Identification and Characterization of Antiyeast Organic Acids Produced by Lactiplantibacillus plantarum 3121M0s Derived from Mongolian Traditional Fermented Milk, Airag.

Lactic acid bacteria are beneficial for food biopreservation by inhibiting not only bacteria but also fungi. However, reports on the control of fungi, especially yeasts, by lactic acid bacteria are limited. In this study, strain 3121M0s derived from Mongolian traditional fermented milk, airag, was selected with relatively high antiyeast activity among 236 strains, and identified as Lactiplantibacillus plantarum. The activity was exhibited under acidic conditions and remained stable after heating. It was also highly resistant to catalase and proteases, indicating that the primary antiyeast subs

Development of a Method for in vivo Direct Monitoring of Stored Fat Burning Using Stable Isotope-labeled Fatty Acids.

We developed a novel approach for direct and rapid evaluation of the fat-burning effects of various food ingredients in vivo. Male ICR mice were fed a diet containing stable isotope-labeled palmitic acid, and fat oxidation was quantified by measuring the production of labeled carbon dioxide (13CO2) generated during β-oxidation. By evaluating the 24-h fat burning profile, we demonstrated the immediate fat-burning effect of caffeine and the gradual fat-burning effect of curcumin. This methodology allows fat-burning-specific detection rather than fat absorption inhibition, thereby providing a pow

Standardized ileal digestibility of amino acids in corn distillers dried grains with solubles fed to gestating and lactating sows.

This study determined the standardized ileal digestibility (SID) of amino acids (AA) and crude protein (CP) in seven corn distillers dried grains with solubles (DDGS) fed to mid-gestating, late-gestating, and lactating sows. Seven representative corn DDGS samples were collected from ethanol plants in seven different provinces across China. Sixteen sows (Landrace × Yorkshire; parity 4) with a T-cannula in the distal ileum were assigned to a repeated 8 × 3 incomplete Latin square design (three periods and eight diets) during mid-gestation, late-gestation, and lactation stages. The diets included

Optimization of protocol for analysis of dicarboxylic acids in ambient aerosol samples using GC-MS: method comparison and application.

Dicarboxylic acids (DCAs), with their deliquescence and hygroscopic nature, can function as cloud condensation nuclei (CCN) and ice nuclei (IN), affecting rainfall patterns. DCA analysis can serve as organic molecular markers for anthropogenic and biogenic sources. Very few studies deal with the optimization of the protocol for qualitative and quantitative analysis of DCAs using gas chromatography-mass spectrometry (GC-MS). In this study, we have optimized the extraction of DCAs from aerosol samples by employing several organic solvents with differing relative polarities. Extraction efficienci

Visible Light-Driven Benzylic C(sp3)-H Carboxylation Enables Synthesis of 13C-Labeled (±)-α-Amino Acids with 13C-Formate.

13C-labeled α-amino acids are important molecules in biological studies and drug development. Cost-effective synthesis of α-amino acids with a high level of 13C incorporation under mild conditions remains limited. Herein, we report the development of a benzylic C(sp3)-H carboxylation method to prepare highly 13C-labeled α-amino acids, i.e., arylglycines, using near-stoichiometric and user-friendly sodium 13C-formate. The sequential reaction pathway of in situ 13C-carbon dioxide (CO2) generation followed by C-H carboxylation via 1,2-HAT was suggested by mechanistic studies.

Dynamic and structural insights into hydrogen-bonded interpolymer complexes of poly(2-alkyl-2-oxazolines) with poly(carboxylic acids).

The formation of interpolymer complexes (IPCs) between poly(2-alkyl-2-oxazolines) (PAOx) and poly(methacrylic acid) (PMAA) in solutions has been investigated using polarized luminescence relaxation and small-angle X-ray scattering (SAXS) structural methods. The results reveal that hydrophobic interactions contribute to the formation of IPCs in both aqueous and methanol solutions, influenced by the length of the side group in PAOx. The relaxation times, which can be used to quantify the intramolecular mobility of the luminescently labeled poly(methacrylic acid) (PMAA*), increase from 80 to 680

Non-esterified fatty acids disrupt hepatic lipid metabolism and mitochondrial function via TLR4/MyD88/IRAK2 signaling in bovine hepatocytes.

Excessive non-esterified fatty acids (NEFA) in the blood often lead to ketosis and fatty liver in peripartal cows. The integrity of mitochondrial structure and function plays a key role in the development of lipid metabolism imbalance and inflammation. However, the precise molecular mechanisms of NEFA's impact on these processes remain unclear. To explore how NEFA (0, 0.3, 0.6, 1.2, and 2.4 mM) influence lipid metabolism, inflammation, and mitochondrial function in perinatal cow hepatocytes. Bovine primary hepatocytes were isolated from five 1-day-old Holstein calves using collagenase IV perfu

Fatty acids and physical activity are critical for β-cell mass and insulin sensitivity: Pathways to T2D and prevention.

Existing mathematical models investigating the progression of type 2 diabetes (T2D) over time primarily focus on glucose, insulin, β-cell mass, and other related factors, while often omitting fatty acids (FA) as an explicit variable-despite FA being a major energy source for the body. There exists a complex network of dynamical interactions among glucose, insulin, FA, and β-cell mass. To gain deeper insights into the metabolic dynamics and pathophysiology of T2D, it is essential to incorporate FA into such models. In this paper, we extend the classic Topp's GIβ model by explicitly incorporatin

The composition of branched-chain amino acids modulates response to nucleotide supplementation of low-protein diets in nursery pigs.

A total of 160 pigs (initial body weight [BW], 5.79 ± 0.01 kg) were used to investigate the effects of dietary nucleotides and branched-chain amino acids (BCAA) supplementation of low-protein diets on growth performance, blood metabolites, short-chain fatty acids (SCFA), apparent ileal and total tract digestibility of nutrients (AID and ATTD). Following a 3-d adaptation period on a common diet after weaning, pigs were randomly assigned to 1 of the 5 dietary treatments using a randomized complete block design. Each treatment consisted of 8 replicate pens with 4 pigs per pen. The experimental di

Comparative analysis of amino acids and volatile compounds as markers in Lueyang black-boned chicken and Arbor Acres broilers.

Derived from a long history of artificial and natural selection, Lueyang black-boned chicken is preferred by consumers due to its unique meat characteristics. The primary aim of this study was to systematically identify key amino acids and volatile organic compounds (VOCs) that differentiate Lueyang black-boned chicken (LBC) from AA chicken. The contents of essential amino acids, non essential amino acids and flavor amino acids were higher in the breast muscle of LBC compared to AA chicken. Volatilomic analysis revealed that the meat contains various VOCs, including esters, aldehydes, ketones,

Decarboxylative sulfinamidation of N-sulfinylamines with carboxylic acids via a photochemical iron-mediated ligand-to-metal charge transfer process.

Sulfinamides are valuable four-valent sulfur compounds in medicinal chemistry, yet methods relying on simple and readily available feedstocks are scarce. Herein, a novel and efficient strategy was developed to facilitate the direct decarboxylative sulfinamidation of carboxylic acids via a photoinduced iron-mediated ligand-to-metal charge-transfer process. Notably, this protocol exemplifies an environmentally benign approach to the synthesis of a range of structurally diverse sulfinamides with an excellent functional group compatibility, thereby rendering it suitable for the late-stage modifica

IMpact of therapY using coleSevelam treatment reducing bile acids in patients with fonTan cIrCulation (MYSTIC): Rationale and study design.

RATIONALE: Patients with a Fontan circulation suffer from progressive multiorgan dysfunction, yet central biochemical drivers remain poorly defined. Our recent work exploring metabolomic analyses have identified - elevated circulating bile acids (BAs) in adult Fontan patients compared with healthy controls. Elevated BAs, especially secondary and hydrophobic ones produced by the gut microbiome were found to correlate with worse exercise capacity, greater frailty, and impaired hemodynamics. PRIMARY HYPOTHESIS: Bile acid accumulation may contribute to Fontan pathophysiology. Colesevelam, a nonab

Effects of Clozapine on N-Methyl-D-Aspartate Glutamate Receptor-Related Amino Acids in the Rat Medial Prefrontal Cortex.

Clozapine is an atypical antipsychotic drug used for schizophrenia with treatment-refractory symptoms to other antipsychotics. Since a body of evidence indicates the involvement of the dysfunction of the N-methyl-D-aspartate type glutamate receptor (NMDAR) in the pathophysiology of antipsychotic-resistant and responsive symptoms of schizophrenia, we have evaluated the exact mechanisms underlying the superior clinical efficacy of clozapine by studying its effects on the extracellular signaling of NMDAR-related amino acids in the rat medial prefrontal cortex of freely moving rats using an in viv

Enhancing concurrent production of volatile fatty acids and phosphorus minerals from waste activated sludge via magnesium ferrate pre-oxidation.

This study investigated the strategy of magnesium ferrate (MF) pre-oxidation to enhance acidogenic fermentation of waste activated sludge (WAS), targeting the simultaneous production of volatile fatty acids (VFAs) and phosphorus-minerals (struvite and vivianite). Results showed that such fermentation produced a high-value liquid within four days, achieving a peak VFA content of 241 ± 4 mg COD/ g VSfeed, ammonia nitrogen levels below 350 mg/L of and PO43-P under 3 mg/L. Further investigation revealed that the MF pre-oxidation raised the pH, enhanced key hydrolases activity, enriched acidogens a

Olive-derived bioactive combinations: synergistic angiotensin-I-converting enzyme (ACE) inhibition and distinct mechanisms of triterpenic acids and phenolics.

BACKGROUND: Cardiovascular diseases, largely driven by hypertension, remain the leading cause of mortality worldwide. Angiotensin-I-converting enzyme (ACE) inhibition is a well-established therapeutic approach; however, synthetic inhibitors are frequently associated with adverse effects. Natural bioactive compounds, particularly those derived from olives, have been proposed as potential alternatives or complementary agents. RESULTS: This study evaluated the ACE-inhibitory potential of olive-derived triterpenic acids (oleanolic acid (OA) and maslinic acid (MA)) and phenolic compounds (hydroxyt

Therapeutic strategies for hypertension: exploring the role of microbiota-derived short-chain fatty acids in kidney physiology and development.

Gut microbiota have emerged as a key regulator of systemic health, influencing various physiological processes, including kidney development, function, and blood pressure regulation. This review highlights the role of microbiota-derived short-chain fatty acids (SCFAs), primarily acetate, propionate, and butyrate, in the gut-kidney axis, focusing on their signaling mechanisms, vascular effects, and developmental implications. Evidence suggests that SCFAs modulate kidney development and function and exert anti-inflammatory, antioxidant, and vasoregulatory effects through specific G protein-coupl

Green coffee bean extract rich in chlorogenic acids prevents muscle capillary regression via inhibiting oxidative stress and enhancing angiogenesis.

Reactive oxygen species (ROS) production in disused skeletal muscle induces capillary regression, which disrupts the balance of pro- and anti-angiogenic factors. We tested whether green coffee bean extract containing rich chlorogenic acid, which has antioxidant properties, can prevent capillary regression caused by muscle atrophy. The study included 24 female Sprague-Dawley rats, randomly assigned to four groups: control (CON), 2-week hindlimb unloading (HU), 2-week hindlimb unloading supplemented with coffee extract rich in chlorogenic acids (HU+50%CGA), and 2-week hindlimb unloading suppleme

Keto Acids Attenuate Skeletal Muscle Atrophy in Chronic Kidney Disease via Inhibiting Pyroptosis and Upregulating Irisin Precursor FNDC5 Expression.

It is widely accepted that keto acids supplementation can protect skeletal muscle from atrophy. Pyroptosis has been considered to be one of the new mechanisms of muscle atrophy. This study aimed to explore the effects and mechanisms of keto acids supplementation on chronic kidney disease (CKD)-induced skeletal muscle atrophy. In vitro, C2C12 myoblast cells were treated with indoxyl sulfate (IS, 1 mM) and leucine (Leu, 0 ng/mL, 50 ng/mL or 100 ng/mL). In animal experiment, animals were divided into four groups: normal control (NC) group (wildtype mice), CKD group (wildtype mice with CKD modelin

Extracorporeal photopheresis induces the release of anti-inflammatory fatty acids and oxylipins and suppresses pro-inflammatory sphingosine-1-phosphate.

AIMS: Extracorporeal photopheresis (ECP) is a UVA-based phototherapy of whole blood and well established as a first line or combination therapy for the treatment of cutaneous T-cell lymphoma, systemic sclerosis, graft-versus-host disease and is used to control organ transplant rejection. While the proapoptotic activity on activated T-cells is evident, the clinical efficacy of this treatment also appears to be based on other yet unknown mechanisms. In this study, we aimed to identify novel mechanisms of ECP regardless of the patient's background situation. MAIN METHODS: To better understand th
