The characteristics of cortical activation during gait in children with spastic diplegic cerebral palsy.

INTRODUCTION: Cerebral palsy (CP) is a common movement disorder caused by abnormalities or injury to the developing brain. It affects more than 17 million people worldwide and is associated with substantial impairments in postural balance and gait control, particularly in children with spastic diplegic CP. This study aimed to examine differences in cortical activation during walking between children with spastic diplegic CP and children with typical development (CTD), and to explore the relationship between cortical activation and gross motor performance. METHODS: Functional near-infrared spe

Lung cancer and altered brain structure: A causal inference based on MR.

Lung cancer is one of the most prevalent malignancies globally and imposes a substantial burden on mortality and healthcare costs. Emerging clinical evidence suggests that lung cancer may influence brain morphology, particularly the cerebral cortex. This study employed Mendelian Randomization (MR) to investigate the potential causal relationship between lung cancer and cortical structural alterations. Single nucleotide polymorphisms associated with small cell lung cancer (SCLC) and non-SCLC (NSCLC) were instrumental variables derived from the FinnGen consortium. Summary-level data on brain cor

Mouse Gnal transcripts and transcriptomics in isolated dystonia.

Heterozygous loss-of-function GNAL mutations are one established cause of isolated dystonia and hyposmia. Homozygous GNAL mutations have been reported in siblings with generalized dystonia and intellectual disability. GNAL encodes major [NM_001369387.1; Gα(olf)] and long [NM_182978.4; XLGα(olf)] isoforms. In striatal medium spiny neurons, dopamine D1 receptors and adenosine A2a receptors are coupled to adenylyl-cyclase through a heterotrimeric G-protein complex composed of Gα(olf), Gβ2, and Gγ7 subunits. In the cerebellum, Gα(olf) colocalizes with cell-surface corticotropin-releasing factor re

Sleep Fragmentation and Sleep Stages 1 and 2 Are Associated With Amyloid-Beta Burden in Cognitively Normal Older Adults.

Insomnia increases the risk of Alzheimer's disease and is associated with increased amyloid-β (Aβ) levels, but assessments using rigorous objective measures are lacking. The purpose of this cross-sectional study was to determine the association between sleep characteristics and Aβ burden using polysomnography and positron emission tomography (PET). Cognitively normal older adults (ages 60-85) with symptoms of insomnia underwent PET and overnight polysomnography. Pearson correlations determined the association between sleep variables and the standard uptake values ratio (SUVR) within global cer

Associations of Physical Activity Engagement with Cerebral Amyloid-β and Tau from Midlife.

BACKGROUND: Higher midlife physical activity engagement has been associated with lower dementia risk in late life. However, the underlying mechanisms contributing to the protective effect remain unclear. OBJECTIVE: The goal of the current study was to evaluate the associations of physical activity with cerebral amyloid-β (Aβ) and tau in a predominately middle-aged community-based cohort, as well as to explore whether the associations differ by sex or age. METHODS: Participants from the Framingham Heart Study underwent 11C-Pittsburgh Compound B amyloid and 18F-Flortaucipir tau positron emissi

Mesenchymal Stem Cell (MSC)-Derived Extracellular Vesicles Protect from Neonatal Stroke by Interacting with Microglial Cells.

Mesenchymal stem cell (MSC)-based therapies are beneficial in models of perinatal stroke and hypoxia-ischemia. Mounting evidence suggests that in adult injury models, including stroke, MSC-derived small extracellular vesicles (MSC-sEV) contribute to the neuroprotective and regenerative effects of MSCs. Herein, we examined if MSC-sEV protect neonatal brain from stroke and if this effect is mediated via communication with microglia. MSC-sEV derived from bone marrow MSCs were characterized by size distribution (NanoSight™) and identity (protein markers). Studies in microglial cells isolated from

Acute changes in cerebral blood flow after single-infusion ketamine in major depression: a pilot study.

BACKGROUND: Ketamine provides rapid antidepressant response in those struggling with major depressive disorder (MDD). This study measured acute changes in brain activity over 24 hours after a single infusion of ketamine using arterial spin labeled (ASL) functional magnetic resonance imaging (fMRI) in patients with MDD. ASL is a novel technique that provides quantitative values to measure cerebral blood flow (CBF). METHODS: A single sub-anesthetic dose (0.5 mg/kg) of ketamine was delivered intravenously. Treatment-refractory patients (n=11) were assessed at: Baseline (pre-infusion), and approx

Chronic prostatitis/chronic pelvic pain syndrome increases susceptibility to seizures in rats and alters brain levels of IL-1β and IL-6.

Chronic Prostatitis/Chronic Pelvic Pain Syndrome (CP/CPPS) is a result of interplay between psychological, immune, neurological and genetic factors, manifested by variety of urological, as well as brain-related symptoms. However, its relation with brain excitability has not been addressed. herefore, our aim was to assess susceptibility to seizures in rats with CP/CPPS. We induced CP/CPPS in adult rats by intraprostatic injection of 3% λ-carrageenan. Sham operated rats served as controls (0.9% NaCl, Sham). On day 7 upon injection, rats were treated with lindane (4 mg/kg) and observed for convul

Chronic Cigarette Smoking in Healthy Middle-Aged Individuals Is Associated With Decreased Regional Brain N-acetylaspartate and Glutamate Levels.

BACKGROUND: Cigarette smoking is associated with metabolite abnormalities in anterior brain regions, but it is unclear if these abnormalities are apparent in other regions. Additionally, relationships between regional brain metabolite levels and measures of decision making, risk taking, and impulsivity in smokers and nonsmokers have not been investigated. METHODS: In young to middle-aged (predominately male) nonsmokers (n = 30) and smokers (n = 35), N-acetylaspartate (NAA), choline-containing compounds, creatine-containing compounds (Cr), myo-inositol (mI), and glutamate (Glu) levels in the a

Action of the diabetogenic drug streptozotocin on glycolytic and glycogenolytic metabolism in adult rat brain cortex and hippocampus.

In sporadic Alzheimer's disease (AD), a number of metabolic alterations to the brain have been observed soon after the onset of the initial clinical symptoms. In particular, impairments of glucose utilization and related metabolic pathways are prominent and well-established findings in incipient AD, resembling metabolic abnormalities such as have been found in noninsulin-dependent diabetes mellitus. To mimic these abnormalities, we administered an intracerebroventricular (icv) injection of streptozotocin (STZ) to rats and studied the effects of glucose and glycogen metabolism in the cerebral c

Commonality and specificity of acupuncture tonifying-reducing manipulations based on functional near-infrared spectroscopy.

BACKGROUND: Acupuncture tonifying-reducing manipulations (TRMs) are typically classified into tonifying manipulations (TMs) and reducing manipulations (RMs) according to their tonifying and reducing effects, and further subdivided into single and complex TRMs based on the different amount of stimulation applied. Different TRMs can produce different therapeutic effects, so distinguishing different TRMs is important for precise acupuncture treatment. OBJECTIVE: This study investigated the specificity and commonality of brain responses induced by TRMs in terms of both the different effects of ac

[Neurological diagnosis and therapeutic measures in cerebral embolism].

UNLABELLED: Stroke is caused by intracerebral or subarachnoid hemorrhage in about 15% of clinical presentations and the remaining 85% result from ischemia. About 15% of ischemic strokes are caused by emboli arising from the heart. In younger patients (18 to 50 years) with ischemic strokes or transient ischemic attacks (TIA), the incidence of cardiac embolism is increased to 23 to 36%. DIAGNOSIS: a) SYMPTOMS: Individual neurologic symptoms of stroke do not provide sensitive or specific indications of the underlying mechanism. In 25 to 82% of patients with possible embolic stroke, there is an

Chapter 5--face sensorimotor cortex: its role and neuroplasticity in the control of orofacial movements.

The range and complexity of orofacial movements require sophisticated neural circuitries that provide for the coordination and control of these movements and their integration with other motor patterns such as those associated with breathing and walking. This chapter is dedicated to Jim Lund whose many research studies have made major contributions to our knowledge of the role of brainstem and cerebral cortex in orofacial motor control. Our own investigations using intracortical microstimulation (ICMS), cortical cold block, and single neuron recordings have documented that the face primary mot

Melanocortin Derivatives Induced Vascularization and Neuroglial Proliferation in the Rat Brain under Conditions of Cerebral Ischemia.

Stroke remains the second leading cause of death worldwide. The development of new therapeutic agents focused on restoring vascular function and neuroprotection of viable tissues is required. In this study the neuroprotective activity of melanocortin-like ACTH(4-7)PGP and ACTH(6-9)PGP peptides was investigated in rat brain at 24 h after transient middle cerebral artery occlusion (tMCAO). The severity of ischemic damage, changes in the proliferative activity of neuroglial cells and vascularization of rat brain tissue were analyzed. The administration of peptides resulted in a significant increa

Trivial Neck Bite Resulting in a Stroke in an 18-Month-Old Child: A Case Report.

Stroke is often viewed as a diagnosis found In the elderly with or without comorbidities, but it is vital to not rule it out in a pediatric patient presenting with signs and symptoms of stroke. Here, we present a case of an 18-month-old boy who arrived at the emergency department with left arm weakness and left-sided seizures a few minutes after a right-sided trivial neck bite that was initially overlooked by the parents until symptoms occurred. Urgent imaging further with a computed tomography scan of the brain revealed a hypodense lesion in the area covering the lateral part of the frontal l

An Improved Surgical Approach for Complete Interhemispheric Corpus Callosotomy Combined with Extended Frontoparietal Craniotomy in Mice.

Callosotomy is an invasive method that is used to study the role of interhemispheric functional connectivity in the brain. This surgical approach is technically demanding to perform in small laboratory animals, such as rodents, due to several methodological challenges. To date, there exist two main approaches for transecting the corpus callosum (CC) in rodents: trephine hole(s) or unilateral craniotomy, which cause damage to the cerebral cortex or the injury of large vessels, and may lead to intracranial hemorrhage and animal death. This study presents an improved surgical approach for complet

Amyloid-β impairs mitochondrial dynamics and autophagy in Alzheimer's disease experimental models.

The most accepted hypothesis in Alzheimer's disease (AD) is the amyloid cascade which establishes that Aβ accumulation may induce the disease development. This accumulation may occur years before the clinical symptoms but it has not been elucidated if this accumulation is the cause or the consequence of AD. It is however, clear that Aβ accumulation exerts toxic effects in the cerebral cells. It is important then to investigate all possible associated events that may help to design new therapeutic strategies to defeat or ameliorate the symptoms in AD. Alterations in the mitochondrial physiology

The Neural Correlates of Face-Voice-Integration in Social Anxiety Disorder.

Faces and voices are very important sources of threat in social anxiety disorder (SAD), a common psychiatric disorder where core elements are fears of social exclusion and negative evaluation. Previous research in social anxiety evidenced increased cerebral responses to negative facial or vocal expressions and also generally increased hemodynamic responses to voices and faces. But it is unclear if also the cerebral process of face-voice-integration is altered in SAD. Applying functional magnetic resonance imaging, we investigated the correlates of the audiovisual integration of dynamic faces a
