Diabetes mellitus (DM), a chronic metabolic disorder characterized by persistent hyperglycemia, poses a significant global health burden. Berberine (BBR), a natural isoquinoline alkaloid derived from medicinal plants such as Coptidis Rhizoma, has shown therapeutic promise for DM and its complications. Preclinical and clinical studies have revealed that BBR exerts multi-target effects, including remodeling the gut microbiome, promoting insulin secretion, ameliorating insulin resistance, and inhibiting hepatic gluconeogenesis. Its protective mechanisms against diabetic complications, such as nep

Protein misfolding and aggregation into amyloid fibrils (AFs) are critical issues associated with protein stability during food processing and storage, as well as with pathological damage in organisms. Hen egg white lysozyme (HEWL), a widely used protein in egg products, meat processing, and food preservation, serves as a classical model for investigating amyloidogenesis. Piperlonguminine (PPR), a low-toxicity bioactive alkaloid from Piperis Longi Fructus (a medicinal and food-homologous spice), has not yet been reported to regulate amyloid fibrillation in food proteins. This study explored th

The neonicotinoid insecticide, imidacloprid (IMI), is one of the widely used pesticides with well-documented serious health effects that are noticeable with long-term exposure. However, the long-term effects of IMI on cardiac tissues have not been fully elucidated. Herein, we investigated the mechanisms of IMI-induced cardiotoxicity. Additionally, we examined the potential protective effects of the natural alkaloid, berberine (BBR), against IMI-induced cardiotoxicity. Rats received IMI (45 mg/kg/day, orally) for 30 days, alone or in combination with BBR-loaded liposomes (BBR-Lip) at a dose of

Corydaline (COR), a natural isoquinoline alkaloid, was evaluated for its anticancer potential against doxorubicin-resistant Cal27 (Dox-Cal27) oral squamous carcinoma cells. Cells treated with increasing concentrations of COR (0-5 µM) were assessed by Annexin V/PI staining, cell-cycle profiling, migration/invasion assays, and RT-qPCR. COR induced a concentration-dependent increase in apoptosis and a pronounced G2/M phase arrest, accompanied by significant inhibition of migration and invasion. At the transcriptional level, COR downregulated the anti-apoptotic gene Bcl-2 and metastasis-associated

BACKGROUND: Mitophagy is a critical mitochondrial quality control mechanism that limits neuronal injury following cerebral ischemia/reperfusion injury (CI/RI). Tetramethylpyrazine (TMP), a bioactive alkaloid from Ligusticum chuanxiong Hort., exhibits neuroprotective effects in cerebrovascular disorders. However, whether these effects involve mitophagy regulation remains unclear.
METHODS: CI/RI was induced using a middle cerebral artery occlusion/reperfusion (MCAO/R) model in mice and an oxygen-glucose deprivation/reoxygenation (OGD/R) model in HT22 cells. Neurological function, infarct volume

Targeted enrichment of tocopherols in fungal fermentation systems remains a significant metabolic engineering challenge. This study presented a novel strategy employing homogentisic acid (HGA) modulation to efficiently enhance tocopherol production in the M. purpureus-fermented coix seed (MFCS) system. Three strategies of exogenous HGA supplementation (Exo-HGA), endogenous HGA accumulation (End-HGA), and the combination of exogenous HGA supplementation and endogenous HGA accumulation (Exo + End-HGA) were systematically investigated. While End-HGA optimally promoted lovastatin, pigment, and bio

Galanthamine, a therapeutic Amaryllidaceae alkaloid produced exclusively by species within the Amaryllidoideae subfamily, is a key treatment for early-stage symptoms of Alzheimer's disease. Elucidating its biosynthetic pathway is essential for strategies aimed at enhancing production through metabolic engineering. Galanthamine derives from the metabolic precursor 4'-O-methylnorbelladine, which undergoes cytochrome P450-mediated para-ortho' C-C phenol coupling to yield nornarwedine. Two competing terminal routes have been proposed: (i) reduction of nornarwedine to norgalanthamine, followed by N

Sepsis represents a potentially lethal condition. In this investigation, we demonstrated that berbamine (BBM), a bisbenzylisoquinoline alkaloid, mitigates systemic inflammation and also alleviates acute cardiac impairment in septic mice. The cecal ligation and puncture (CLP)-induced sepsis murine model was developed. BBM at three different doses (25, 50, and 100 mg/kg) was administered before CLP surgery, and a standard control received dexamethasone. The effect of BBM pre-treatment on sepsis mitigation was evaluated using echocardiographic, histopathological, biochemical, and molecular method

BACKGROUND: 5-Methoxy-N,N-dimethyltryptamine (5-MeO-DMT), also officially known as mebufotenin, is a naturally occurring serotonergic psychedelic alkaloid found in the venom of the Sonoran Desert toad ( Incilius alvarius ) and various plant species.
AREAS OF UNCERTAINTY: Trace levels of 5-MeO-DMT may be produced endogenously in humans, but its physiologic role remains unclear. Safety profiles indicate low risk in controlled settings, though longer-term follow-ups with human subjects may be needed. Phenomenological overlaps with near-death experiences are noted but debated.

Scopolamine, also known as hyoscine, is a naturally occurring tropane alkaloid derived from plants of the Solanaceae family. Clinically, the compound has long been used for the prevention of motion sickness and postoperative nausea and vomiting, as well as for ophthalmological procedures requiring mydriasis and cycloplegia. However, beyond these established indications, increasing attention has been directed toward its broader neuropharmacological actions. This narrative review aims to summarise current knowledge regarding the pharmacological properties of scopolamine, with particular emphasis

Sepsis, a life-threatening disorder driven by a dysregulated host response to infection, is frequently accompanied by acute lung injury (ALI), worsening disease severity, and mortality. Excessive macrophage-mediated inflammation is central to its pathogenesis. Given emerging evidence that mitochondria-endoplasmic reticulum (ER) crosstalk drives inflammatory injury, we investigated whether modulating this interaction could mitigate sepsis-induced damage. Using LPS-stimulated THP-1 macrophages and a murine model of LPS-induced sepsis, we evaluated the anti-inflammatory and organ-protective effec
