In vitro and in vivo activities of synthetic trioxolanes against major human schistosome species.

Schistosomiasis is a parasitic disease that remains of considerable public health significance in tropical and subtropical environments. Since the mainstay of schistosomiasis control is chemotherapy with a single drug, praziquantel, drug resistance is a concern. Here, we present new data on the antischistosomal properties of representative synthetic 1,2,4-trioxolanes (OZs). Exposure of adult Schistosoma mansoni for 24 h to a medium containing 20 mug/ml OZ209 reduced worm motor activity, induced tegumental alterations, and killed worms within 72 h. While exposure of S. mansoni to OZ78 had no ap

Cellular thermal shift assay of subcellular isolates to evaluate drug-membrane target interactions.

The cellular thermal shift assay (CETSA) is a tool for target identification and validation in drug discovery. It relies on thermal melting curves to indicate drug binding and is performed in cells, cell lysates, or purified protein. These approaches can disrupt the structural integrity of membrane proteins, hindering drug-target engagement. We describe the first application of CETSA in isolated mitochondria and show the effects of this approach on the analysis of the compound UK5099 and its known binding target, the mitochondrial pyruvate carrier (MPC), a mitochondrial inner membrane-localize

The acute effects of psychoactive drugs on emotional episodic memory encoding, consolidation, and retrieval: A comprehensive review.

Psychoactive drugs modulate learning and emotional processes in ways that could impact their recreational and medical use. Recent work has revealed how drugs impact different stages of processing emotional episodic memories, specifically encoding (forming memories), consolidation (stabilizing memories), and retrieval (accessing memories). Drugs administered before encoding may preferentially impair (e.g., GABAA sedatives including alcohol and benzodiazepines, Δ9-tetrahydrocannabinol or THC, ketamine), enhance (e.g., dextroamphetamine and dextromethamphetamine), or both impair and enhance (i.e.

A high-throughput screening assay for the identification of flavivirus NS5 capping enzyme GTP-binding inhibitors: implications for antiviral drug development.

There are no effective antivirals currently available for the treatment of flavivirus infection in humans. As such, the identification and characterization of novel drug target sites are critical to developing new classes of antiviral drugs. The flavivirus NS5 N-terminal capping enzyme (CE) is vital for the formation of the viral RNA cap structure, which directs viral polyprotein translation and stabilizes the 5' end of the viral genome. The structure of the flavivirus CE has been solved, and a detailed understanding of the CE-guanosine triphosphate (GTP) and CE-RNA cap interactions is availab

The inhibitory effect of anticonvulsants on human sperm motility: measured with a trans-membrane migration method.

The in-vitro effects of four anticonvulsant drugs (phenytoin, phenobarbitone, carbamazepine and valproate) on human sperm motility were studied with a trans-membrane migration method. Sperm motility was measured either immediately after semen had been mixed with the drug or after a 2-hour pre-incubation at 37 degrees C. When the drug effect was evaluated after the semen-drug mixture had been pre-incubated for 2 hours, the concentration of phenytoin, carbamazepine and valproate that inhibited sperm motility to 50% of control (EC50) was 1.59, 4.23, and 5.00 mM, respectively. If sperm motility wa

Exosomes and Extracellular Vesicles in Oncology: Next-Generation Smart Nanocarriers for Precision Cancer Therapy.

Exosomes are an evolved sub-type of extracellular vesicles (EVs) that have gained popularity in recent years as a novel biological nanocarrier system for targeted precision therapy in cancer owing to their excellent biocompatibility, low immunogenicity, and high stability. While other artificial systems can only deliver specific types of drug, exosomes have been found to effectively carry different types of therapeutic cargo, such as chemotherapeutic agents, proteins, peptides, mRNA, siRNA, miRNA, and even CRISPR/Cas gene editing elements. Recent progress in exosome engineering technologies, s

Targeting epigenetic regulators: In-silico discovery of natural inhibitors against histone demethylase KDM4C.

Cancer is a multifaceted disease driven by genetic mutations and epigenetic dysregulation. Among epigenetic modifiers, histone demethylases like KDM4C (lysine demethylase 4C) play a pivotal role in tumor progression by removing repressive methylation mark at Histone H3K9/H3K36 and altering chromatin structure and gene expression. Overexpression of KDM4C has been implicated in various malignancies, including breast, prostate, colorectal, and hepatocellular carcinomas, hence it is promising drug target. This study employs a structure-based drug discovery strategy to identify natural polyphenolic

Colon-targeted pH-responsive tofacitinib beads improve ulcerative colitis outcomes with lower systemic exposure.

Ulcerative colitis (UC) is a chronic inflammatory bowel disease resulting in mucosal inflammation and ulceration. Although tofacitinib, a Janus kinase inhibitor, is effective when administered systemically, its clinical use may be limited by systemic adverse effects. Thus, this study aimed to design and evaluate a colon‑targeted tofacitinib delivery system based on sodium alginate beads coated with Eudragit® S‑100 in a dextran sulfate sodium (DSS) induced colitis rat model. Tofacitinib was encapsulated within alginate beads and subsequently coated with Eudragit® S-100 for pH-dependent release.

A Comprehensive Review on Extracellular Vesicles, Liposomes, and Biohybrid Nanocarriers for Enhanced Wound Healing and Precision Drug Delivery.

Nanomedicine has advanced drug delivery by addressing key challenges such as poor solubility, instability, and off-target effects. Yet, despite its promise, clinical translation remains limited, with few FDA-approved formulations and ongoing biocompatibility concerns. Extracellular vesicles (EVs), particularly those derived from mesenchymal stem cells, offer natural advantages as nanocarriers, including biocompatibility, immunomodulatory effects, and regenerative properties. However, their therapeutic application is constrained by low drug-loading capacity, rapid clearance, and batch-to-- batc

Bioactive hydroxyl-terminated phosphorus dendrimers mediate protein/drug co-delivery for enhanced multi-target ischemic stroke therapy.

The treatment of ischemic stroke (IS) faces significant challenges due to the complex pathophysiology, which encompasses oxidative stress, neuroinflammation, and blood-brain barrier (BBB) dysfunction. Here, we report the development of a bioactive per se hydroxyl-terminated phosphorus dendrimer-based nanoplatform for protein/drug co-delivery to the ischemic brain. We show that through sequential physical complexation and loading, nanocomplexes (NCs) composed of phosphorus dendrimers, a protein drug of fibronectin (FN) with anti-inflammatory/antioxidant/angiogenic properties and a small molecul

Computational Chemistry Approach in the Assessment of Potential Acyl Glucuronide-mediated Toxicity.

INTRODUCTION: Acyl glucuronides are common phase II metabolites of xenobiotics and can sometimes contribute to idiosyncratic toxicities. Their reactivity is primarily mediated through acyl migration and/or nucleophilic displacement, and shorter acyl glucuronide halflives are associated with increased reactivity. This reactivity can lead to metabolite-induced toxicity, posing a significant risk during drug development. METHODS: We developed regression models trained on features derived from Density Functional Theory (DFT) calculations to predict the half-lives of acyl glucuronide metabolites.

Structure-based design and molecular modelling of n-propylcarbazole-1,3-thiazole hybrids as potent α-glucosidase inhibitors: Kinetic, in vitro, and in vivo evaluation.

A series of seventeen novel N-propylcarbazole-1,3-thiazole derivatives (5a-5q) were synthesised via a five-step route and fully characterised by FT-IR, 1H/13C NMR, and HR-MS. The compounds were evaluated for antibacterial, antifungal, α-glucosidase inhibitory, and anti-inflammatory (NO inhibition) activities. The compounds were primarily evaluated for their α-glucosidase inhibitory activity as potential antidiabetic agents. Among them, compound (5k) exhibited outstanding inhibitory potency (IC50 = 6.25 ± 0.17 μM), significantly surpassing the reference drug acarbose (IC50 = 118.09 ± 8.39 μM).

Transporter Interactions of Methoxyflavones and Their In Vivo Pharmacokinetic Implications.

Flavonoids are widely consumed dietary polyphenols that modulate drug disposition via membrane transporters. Increasing attention has been paid to methoxyflavones (MeO-FLVs) because methoxylation can increase membrane permeability and metabolic stability, potentially elevating transporter-mediated drug-drug interaction risk; however, position-specific effects are poorly defined. We profiled a series of structurally related flavones (3-OH-FLV, 3-MeO-FLV, 5-MeO-FLV, 6-MeO-FLV, and 7-MeO-FLV) across major uptake and efflux transporters. In vitro uptake and bidirectional transport assays demonstra

Quality by Design for Bilayer Tablets: Integrating Formulation, Manufacturing, and Process Analytical Technology.

Bilayer fixed-dose combination tablets are increasingly used to improve adherence and tailor drug-release profiles, but their development remains challenging because multiple interacting formulation and process variables strongly affect product performance. Quality by Design (QbD) offers a systematic framework to address these challenges, yet its implementation for bilayer tablets and associated analytical methods remains fragmented. In this review we map and critically analyze published applications of QbD to the formulation design, manufacturing process, and testing of bilayer tablets. Using

New co-drugs of O-demethylvenlafaxine and NSAIDs exert antidepressant effects by alleviating inflammation, oxidative stress, and increasing neurotransmitter.

Depression and anxiety disorders are highly comorbid, yet their complex pathogenesis often limits the efficacy of monotherapy. Growing evidence implicates neuroinflammation in their pathogenesis. Co-drugs that linked two active molecules into a single compound and released the drugs after administration, which offering improved efficacy and tolerability than individual drug mixtures or monotherapy. In this work, five new co-drugs ODV-NSAIDs were synthesized from O-desmethylvenlafaxine (ODV) with non-steroidal anti-inflammatory drugs (NSAIDs) to achieve synergistic antidepression and anxiolytic
