Mobile genetic elements frequently encode defense mechanisms to protect their bacterial hosts from viral attack. In Vibrio cholerae, these defensive elements include phage-inducible chromosomal island-like elements (PLEs), which are phage satellites that act as highly specialized parasites of the lytic phage ICP1. While PLE transcriptional activation upon ICP1 infection is known to be temporally regulated, the underlying regulatory logic and dependencies on the progression of the phage's developmental program required for activation remain unclear. In this study, we took a novel approach to de

Theileria orientalis is a tick-borne intraerythrocytic protozoan parasite of cattle causing oriental or benign theileriosis. It has a worldwide distribution and is not considered a zoonotic agent, while the disease symptoms range from subclinical to severe. Theileria spp. are very common co-infecting agents with other tick borne pathogens such as Babesia spp., Anaplasma spp. including zoonotic Anaplasma phagocytophilum with rising prominence. This study is an exploratory investigation of the occurrence of Anaplasma spp. and other blood parasites in cattle from Gacko and Bileća, Herzegovina. Bl

Fungal endophthalmitis (FE), although less common than bacterial endophthalmitis, carries a disproportionately high risk of irreversible blindness. Clinical observations show that some patients continue to experience progressive visual loss even after successful microbiological clearance, suggesting that disease outcomes are strongly influenced by excessive host immune-inflammatory injury rather than pathogen burden alone. Focusing on the retinal pigment epithelium (RPE), a key component of the blood-retinal barrier, this review summarizes recent advances in intraocular microenvironmental alte

Background: There is growing evidence that uterine microbiota might be linked to endometrial receptivity (ER) and affect the outcome of assisted reproductive technology (ART) procedures. Owing to the invasive nature of endometrial sampling, the evaluation of microbiota in this biomaterial is only possible outside the embryo transfer (ET) cycle. However, menstrual blood might be the key to overcoming this challenge as it can be safely aspirated from the uterine cavity at the beginning of the target ET cycle. This study aimed to evaluate the association of the microbiological profiles of menstru

Torque teno virus (TTV) replication is tightly modulated by the host immune response. As such, TTV viremia has been investigated as a biomarker of immune competence for monitoring the risks of allograft rejection and opportunistic infections in solid organ transplant recipients. We aimed to investigate the associations between TTV levels in bronchoalveolar lavage fluid samples (BALF) and pathogen detection in patients with hematological diseases with suspicion of lower respiratory tract infection (LRTI). TTV DNA was quantified in BALF using qPCR. Linear and logistic regressions were used to st

BACKGROUND: Malaria is typically diagnosed using rapid antigen tests and microscopy. This case describes an unusual presentation of Plasmodium falciparum, initially detected via 16S rDNA testing of pericardial fluid identifying apicoplast DNA-a finding not previously reported in the literature.
CASE SUMMARY: A septuagenarian Nigerian man visiting Canada presented with shortness of breath, tachycardia, tachypnea, anemia, and a pericardial effusion. Pericardiocentesis yielded serosanguineous fluid, and imaging revealed multifocal opacities. Blood and fluid cultures were negative for bacterial o

Nucleus-forming phages are a class of jumbo phages with the capacity to form a protein nuclear shell during infection within a bacterial host. This protein nuclear shell protects the replication of the phage genome by excluding host defense factors. However, the lack of genetic characterization of these phages limits our understanding of nucleus-forming phages. Here, we used HMMER and RoseTTAFold to identify a proposed dataset of nucleus-forming phages, based on the presence of chimallin and PhuZ tubulin genes, containing the 1103 phage genomes (including 406 high quality genomes) from 16 mill

The rise of drug-resistant Mycobacterium tuberculosis underscores the urgent need for safe and bioavailable compounds that target essential bacterial pathways. ATPase domain of DNA gyrase subunit B (GyrB) is a mechanistically distinct and druggable site involved in DNA supercoiling and energy metabolism. In this study, we employed a structure- and system-guided computational pipeline to identify natural flavonoids with multi-target inhibitory potential against GyrB. Naringenin, taxifolin, and quercetin were selected from an initial flavonoid library based on their predicted ATPase inhibition,

CysB is a member of the large bacterial LysR-type transcriptional regulator (LTTR) protein family. Like the majority of LTTRs, CysB functions as a homotetramer in which each subunit has an N-terminal winged-helix-turn-helix (wHTH) DNA-binding domain connected to an effector-binding domain by a helical hinge region. CysB is best known for its role in regulating the expression of genes associated with sulfur uptake and biosynthesis of cysteine in Gram-negative species such as Escherichia coli and Salmonella enterica. Activation of CysB target genes generally requires the effector N-acetyl-L-seri

INTRODUCTION: Neisseria gonorrhoeae is a notorious superbug responsible for causing 'Gonorrhoea' in humans. Recently, it has been classified as a high-priority pathogen by the World Health Organization due to its increasing resistance to available antibiotics. A multi-prolonged approach is needed to combat the growing problem of drug resistance caused by N. gonorrhoeae. This study evaluates Glutamate Racemase (GR), a moonlight protein of N. gonorrhoeae (Ng-GR), as a novel therapeutic target with potential for both inhibitor design and peptide vaccine development. Ng-GR plays a crucial role in

Urban stray cats are often associated with ectoparasites and zoonotic pathogens due to their unsanitary living conditions and lack of veterinary care. Fleas, especially Ctenocephalides spp., are competent vectors of Bartonella spp., a genus of emerging bacterial pathogens with both public health and veterinary relevance. This study investigated the presence of Bartonella DNA in ectoparasitic fleas infesting stray cats in various urban habitats within the Klang Valley, Malaysia. A total of 204 fleas were collected from 89 stray cats. Fleas were identified morphologically using established taxon

This study investigates the effects of bariatric surgery on non-alcoholic fatty liver disease (NAFLD) by examining the interplay between gut microbiota, epigenetics, and metabolic health. A cohort of 22 patients undergoing sleeve gastrectomy (SG) was analyzed for changes in gut microbial composition and DNA methylation profiles before and six months after surgery. Correlations between gut microbial abundance and clinical markers at baseline revealed that certain genera were associated with worse metabolic health and liver markers. Following SG, significant improvements were observed in the cli

Bovine respiratory disease (BRD) is an important health and economic burden to the cattle industry worldwide. Three bacterial pathogens frequently associated with BRD (Mannheimia haemolytica, Pasteurella multocida, and Histophilus somni) can possess integrative and conjugative elements (ICEs), a diverse group of mobile genetic elements that acquire antimicrobial resistance (AMR) genes (ARGs) and decrease the therapeutic efficacy of antimicrobial drugs. We developed a duplex recombinase polymerase amplification (RPA) assay to detect up to two variants of ICEs in these Pasteurellaceae. Whole gen

Metabolically associated fatty liver disease, formerly called nonalcoholic fatty liver disease, is the most common liver disease globally, representing the third cause of liver transplantation. Metabolically associated fatty liver disease is defined as having more than 5% lipid droplets in hepatocytes without other concomitant liver diseases. Various stimuli such as the secretion of inflammatory cytokines, mitochondrial and endoplasmic reticulum dysfunction due to oxidative stress, alteration of the intestine-liver axis, bacterial dysbiosis, as well as genetic and epigenetic factors can modify

Accelerated cellular senescence may be a crucial process for periodontitis progression because it integrates the damaging effects of major periodontitis risk factors. Cellular senescence is a manifestation of aging at the cellular level. However, recent technological advances have shown that healthy young cells continuously exposed to sublethal oxidative stress undergo accelerated senescence. Although accumulation of senescent cells is normal in aged tissues, persistent bacterial infection, chronic inflammation, diabetes, and smoking promote the early onset of senescence by causing DNA damage.

Drug development process demands validation of specific drug target impeding the Multi Drug Resistance (MDR). DNA gyrase, as a bacterial target has been in trend for developing newer antibacterial candidates due to its absence in higher eukaryotes. The fluoroquinolones are the leading molecules in the drug discovery pipeline for gyrase inhibition due to its diversity. The fluoroquinolones like levofloxacin and moxifloxacin have been listed in class A drugs for treating MDR. Gatifloxacin and ciprofloxacin also proved its efficacy against MDR TB and MDR enteric fever in adults, whereas nemonoxac
