Cryo-Electron Microscopy Structure and Interactions of the Human Cytomegalovirus gHgLgO Trimer with Platelet-Derived Growth Factor Receptor Alpha.

Human cytomegalovirus (HCMV) is a herpesvirus that produces disease in transplant patients and newborn children. Entry of HCMV into cells relies on gH/gL trimer (gHgLgO) and pentamer (gHgLUL128-131) complexes that bind cellular receptors. Here, we studied the structure and interactions of the HCMV trimer, formed by AD169 strain gH and gL and TR strain gO proteins, with the human platelet-derived growth factor receptor alpha (PDGFRα). Three trimer surfaces make extensive contacts with three PDGFRα N-terminal domains, causing PDGFRα to wrap around gO in a structure similar to a human hand, expla

Rare Earth Element Biosensing in Chemically Complex Waters: Interference Mechanisms, Operating Windows, and Mitigation for a Lanmodulin Enzymatic Switch.

Rare earth elements (REE) are increasingly mobilised from mine residues and other industrial sources, creating a need for rapid screening of chemically complex waters where conventional ICP-based analysis is impractical on site. Here, we evaluate a lanthanide-gated enzymatic switch biosensor, for matrix-aware monitoring of mine-influenced waters. We quantified REE-dependent activation and systematic interference from co-occurring metals and anions typical of mine waters, and defined operating windows that preserve lanthanide responsiveness. REE activation was readily detectable at ~100nM, wher

Sphingosine-1-Phosphate Receptor 1 regulates competition dependent astrocyte morphogenesis and tiling in murine cortex.

Astrocytes are highly abundant in the mammalian brain and coordinate with neurons and other glial cells to regulate neural circuit structure, function, and blood brain barrier integrity among many to maintain proper brain homeostasis. Astrocytes perform most of these functions owing to their highly complex morphologies and hundreds of thousands of fine processes that are important in contacting neuronal synapses and other glial cells. In fact, the morphological complexity of astrocytes is regulated by the presence and activity of neurons and helps establish astrocyte territory/tiling in a non-

Small-molecule cocktail 2SM promotes heart regeneration via inducing embryonic-like cardiomyocyte state.

Adult mammalian cardiomyocytes exhibit limited regenerative capacity, which remains a major challenge for functional recovery after myocardial infarction (MI). Our previous study identified a cocktail of five small molecules (5SM) that promotes cardiomyocyte proliferation and heart regeneration; however, the complexity of this five-compound mixture poses significant barriers to clinical translation and mechanistic studies. Here, we identified a simplified dual-compound cocktail (2SM) composed of phenylephrine hydrochloride (PE) and harmine (HM), which also robustly enhanced cardiomyocyte proli

Insulin preservatives trigger neutrophil extracellular trap formation through SYNE1-mediated nuclear disassembly.

Neutrophil extracellular traps (NETs) are critical effector molecules in sterile inflammation, yet the molecular mechanisms by which xenobiotic chemical exposures trigger NETosis remain poorly defined. Here, using phenolic preservatives present in all FDA-approved insulin formulations as a discovery platform, we show that these compounds induce NETosis in primary human neutrophils (34.3 ± 5.0% vs. 2.8 ± 0.9% for preservative-free insulin; p < 0.001) via a mechanism distinct from canonical PKC- and calcium-dependent pathways. Data-independent acquisition mass spectrometry (n = 6 donors) reveals

Interactions between Notch and MAPK signaling in neurovascular development: Implications for advanced disease modeling and tissue engineering.

Neurovascular development is a tightly regulated, highly coordinated process essential for the concurrent growth and patterning of the central nervous system and its vascular network during embryogenesis and early postnatal life. This intricate process relies on dynamic cellular crosstalk that guides the invasion of blood vessels into developing neural tissues, culminating in the establishment of the blood-brain barrier and the regulation of cerebral blood flow. Among the key signaling cascades orchestrating neurovascular development, the Notch and Mitogenactivated Protein Kinase pathways emer

Policy approach to optimize multiple myeloma treatment sequencing practices.

INTRODUCTION: As a consequence of major therapeutic advances that have transformed multiple myeloma (MM) into a manageable chronic condition, treatment sequencing has become highly complex. According to current practices, novel therapies are predominantly reserved for later disease stages, yet limited evidence suggests that this approach yields optimal long-term outcomes. Understanding the drivers of current practices and identifying strategies to strengthen evidence-informed sequencing are therefore critical. AREAS COVERED: This paper synthesizes insights from a multi-stakeholder discussion

An orally administered biohybrid complex leveraging probiotic guidance to treat colorectal cancer via in situ vaccination.

Oral therapy for colorectal cancer (CRC) holds inherent promise for enhancing patient compliance and enabling gastrointestinal targeting, while its application is hampered by low oral drug utilization efficiency and precise colonic localization. This study developes an orally delivered biohybrid complex, CS@CB-Lipo@5-FU/R837, which leverages the hypoxic tropism of the probiotic Clostridium butyricum (CB) for active tumor targeting. The core of the system consists of nanoliposomes co-loaded with 5-fluorouracil (5-FU) and the TLR7 agonist, R837, which are site-specifically anchored to CB via bio

Impact of process impurities on developability assessments of complex antibody formats: the striking role of leachable copper.

The assessment of developability and physical stability of antibody therapeutics is a cornerstone of modern biopharmaceutical discovery, increasingly relying on high-throughput automation to screen candidates to identify the most suitable lead candidate. In this study, we report a severe, anomalous fragmentation event observed in therapeutic antibodies during routine automated buffer exchange into phosphate-buffered saline (PBS) at physiological pH. Through a systematic investigation, we demonstrate that this instability is not intrinsic to the proteins, but is driven by the specific physicoch

Scaling Phage-Bacteria Interaction Contexts: From Pairwise Mechanisms to Community Dynamics.

Bacteriophages (phages), viruses that parasitize bacteria, hold tremendous potential as antimicrobial agents, microbiome modulators, and industrial biocontrol tools; yet clinical and environmental applications remain frustratingly inconsistent. Decades of research on isolated phage-bacteria pairs have revealed fundamental mechanisms governing infection specificity, coevolutionary arms races, and resistance trade-offs. These foundational studies, however, do not help predict outcomes when phages encounter multi-species assemblages characteristic of natural ecosystems. In this review, we integra

Three-Dimensional Scaffolds for Bone, Soft Tissue, and Periodontal Regeneration.

The oral cavity presents three distinct regenerative challenges: restoration of alveolar bone volume and stability, augmentation of soft mucogingival tissues, and regeneration of the tooth-supporting periodontium. In each of these clinical contexts, conventional approaches remain limited by the morbidity associated with autologous tissue harvesting and by the difficulty in predictably reconstructing the complex architecture and functional integration required for the long-term stability of these distinct tissues. Although currently available biomaterials have demonstrated clinical effectivenes

Recent advances in directing group rearrangement strategies in transition-metal-catalyzed C-H activation.

After more than two decades of continuous development, transition-metal-catalyzed C-H activation cascade reactions have gradually become an important methodology and strategy for the construction of complex molecules. This transformation is typically achieved by installing a functional group with a directing effect onto the substrate to enable C-H activation at a specific site. Such directing groups must possess adequate coordination ability, oxidation capability, and transformability. In most reported reactions, C-H activation occurs first, followed by the participation of the directing group

PagCOBL5 Integrates Transcriptional and Spatial Cues to Confer G-Layer Deposition in Poplar.

Angiosperm trees reorient their stems by producing tension wood (TW), a specialised xylem that generates contractile force. TW fibres develop a gelatinous (G) layer rich in cellulose, yet the molecular mechanisms governing G-layer formation remain poorly understood. COBRA-like genes encode glycosylphosphatidylinositol (GPI)-anchored proteins known to regulate cellulose deposition. Here, we functionally characterised PagCOBL5 in hybrid poplar (Populus alba × P. glandulosa) by identifying its upstream regulator and interaction partner. PagCOBL5 was preferentially expressed in mature xylem fibres

The genomic origins of cyanobacterial morphological diversity.

Cyanobacteria are the only prokaryotes to have evolved oxygenic photosynthesis, transforming the geochemistry and biology of the planet. They exhibit remarkable morphological diversity and represent one of the earliest known multicellular organisms, including filamentous growth and cellular differentiation. However, the genomic mechanisms underlying their repeated transitions between unicellularity and multicellularity remain unresolved. Here, we implement phylogenomic and Bayesian molecular clock analyses, estimate ancestral protein-coding gene contents, and examine gene gains and losses asso

Oxophilicity of Silicon(IV) in Silanethiols Facilitates the Generation of Thionitrite and Perthionitrite from a Binuclear Zinc(II)-bis(nitrito) Complex.

A detailed study of the reactivity of a Zn(II)-bis(nitrito) complex, [Zn2(BPMP)(ONO)(κ-NO2)]1+ (2), toward R3SiSH/R3SiS- and R3SiCl (R = Ph, iPr) in comparison to R1SH/R1S- (R1 = Ph3C, PhCH2) and PhCH2Br is presented. Unlike the negligible (for R1SH/R1S-) or less reactive (for PhCH2Br) nature of their carbon analogues, the stronger oxophilic nature of Si(IV) in R3SiSH/R3SiS- and R3SiCl allows facile reactions with the coordinated NO2- in 2 to generate the physiologically relevant S/N "crosstalk" species, SNO- and SSNO-, and R3SiONO, respectively, which in turn produces NO in high yields. Jasmonic acid signaling in plants: regulatory mechanisms in development and stress responses.

Jasmonic acid (JA), one of the most pivotal plant hormones, has emerged as a central focus in plant biology research due to its extensive and diverse biological functions. This review synthesizes the significant advancements made over the past decade in understanding JA's role in regulating plant development and mediating responses to environmental stresses, areas that lacked systematic review in previous years. We provide a concise overview of JA's biosynthetic and signal transduction pathways, with particular emphasis on the key regulatory complex comprising CORONATINE INSENSITIVE 1 (COI1),

Glutathione-Triggered Nanodrug Disassembly for Enhanced Combined Ferroptosis and Photodynamic-Chemotherapy of Tumors.

Cancer treatment remains challenging due to tumor heterogeneity, drug resistance, and the complex microenvironment that limits monotherapy efficacy. Herein, we report a glutathione (GSH)-responsive multifunctional nanodrug FeCe6@SKN that is self-assembled from the complex formed by the coordination of the photosensitizer Ce6, the natural anticancer agent Shikonin (SKN), and ferric ions (Fe3+). After tail vein injection, FeCe6@SKN can effectively accumulate at the tumor site through the EPR effect and subsequently disassemble by high concentration of GSH, leading to the controlled release of Fe
