GWAS reveals loci associated with velopharyngeal dysfunction.

Velopharyngeal dysfunction (VPD) occurs when the muscular soft palate (velum) and lateral pharyngeal walls are physically unable to separate the oral and nasal cavities during speech production leading to hypernasality and abnormal speech reduction. Because VPD is often associated with overt or submucous cleft palate, it could be present as a subclinical phenotype in families with a history of orofacial clefting. A key assumption to this model is that the overt and subclinical manifestations of the orofacial cleft phenotype exist on a continuum and therefore share common etiological factors. W

Measurement of regional variation of GABA in the human brain by optimized point-resolved spectroscopy at 7 T in vivo.

The (1)H resonances of γ-aminobutyric acid (GABA) in the human brain in vivo are extensively overlapped with the neighboring abundant resonances of other metabolites and remain indiscernible in short-TE MRS at 7 T. Here we report that the GABA resonance at 2.28 ppm can be fully resolved by means of echo time optimization of a point-resolved spectroscopy (PRESS) scheme. Following numerical simulations and phantom validation, the subecho times of PRESS were optimized at (TE, TE2) = (31, 61) ms for detection of GABA, glutamate (Glu), glutamine (Gln), and glutathione (GSH). The in vivo feasibility

TCRA, P2RY11, and CPT1B/CHKB associations in Chinese narcolepsy.

OBJECTIVES: Polymorphisms in the TCRA and P2RY11, two immune related genes, are associated with narcolepsy in Caucasians and Asians. In contrast, CPT1B/CHKB polymorphisms have only been shown to be associated with narcolepsy in Japanese, with replication in a small group of Koreans. Our aim was to study whether these polymorphisms are associated with narcolepsy and its clinical characteristics in Chinese patients with narcolepsy. METHODS: We collected clinical data on 510 Chinese patients presenting with narcolepsy/hypocretin deficiency. Patients were included either when hypocretin deficienc

Activation-dependent descending reflex evacuation motority of anal canal in rat model.

The evacuative motor responses of the anal canal and recto-anal reflexes during defecation were studied in an isolated rat recto-anal model preparation using (i) partitioned organ bath, (ii) electrical stimulation, (iii) balloon distension and (iv) morphological techniques. Electrical field stimulation applied to the anal canal or to the distal part of the rectum elicited tetrodotoxin (10(-7) M)-sensitive frequency-dependent local or descending contractions of the anal canal and the local responses were bigger in amplitude (14.9 ± 1.35 mN) than the descending contractions (5.3 ± 0.7 mN at freq

Inhibition of adenylate cyclase attenuates muscarinic Ca²(+) signaling by a PKA-independent mechanism in rat carotid body Type I cells.

Carotid body (CB) Type I cells respond to hypoxia by releasing excitatory and inhibitory neurotransmitters. This mechanism leads to increased firing of the carotid sinus nerve (CSN) which alters breathing to maintain blood gases within the physiological range. Acetylcholine targets both muscarinic and nicotinic receptors in the rat CB, acting postsynaptically on CSN and presynaptically on Type I cells. Muscarinic Ca²(+) signaling is inhibited by the activation of G(i)-coupled receptors including histamine H3 receptors. Here inhibition of adenylate cyclase with SQ22536 mimicked H3 receptor acti

Long-term follow-up in patients with congenital myasthenic syndrome due to CHAT mutations.

BACKGROUND: Congenital myasthenic syndromes (CMSs) are a group of clinically and genetically heterogeneous inherited disorders of the neuromuscular junction. Mutations in the acetylcholine transferase (CHAT) gene cause a pre-synaptic CMS, typically associated with episodic apnoea and worsening of myasthenic symptoms during crises caused by infections, fever or stress. Between crises symptoms may be mild and variable. Acetylcholinesterase - inhibitor therapy is reported to improve clinical symptoms and reduce crises. PATIENTS AND METHODS: We present data on the long-term follow-up of 11 patien

Melatonin is protective in necrotic but not in caspase-dependent, free radical-independent apoptotic neuronal cell death in primary neuronal cultures.

To assess the neuroprotective potential of melatonin in apoptotic neuronal cell death, we investigated the efficacy of melatonin in serum-free primary neuronal cultures of rat cortex by using three different models of caspase-dependent apoptotic, excitotoxin-independent neurodegeneration and compared it to that in necrotic neuronal damage. Neuronal apoptosis was induced by either staurosporine or the neurotoxin ethylcholine aziridinium (AF64A) with a delayed occurrence of apoptotic cell death (within 72 h). The apoptotic component of oxygen-glucose deprivation (OGD) unmasked by glutamate antag

MIND-NL diet adherence moderates the relation of low-grade systemic inflammation with neuroinflammatory metabolites and cognitive functioning: an exploratory cross-sectional study in older adults.

BACKGROUND: Observational studies have linked high adherence to the “Mediterranean-Dietary Approaches to Stop Hypertension Intervention for Neurodegenerative Delay” (MIND) diet to improved cognitive functions in older adults. The underlying peripheral and central mechanisms of this association remain poorly understood, although multiple nutrients in the MIND diet are known for their anti-inflammatory effects. Therefore, we explored the cross-sectional relation between MIND diet adherence (Dutch version), systemic inflammation, neuroinflammation, and cognitive functioning in older adults. In ad

Integrated Proteomics and Metabolomics Analysis Provides Insights into Metformin Down-regulating GLUL and SP1 to Inhibit Esophageal Squamous Cell Carcinoma Migration and Invasion.

INTRODUCTION: Esophageal Cancer (EC) is a prevalent gastrointestinal malignancy. Despite significant advances in diagnostic and therapeutic approaches, the prognosis of Esophageal Squamous Cell Carcinoma (ESCC) remains poor. The development of new therapeutic agents for ESCC would greatly benefit patients and their families. Metformin has been found to be useful in other tumours, but its role and mechanism in ESCC have not been investigated. METHODS: The effect of metformin on inhibiting invasive metastasis in ESCC cells was explored by in vitro cytology experiments. 4D-DIA proteomics and unt

Graph Theory of Deep Eutectic Solvents: From Typed Interaction Networks to Property Prediction.

Deep eutectic solvents (DES) present a formidable challenge for property prediction due to the complex interplay of hydrogen bonding, Coulombic, dispersion, and coordination interactions that define their macroscopic behavior. Existing approaches─group contribution methods, COSMO-RS, and brute-force molecular dynamics─either lack transferability, require prohibitive computational cost, or obscure the physical origins of observed trends. Here we propose a graph-theoretic framework in which a DES mixture is represented as a directed, weighted, typed multigraph whose nodes are mixture components

Nutrient gaps are prevalent among women experiencing infertility, a cross-sectional analysis of the National Health and Nutrition Examination Survey (NHANES), 2013-2020.

BACKGROUND: Adequate nutrient intake is important for supporting reproductive health. Few studies have examined the role of nutrients for fertility among women, resulting in a critical evidence gap. The aim of this study was to explore the usual intake and prevalence of nutrient inadequacies from foods only and foods plus dietary supplements among women of child bearing age with and without infertility. METHODS: This cross-sectional study used data from the National Health and Nutrition Examination Survey (NHANES), a national survey in the United States. Participants included women aged 18-44

Reproducibility of Metabolic Mapping Using 2D Multi-Slice Short-TE and GABA-Edited Spin-Echo MRSI at 3T in a Single Protocol.

MRSI is a non-invasive tool for mapping metabolic distributions in multi-focal or other diseases where the location of abnormalities may be uncertain. High-concentration metabolites can be investigated at 3T using non-edited MRSI, whereas lower-concentration metabolites, such as GABA, typically require specialized editing techniques because of spectral overlap. This study reports on the reproducibility of a protocol containing both co-localized short-TE and GABA-edited multi-slice spin-echo 2D MRSI. Multi-slice, short-TE (TE 20 ms) and GABA-edited (TE 68 ms) MRSI at a nominal spatial resolutio

Layer-specific neuroanatomical architecture of the quadratus lumborum: a quantitative study using 3D micro-CT and immunofluorescence staining.

PURPOSE: The quadratus lumborum (QL) is increasingly recognized as an anatomically important component of the posterior abdominal wall, yet its internal organization remains incompletely defined. This study aimed to characterize the layer-specific architecture of the QL and to quantitatively analyze the distribution of immunolabeled axonal components across its layers. METHODS: Ten embalmed adult cadavers (mean age, 82.1 ± 9.9 years) were examined. Gross dissection and morphometric analysis were performed in eight cadavers (16 sides) to identify layer-specific organization and measure width a

Primary glioblastoma with spontaneous transdural invasion and contiguous calvarial/mastoid osteolysis: a case report and literature review.

BACKGROUND: Glioblastoma (GBM) is the most common and most aggressive primary malignant tumor of the central nervous system in adults. Although GBM is characterized by highly infiltrative growth within the brain parenchyma, spontaneous transdural extension with direct invasion of the calvarium and skull base at initial presentation is exceptionally rare. Such cases may radiologically mimic metastatic skull tumors or other extra-axial lesions, creating diagnostic and therapeutic challenges. CASE PRESENTATION: We report the case of a 63-year-old man who presented with dizziness, slowed response

Total flavonoids from Abrus cantoniensis alleviate fatty liver hemorrhagic syndrome in laying hens by regulating inflammation, oxidative stress, and cecal metabolites and microbiota.

Fatty liver hemorrhagic syndrome (FLHS) is a metabolic disease in laying hens. Total flavonoids from Abrus cantoniensis (TFAC) comprise multiple bioactive compounds with potential benefits against FLHS. This study aimed to explore the effects and mechanism of TFAC in improving FLHS. Firstly, analysis by liquid chromatography-tandem mass spectrometry identified 20 flavonoid compounds in the TFAC, including vicenin-3 and acacetin. Subsequently, 144 laying hens at 28 weeks of age with similar body weight (1.45 ± 0.03 kg) were randomly divided into six groups (eight replicates per group and three
