A Novel Mutation in the FSH Receptor (I423T) Affecting Receptor Activation and Leading to Primary Ovarian Failure.

CONTEXT: Follicle-stimulating hormone (FSH) plays an essential role in gonadal function. Loss-of-function mutations in the follicle-stimulating hormone receptor (FSHR) are an infrequent cause of primary ovarian failure. OBJECTIVE: To analyze the molecular physiopathogenesis of a novel mutation in the FSHR identified in a woman with primary ovarian failure, employing in vitro and in silico approaches, and to compare the features of this dysfunctional receptor with those shown by the trafficking-defective D408Y FSHR mutant. METHODS: Sanger sequencing of the FSHR cDNA was applied to identify th

A Force-Kernel Reformulation of the Extended-System Adaptive Biasing Force for Free-Energy Calculations.

We introduce force-kernel extended-system adaptive biasing force (FK-eABF), a force-based kernel reformulation of eABF that replaces the histogram-based mean-force accumulator of conventional eABF with a sparse population of Gaussian kernels storing local running-mean forces. Biasing forces are recovered by Nadaraya-Watson regression, yielding smooth estimates from the earliest stages of a simulation without a minimum-count threshold, while the same kernel population also defines an auxiliary, self-attenuating exploration force that requires no prior knowledge of barrier heights. Exploring the Self-Assembly of Glycolipids into Three-Dimensional Network Phases.

Glycolipids are sugar-based amphiphiles that play crucial roles in many biological processes. Under thermotropic and lyotropic conditions, glycolipids self-assemble into a variety of mesophases, including cocontinuous network phases, such as the double gyroid. In this work, a two-stage molecular dynamics simulation workflow is developed to probe network formation for solvent-free amphiphiles at different temperatures. In the first stage, the structural evolution of systems initiated in lamellar and hexagonally packed cylinder arrangements is examined as an indicator of the likelihood of networ

Simulation Investigation on the Influence Mechanism of Toluene and Heptane on the Aggregation of Asphaltene Molecules.

Asphaltenes are a group of compounds that are soluble in benzene and toluene but insoluble in nonpolar small molecule n-alkanes. The asphaltene aggregation in the asphaltene-heptane-toluene system was studied using molecular dynamics (MD) simulation, and the interaction between asphaltene molecules during this process was also revealed from the evolution of the density field, radial distribution function (RDF), and intermolecular distance of asphaltenes. Three main findings were made: (1) more asphaltene precipitates (heptane) were contained, and more asphaltene dimers or trimers were formed d

Size-Dependent Mechanical Properties of Amorphous SiO2 Nanowires: A Molecular Dynamics Study.

Uniaxial tension tests were performed for amorphous SiO2 nanowires using molecular dynamics simulation to probe the size effect on the mechanical properties and plastic deformation by varying the length of nanowires. The simulation results showed that the Young's modulus of SiO2 nanowires increased with the decrease of nanowires length due to its higher surface stress. The corresponding deformation of SiO2 nanowires during tension exhibited two periods: atomic arrangement at small strain and plastic deformation at large strain. During the atomic arrangement period, the percentage variations of

Bioactive compounds from marine algae targeting MMP-9: A computational approach to rheumatoid arthritis treatment.

Rheumatoid arthritis (RA) is a chronic autoimmune systemic inflammatory condition that significantly damages bone and cartilage, leading to severe joint deformities. Matrix metalloproteinase(s) (MMP-9) plays a significant role in both the pathogenesis and management of RA, and its status as a biomarker and a target for inhibition is important in understanding and treating the disease. This work aims to examine the interactions between the molecules of MMP-9 and bioactive compounds derived from marine algae using molecular docking and absorption, distribution, metabolism, excretion, and toxicit

Bisphenol S disrupts human sertoli cell function through NR1H3 degradation: Mechanistic insights from integrated bulk and single-cell transcriptomics.

Male infertility, often linked to impaired spermatogenesis, is increasingly associated with environmental pollutants such as bisphenol S (BPS), a common bisphenol A substitute, yet its molecular mechanisms in human Sertoli cells remain unclear. In this study, immortalized human Sertoli cells were exposed to BPS, and cell viability, proliferation, and transcriptomic changes were assessed, with bulk RNA sequencing integrated with single-cell transcriptomic profiles from non-obstructive azoospermia (NOA) testes to identify key regulatory factors. Potential BPS targets were predicted via pharmacop

Estrogen receptors in urogenital schistosomiasis and bladder cancer: Estrogen receptor alpha-mediated cell proliferation.

Estrogen-like metabolites have been identified in S. haematobium, the helminth parasite that causes urogenital schistosomiasis (UGS) and in patients´ blood and urine during UGS. Estrogen receptor (ER) activation is enriched in the luminal molecular subtype bladder cancer (BlaCa). To date, the significance of ER to these diseases remains elusive. We evaluated ERα and ERβ expression in UGS-related BlaCa (n = 27), UGS-related non-malignant lesions (n = 35), and noninfected BlaCa (n = 80). We investigated the potential of ERα to recognize S. haematobium-derived metabolites by docking and molecular

A ReaxFF molecular dynamics study of molecular-level interactions during binder jetting 3D-printing.

In the present work, we study one of the major additive manufacturing processes, i.e., the binder jetting printing (BJP) process, at the molecular level through atomistic-scale level representations of powders and binder solutions with chromium-oxide (Cr-oxide) nanoparticles and water-based diethylene glycol solutions, respectively. The results show that both diethylene glycol and water contribute to the bonding of Cr-oxide particles during the print and curing stages by forming a hydrogen bond network. Heating the system to the burn-out temperature results in the oxidation of diethylene glyco

Dynamics of the conformational transitions during the dimerization of an intrinsically disordered peptide: a case study on the human islet amyloid polypeptide fragment.

Amyloid deposits of human islet amyloid polypeptide (hIAPP) are identified in 95% of type II diabetes patients. The oligomers during the early stage of hIAPP aggregation are believed to be more cytotoxic than the mature fibrils. However, the structural details during the initial stage of hIAPP aggregation are still under debate experimentally. To understand its initial nucleation mechanism, we investigate the thermodynamics and kinetics of hIAPP(11-25) dimerization, which is the first manifestation of the interplay between intra- and inter-molecular interactions, via the construction of Markov

Molecular dynamics of leucine and dopamine transporter proteins in a model cell membrane lipid bilayer.

The dopamine transporter (DAT) operates via facilitated diffusion, harnessing an inward Na(+) gradient to drive dopamine from the extracellular synaptic cleft to the neuron interior. The DAT is relevant to central nervous system disorders such as Parkinson disease and attention-deficit hyperactivity disorder and is the primary site of action for the abused psychostimulants cocaine and amphetamines. Crystallization of a DAT homolog, the bacterial leucine transporter LeuT, provided the first reliable 3-D DAT template. Here, the LeuT crystal structure and the DAT molecular model have been combine

Structural insights into the binding potential of natural therapeutic leads as antiviral agents against human Metapneumovirus nucleoprotein.

Human metapneumovirus (hMPV), a member of the family Pneumoviridae and the genus Metapneumovirus, is a prevalent cause of respiratory tract infections, leading to severe complications and even death. As of now, there are no approved specific antiviral drugs or vaccines for hMPV, underscoring the need for potential therapeutic candidates. Hence, the present study employed a structure-based virtual screening (SBVS) of prepared ligands from CMNPD and SPECS databases against the hMPV nucleoprotein. Multi-level precision screening identified CMNPD3066, CMNPD12667, CMNPD25748, SPECS-AE-765, and SPEC

Ab initio calculations on the formation and rearrangement of spiropentane.

The formation of spiropentane, by addition of singlet (1A1) methylene to methylenecyclopropane, and the unimolecular reactions of spiropentane have all been studied computationally. Benchmark calculations on two key biradicals were conducted by the multireference Mukherjee's coupled-cluster (MkCC) method. Various single-reference coupled-cluster methods and multireference second-order perturbation theory were then compared for accuracy against experimental data and the MkCC results. The object of the exercise was to get the best possible description of the potential energy surface for formatio

Synergistic effect of modified konjac glucomannan and biological enzyme induced carbonate precipitation on the biocementation of soil fugitive dust.

Soil fugitive dust poses a serious threat to human health, the plant-derived polysaccharide konjac glucomannan was modified to obtain konjac glucomannan-acrylic acid graft copolymer (KGMP), which was combined with enzyme induced carbonate precipitation (EICP) to control it. The performance of KGMP-EICP was evaluated. Microstructure characterization was performed on the treated soil fugitive dust samples, and molecular dynamics simulations were employed to investigate how KGMP regulates the nucleation of calcium carbonate, thereby elucidating the fugitive dust control principle of KGMP-EICP. Computational design and experimental validation of peptide inhibitors to disrupt urease enzyme maturation in pathogenic bacteria Proteus mirabilis.

Urease, an essential virulence factor in pathogens such as Proteus mirabilis, requires nickel incorporation into its active site for maturation. This process is mediated by accessory proteins, with a key role of UreE-UreG interaction representing an early step in nickel delivery. Disruption of this interaction impairs enzyme maturation, reduces urease secretion, and limits microbial survival under acidic conditions. In this study, peptide inhibitors were designed and evaluated to target the protein-protein interaction within the UreE-UreG complex. Protein structures were predicted using AlphaF
