Cytogenetics of genetic counseling patients in Pelotas, Rio Grande do Sul, Brazil.

Influenza A virus has been circulating in birds in Eurasia for more than 146 years, but human infection has been sporadic. H5N1 (clade 2.3.4.4b) has recently infected hundreds of species of wild and domestic birds and mammals in North America. Infections include 71 people in the United States. There have been 2 human fatalities (United States and Mexico). We have integrated time-series analysis, molecular phylogenetics, and structural biology to understand how H5N1 is circulating in North America and adapting to new hosts. Our time-series analysis reveals that the circulation of H5N1 follows a

Comparative cytotoxicity of dexamethasone and prednisolone in childhood acute lymphoblastic leukemia.

PURPOSE: The relative cytotoxicity of prednisolone and dexamethasone in acute lymphoblastic leukemia (ALL) is controversial. We therefore compared the direct antileukemic activities of these compounds in stroma-supported cultures of leukemic lymphoblasts. MATERIALS AND METHODS: Bone marrow samples from children with B-lineage ALL were cultured on allogeneic bone marrow-derived stromal layers and exposed to various concentrations of glucocorticoids. After 4 days of culture, the number of viable leukemic cells was counted by flow cytometry and compared with that in parallel cultures without dru

Contemporary enterovirus-D68 isolates infect human spinal cord organoids.

Enterovirus D68 (EV-D68) is a nonpolio enterovirus associated with severe respiratory illness and acute flaccid myelitis (AFM), a polio-like illness causing paralysis in children. AFM outbreaks have been associated with increased circulation and genetic diversity of EV-D68 since 2014, although the virus was discovered in the 1960s. The mechanisms by which EV-D68 targets the central nervous system are unknown. Since enteroviruses are human pathogens that do not routinely infect other animal species, establishment of a human model of the central nervous system is essential for understanding path

The role of sdc-1 in the sex determination and dosage compensation decisions in Caenorhabditis elegans.

Our previous work demonstrated that mutations in the X-linked gene sdc-1 disrupt both sex determination and dosage compensation in Caenorhabditis elegans XX animals, suggesting that sdc-1 acts at a step that is shared by the sex determination and dosage compensation pathways prior to their divergence. In this report, we extend our understanding of early events in C. elegans sex determination and dosage compensation and the role played by sdc-1 in these processes. First, our analysis of 14 new sdc-1 alleles suggests that the phenotypes resulting from the lack of sdc-1 function are (1) an incomp

Reporter-guided photoreceptor differentiation in 2D culture using PGP1-hIPSCs: A comparable alternative to 3D organoid systems?

PURPOSE: To develop and characterize a xeno-free, two-dimensional (2D) differentiation protocol for directing human induced pluripotent stem cells (hIPSCs) toward photoreceptor (PhR)-like cells, using a live-reporter system and transcriptomic analysis to evaluate lineage fidelity and maturation compared to the three-dimensional (3D) culture paradigm. METHODS: A CRISPR/Cas9-engineered PGP1 hIPSC line expressing fluorescent reporters for retinal markers (VSX2, BRN3B, and RCVRN) was differentiated in adherent culture using chemically defined media supplemented with small molecules (T3, DAPT, tau

Advances in stem-cell--generated transplantation therapy for Parkinson's disease.

INTRODUCTION: Human pluripotent stem cells have the potential to differentiate into different cell lineages of the human body, including dopaminergic (DA) neurons. Previous studies have shown that stem-cell--derived DA neurons can improve the motor deficits of Parkinson's disease (PD) animal models. That is why current research interests focus on the development of stem-cell--derived neural cells for transplantation therapies for PD patients. AREAS COVERED: This review article emphasizes the safety and efficacy requirements of human pluripotent stem-cell--derived neural cells and usage of rel

Patient-Derived iPSC-MSC Modelling Reveals Reversible GSK3-Mediated Suppression of Mesenchymal Lineage Differentiation in Legg-Calvé-Perthes Disease.

Legg-Calvé-Perthes disease (LCPD) is an idiopathic paediatric osteonecrosis characterized by avascular injury to the femoral head, resulting in structural collapse and early osteoarthritis. Although mesenchymal stem cell (MSC) dysfunction has been implicated, the underlying mechanisms remain unclear due to the absence of human disease models. Here, we generated patient-specific induced pluripotent stem cells (iPSCs) from dermal fibroblasts of LCPD patients and differentiated them into mesenchymal stem cells (iMSCs) to establish an in vitro platform for mechanistic interrogation. LCPD-iMSCs exh

Exploiting the Role of Hypoxia-Inducible Factor 1 and Pseudohypoxia in the Myelodysplastic Syndrome Pathophysiology.

Myelodysplastic syndromes (MDS) comprise a heterogeneous group of clonal hematopoietic stem (HSCs) and/or progenitor cells disorders. The established dependence of MDS progenitors on the hypoxic bone marrow (BM) microenvironment turned scientific interests to the transcription factor hypoxia-inducible factor 1 (HIF-1). HIF-1 facilitates quiescence maintenance and regulates differentiation by manipulating HSCs metabolism, being thus an appealing research target. Therefore, we examine the aberrant HIF-1 stabilization in BMs from MDS patients and controls (CTRLs). Using a nitroimidazole-indocyani

A potential role of galectin-1 in promoting mouse trophoblast stem cell differentiation.

Galectin-1 is highly expressed in blastocysts and trophoblast giant cells during implantation, and dysregulated galectin-1 is associated with many pregnancy-related abnormalities. Elevated galectin-1 contributes to cancer cells invasion. Here, we found that galectin-1 is expressed in mouse oocytes, preimplantation embryos (all stages), and trophoblast stem (TS) cells. Peak levels of galectin-1 mRNA and protein were detected on day 4 and day 5 after the induction of TS cells differentiation. Overexpression of galectin-1 increased TS cells migration and invasion, whereas knockdown of galectin-1

Cells of origin of squamous epithelium, dysplasia and cancer in the head and neck region after bone marrow transplantation.

Secondary solid tumors that occur after hematopoietic stem cell transplantation (HSCT) are late complications of HSCT. Previously, secondary solid tumors were considered to be recipient-derived cells because transplanted cells do not contain epithelial cells. Recently, however, not only donor‑derived epithelial cells but also donor-derived secondary solid tumors have also been reported in mice and humans. It means that circulating bone marrow-derived stem cells (BMDCs) including hematopoietic stem cells include the stem cells of many tissue types and the precancerous cells of many solid tumors

Rapid engraftment of mast cells of donor origin in a case of acute myeloid leukemia with mast cell leukemia after allogeneic stem cell transplantation.

Mastocytosis is a rare disease characterized by an abnormal increase of mast cells in tissues. We report a case of acute myeloid leukemia (AML) with t(8;21) and mast cell leukemia (MCL) in which the mastocytosis persisted after standard chemotherapy and allogeneic stem cell transplantation, although the myeloid leukemia achieved molecular complete remission soon after induction chemotherapy. Donor-type mast cells were noted on d31 after transplant. No c-kit mutation was found before or after the transplant. This represents the first reported case in which rapid engraftment of mast cells of don

Undifferentiated small round cell tumor of the oral cavity with non-contributory immunohistochemistry: a diagnostic pitfall.

The lack of lineage-specific markers and overlapping shape make undifferentiated round cell cancers of the oral cavity extremely difficult to diagnose. We describe a 47-year-old female patient who had a submucosal lesion affecting her mandibular gingiva and labial mucosa. Histopathological analysis showed that the stroma was infiltrated by sheets of monotonous round cells with hardly noticeable nucleoli without any epithelial involvement. The results of a comprehensive immunohistochemistry panel that included cytokeratin, CD45, S100, desmin, synaptophysin, CD99, CD117, MyoD1, CK20, TTF-1, ER,

RBPJ regulates differentiation and effector programming of intestinal γδ intraepithelial lymphocytes.

Intestinal γδ intraepithelial lymphocytes (IELs) are heterogeneous tissue-resident T cells that contribute to epithelial homeostasis and host defense, but the mechanisms regulating their differentiation and functional states remain incompletely understood. Here, we investigated the role of recombination signal binding protein for immunoglobulin kappa J region (RBPJ) in intestinal γδ IELs using mice with CD8 lineage-specific deletion of Rbpj. RBPJ deficiency did not markedly affect the overall proportion of γδ IELs but altered the distribution of Thy1-defined γδCD8αα IEL subsets in the small in

Neurodevelopmental Blueprint in Gliomas: Toward an Ontogenetic Multimodal Integrated Therapy.

Neuroepithelial tumors, including diffuse gliomas and embryonal neoplasms, remain among the most lethal cancers despite major advances in molecular diagnostics and some novel targeted therapies for subsets of patients. A persistent barrier to progress is the lack of a unifying framework that integrates their cellular hierarchy, spatial heterogeneity, and distinctive patterns of progression. In this review, we synthesize converging evidence that neuroepithelial tumor biology represents a pathologic re-enactment of brain ontogeny. Integrating molecular, single-cell, histoarchitectonic, and anato

Notch signalling governs human enteric nervous system progenitor dynamics.

The enteric nervous system (ENS) is the main branch of the peripheral nervous system that innervates the gastrointestinal tract controlling vital functions. It arises during embryogenesis via migration, proliferation and differentiation of neural crest-derived ENS progenitors. Perturbation of these processes, caused by mutations in key signalling pathway components and transcription factors, prevents progenitor colonisation of the distal gut causing aganglionic phenotypes and enteric neuropathies such as Hirschsprung (HSCR) disease. While animal models implicate Notch signalling in ENS specifi

Lipopolysaccharides Drive Proinflammatory Extracellular Vesicle Secretion in Coronary Artery Endothelial Cells via Noncanonical Inflammasome Activation.

Intestinal dysbiosis and impaired gut barrier function lead to increased blood lipopolysaccharide (LPS) load, a condition known as endotoxemia. We investigated whether intracellular LPS induces activation of the noncanonical caspase-4 and -5 inflammasome in human coronary artery endothelial cells (HCAECs) and whether this contributes to arterial wall inflammation. Cytosolic LPS induced activation of the noncanonical inflammasome in endothelial cells, leading to marked increase in extracellular vesicle (EV) secretion. These EVs were enriched with biologically active proteins, particularly kinas

Current evidence on the cell of origin of meningiomas disputes the arachnoid cap cell dogma.

BACKGROUND: For over a century, the dominant hypothesis has maintained that meningiomas arise exclusively from arachnoid cap cells. However, emerging evidence from single-cell transcriptomics, spatial sequencing, developmental biology, and transgenic animal models suggests a significantly more complex and heterogeneous cellular origin. METHODS: This mini-review revisits historical perspectives and critically evaluates the limitations of the traditional arachnoid-based paradigm. By synthesizing classical histological insights with contemporary molecular and proteomic data, the study explores a

Pregnancy-induced tissue-resident memory-like T cells contribute to tumor control in breast cancer.

Women who have full-term pregnancies have a reduced risk of breast cancer, although the underlying mechanisms are not well understood. Here we show that tissue-resident memory-like T (TRM-like) cells are enriched in the breasts of parous women and mice compared to nulliparous individuals. These cells develop during mid-gestation, persist after lactation and are closely associated with mammary epithelial cells, suggesting dependence on epithelial-derived signals. Impaired alveolar differentiation or a deficiency in epithelial cell-derived cytokines interleukin (IL)-15 and transforming growth fa

Serum exosomes protect heat-stressed mouse 2-cell embryos: Enhanced antioxidant defense and base excision repair of oxidative DNA damage.

High temperatures pose a significant threat to human health; however, there are limited measures to address infertility resulting from elevated temperatures. This study aimed to evaluate the protective effects of exosomes on heat-stressed mouse 2-cell Embryos. The results indicated that exposure to high environmental temperatures caused substantial damage to the developmental potential of embryos, while the inclusion of serum exosomes during early pregnancy demonstrated a beneficial impact. Specifically, high environmental temperatures lead to embryonic retardation and increase oxidative stres

Periosteal mitochondria DNA structures drive aging-associated poor skeletal repair.

Insufficient skeletal repair is the primary threat of health span and lifespan in elders with increasingly vast global burden; yet, to date, the knowledge of resolving this crisis remains limited. In this study, we addressed the specific mechanisms underlying aging-associated poor bone repair, which are driven by the mitochondrial DNA structures mitochondrial G-quadruplex (mtG4). We found that mtG4 is spatiotemporal-wisely accumulated within Pdgfra+ periosteal mesenchymal stromal/stem cells (PPM) both in healthy and premature aging, which substantially increases cellular senescence and the deg
