Targeting downstream subcellular YAP activity as a function of matrix stiffness with Verteporfin-encapsulated chitosan microsphere attenuates osteoarthritis.

Changes in the stiffness of chondrocyte extracellular matrix (ECM) are involved in the pathological progression of osteoarthritis (OA). However, the downstream responses of cartilage ECM stiffness are still unclear. YAP (Yes-associated protein) has been extensively studied as a mechanotransducer, we thus hypothesized that by targeting the downstream molecule activity of ECM stiffness could maintain chondrocyte phenotype and prevent cartilage degeneration in OA. Here, we showed that human cartilage matrix stiffened during pathological progression of OA, and the chondrocyte YAP activity was asso

An optical probe for detecting chondrocyte apoptosis in response to mechanical injury.

Cartilage injury induced by acute excessive contact stress is common and mostly affects young adult. Although early detection of cartilage injury may prevent serious and lifelong arthritic complications, early detection and treatment is not possible due to the lack of a reliable detection method. Since chondrocyte injury and subsequent cell death are the early signs of cartilage injury, it is likely that cartilage cell apoptosis can be used to predict the extent of injury. To test this hypothesis, a near infrared probe was fabricated to have high affinity to apoptotic cells. In vitro tests sho

Ear reconstruction after auricular chondritis secondary to ear piercing.

The recent fad of high ear piercing in the pinna has led to an increased incidence of auricular chondritis, which leads to dissolution of the cartilage and residual ear deformity. The typical postpiercing chondritis deformity presents as a structural collapse of the superior helical rim, scaphal cartilage, and the adjacent antihelix. The skin envelope is usually preserved, but it may be scarred from the infectious process and from previous drainage incisions. In the present article, the authors present a systematic approach to reconstruction of these acquired ear deformities. Careful assessmen

Bovine achondrogenesis: evidence for defective chondrocyte differentiation.

A survey study of growth cartilage abnormalities in bovine bone dysplasias revealed that a disorder in Holstein cattle called bulldog calf closely resembles human achondrogenesis Type II. Substantial amounts of Type I collagen and other non Type II collagens were detected in the bulldog cartilage which was comprised primarily of extensive vascular canals and cells having the characteristics of hypertrophic and degenerative chondrocytes normally found in the growth plate. It is proposed that chondrocytes throughout the bulldog growth cartilage prematurely differentiate into hypertrophic cells t

A histologic study of the growth of the subglottis after endolaryngeal trauma.

In young rabbits the histologic features of the growth of the subglottis were studied after an endolaryngeal trauma. This lesion of the larynx leading to a subglottic stenosis, caused specific pathologic changes of the cricoid cartilage and the subepithelial tissues. Activation of chondrocytes resulted in proliferation and regeneration of destructed cartilage. The normal subepithelial structures were replaced by granulation tissue differentiating into a thick fibrous scar and into cartilage. This ectopic cartilage together with the repaired cricoid formed a composite thickened 'ring'-like cart

Curcumin ameliorates experimentally induced knee osteoarthritis in rats via regulation of synovial inflammatory.

This study was designrd to investigate the effect of curcumin on chondrosynovitis in knee osteoarthritis (KOA) rat models and to explore its mechanism of action. The damage of knee cartilage tissue was detected by haematoxylin and eosin (HE) staining, toluidine blue, safranin O-fast green and a scoring system by Mankin were used to assess the histopathological features of cartilage. Chondrocytic cells of apoptosis was visualized by TUNEL staining; inflammatory cytokines (interleukin-6, interleukin-1beta (IL-1β) and tumor necrosis factor alpha (TNF-α)) in the synovial fluid were examined by ELI

Mesenchymal stromal cells donate mitochondria to articular chondrocytes exposed to mitochondrial, environmental, and mechanical stress.

Articular cartilage has limited healing capacity and no drugs are available that can prevent or slow the development of osteoarthritis (OA) after joint injury. Mesenchymal stromal cell (MSC)-based regenerative therapies for OA are increasingly common, but questions regarding their mechanisms of action remain. Our group recently reported that although cartilage is avascular and relatively metabolically quiescent, injury induces chondrocyte mitochondrial dysfunction, driving cartilage degradation and OA. MSCs are known to rescue injured cells and improve healing by donating healthy mitochondria

Knee loading inhibits osteoclast lineage in a mouse model of osteoarthritis.

Osteoarthritis (OA) is a whole joint disorder that involves cartilage degradation and periarticular bone response. Changes of cartilage and subchondral bone are associated with development and activity of osteoclasts from subchondral bone. Knee loading promotes bone formation, but its effects on OA have not been well investigated. Here, we hypothesized that knee loading regulates subchondral bone remodeling by suppressing osteoclast development, and prevents degradation of cartilage through crosstalk of bone-cartilage in osteoarthritic mice. Surgery-induced mouse model of OA was used. Two week

[Plica medialis, patellar shape and chondromalacia].

The influence of the shape of the patella and of the plica medialis parapatellaris on cartilage changes was studied in 161 knee joints (autopsy material). The cartilage changes were cartographically recorded and transferred to a computer matrix. The type and size of the plica were documented. The material studied was classified according to age, patellar shape, and degree of prominence of the plica. Cartilage lesions were found on the odd facet even in younger material. The cartilage damage depended on the shape of the patella. The more convex the medial facet, the less cartilage damage had oc

Quantification of morphometric changes in murine experimental osteoarthritis using image analysis.

The aim of this study was to evaluate a method for the quantification of cartilage erosions and osteophyte sizes in a murine model of osteoarthritis (OA). Mice in which OA was induced in the knee joint by intra-articular injection of bacterial collagenase were used. With an interactive image analysis system, the areas occupied by osteophytes and the areas of erosions of the articular cartilage were measured on histological sections by two independent observers at two time points. Measurements of osteophyte areas and cartilage loss at the tibial plateau showed good reproducibility, whereas meas

Changes of the subchondral bone microchannel network in early osteoarthritis.

OBJECTIVE: We have identified a 3D network of subchondral microchannels that connects the deep zone of cartilage to the bone marrow (i.e., cartilage-bone marrow microchannel connectors; CMMC). However, the pathological significance of CMMC is largely unknown. Here, we quantitatively evaluated how the CMMC microarchitecture is related to cartilage condition, as well as regional differences in early idiopathic osteoarthritis (OA). METHODS: Two groups of cadaveric female human femoral heads (intact cartilage vs early cartilage lesions) were identified, and a biopsy-based high-resolution micro-CT

A new method for investigating osteoarthritis using Fast Field-Cycling nuclear magnetic resonance.

Osteoarthritis in synovial joints remains a major cause of long-term disability worldwide, with symptoms produced by the progressive deterioration of the articular cartilage. The earliest cartilage changes are thought to be alteration in its main protein components, namely proteoglycan and collagen. Loss of proteoglycans bound in the collagen matrix which maintain hydration and stiffness of the structure is followed by collagen degradation and loss. The development of new treatments for early osteoarthritis is limited by the lack of accurate biomarkers to assess the loss of proteoglycan. One p

Co-expression of DKK-1 and Sclerostin in Subchondral Bone of the Proximal Femoral Heads from Osteoarthritic Hips.

BACKGROUND: Osteoarthritis (OA) is a progressively degenerative joint disease influenced by structural and metabolic factors. There is growing evidence that subchondral bone is involved in both symptomatic and structural progression in OA. The Wnt pathway has been implicated in the progression of OA but the expression and function of the Wnt inhibitors, Dikkopf (DKK-1) and sclerostin (SOST), are unclear. METHODS: We examined the regional distribution of DKK-1 and SOST in subchondral bone of the femoral head using resection specimens following arthroplasty in patients presenting with end-stage

Unilateral condylar hyperactivity: a histopathologic analysis of 47 patients.

PURPOSE: To perform a histopathologic analysis of condyles that were resected because of unilateral condylar hyperactivity and compare the results of the bone scan with the histopathologic findings. PATIENTS AND METHODS: A total of 47 resected condyles were histopathologically examined. In 29 cases, a single photon emission computed tomography (SPECT) bone scan was available. For all condylar specimens, a standardized histologic scoring system was used to assess the number of cartilage islands and the thickness of the cartilage layer. The SPECT scans were analyzed by calculating the differenc

Therapeutic Window of Laser Dosimetry for the Treatment of Knee Osteoarthritis.

OBJECTIVES: We propose a non-destructive laser method that uses controlled thermomechanical action to treat degenerative cartilage changes. The technique aims to reduce pain, offers a minimally invasive option for early- to mid-stage degeneration, and lessens the need for joint replacement. METHODS: An ex vivo study on laser treatment of articular cartilage involves theoretical modeling of laser-induced heating, stress relaxation, and cartilage denaturation. Theoretical model simulates the laser treatment of a multilayer system using modulated infrared radiation. It integrates the 3D heat dif

Wood-Derived Hydrogels for Osteochondral Defect Repair.

Repairing cartilage tissue is a serious global challenge. Herein, we focus on wood skeletal structures that are highly porous for cell penetration yet have load-bearing strength, and aim to synthesize wood-derived hydrogels with the ability to regenerate cartilage tissues. The hydrogels were synthesized by wood delignification and the subsequent intercalation of citric acid (CA), which is involved in tricarboxylic acid cycles and essential for energy production, and N-acetylglucosamine (NAG), which is a cartilage glycosaminoglycan, among cellulose microfibrils. CA and NAG intercalation increas

Self-Assembly Culture Model for Engineering Musculoskeletal Tissues.

The goal of a self-assembly tissue engineering is to create functional tissue following a natural cell-driven process that mirrors natural development. This approach to tissue engineering has tremendous potential for the development of reparative strategies to treat musculoskeletal injuries and diseases, especially for articular cartilage which has poor regenerative capacity. Additionally, many bioengineering and culture methods fail to maintain the chondrocyte phenotype and contain the correct matrix composition in the long term. Existing cartilage-engineering approaches have been developed,

Multiple roles of ALK3 in osteoarthritis.

Osteoarthritis (OA) is a chronic degenerative joint disease characterized by progressive cartilage degradation, synovial membrane inflammation, osteophyte formation, and subchondral bone sclerosis. Pathological changes in cartilage and subchondral bone are the main processes in OA. In recent decades, many studies have demonstrated that activin-like kinase 3 (ALK3), a bone morphogenetic protein receptor, is essential for cartilage formation, osteogenesis, and postnatal skeletal development. Although the role of bone morphogenetic protein (BMP) signalling in articular cartilage and bone has been
