Engineering a Multifunctional Sprayable Curcumin-Modified Polyitaconate-Based Hydrogel to Accelerate Methicillin-Resistant Staphylococcus aureus-Infected Wound Healing.

Hydrogel dressings that can simultaneously curb infection and dampen inflammation are particularly attractive for accelerating multidrug-resistant bacteria-infected wound healing. However, clinical translation of many reported hydrogels is often constrained by multi-component formulations, complicated preparation procedures, and insufficient sprayability, which limit conformal coverage on irregular wound beds. Polyitaconate possesses intrinsic antibacterial and anti-inflammatory activities, yet its weak antioxidative properties and lack of thermogelling properties limit its utility in infected

Prevalence of Streptococcus pneumoniae and Staphylococcus aureus co-colonization in healthy children under five years: a systematic review.

BACKGROUND: Nasal colonization is asymptomatic but facilitates transmission and can precede infection. Staphylococcus aureus and Streptococcus pneumoniae commonly colonize young children and contribute to substantial morbidity. Although pneumococcal conjugate vaccines (PCVs) have reduced vaccine-type (VT) carriage, serotype replacement and altered colonization patterns have emerged. This systematic review evaluated S. pneumoniae–S. aureus co-colonization in healthy children under five and the relationship between these colonization patterns. METHODS: Following PRISMA guidelines and registered

In vitro activity of recently introduced Gram-positive-specific antimicrobial agents against Australian methicillin-resistant Staphylococcus aureus isolates.

Existing therapeutic options are limited for severe methicillin-resistant Staphylococcus aureus (MRSA) infections. Newer antimicrobials with anti-MRSA activity have been introduced, but evaluation of pre-existing resistance in the Australian setting is limited. Our objectives are to characterise the antimicrobial susceptibility profile of the common MRSA sequence types (STs) in Australia relating to newer antimicrobial agents to determine the association between non-susceptibility and ST, trends in minimum inhibitory concentration (MIC) for antimicrobial classes, and genomic associations with

A Dual Setback: Staphylococcus aureus Biofilms From Severe Chronic Rhinosinusitis Show Enhanced Virulence Capacity and Tolerance to Antibiotics.

BACKGROUND: Staphylococcus aureus biofilms play a crucial role in chronic rhinosinusitis (CRS), leading to the persistence of symptoms. Severe CRS patients are frequently infected with S. aureus strains that exhibit higher biofilm properties (e.g., biomass, exoprotein production) compared to S. aureus from controls. S. aureus biofilms resist antibiotic treatment; however, the relationship between bacterial biofilm properties, antibiotic susceptibility, and CRS severity has not yet been defined and is the subject of this study. METHODS: S. aureus clinical isolates and reference strains, and ma

Synthetic short peptide mimicking IsdB and IsdH conserved motifs selectively bind and enable detection of Staphylococcus aureus infection.

BACKGROUND: Staphylococcus aureus (S. aureus) is the most common pathogen in musculoskeletal infections (MSKI). Application of antibody responses to S. aureus virulence factors have suggested to improve diagnostic specificity. Iron acquisition plays a crucial role in S. aureus pathogenesis. The bacterium scavenges iron from the host by expressing cell surface receptors that bind heme and hemoglobin through the iron-regulated surface determinant (Isd) system. These heme and hemoglobin binding motifs are essential for bacterial survival and virulence. In particular, the highly conserved region o

Staphylococcus aureus superantigens drive a concentration-dependent neutrophilic or eosinophilic pulmonary inflammation in mouse models of cystic fibrosis.

Staphylococcus aureus (SA), the most common cystic fibrosis (CF) respiratory pathogen, is uniquely capable of producing superantigen (SAg) exotoxins, which are recognized as the most potent activators of the immune system and inducers of inflammation. Although a high frequency of CF SA isolates can produce SA-SAgs, their role in the immunopathogenesis of CF lung disease is unexplored. Therefore, to investigate the role of SA-SAgs in pulmonary inflammation in vivo, we crossed HLA-DR3 transgenic mice with CF gut corrected (HLA-DR3.CFGC) mice and βENaC transgenic (HLA-DR3.ENAC) mice expressing th

Construction of a linezolid-resistant strain of methicillin-susceptible Staphylococcus aureus and its multi-omics based mechanism study.

BACKGROUND: The emergence of linezolid-resistant methicillin-susceptible Staphylococcus aureus (MSSA) is a growing clinical threat, but the underlying mechanisms are unclear. Here, we generated stable resistant derivatives and used a multi-omics strategy to investigate the transcriptional and metabolic regulation underlying linezolid resistance in MSSA. METHODS: A clinical isolate of linezolid-susceptible S. aureus (strain S. aureus_97) was used as the parental strain. A stable linezolid-resistant derivative (designated LZR) was generated through stepwise induction with subinhibitory concentr

Genomic insights into specialized and primary metabolism gene clusters in methicillin-resistant Staphylococcus aureus from Egyptian community settings.

Community-acquired methicillin-resistant Staphylococcus aureus (CA-MRSA) remains a major public health concern. Beyond canonical virulence determinants, biosynthetic gene clusters (BGCs) for specialized metabolites (SMs) and primary metabolic adaptations may shape fitness in community settings. In this study, we analyzed 123 CA-MRSA isolates from Egypt (2020-2021) alongside three reference genomes using a combined genomics workflow: antiSMASH and PRISM for homology-based specialized BGCs and predicted structures, GECCO for de novo BGC detection, BiG-SCAPE for gene-cluster family (GCF) networks

Molecular typing of PVL-positive Staphylococcus aureus isolates from Lithuania and genome sequencing of a local outbreak strain.

Staphylococcus aureus is a major human commensal and pathogen, with the Panton-Valentine leukocidin (PVL) genes being associated with increased virulence. Rapid detection and molecular typing of such isolates are essential for effective epidemiological surveillance and infection control. Studies from Lithuania indicate that PVL is relatively common among S. aureus isolates, but detailed molecular typing data are still lacking. Clinical S. aureus isolates were collected from two hospitals in Vilnius in 2018-2019 and 2024, and from healthy volunteers between 2012 and 2020. Isolates were screened

The metabolic trap: Candida parapsilosis inhibits Staphylococcus aureus biofilm maturation by disrupting pH homeostasis and inducing premature exodus.

Introduction. Hospital-acquired infections (HAIs) frequently manifest as device-related biofilms that exhibit enhanced tolerance to conventional therapies contributing to antimicrobial resistance. Polymicrobial biofilms involving Candida and Staphylococcus species are a major cause of persistent nosocomial infections. However, while the synergism between Candida albicans and Staphylococcus aureus is well-characterized, the interactions involving non-albicans Candida remain poorly understood.Hypothesis/Gap Statement. The specific interactions between Candida parapsilosis and S. aureus were enti

Global Transcriptional Differences in Staphylococcus aureus Biofilm-Associated Genes in a brpR Mutant Compared to Wild-Type Strain.

Background: Staphylococcus aureus causes bloodstream and skin infections in humans. The prevalence of multidrug-resistant S. aureus strains means new antibiotics are needed. A novel antimicrobial drug named SK-03-92, a synthetic aromatic organic stilbenoid compound, kills S. aureus cells within 30 min, but an increase in both biofilm formation and persister cells occurs. SK-03-92 treatment downregulates transcription of the biofilm regulating protein regulator (brpR) gene and biofilm regulating protein sensor (brpS) gene in S. aureus. BrpR/BrpS system may be a LytTR regulatory system tied to b

Polyvinyl Alcohol/Lignin Composite Films Crosslinked with Citric Acid: Physicochemical Properties and Anti-Adhesive Activity Against Staphylococcus aureus.

Antibiotic-resistant infections, particularly in hospital settings, are increasingly associated with biofilm formation on biomedical devices. An effective strategy to address this problem is the development of biopolymeric films with antibacterial and anti-adhesive properties. In this work, polyvinyl alcohol (PVA), a synthetic biodegradable polymer, was composited with lignin (LG), a biomass-derived polymer, and crosslinked with citric acid (CA) to obtain PVA-LG/CA gels/films. Materials were prepared with fixed PVA content (10% w/v) and varying LG (6.6 and 10% w/v) and CA (1-4% w/v) contents.
