PTX3 induces the expression of PD-L1 and promotes breast cancer immune escape.

BACKGROUND: Pentraxin 3 (PTX3) is an inflammatory mediator involved in tumorigenesis; however, its role in tumor immune evasion remains elusive. Herein, we demonstrate that PTX3 promotes immune evasion in breast cancer by upregulating PD-L1. METHODS: RT-qPCR and Western blot were performed to assess PD-L1 expression in human and murine breast cancer cells. Cell surface PD-L1 was assayed by flow cytometry. Small interfering RNA and CRISPR-Cas9 were used to inhibit PTX3 expression. Co-culture experiments were conducted to evaluate the suppressive effects of tumor cells on CD8 + T cell activatio

Dual orexin receptor antagonism with lemborexant enhances microglial clearance of β-amyloid in mice.

BACKGROUND: Sleep disturbances elevate brain amyloid-beta (Aβ) levels and represent a modifiable risk factor for Alzheimer's disease (AD). The orexin/hypocretin system regulates sleep-wake behavior and has emerged as a therapeutic target in AD; however, the effects of FDA-approved dual orexin receptor antagonists (DORAs) on amyloid pathology remain unclear. We compared lemborexant, an FDA-approved DORA, to doxepin, an antihistaminergic sleep medication, on amyloid pathology and microglial responses in PSAPP mice. METHODS: PSAPP mice received lemborexant (10 or 30 mg/kg/day), doxepin (35 mg/kg

Rescue a Reassortant Human-Bovine Rotavirus and a Recombinant NSP3-EGFP Rotavirus Serotype G1.

BACKGROUND: Human group A rotaviruses (RVs), with an 11-segmented double-stranded RNA genome, are a cause of viral gastroenteritis in infants and young children. Despite the availability of effective live-attenuated vaccines, RV infection is still a global health challenge. Hence, a reverse genetics system enabling the manipulation of RV segments, such as genome reassortment, is essential for developing new RV vaccines. Herein, a reverse genetics system was used to rescue a reassortant human-bovine RV, incorporating the VP7 gene segment from the human RV strain Wa (serotype G1) and a recombina

HNRNPC Succinylation Influences the Neurodegeneration of Alzheimer's Disease Through YME1L1-Mediated Mitochondrial Metabolism.

Mitochondrial dysfunction and abnormal energy metabolism are important pathological features of Alzheimer's disease (AD). This study investigates how mitochondrial protease YME1L1 affects mitochondrial function and its upstream regulation in the pathogenesis of AD. The AD model was established by using APP/PS1 transgenic mice, primary neurons treated with Aβ1-42, and HT22 cells. The silencing of YME1L1 was achieved to evaluate its effects on mitochondrial function and OPA1 protein hydrolysis. RIP-qPCR and RNA pull-down test were used to evaluate the interaction between HNRNPC and YME1L1 mRNA.

Indiscriminate Trans-Cleavage Activity of CRISPR/SuCas12a2 Enables Sensitive Detection of SARS-CoV-2.

Sensitive detection of SARS‑CoV‑2 remains critical for controlling COVID‑19 outbreaks and guiding patient care. Although reverse transcription-polymerase chain reaction (RT‑PCR), the gold standard for detecting SARS-CoV-2, is highly sensitive, the need for specialized equipment and trained personnel limits its widespread application in low or middle-resource settings. Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR) technology could overcome these limitations by providing simplicity, low cost, and high specificity. However, current CRISPR-based diagnostics can simultaneously

Endothelial-targeted modification of ginseng-derived exosomes for IL-6 SiRNA delivery ameliorates hepatic ischemia-reperfusion injury.

Liver ischemia-reperfusion injury (IRI) serves as a critical pathological basis for post-hepatectomy liver failure and graft dysfunction following liver transplantation. Excessive inflammatory responses, oxidative stress, and cell death are key mechanisms underlying IRI. The lack of multi-targeted therapies contributes to the current insufficiency in clinical IRI management. This study developed endothelial-targeting VHPKQHR peptide (VHP)-modified ginseng-derived exosomes (G-Exos) loaded with IL-6 small interfering RNA (Si-IL6) (siRNA@VG-Exos) to mitigate liver IRI. VHP modification facilitate

A Neurotensin Brake on Exploratory Drive under Persistent Threat.

Avoidance behavior is an adaptive response that delays exploration to promote survival. Avoidance is enhanced by psychological stress and is a hallmark of many neuropsychiatric disorders. Neural circuits that control avoidance by integrating stressful stimuli and modulating exploratory behavior remain underexplored. Elucidating the functional dynamics of this highly conserved phenomenon and the underlying neural mechanism of avoidance is an important open-ended question, with relevance to understanding both innate behaviors and neuropsychiatric disorders. Using predator odor as an innate, chro

A Forward Genetic Screen in Caenorhabditis elegans for Genes that Modulate α-synuclein-Induced Neurodegeneration through the Mitochondrial UPR.

Overexpression of α-synuclein (α-syn), an inherently disordered protein, triggers chronic activation of the mitochondrial unfolded protein response (UPRmt) pathway in Caenorhabditis elegans with enhanced dopaminergic (DAergic) neurodegeneration. Introduction of a loss-of-function(lf) mutation in atfs-1, the main transcriptional regulator of the UPRmt, into α-syn nematodes results in significant neuroprotection from α-syn-induced DA neuron loss, indicating that compensatory mechanisms provide neuroprotection. We performed a F3 forward genetic screen in C. elegans atfs-1(lf) mutants to identify

The Metabolic Reprogramming Mechanism of Astragalus Polysaccharide-Induced Immune Memory in HD11 Cells: Integrated Analysis Based on Glycolysis and the TLR2/mTOR Pathway.

This study aimed to investigate the immunomodulatory effects of APS on HD11 cells and its potential mechanisms. This study aimed to investigate the immunomodulatory effects of APS on HD11 cells and its underlying mechanisms. CCK-8 assays revealed that APS significantly promoted HD11 cell proliferation in a dose-dependent manner. Griess and lactate dehydrogenase (LDH) assays indicated that APS dose-dependently increased nitric oxide (NO) release and LDH activity, although these levels remained lower than those in the lipopolysaccharide (LPS) group. RT-qPCR analysis demonstrated that APS signifi

Mesenchymal stem cell-based therapeutic strategies for female infertility: current evidence and emerging targeting approaches.

Reproductive disorders remain a major cause of subfertility and infertility in human and animals, encompassing ovarian dysfunction, endometritis, impaired endometrial receptivity, and treatment-induced gonadotoxicity. These conditions substantially reduce reproductive efficiency and quality of life. In recent years, advances in translational medicine have shifted focus toward regenerative strategies, particularly stem cell-based therapies, as alternatives to conventional pharmacological or surgical interventions that often yield limited or transient benefits. Among these, mesenchymal stem/stro

Myeloperoxidase aggravates thoracic aortic aneurysm formation in Marfan disease.

AIMS: Patients suffering from Marfan syndrome (MFS), the most prevalent inherited connective tissue disorder, face premature mortality due to dissection and rupture of thoracic aortic aneurysms. Here, we questioned whether myeloperoxidase (MPO), a leucocyte-derived enzyme with high affinity to the vessel wall, contributes to aortic remodelling in MFS. METHODS AND RESULTS: Plasma MPO levels were evaluated in MFS patients and healthy controls. Thoracic aortic aneurysm formation was determined in heterozygous transgenic Fbn1C1041G/+ (MFS) mice, MPO-deficient MFS mice (MFSxMpo-/-), and MFS mice t

PD-1 Blockade-Induced DKK1 Expression by CD8+ T Cells Promotes Blood-Brain Barrier Permeabilization.

UNLABELLED: Anti-PD-1 therapy benefits a subset of patients with brain metastasis (BrM); however, heterogeneous responses imply an incomplete understanding of the brain-immune ecosystem. To elucidate host-driven determinants of this variability, we performed single-cell RNA sequencing to characterize the brain microenvironment. Although anti-PD-1 induced robust antitumor immune activation, it uniquely, among all immune checkpoint inhibitors (ICI) tested, compromised blood-brain barrier (BBB) integrity. This permeabilization was mediated by DKK1-expressing activated CD8+ T cells through the ind

Low-dose lithium supplementation promotes musculoskeletal and metabolic health in ovariectomized female mice.

Postmenopausal women have an increased risk for age-related conditions like sarcopenia, osteoporosis and type 2 diabetes. Here we examined the effects of low-dose lithium (Li) supplementation, a well-known glycogen synthase kinase 3 (GSK3) inhibitor, on ovariectomized (OVX) mice, focusing on muscle strength and endurance, bone mineral density (BMD) and insulin sensitivity. 28-week-old female sham and OVX C57BL/6J mice were divided into three groups: sham, OVX and OVX-Li, with the latter receiving 50 mg/kg/day of Li in their drinking water for 8 weeks. Li supplementation enhanced isometric spec
