The Effect of Aging on Auditory Nerve Function: Insights from Intraoperative eCAP Recordings in Cochlear Implant Users.

BACKGROUND: Advanced age is associated with poorer speech perception outcomes in cochlear implant (CI) users. In an effort to improve our understanding of the basis for this decline, this study focused on the contributions from peripheral auditory processing, using the electrically evoked compound action potential (eCAP). OBJECTIVES: To investigate the effect of aging on intraoperative, suprathreshold eCAP responses (amplitude growth function [AGF] slopes, eCAP maximum amplitudes, and N1 latencies) across the electrode array, in a large cohort of recipients of newer generation devices, who me

Cumulative effects of subsequent concussions on the neural patterns of young rugby athletes: data from event-related potentials.

Our study aimed at detecting a potential cumulative effect of subsequent concussions on the neural activation patterns of young rugby athletes with or without concussion history. Event-related brain potential (ERP) data from 24 rugby players, 22-year-old on average, were retrospectively examined. All underwent a Sport Concussion Assessment Tool (SCAT2) during preseason and an on-site ERP task (P300) following a recent concussion event (<48 hours). Sixteen players suffered at least one concussion in the previous 3 years and eight were without self-reported past concussion. While no differences

Impaired motor cortical plasticity associated with cannabis use disorder in young adults.

Maladaptive cortical plasticity has been described in individuals with heroin and methamphetamine addiction and may mediate other substance abuse disorders. It is unknown whether cannabis dependence in humans alters the capacity for induction of cortical plasticity. The aim of this study was to non-invasively investigate cortical plasticity with transcranial magnetic stimulation in young adults who meet DSM-5 criteria for cannabis use disorder (CUD). Thirty men (ages 20- 30) who used cannabis daily over the previous 6 months (15 diagnosed of CUD) and 15 demographically matched non-users were e

Temporary Versus Permanent Synaptic Loss from Repeated Noise Exposure in Guinea Pigs and C57 Mice.

A single brief noise exposure can cause a significant loss of cochlear afferent synapses without causing permanent threshold shift. Previously we reported that the initial synaptic loss is partially reversible in Guinea pigs, indicating that synaptic loss can be categorized as either temporary or permanent. Since synaptic loss is biased to innervating auditory nerve fibers (ANFs) with low spontaneous spike rates (SSR), which are critical to the coding of in-background noise, coding-in-noise deficits (CIND) have been predicted to result from noise-induced synaptic damage. However, recent study

Boosting the LTP-like plasticity effect of intermittent theta-burst stimulation using gamma transcranial alternating current stimulation.

BACKGROUND: Transcranial Alternating Current Stimulation (tACS) consists in delivering electric current to the brain using an oscillatory pattern that may entrain the rhythmic activity of cortical neurons. When delivered at gamma frequency, tACS modulates motor performance and GABA-A-ergic interneuron activity. OBJECTIVE: Since interneuronal discharges play a crucial role in brain plasticity phenomena, here we co-stimulated the primary motor cortex (M1) in healthy subjects by means of tACS during intermittent theta-burst stimulation (iTBS), a transcranial magnetic stimulation paradigm known t

Stimulation strategies for selective activation of retinal ganglion cell soma and threshold reduction.

OBJECTIVE: Retinal prosthetic implants restore partial vision to patients blinded due to outer retinal degeneration, using a camera-guided multielectrode array (MEA) that electrically stimulates surviving retinal neurons. Commercial epi-retinal prostheses use millisecond-scale charge-balanced, symmetric, cathodic-first biphasic pulses to depolarize retinal ganglion cells (RGCs) and bipolar cells (BCs), frequently creating oblong perceptions of light related to axonal activation of RGCs. Stimulation strategies that avoid axonal stimulation and decrease the threshold of targeted neurons may sign

Inhibition in the auditory brainstem enhances signal representation and regulates gain in complex acoustic environments.

Inhibition plays a crucial role in neural signal processing, shaping and limiting responses. In the auditory system, inhibition already modulates second order neurons in the cochlear nucleus, e.g. spherical bushy cells (SBCs). While the physiological basis of inhibition and excitation is well described, their functional interaction in signal processing remains elusive. Using a combination of in vivo loose-patch recordings, iontophoretic drug application, and detailed signal analysis in the Mongolian Gerbil, we demonstrate that inhibition is widely co-tuned with excitation, and leads only to mi

Cognitive reappraisal fails when attempting to reduce the appetitive value of food: an ERP study.

This event-related potential (ERP) study investigated neural correlates of cognitive reappraisal during the exposure to food cues. Thirty-three healthy, normal-weight women viewed images of high-caloric food and non-food items after an overnight fast. The participants were instructed to either passively look at the pictures, or to change (increase, decrease) the appetitive value of the food items. The P300 and the late positive potential (LPP) were higher across all conditions for food relative to non-food pictures. In the 'increase condition' the food images were rated as more appetizing and

Nicotine-evoked cytosolic Ca(2+) increase and cell depolarization in capillary endothelial cells of the bovine adrenal medulla.

Endothelial cells are directly involved in many functions of the cardiovascular system by regulating blood flow and blood pressure through Ca(2+) dependent exocitosis of vasoactive compounds. Using the Ca(2+) indicator Fluo-3 and the patch-clamp technique, we show that bovine adrenal medulla capillary endothelial cells (B AMCECs) respond to acetylcholine (ACh) with a cytosolic Ca(2+) increase and depolarization of the membrane potential (20.3+/-0.9 mV; n=23). The increase in cytosolic Ca(2+) induced by 10microM ACh was mimicked by the same concentration of nicotine but not by muscarine and was

Suppression of axonal conduction by sinusoidal stimulation in rat hippocampus in vitro.

Deep brain stimulation (DBS), also known as high frequency stimulation (HFS), is a well-established therapy for Parkinson's disease and essential tremor, and shows promise for the therapeutic control of epilepsy. However, the direct effect of DBS on neural elements close to the stimulating electrode remains an important unanswered question. Computational studies have suggested that HFS has a dual effect on neural elements inhibiting cell bodies, while exciting axons. Prior experiments have shown that sinusoidal HFS (50 Hz) can suppress synaptic and non-synaptic cellular activity in several in

Additivity of loud-sound--induced threshold losses in the cat under conditions of active or inactive cochlear efferent-mediated protection.

1. An additivity model for the accretion of cochlear sensorineural hearing losses has been described from studies in the guinea pig cochlea. Among other aspects, the model allows determination of how residual hearing losses after an initial exposure (E1) affect hearing losses to be expected to a subsequent second exposure (E2). In the present study, the model was applied to temporary hearing losses produced in the cat cochlea by loud pure tones at a frequency from 3 to 15 kHz, affecting regions from 2 to 28 kHz. Successive identical exposures, generally with an interexposure interval of approx

Bilateral electrocochleographic findings in unilateral Menière's disease.

At present, electrocochleography is the only proven investigation that can demonstrate objectively the presence of endolymphatic hydrops. The electrophysiologic recordings in response to sound stimuli show an enhancement of the negative summating potential in these cases. It is well established that patients with unilateral Meniere's disease have a high likelihood of development of the disease bilaterally in the fullness of time. Using transtympanic electrocochleography in 40 patients who manifested unilateral clinical Meniere's disease, we have recorded bilateral abnormalities indicative of e

Dorsal column input to thalamic VL neurons: an intracellular study in the cat.

This study investigated the role of the ventral lateral (VL) nucleus of the thalamus as a lemniscal relay to motor cortex. Intracellular recordings were obtained from thalamic VL relay neurons in cats anesthetized with chloralose, following stimulation of the dorsal column nuclei. VL neurons were identified by their short-latency input from the cerebellar nuclei, their antidromic activation from motor cortex and their anatomical location. A total of 105 neurons was studied. The occurrence of temporal facilitation to double volleys was also examined. It was found that 80/105 (75%) neurons respo

Longitudinal distribution of cochlear potentials and the K+ concentration in the endolymph after acoustic trauma.

Guinea pigs were exposed to 142 dB third-octave band of noise control at 1 kHz for 1 h. At different times after exposure the endocochlear potential (EP), the anoxic negative endocochlear potential (-EP), the concentration of K+ (K+e) and microphonic potentials were recorded in scala media in four cochlear turns. The remaining hair cells were counted in each animal. Immediately after the exposure, the EP and K+e decreased evenly in all four cochlear turns and gradually returned to normal physiological values in 5-20 days. When measured 20 days after the exposure, essentially normal EP and K+e

Effects of acute and chronic amphetamine treatment on Purkinje neuron discharge in rat cerebellum.

The spontaneous discharge of the cerebellar Purkinje neuron in urethane-anesthetized rats is used as an indicator of central noradrenergic activity during the acute and chronic administration of d-amphetamine. Acute parenteral administration of amphetamine causes a dose-dependent slowing of the discharge. Since this effect is not seen in animals in which the catecholaminergic innervation has been selectively removed by previous treatment with 6-hydroxydopamine, it is hypothesized that the slowing is caused by the increased release and/or blocked reuptake of endogenous norepinephrine. Acute adm

Control of membrane N+ permeability in a hyperpolarizing photoreceptor: similar effect of light and metabolic inhibitors.

In the hyperpolarizing photoreceptors of the scallop Pecten irradians the metabolic inhibitors cyani 'e and 2,4-dinitrophenol cause a rapid hyperpolarization and increase in membrane permeability to potassium ions, similar to the effect of light. Cellular metabolism appears important in maintaining the low permeability to potassium ions necessary to keep the membrane depolarized in darkness, possibly by regulating the intracellular calcium ion concentration.
