruptive events presenting with variable imaging findings. However, rare familial occurrences have been reported. We describe five patients (two sib pairs and one sporadic) with congenital severe microcephaly, seizures, and profound intellectual disability. Brain magnetic resonance imaging (MRI) revealed unique and uniform picture of underdeveloped cerebral hemispheres with increased extraxial CSF, abnormal gyral pattern (polymicrogyria-like lesions in two sibs a

POMT2 intragenic deletions and splicing abnormalities causing congenital muscular dystrophy with mental retardation.

BACKGROUND: Alpha-dystroglycanopathies are a group of congenital muscular dystrophies (CMDs) with autosomal recessive inheritance characterized by abnormal glycosylation of alpha-dystroglycan. Although six genetic causes have been identified (FKTN, POMGNT1, POMT1, POMT2, FKRP, and LARGE) many alpha-dystroglycanopathy patients remain without a genetic diagnosis after standard exon sequencing. To date POMT2 mutations have been identified in CMD cases with a wide range of clinical severities from Walker-Warburg syndrome to limb girdle muscular dystrophy without structural brain or ocular involvem

Prenatally diagnosed hypoplastic left heart syndrome--outcomes after postnatal surgery.

OBJECTIVE: To identify prenatally diagnosed cases of hypoplastic left heart syndrome (HLHS) and then to determine postnatal outcomes after surgical interventions. METHODS: An ultrasound and pediatric cardiology database was used to identify all fetuses diagnosed prenatally from 1991-1996 with HLHS. Fetal karyotypes were performed on cultured amniocytes. After diagnosis, parents were given several management options: pregnancy termination before 22 weeks, postnatal hospice care, or surgery using the Norwood procedure or cardiac transplantation. Ultrasound and echocardiography findings were lat

[Neonatal Basedow's disease in twins from a mother with severe T3 hyperthyroidism].

HISTORY AND CLINICAL FINDINGS: Dizygotic twin sisters were born to a woman who, shortly before becoming pregnant, had developed Graves' disease with markedly elevated triiodothyronine (T3) levels and highly positive TSH receptor antibody titres (TRAb: 169 mU/ml). From the second week of life onwards they had a goitre and hyperexcitability, tachycardia and failure to thrive were noted. In addition, twin I had mild exophthalmos. As thyrostatic treatment of the mother was very difficult, intrauterine hypothyroidism or transitory hyperthyroidism had presumably occurred in the twins. INVESTIGATION

Human brain evolution: Role of SINE-VNTR-Alu retrotransposons and SINE-VNTR-Alu long noncoding RNA AK057321.

Humans differ remarkably from great apes, with advanced cognitive abilities correlated to enlarged brains resulting from prolonged neurodevelopment. The SINE-VNTR-Alu (SVA) retrotransposons are candidate genetic drivers of these differences. Human genes with intronic SVAs are enriched for neurodevelopmental diseases; the microcephaly gene CDK5 regulatory subunit associated protein 2 (CDK5RAP2) and epilepsy/intellectual disability gene SCN8A coding for sodium channel subunit contain human-specific, intronic SVAs. Deleting gene ZNF91 coding for the SVA-binding zinc finger protein 91 or the intro

Ecotoxicological Assessment of a Glyphosate-Based Commercial Formulation and a Novel Experimental Formulation by Standardized Amphibian Bioassays.

Herbicide effectiveness depends on the adjuvants in the formulation, with surfactants being crucial for systemic ones such as glyphosate, but surfactants are often more toxic than the pesticide itself to nontarget organisms, such as amphibians. Previously, a novel experimental formulation (EF) of glyphosate has been developed with natural products (alkylglucosides and saponin) with great efficiency in field tests. The objective of this study was to compare the toxicity of the EF with a commercial formulation of glyphosate (Roundup Full II, CF) by standardized bioassays (AMPHITOX). For this, em

[The physiological roles of primary ciliary resorption and its potential as therapeutic targets].

Primary cilia are cellular antennae protruding from the surface of nearly all cell types during the G0 phase. They control the organization and maturation of systemic tissues and organs by receiving selective signals. Ciliopathies are heritable disorders that cause dysplasia of various tissues and organs, as well as the progression of obesity. In contrast, recent studies using cultured cells have revealed that cilia were resorbed upon receiving selective proliferative stimuli, thereby reentering the cell cycle into the G1/S phase. However, the physiological significance and molecular mechanism

Nabais Sá-de Vries Syndrome Type 1 in a Mexican Girl: A Case Report.

Nabais Sá-de Vries syndrome (NSDVS) is a rare disease caused by a heterozygous mutation in the SPOP gene (17q21), which is involved in protein degradation via the ubiquitin-proteasome pathway. Different germline variants within this gene cause two distinct phenotypes (allelic heterogeneity) that affect multiple organs and systems (pleiotropy). These variants are clinically associated with intellectual disability, neurological disorders, and dysmorphic features that exhibit variable expressivity. Here, we present the case of a five-year-old girl who was conceived through assisted reproductive t

Cortical Auditory Neural Responses to Speech and Communicative Functioning in Normocephalic Children With Prenatal Exposure to the Zika Virus.

OBJECTIVES: Prenatal exposure to the Zika virus may have an adverse effect on communicative functioning in children, even in the absence of microcephaly associated with Congenital Zika Syndrome. This study characterized cortical auditory neural function and communication abilities in normocephalic children with and without prenatal Zika virus exposure. DESIGN: Cortical auditory evoked potentials were recorded in response to naturally spoken consonant-vowel syllables in 117 Brazilian children with prenatal exposure to the Zika virus (M age: 6.29 ± 0.42 years) and a comparison group of 90 non-e

A Novel Intracellular Domain Missense Variant in GRM7 Associated with Early-Onset Neurodevelopmental Encephalopathy.

INTRODUCTION: Biallelic pathogenic variants in the GRM7 gene, which encodes the metabotropic glutamate receptor 7 (mGlu7), have been associated with early-onset neurodevelopmental disorders characterized by epilepsy, developmental delay, and white matter abnormalities. However, reported cases predominantly involve variants affecting the extracellular or transmembrane domains, and the full phenotypic spectrum remains unclear. CASE PRESENTATION: We present the case of a male patient exhibiting early-onset focal epilepsy, global developmental delay, and neuroimaging evidence of corpus callosum t

Identification and growth features of developmental delay with macrocephaly caused by a novel TRIO variant affecting the second SH3 domain.

BACKGROUND: Trio Rho guanine nucleotide exchange factor (TRIO) encodes the guanine nucleotide exchange factor (GEF) for RHOA and RAC1 GTPases, which plays a critical role in neurodevelopment. Pathogenic variants in the TRIO gene are predominantly associated with two autosomal dominant neurodevelopmental disorders: intellectual developmental disorder 44 with microcephaly (MRD44) and intellectual developmental disorder 63 with macrocephaly (MRD63). However, the genotype-phenotype correlation of TRIO-related disorders and the impacts of TRIO variants on early-life disease progression remain uncle
