Exploring the shared genetic architecture of migraine and autoimmune diseases.

BACKGROUND: Migraine (MIG) frequently co-occurs with autoimmune diseases (ADs), but the shared genetic basis underlying this comorbidity remains unclear. METHODS: Using publicly available GWAS summary statistics from European populations, we applied an integrated framework to investigate the shared genetic architecture between MIG and seven ADs (SLE, UC, CD, PSC, T1D, RA, MS). Genetic correlations and polygenic overlap were evaluated using LDSC, HDL, and MiXeR. Shared pleiotropic loci were identified via PLACO and annotated with FUMA. Gene-level analyses were conducted using MAGMA, followed b

Nationwide epidemiological survey of autoimmune pancreatitis in Japan in 2021.

BACKGROUND: To update the clinic-epidemiological features of autoimmune pancreatitis (AIP) in Japan, we conducted the fifth nationwide survey. METHODS: We performed a two-stage survey of patients with AIP who were treated at selected hospitals in 2021. The first-stage survey estimated the total number of patients with AIP, and the second-stage survey analyzed detailed clinical characteristics. RESULTS: The estimated number of patients with AIP in 2021 was 16,750 (prevalence, 13.3 per 100,000 persons), representing a 25% increase from 2016. The number of newly diagnosed cases was 4210 (annual

Exploring the Role of Autoimmune Conditions in Acute Interstitial Pneumonia.

BACKGROUND: Clinical factors influencing outcomes of patients hospitalized with acute interstitial pneumonia (AIP) are not well described because of the condition's rarity. RESEARCH QUESTION: Do patients with AIP and autoimmune conditions differ from patients with AIP without autoimmune conditions in mortality, recurrence, mechanical ventilation, discharge disposition, and length of hospital stay? STUDY DESIGN AND METHODS: This retrospective cohort study used US MarketScan and Medicare claims (2016-2023). Diagnoses were identified using International Classification of Diseases, 10th Revision

Effectiveness and Safety of Immunosuppressive Therapy in Recurrent Autoimmune Pancreatitis.

Background Autoimmune pancreatitis (AIP) is a distinct type of chronic pancreatitis that responds well to glucocorticoid therapy. However, some patients experience frequent relapses or become glucocorticoid-dependent. In Western countries, immunosuppressive agents such as azathioprine are widely used to maintain remission. In contrast, evidence for the use of immunosuppressive agents for AIP is limited in Japan and other Asian countries. Therefore, this study aimed to evaluate the efficacy and safety of immunosuppressive agents in patients with refractory AIP. Methods We retrospectively analyz

A two-hit mechanism triggers autoimmune hair loss.

Autoimmune diseases arise from a breakdown of immune tolerance, yet the mechanisms that initiate tissue-specific autoimmunity remain poorly understood1-4. Here we identify a two-hit mechanism in which immune dysregulation and physiological neural activation converge to trigger autoimmune alopecia. We show that transient depletion of regulatory T (Treg) cells during the anagen phase of the hair cycle, but not during telogen, induces CD8+ T-cell-mediated destruction of immune-privileged hair follicle stem cells (HFSCs), resulting in hair loss. This anagen-restricted susceptibility depends on sym

Incidence of autoimmune diseases in children with atopy treated with dupilumab.

BACKGROUND: The type 2 inflammation-blocking agent dupilumab has gained traction as an effective treatment of multiple atopic diseases. New-onset autoimmune manifestations have been infrequently reported in dupilumab-treated atopic patients, primarily among adults. Autoimmune outcomes in atopic children on dupilumab are less clear. OBJECTIVE: We aimed to determine if dupilumab treatment of atopic children is associated with increased autoimmune or autoinflammatory disease diagnosis risk. METHODS: Using a multi-institutional electronic health record database composed of 10 PEDSnet academic he

Autonomic Dysfunction and Sleep Disorder in 35 Autoimmune Encephalitis.

INTRODUCTION: Considering that sleep disorders have garnered limited attention in patients with Autoimmune Encephalitis, often eclipsed by other prevailing neurological and psychiatric symptoms within this subgroup, identifying and addressing underlying sleep disturbances could alleviate AE-related complications and improve long-term outcomes. Consequently, the present study aims to ascertain the prevalence of sleep disorders and autonomic dysfunction in individuals grappling with autoimmune encephalitis within the confines of Isfahan City. METHODS: This descriptive-analytical study initially

Autoimmune Progesterone Dermatitis: A Case Report and Review of Literature.

Autoimmune progesterone dermatitis (APD) is a rare hypersensitivity reaction triggered by progesterone, predominantly affecting women in the luteal phase of the menstrual cycle. This case describes a 50-year-old woman with hypothyroidism and prolonged use of oral contraceptives, who presented with recurrent pruritic skin lesions occurring 3-10 days before menstruation and resolving post-menses. Clinical examination identified the lesions as urticarial plaques. After ruling out other conditions, such as urticarial vasculitis and autoinflammatory syndromes, a diagnosis of APD was established. The burden of autoimmune pulmonary alveolar proteinosis: a systematic review.

BACKGROUND: Autoimmune pulmonary alveolar proteinosis (aPAP) is characterised by abnormal alveolar surfactant accumulation and reduced pulmonary gas transfer. Disease severity and progression depend on pulmonary surfactant accumulation, the rate of which varies widely among patients. Currently, whole-lung lavage (WLL) is the most widely accepted therapy. This review addresses the burden of aPAP on patients, caregivers and society. METHODS: MEDLINE and Embase databases were systematically searched for reports on the manifestations, treatment burden, caregiver impact and healthcare costs of aPA

Recent advances in the mechanisms and treatment of cuproptosis in autoimmune diseases.

Autoimmune diseases, characterized by the immune system's erroneous attack on the body's own tissues, are highly challenging to treat. Recently discovered cuproptosis, triggered by excess copper ions, leads to the abnormal accumulation of acetylated proteins and mitochondrial dysfunction, and has become a research hotspot in this field. Recent studies have shown that cuproptosis-related genes (CRGs) play a potential role in various autoimmune diseases, such as systemic lupus erythematosus (SLE), rheumatoid arthritis (RA), Primary Sjögren's syndrome (pSS), and ankylosing spondylitis (AS), and h

Cardiac Troponin in Autoimmune Rheumatic Diseases: Lights and Shadows.

Cardiovascular involvement is a leading cause of morbidity and mortality in autoimmune rheumatic diseases (ARDs), arising through diverse mechanisms such as microvascular dysfunction, inflammation, fibrosis, in addition to traditional cardiovascular risk factors. Cardiac troponin (cTn), a sensitive and specific marker of myocardial injury, is increasingly investigated for its diagnostic and prognostic value. However, interpretation in ARDs is complex, as cTn elevations may reflect primary cardiac involvement, comorbidities or analytical interferences. This review explores the value of cTn as a

Autoimmune disease-associated lymphomas: research progress and review.

Autoimmune diseases (ADs) are strongly associated with a significantly increased risk of lymphoma, with the standardised incidence ratio (SIR) markedly elevated in certain conditions, most notably in Sjögren's disease (SjD) where an SIR as high as 18.8 has been reported. The risk is particularly prominent for diffuse large B-cell lymphoma (DLBCL) and mucosa-associated lymphoid tissue (MALT) lymphoma. This review systematically elucidates the epidemiological features, pathological mechanisms, risk factors, and therapeutic strategies of ADs-associated lymphomas. Epidemiological studies have conf

Cleavage-Resistant CYLD Protects Against Autoimmune Hepatitis.

Autoimmune hepatitis (AIH) is an immune-mediated liver disease that can progress to fibrosis, cirrhosis, and hepatocellular carcinoma. However, the pathogenic mechanisms underlying AIH remain poorly understood, limiting the development of effective therapies. Here, using a concanavalin A-induced murine model of experimental autoimmune hepatitis (EAH), proteolytic cleavage of the deubiquitinase cylindromatosis (CYLD) at Asp215 is identified as a critical molecular event that promotes disease progression. Mice harboring a macrophage-specific, cleavage-resistant CyldD215A/D215A mutation are marke

[Exploring the research on the molecular mechanisms of autoimmune liver diseases].

Autoimmune liver diseases include subtypes such as autoimmune hepatitis and primary biliary cholangitis, which are caused by abnormal immune system attacks on hepatocytes or bile ducts. This article reviews their molecular mechanisms from three aspects: genetics, environment, and immunity. Human leukocyte antigen (HLA) gene polymorphisms such as DRB10301 and DRB10405 show regional specificity, and non-HLA gene variants such as CTLA4 and FAS also contribute to disease development in terms of genetic factors. Cross-immune responses are triggered through 'molecular mimicry," disturbances in bile

Revisiting CCL22 in autoimmune diseases: pathogenesis and clinical implications.

CCL22 (macrophage-derived chemokine, MDC) is an important member of the CC chemokine family. By binding to its receptor CCR4, it regulates immune cell trafficking, plays a dual role in maintaining immune homeostasis and promotes immune-mediated inflammation. The biological effects of CCL22-CCR4 signaling in autoimmune diseases are highly dependent on the tissue microenvironment. Under steady-state conditions, CCL22 secreted by dendritic cells and macrophages recruits regulatory T-cells (Tregs) to peripheral tissues, suppresses excessive immune responses, and maintains immune tolerance. When CC

Albendazole-Induced Liver Injury Mimicking Autoimmune Hepatitis.

Drug-induced liver injury is one of the leading causes of acute liver injury and may mimic other liver diseases, including idiopathic autoimmune hepatitis. Although albendazole is generally a safe antiparasitic agent, it can be associated with rare cases of clinically significant liver injury; it can even cause a mixture of both types of injuries, or a type of hepatitis that is similar to an autoimmune disease, such as positive autoantibodies and elevated IgG, making differential diagnosis more difficult. We present a case of albendazole-induced liver injury mimicking autoimmune hepatitis in a
