Chronic restraint stress causes anxiety- and depression-like behaviors, downregulates glucocorticoid receptor expression, and attenuates glutamate release induced by brain-derived neurotrophic factor in the prefrontal cortex.

Stress and the resulting increase in glucocorticoid levels have been implicated in the pathophysiology of depressive disorders. We investigated the effects of chronic restraint stress (CRS: 6 hours × 28 days) on anxiety- and depression-like behaviors in rats and on the possible changes in glucocorticoid receptor (GR) expression as well as brain-derived neurotrophic factor (BDNF)-dependent neural function in the prefrontal cortex (PFC). We observed significant reductions in body weight gain, food intake and sucrose preference from 1 week after the onset of CRS. In the 5th week of CRS, we conduc

Association study of RGS2 gene polymorphisms with panic disorder in Japanese.

Genetic factors for panic disorder have been consistently observed in family and twin studies. Regulators of G-protein signaling (RGS) is a family of proteins that negatively regulate the intracellular signaling of G protein-coupled receptors such as dopamine and serotonin receptors. RGS2, one of the RGS families, has been suggested to plays a role in anxiety and/or aggressive behavior. Polymorphisms in the RGS2 gene were recently associated with panic disorder, trait anxiety, suicidal behavior, and generalized anxiety disorder. From these findings, we tried to replicate an association between

Development of the 2nd generation neurokinin-1 receptor antagonist LY686017 for social anxiety disorder.

The neurokinin-1 (NK-1) antagonist LY686017 showed activity in preclinical anxiety models. The clinical development of LY686017 included a PET study and a proof-of-concept in social anxiety disorder (SAD). [(11)C]GR205171 was used healthy volunteers receiving 1-100mg/d LY686017 for 28 days to determine brain receptor occupancy (RO). The mean NK-1 RO increased ranged from 25% with 1mg to 93% with 100mg. Subsequently, a 12-week randomized clinical trial tested LY686017 vs. paroxetine, or placebo in SAD. Pharmacokinetic (PK)/RO modeling based on the PET results predicted that once daily dosing of

[Serum serotonin in patients with tension headaches].

Tension headache (TH) is one of the most frequent types of idiopathic headaches. The leading role in its pathogenesis is played by depression and dysmetabolism of the neurotransmitter serotonin. The subjects were 100 patients with TH. The examination included study of headache intensity, complex psychometric testing, and immune-enzyme measurement of serotonin serum level, performed before and after treatment with the anti-depressant prodep. All the patients had moderate pain syndrome, depression, and moderate or severe anxiety, which demonstrated negative correlation with serotonin serum level

Assessment of Self-Medication Behaviour in Response to Dental Pain in Two Populations, France.

INTRODUCTION AND AIMS: Self-medication appears to be a common practice for dental pain. However, in France, its prevalence and patterns in dentistry have never been studied. The primary objective was to assess the prevalence and self-medication behaviours in two at-risk populations: patients consulting for acute pulpal or periapical pain and patients with dental anxiety requiring treatment under general anaesthesia. The secondary objective was to examine the influence of socio-behavioural factors on these practices. METHOD: Between April 2021 and May 2023, the behaviours of two at-risk popula

Psychological distress in patients with detected pulmonary nodules from lung cancer screening.

BACKGROUND: Low-dose computed tomography (LDCT) has improved early detection of lung cancer, but the identification of indeterminate pulmonary nodules (PNs) often triggers psychological distress. Evidence remains limited regarding longitudinal psychological changes following different management approaches and the identification of patients at risk for persistent distress. This study aimed to longitudinally evaluate psychological outcomes and identify predictors of symptom change in patients with LDCT-detected PNs managed by surgical resection/biopsy or radiological surveillance. METHODS: A p

A network analysis of post-traumatic stress disorder, anxiety, and depression in women with pregnancy loss.

BACKGROUND: Pregnancy loss represents a significant public health concern within the realm of female reproductive health. In the current landscape of research, post-traumatic stress disorder (PTSD), anxiety, and depression are often investigated as an undifferentiated whole. However, this approach overlooks the unique symptomatic relationships and nuances that may exist among these mental health conditions in women who have endured pregnancy loss. OBJECTIVE: This study aimed to dissect the symptomatic interconnections among PTSD, anxiety, and depression in women who have experienced pregnancy

Internet-based therapy for social anxiety in adults in Portugal (PORTiSOFIE) and its impact on sexual and relationship satisfaction: study protocol for a randomized controlled trial.

BACKGROUND: Social anxiety disorder (SAD) is a common condition that affects intrapersonal (e.g., distress and impairment) and interpersonal functioning (e.g., difficulties in amorous relationships). People with SAD tend to have difficulties in communicating feelings to partners and report lower levels of sexual and relationship satisfaction. Cognitive behavioral therapy (CBT) for SAD has been found to reduce symptoms of social anxiety, but less is known about its impact on relationship functioning. Internet-based CBT (iCBT) has gained attention for its primary advantage: bridging distances an

The influence of postural threat-induced anxiety on locomotor learning and updating.

The ability to adapt our locomotion in a feedforward (i.e., "predictive") manner is crucial for safe and efficient walking behavior. Equally important is the ability to quickly deadapt and update behavior that is no longer appropriate for the given context. It has been suggested that anxiety induced via postural threat may play a fundamental role in disrupting such deadaptation. We tested this hypothesis, using the "broken escalator" phenomenon: Fifty-six healthy young adults walked onto a stationary walkway ("BEFORE" condition, 5 trials), then onto a moving walkway akin to an airport travelat

Beneficial effects of tannic acid on comorbid anxiety in cecal ligation and puncture-induced sepsis in rats and potential underlying mechanisms.

Sepsis-associated encephalopathy (SAE), a neurological dysfunction caused by sepsis, is the most common complication among septic ICU patients. Given the major role of inflammation in the pathophysiology of sepsis-induced anxiety, an extreme and early manifestation of SAE, the present study examined whether tannic acid, as an anti-inflammatory agent, has anxiolytic effects in cecal ligation and puncture (CLP)-induced sepsis. Forty male Wistar rats were assigned to four groups: (1) sham; (2) sham + tannic acid; (3) sepsis and (4) sepsis + tannic acid. Sepsis was induced by cecal ligation and pu

Impact of Preoperative Psychiatric Profile in Bariatric Surgery on Long-term Weight Outcome.

BACKGROUND: Conflicting results have been reported regarding the predictive value of preoperative psychological assessment and weight outcome after bariatric surgery. This might be attributed to different factors affecting early weight loss and long-term weight loss. Herein, we investigated whether preoperative psychiatric profile was associated with preoperative BMI and with both early (1 year) and long-term (5 years) weight loss after Roux-en-Y gastric bypass (RYGB). METHODS: Prospective observational cohort study of patients undergoing RYGB between 2013 and 2019. Symptoms related to anxiet

Expression levels of miR-132, miR-134, and miR-195 in treatment-naïve children and adolescents with obsessive-compulsive disorder: a case-control study.

BACKGROUND: Obsessive–compulsive disorder (OCD) is a chronic neuropsychiatric condition characterized by obsessions and/or compulsions and arises from a complex interaction of genetic and environmental factors. Emerging evidence suggests that microRNAs (miRNAs) may play a role in the neurobiological mechanisms underlying psychiatric disorders; however, their involvement in pediatric OCD remains unclear. This study aimed to compare serum expression levels of selected miRNAs between children and adolescents with OCD and healthy controls. METHODS: This study included 50 children and adolescents w
