Structure-guided design and evaluation of oxazolone-based PIM-1 kinase inhibitors with promising anticancer activity.

PIM-1 kinase is a crucial modulator of cellular processes, including proliferation, survival and programmed cell death, positioning it as a compelling target for anticancer therapies. Using a structure-guided drug design strategy, we have designed novel oxazolone derivatives as potential PIM-1 inhibitors. Out of 42 designed oxazolone derivatives, molecular docking identified 30 candidates with favorable binding profiles. In this study, synthetic oxazolone derivatives (O20, O23 and O29) were evaluated for their inhibitory potential against PIM-1 through biochemical and biophysical approaches. E

State of the Art Review: Thiazide Diuretics Exploit the Endocannabinoid System via NAPE-PLD.

The present State of the Art Review will take stock of targeting the endocannabinoid system (ECS) in the management of hypertension and vascular diseases. Major efforts have been made in the last thirty years to develop compounds modulating the ECS for diseases, both in the central and peripheral tissues. Agonists of the cannabinoid receptor CB1 elicited hypotension but were at strong risks of inducing tachycardia, heart and kidney damage. The clinical translation of CB2 agonists, as well as antiobesity CB1 blockers, proposed as promising treatments for cardiovascular risk factors, was complic

T-2 toxin induces ER stress-dependent liver injury via mitophagy-mediated ER-phagy suppression: Berbamine blocks SNARE complex for hepatoprotection.

T-2 toxin is a persistent, bioaccumulative environmental contaminant that poses major health threats to humans and animals. Endoplasmic reticulum (ER) stress and autophagy are two interconnected stress responses critical for maintaining cellular homeostasis. Berbamine (BBM) is an important member of bis-benzy lisoquinoline alkaloid with diverse biological activities. This study aimed to identify the molecular target of BBM against T-2 toxin-induced hepatotoxicity, focusing on autophagy-ER stress crosstalk. We systematically evaluated autophagy and ER stress in human HepaRG cells using immunobl

TIGAR maintains intestinal epithelial regeneration by stabilizing HMGCL and promoting β-catenin β-hydroxybutyrylation in burn-induced sepsis.

Burn-induced sepsis triggers profound intestinal injury, contributing to systemic inflammation and organ damage. Beta-hydroxybutyrate (BHB), a major ketone body, acts as a key regulator of intestinal epithelial regeneration. Its metabolic dysregulation has been implicated in impaired cell proliferation and the maintenance of intestinal stem cells (ISCs). However, the dynamic regulatory mechanisms underlying BHB fluctuation during burn sepsis-induced intestinal injury remain elusive. In this study, we demonstrate that TIGAR expression is markedly reduced in small intestinal crypts of burn sepsi

Endophyte-mediated pest resistance: mechanisms, evidence gaps, and translational opportunities.

Escalating crop losses caused by insect pests, together with pesticide resistance, environmental persistence, and increasing regulatory scrutiny of conventional agrochemicals, have intensified interest in biologically based pest-management strategies. Endophytic bacteria and fungi represent a promising avenue for crop protection because they can influence plant defense, pest performance, and multitrophic interactions within plant tissues. This review critically synthesizes the 'landscape' of endophyte-mediated pest resistance-the 'journey' of microbes from colonization to field protection-thro

Enzyme-Triggered In Situ Assembly of Fe3O4 Nanozyme Synthesis Enables Portable Point-of-Care Detection of Acid Phosphatase.

Acid phosphatase (ACP) is a clinically important enzyme whose early-stage detection is hindered by its extremely low abundance, nonspecific tissue distribution, and rapid loss of activity under conventional analytical conditions. Herein, we present a target-driven in situ nanozyme synthesis strategy that enables rapid and ultrasensitive point-of-care testing (POCT) of ACP. In this approach, ACP catalyzes the hydrolysis of L-ascorbic acid 2-phosphate sesquimagnesium (AAPS), producing ascorbic acid (AA). The generated AA partially reduces Fe3+ ions to Fe2+, thereby initiating alkaline co-precipi

USP52 impedes malignant progression and cell stemness in prostate cancer by deubiquitinating RBM5 to down-regulate NCAPG2.

Ubiquitin-specific protease 52 (USP52) is vital to cancer progression by mediating the deubiquitination; however, its biological role and mechanism in prostate cancer (PCa) remain unexplored. Herein, this study aimed to discover the functional regulation of USP52 with RNA binding motif protein 5 (RBM5) and non-SMC condensin II complex subunit G2 (NCAPG2) in PCa development and stemness. RT-qPCR and Western blot were applied for expression analysis. Proliferation was assessed by colony formation and EdU assays. Cell metastasis was measured by wound healing migration assay and transwell invasion

Endothelial glycocalyx in perioperative medicine current understanding and future direction.

The endothelial glycocalyx (EG) is a dynamic, gel-like layer that lines the luminal surface of blood vessels, playing a crucial role in vascular biology. Previously considered a passive barrier, it is now recognized as a key regulator of vascular tone, permeability, inflammation, and coagulation. Composed mainly of proteoglycans, glycoproteins, and glycosaminoglycans, the EG serves as a semipermeable interface between blood and the endothelium, maintaining microvascular flow and modulating nitric oxide (NO) production while protecting against oxidative and inflammatory damage. This review high

Diphenylpiperazine attached to different triazole acetamide derivatives as a potent α-glucosidase inhibitor, new insight into the in silico study of an uncompetitive inhibitor.

Type 2 diabetes mellitus (T2DM) is considered as one of the major global health challenges, and inhibition of α-glucosidase is an effective strategy for controlling postprandial hyperglycemia. Therefore, this study aimed to design and develop novel α-glucosidase inhibitors. This study synthesized thirteen di-phenyl piperazine-based derivatives attached to different triazole acemamide (8a-m) via a multi-step procedure involving a Cu(I)-catalyzed click reaction and evaluated them as α-glucosidase inhibitors for type 2 diabetes mellitus (T2DM) management. All derivatives were screened as α-glucos

Combined calcium chloride and heat treatment delays softening of fresh-cut jackfruit by activating the Ca2+ signaling-NAC29 transcriptional regulatory module.

Fresh-cut jackfruit is highly susceptible to rapid softening during storage, representing a major quality deterioration that limits its commercial distribution. This study elucidated the potential mechanisms by which combined calcium chloride and heat treatment (CH) mitigates softening in fresh-cut jackfruit. CH treatment mitigated cell wall degradation, inhibited the accumulation of water-soluble pectin (WSP), and delayed the degradation of chelator-soluble pectin (CSP) and cellulose, while significantly suppressing the activities of key cell wall-degrading enzymes, including polygalacturonas

Therapeutic targeting of cancer-associated PKM2 mutants by shikonin via mutation-induced pocket recognition.

Pyruvate kinase (PKM2) is a key glycolytic enzyme involved in cancer metabolic reprogramming and frequently harbors oncogenic missense mutations that alter its structural and functional dynamics. Here, we investigate the molecular basis of recognition of the anti-cancer drug shikonin in wild type (WT) and six PKM2 mutants. Molecular docking identified three mutants (R246S, P117L, and H464A) exhibiting favourable binding of shikonin within a newly identified pocket adjacent to the allosteric site, while the WT accommodated shikonin in the known active site pocket, consistent with previous liter

Archaeal HhH-GPD proteins expand the functional repertoire of DNA glycosylases in base excision repair.

Genomic DNA continuously sustains oxidative, alkylative and deaminative lesions that compromise genome stability in all three domains of life. Base excision repair (BER), the major conserved pathway eliminating small non-bulky base damage, is initiated by substrate-specific DNA glycosylases. The helix-hairpin-helix-Gly/Pro/Asp (HhH-GPD) superfamily comprises ancient, functionally diverse glycosylases ubiquitous across Bacteria, Eukarya and Archaea. Prior cross-kingdom reviews outline general HhH-GPD features, but no dedicated work systematically catalogues and phylogenetically analyzes archaea

Design and development of 1,5-diarylpyrazole-based multitarget-directed ligands as dual COX-2/HDAC6 inhibitors for Alzheimer's disease therapy: Molecular dynamics and experimental insights.

Alzheimer's disease (AD) is a multifactorial neurodegenerative disorder, requiring multitarget-directed ligands capable of modulating interconnected pathological pathways. Here, a pharmacophore-hybridization strategy combining COX-2-selective 1,5-diarylpyrazole scaffolds with a zinc-binding group was used to design dual COX-2/HDAC6 inhibitors. Twenty hybrids were synthesized and evaluated, identifying 10a and 11e as the most potent compounds, with IC50 values of 0.18 and 0.66 μM for COX-2, and 0.15, 0.12 μM for HDAC6, respectively. Molecular dynamics simulations confirmed stable binding of 11e

Effects of a microbial composite amendment and ryegrass on multi-metal(loid) stabilization and nutrient cycling functional potential in smelting-contaminated soil.

Multi-metal(loid) contamination from nonferrous smelting threatens soil safety and weakens microbial nutrient cycling, yet field remediation is assessed mainly by metal immobilization. Whether microbial amendments and vegetation can simultaneously reduce metal risk and improve the cycling functional potential of C, N, P, and S remains poorly understood under field conditions. A 300-day in situ experiment at a Pb-Zn smelting site evaluated four treatments: control (CK), microbial amendment (M), ryegrass (P), and combined (MP). MP treatment reduced Pb, Cd, and Zn bioavailability by 42-52% and sh

Heat Shock Proteins in Cancer: Mechanisms and Therapeutic Targeting.

Heat shock proteins (HSPs) are molecular chaperones that couple proteostasis to cancer metabolic reprogramming under oncogenic and microenvironmental stress. This Review integrates structural, biochemical, preclinical, and clinical evidence on HSP90, HSP70, HSP60, HSP40, HSP110, and small HSPs. We explain how ATPase cycles, chaperonin cages, co-chaperone networks, and organelle-specific localization stabilize metabolic enzymes, glucose transporters, hypoxia-responsive factors, and electron-transport-chain components, thereby coordinating glycolysis, mitochondrial bioenergetics, redox homeostas

Loss of CSE couples with BRAF V600E to fuel thyroid cancer metastatic progression through zinc-dependent activation of the EMT machinery.

BACKGROUND: BRAF V600E, the principal oncogenic driver in papillary thyroid carcinoma (PTC), strongly correlated with lymph node metastasis, yet the underlying molecular mechanisms remain elusive. METHODS: This study screened effector related to the BRAF V600E mutation and PTC progression through bioinformatics analysis. The precise functional roles and regulatory networks of cystathionine γ-lyase (CSE), a primary hydrogen sulfide (H₂S)-producing enzyme, in thyroid cancer metastasis were then investigated in cell and mice models. Finally, these findings were further validated in a cohort of c

m6A RNA methylation orchestrates spermatogenesis: decoding the synergistic regulatory network of signaling pathways and ubiquitination modifications.

N6-methyladenosine (m6A), the most prevalent epitranscriptomic modification in eukaryotic mRNA, dynamically regulates mRNA stability, splicing, and translational efficiency through a sophisticated "writer-eraser-reader" network. This system comprises methyltransferase writers (e.g., METTL3/14, METTL16, WTAP), demethylase erasers (e.g., ALKBH5, FTO), and recognition readers (e.g., YTHDC2, YTHDF2, hnRNPC). This precise regulatory mechanism governs spermatogonial stem cell proliferation, meiotic initiation, and spermatid maturation by orchestrating cell cycle progression, meiotic timing, and sper

Oncogenic VEGFR2 rewires the extracellular matrix to impair endothelial mechanics and destabilize tumor vasculature.

Solid tumors are highly dynamic environments, in which endothelial cells (ECs) interface with blood, cancer cells, and the extracellular matrix (ECM), thereby regulating tumor behavior and metastatic dissemination. We recently showed that aberrant Vascular Endothelial Growth Factor Receptor 2 (VEGFR2) activation promotes tumor progression through ECM remodeling. Here we investigated whether VEGFR2-driven ECM remodeling affects EC behavior and vessel stability. Using decellularized ECM from Sk-Mel-31 melanoma cells expressing either VEGFR2WT or the aberrant VEGFR2R1032Q mutant, we found that VE

Acute Pancreatitis Associated With Anaphylactic Shock: A Case Report and Literature Review.

Acute pancreatitis is most commonly caused by gallstones or alcohol abuse, although rare etiologies such as drug-induced anaphylaxis have been described. Pancreatic involvement during anaphylactic shock remains exceptional and is likely underdiagnosed. We report the case of a 58-year-old woman who developed acute circulatory shock shortly after the topical application of a hemorrhoidal ointment (TriAnal). She presented with hypotension, tachycardia, pruritus, and erythematous urticarial lesions. Laboratory investigations revealed elevated serum lipase and tryptase. Abdominal computed tomograph
