Sea Cucumber Saponins Derivatives Alleviate Hepatic Lipid Accumulation Effectively in Fatty Acids-Induced HepG2 Cells and Orotic Acid-Induced Rats.

The sulfated echinoside A (EA) and holothurin A (HA) are two prominent saponins in sea cucumber with high hemolytic activity but also superior lipid-lowering activity. Deglycosylated derivatives EA2 and HA2 exhibit low hemolysis compared to EA and HA, but their efficacies on lipid metabolism regulation remains unknown. In this study, fatty acids-treated HepG2 cells and orotic acid-treated rats were used to investigate the lipid-lowering effects of sea cucumber saponin derivatives. Both the saponin and derivatives could effectively alleviate lipid accumulation in HepG2 model, especially EA and

Dietary omega-6/omega-3 ratio is not associated with gut microbiota composition and disease severity in patients with nonalcoholic fatty liver disease.

In this cross-sectional study, we hypothesized that a high dietary ratio of omega-6 (n-6) to omega-3 (n-3) fatty acids could be associated with an altered gut bacterial composition and with the disease severity in patients with nonalcoholic fatty liver disease (NAFLD). A total of 101 NAFLD patients were included in the study, of which 63 underwent a liver biopsy. All 101 patients completed a 14-day food and activity record. Ebispro 2016 professional software was used to calculate individual macronutrients and micronutrients consumed. Patients were grouped into 3 quantiles (Q) according to a lo

A pilot study of microbial signatures of liver disease in those with HIV mono-infection in Rio de Janeiro, Brazil.

OBJECTIVE: The rectal microbiome was examined to assess the relationship between the microbiome and liver disease in HIV-infection. DESIGN: Eighty-two HIV-1 mono-infected individuals from the PROSPEC-HIV-study (NCT02542020) were grouped into three liver health categories based on results of controlled attenuation parameter (CAP) and liver stiffness measurement (LSM) of transient elastography: normal (n = 30), steatosis (n = 30), or fibrosis (n = 22). METHODS: Liver steatosis and fibrosis were defined by CAP at least 248 dB/m and LSM at least 8.0 kPa, respectively. 16S rRNA gene and whole gen

Dysregulated hepatic lipid metabolism and gut microbiota associated with early-stage NAFLD in ASPP2-deficiency mice.

BACKGROUND: Growing evidence indicates that lipid metabolism disorders and gut microbiota dysbiosis were related to the progression of non-alcoholic fatty liver disease (NAFLD). Apoptosis-stimulating p53 protein 2 (ASPP2) has been reported to protect against hepatocyte injury by regulating the lipid metabolism, but the mechanisms remain largely unknown. In this study, we investigate the effect of ASPP2 deficiency on NAFLD, lipid metabolism and gut microbiota using ASPP2 globally heterozygous knockout (ASPP2+/-) mice. METHODS: ASPP2+/- Balb/c mice were fed with methionine and choline deficient

Loss of lysophosphatidic acid receptor 1 in hepatocytes reduces steatosis via down-regulation of CD36.

Nonalcoholic steatohepatitis is a major public health concern and is characterized by the accumulation of triglyceride in hepatocytes and inflammation in the liver. Steatosis is caused by dysregulation of the influx and efflux of lipids, lipogenesis, and mitochondrial β-oxidation. Extracellular lysophosphatidic acid (LPA) regulates a broad range of cellular processes in development, tissue injury, and cancer. In the present study, we examined the roles of LPA in steatohepatitis induced by a methionine-choline-deficient (MCD) diet in mice. Hepatocytes express LPA receptor (Lpar) 1-3 mRNAs. Stea

SMN Depleted Mice Offer a Robust and Rapid Onset Model of Nonalcoholic Fatty Liver Disease.

BACKGROUND & AIMS: Nonalcoholic fatty liver disease (NAFLD) is considered a health epidemic with potential devastating effects on the patients and the healthcare systems. Current preclinical models of NAFLD are invariably imperfect and generally take a long time to develop. A mouse model of survival motor neuron (SMN) depletion (Smn2B/- mice) was recently shown to develop significant hepatic steatosis in less than 2 weeks from birth. The rapid onset of fatty liver in Smn2B/- mice provides an opportunity to identify molecular markers of NAFLD. Here, we investigated whether Smn2B/- mice display

Role of Cholesterol-Associated Steatohepatitis in the Development of NASH.

The rising prevalence of nonalcoholic fatty liver disease (NAFLD) and NAFLD-related cirrhosis in the United States and globally highlights the need to better understand the mechanisms causing progression of hepatic steatosis to fibrosing steatohepatitis and cirrhosis in a small proportion of patients with NAFLD. Accumulating evidence suggests that lipotoxicity mediated by hepatic free cholesterol (FC) overload is a mechanistic driver for necroinflammation and fibrosis, characteristic of nonalcoholic steatohepatitis (NASH), in many animal models and also in some patients with NASH. Diet, lifest

Curcumin and liver disease.

Liver diseases pose a major medical problem worldwide and a wide variety of herbs have been studied for the management of liver-related diseases. In this respect, curcumin has long been used in traditional medicine, and in recent years it has been the object of increasing research interest. In combating liver diseases, it seems clear that curcumin exerts a hypolipidic effect, which prevents the fatty acid accumulation in the hepatocytes that may result from metabolic imbalances, and which may cause nonalcoholic steatohepatitis. Another crucial protective activity of curcumin, not only in the c

Diacylglycerol activation of protein kinase Cε and hepatic insulin resistance.

Nonalcoholic fatty liver disease (NAFLD) is now the most frequent chronic liver disease in Western societies, affecting one in four adults in the USA, and is strongly associated with hepatic insulin resistance, a major risk factor in the pathogenesis of type 2 diabetes. Although the cellular mechanisms underlying this relationship are unknown, hepatic accumulation of diacylglycerol (DAG) in both animals and humans has been linked to hepatic insulin resistance. In this Perspective, we discuss the role of DAG activation of protein kinase Cε as the mechanism responsible for NAFLD-associated hepat

Liver fibrosis in paediatric liver diseases.

Numerous paediatric liver diseases from different origins may be complicated by development of liver fibrosis and progression to cirrhosis. Although fibrogenesis, which represents a major driving force for the development of liver fibrosis, has common tracts whatever the aetiology, liver fibrosis has different histopathological patterns in paediatric liver disease. In these diseases management choices may depend upon the stage of liver fibrosis. Thus, the accurate estimation of histological pattern of liver fibrosis is important for the prevention of the subsequent complications. Liver biopsy

Tetrahydrobiopterin is synthesized from 6-pyruvoyl-tetrahydropterin by the human aldo-keto reductase AKR1 family members.

Tetrahydrobiopterin (BH(4)) is a cofactor for aromatic amino acid hydroxylases and nitric oxide synthase. The biosynthesis includes two reduction steps catalyzed by sepiapterin reductase. An intermediate, 6-pyruvoyltetrahydropterin (PPH(4)) is reduced to 1(')-oxo-2(')-hydroxypropyl-tetrahydropterin (1(')-OXPH(4)) or 1(')-hydroxy-2(')-oxopropyl-tetrahydropterin (2(')-OXPH(4)), which is further converted to BH(4). However, patients with sepiapterin reductase deficiency show normal urinary excretion of pterins without hyperphenylalaninemia, suggesting that other enzymes catalyze the two reduction

The interplay of plant polysaccharide structure, gut microbiota metabolism, and host health: Mechanisms and perspectives.

Plant polysaccharides, as natural biomolecules, possess the capability to modulate the structure and function of gut microbiota, subsequently influencing host health. Nevertheless, the molecular mechanisms underlying their interactions remain insufficiently understood. This article presents a systematic review of the impact of the chemical structure of plant polysaccharides, including monosaccharide composition and types of glycosidic bonds, on the metabolic preferences of gut microbiota. It emphasizes how structural variations in polysaccharides can promote the proliferation of specific gut m

Enhancement in the nutritional profile and hepatoprotective activity of pine pollen via Aspergillus niger fermentation.

Fermentation with Aspergillus niger significantly enhances the hepatoprotective effects of pine pollen. This study evaluated the effects of Aspergillus niger-fermented pine pollen (FPP) against alcoholic fatty liver disease (AFLD) using ethanol-induced HepG2 cells and an AFLD mouse model. In AFLD mice, FPP markedly reduced hepatic triglyceride levels from 1.47 ± 0.13 to 0.90 ± 0.11 mg/g and serum alanine aminotransferase (ALT) from 2.27 ± 0.63 to 1.02 ± 0.15 U/mL, indicating significant alleviation of lipid accumulation and liver injury. Mechanistically, FPP suppressed lipogenesis by downregul

Lactobacillaceae in Acute and Chronic Liver Diseases: From Microbiota Modulation to Therapeutic Potential.

Liver diseases, including metabolic-associated fatty liver disease (MAFLD), alcoholic liver disease (ALD), and viral hepatitis, are highly prevalent and constitute a major global health burden. Accumulating evidence indicates that dysbiosis of the gut microbiota is closely associated with the development and progression of various liver diseases. Among microbial regulators, Lactobacillaceae, a family of probiotic lactic acid bacteria, has attracted considerable attention for its capacity to reshape the gut microbiota, strengthen mucosal barrier integrity, modulate nutrient metabolism, and regu

Application of probiotic therapy in nonalcoholic fatty liver disease: mediating mechanism and future perspective.

Nonalcoholic fatty liver disease (NAFLD) has a global prevalence of 20%-33%, and has become the main cause of chronic liver disease. Apart from lifestyle modification therapy, there is currently no definitive pharmacological treatment, thus there is an urgent need to find effective intervention strategies to treat NAFLD. With the discovery of the important role of gut microbes in the pathogenesis of NAFLD, research on the prevention and treatment of non-alcoholic fatty liver disease by probiotics is increasing. At present, many studies have confirmed the role of probiotic regulation in the tre

Hepatic lipopolysaccharide binding protein partially uncouples inflammation from fibrosis in MAFLD.

Non-alcoholic fatty liver disease (NAFLD), recently renamed metabolic-associated fatty liver disease (MAFLD), is the most common liver disease worldwide. The progression to fibrosis, occurring against a backdrop of hepatic steatosis and inflammation, critically determines liver-related morbidity and mortality. Inflammatory processes contribute to various stages of MAFLD and thought to instigate hepatic fibrosis. For this reason, targeting inflammation has been heavily nominated as a strategy to mitigate liver fibrosis. Lipopolysaccharide binding protein (LBP) is a secreted protein that plays a

PGE2 synthesis and signaling in the liver physiology and pathophysiology: An update.

The liver plays a central role in systemic metabolism and drug degradation. However, it is highly susceptible to damage due to various factors, including metabolic imbalances, excessive alcohol consumption, viral infections, and drug influences. These factors often result in conditions such as fatty liver, hepatitis, and acute or chronic liver injury. Failure to address these injuries could promptly lead to the development of liver cirrhosis and potentially hepatocellular carcinoma (HCC). Prostaglandin E2 (PGE2) is a metabolite of arachidonic acid that belongs to the class of polyunsaturated f
