Ser/Thr Kinase-Dependent Phosphorylation of the Peptidoglycan Hydrolase CwlA Controls Its Export and Modulates Cell Division in Clostridioides difficile.

Cell growth and division require a balance between synthesis and hydrolysis of the peptidoglycan (PG). Inhibition of PG synthesis or uncontrolled PG hydrolysis can be lethal for the cells, making it imperative to control peptidoglycan hydrolase (PGH) activity. The synthesis or activity of several key enzymes along the PG biosynthetic pathway is controlled by the Hanks-type serine/threonine kinases (STKs). In Gram-positive bacteria, inactivation of genes encoding STKs is associated with a range of phenotypes, including cell division defects and changes in cell wall metabolism, but only a few ki

Molecular Characteristics, Ecology, and Zoonotic Potential of Escherichia coli Strains That Cause Hemorrhagic Pneumonia in Animals.

Hemorrhagic pneumonia (HP) is a rare but highly lethal disease, mainly of dogs and cats, caused by hemolytic Escherichia coli strains that contain cnf1 (encoding cytotoxic necrotizing factor 1). After encountering fatal HP in two dogs, we used contemporary molecular methods, including multilocus sequence typing and whole-genome sequencing, to compare the corresponding case isolates with published HP clinical isolates and newly obtained fecal E. coli isolates from 20 humans and animals in the index HP case household. We also compared the aggregated HP clinical isolates, which represented 13 dis

Critical Role of Cytosolic DNA and Its Sensing Adaptor STING in Aortic Degeneration, Dissection, and Rupture.

BACKGROUND: Sporadic aortic aneurysm and dissection (AAD), caused by progressive aortic smooth muscle cell (SMC) loss and extracellular matrix degradation, is a highly lethal condition. Identifying mechanisms that drive aortic degeneration is a crucial step in developing an effective pharmacologic treatment to prevent disease progression. Recent evidence has indicated that cytosolic DNA and abnormal activation of the cytosolic DNA sensing adaptor STING (stimulator of interferon genes) play a critical role in vascular inflammation and destruction. Here, we examined the involvement of this mecha

Fatty Acid Synthase and Acetyl-CoA Carboxylase Are Expressed in Nodal Metastatic Melanoma But Not in Benign Intracapsular Nodal Nevi.

BACKGROUND: Melanoma is a potentially lethal form of skin cancer for which the current standard therapy is complete surgical removal of the primary tumor followed by sentinel lymph node biopsy when indicated. Histologic identification of metastatic melanoma in a sentinel node has significant prognostic and therapeutic implications, routinely guiding further surgical management with regional lymphadenectomy. While melanocytes in a lymph node can be identified by routine histopathologic and immunohistochemical examination, the distinction between nodal nevus cells and melanoma can be morphologic

Deep-proteome mapping of WM-266-4 human metastatic melanoma cells: From oncogenic addiction to druggable targets.

Cutaneous melanoma is a malignant tumor of skin melanocytes that are pigment-producing cells located in the basal layer (stratum basale) of epidermis. Accumulation of genetic mutations within their oncogenes or tumor-suppressor genes compels melanocytes to aberrant proliferation and spread to distant organs of the body, thereby resulting in severe and/or lethal malignancy. Metastatic melanoma's heavy mutational load, molecular heterogeneity and resistance to therapy necessitate the development of novel biomarkers and drug-based protocols that target key proteins involved in perpetuation of the

Induction of Protective Immunity to Cryptococcal Infection in Mice by a Heat-Killed, Chitosan-Deficient Strain of Cryptococcus neoformans.

UNLABELLED: Cryptococcus neoformans is a major opportunistic fungal pathogen that causes fatal meningoencephalitis in immunocompromised individuals and is responsible for a large proportion of AIDS-related deaths. The fungal cell wall is an essential organelle which undergoes constant modification during various stages of growth and is critical for fungal pathogenesis. One critical component of the fungal cell wall is chitin, which in C. neoformans is predominantly deacetylated to chitosan. We previously reported that three chitin deacetylase (CDA) genes have to be deleted to generate a chitos

Epistatic rescue of Nkx2.5 adult cardiac conduction disease phenotypes by prospero-related homeobox protein 1 and HDAC3.

RATIONALE: Nkx2.5 is one of the most widely studied cardiac-specific transcription factors, conserved from flies to man, with multiple essential roles in both the developing and adult heart. Specific dominant mutations in NKX2.5 have been identified in adult congenital heart disease patients presenting with conduction system anomalies and recent genome-wide association studies implicate the NKX2.5 locus, as causative for lethal arrhythmias ("sudden cardiac death") that occur at a frequency in the population of 1 in 1000 per annum worldwide. Cloning of Syrian hamster (Mesocricetus auratus) cytokine cDNAs and analysis of cytokine mRNA expression in experimental visceral leishmaniasis.

The Syrian golden hamster (Mesocricetus auratus) is uniquely susceptible to a variety of intracellular pathogens and is an excellent model for a number of human infectious diseases. The molecular basis for this high level of susceptibility is unknown, and immunological studies related to this model have been limited by the lack of available reagents. In this report we describe the cloning and sequence analysis of portions of the Syrian hamster interleukin 2 (IL-2), IL-4, gamma interferon (IFN-gamma), tumor necrosis factor alpha, IL-10, IL-12p40, and transforming growth factor beta cDNAs. Developmental toxicities of albendazole and its metabolites in zebrafish (Danio rerio) embryos.

Albendazole (ABZ) is a benzimidazole anthelmintic medicine widely used in livestock and, especially, aquaculture industries. Given its widespread use, ABZ commonly enters waterways, negatively impacting aquatic organisms. Classified as Pregnancy Category C because of its teratogenic properties, ABZ poses a high risk of developmental toxicity. In this study, zebrafish (Danio rerio) embryos were used to assess the developmental toxicity of ABZ and its two metabolites, ABZ-sulfoxide and ABZ-sulfone. Among these, only ABZ exhibited strong lethality, demonstrating an LC50 value of 83.3 μg/L, and in

BACKGROUND: Pancreatic ductal adenocarcinoma (PDAC), the main pancreatic cancer type, was highly aggressive and lethal. Studies showed that the epidermal growth factor receptor pathway substrate 8-Like protein 3 (EPS8L3) was significantly upregulated in PDAC. This study aimed to explore how EPS8L3 promoted PDAC progression. METHODS: First, bioinformatics analysis, qRT-PCR, and western blot (WB) techniques were utilized to ascertain the expression profile of EPS8L3 in clinical samples and cells of PDAC. Subsequently, EdU proliferation assays, cell apoptosis detection, glycolysis assay kits, in

Reprogramming resistance in advanced lung cancer: epigenetic modulation to restore therapeutic vulnerability.

Lung cancer is the most prevalent cancer and cause of death; most patients present themselves at an advanced stage and continuously acquire resistance to targeted agents, antibody-drug conjugates, chemotherapy, and immune checkpoint inhibitors. In addition to secondary mutations, epigenetically driven cellular plasticity, including DNA methylation, histone modification, chromatin remodeling, RNA (m 6A)-marks, and non-coding RNAs, facilitates resistance coordination, EMT/drug-tolerant persisters, lineage switching (e.g., NSCLC to NSCLC), bypass signaling, and immune evasion by tumor cells. Proteasomal and lysosomal protein signatures in surviving Meretrix meretrix after acute ammonia exposure.

Ammonia stress can disturb cellular protein homeostasis, but the coordinated response of protein quality-control systems in the bivalve hepatopancreas is not well understood. Hard clams (Meretrix meretrix) were exposed for 96 h to nominal total ammonia nitrogen (TAN) concentrations of 0 (Ctrl), 1.17 (Low), or 11.7 mg L-1 (High). The High concentration was based on an extrapolated 96 h median lethal concentration estimate: the estimate of 11.7 mg L-1 was below the lowest non-control concentration tested (12.5 mg L-1). The experimental unit was the tank (n = 3 per treatment), and hepatopancreata

Upregulation of limb-bud and heart (LBH) drives liver cancer progression by interacting with the oncoprotein Mortalin.

PURPOSE: Hepatocellular carcinoma (HCC), the major subtype of primary liver cancer, is one of the most prevalent and lethal malignancies globally. Limb-bud and heart (LBH), a highly conserved transcriptional cofactor, regulates embryonic development and is implicated in various human diseases, including cancer. However, whether and how LBH relates to liver cancer progression remains elusive. METHODS: The expression patterns and prognostic value of LBH in HCC were evaluated using data from The Cancer Genome Atlas (TCGA) database. The role of LBH during HCC progression was characterized through

Comparative analysis of viral biological characteristics and pathogenicity of representative prevalent avian reovirus strains from genotypes I to V.

The variable pathogenicity of avian reovirus (ARV), driven by genotypic diversity, poses significant control challenges to the global poultry industry. However, biological and pathogenic distinctions among genotypes remain poorly characterized. We performed a comprehensive comparison of 14 ARV strains (genotypes I-V), evaluating viral biological characteristics and pathogenicity indicators. Phylogenetic analysis of these isolates and field strains in China based on the σC gene revealed co-circulation of multiple genotypes (predominantly I and II), with epidemic strains actively evolving and ge

Protein phosphatase 2 phosphatase activator (PTPA) promotes oncogene-induced senescence and carboplatin response in human malignant pleural mesothelioma cells.

PURPOSE: Malignant pleural mesothelioma (MPM) is a rare cancer of the pleura, frequently related to asbestos exposure. It is characterized by a grim prognosis, as few therapeutic options are available. Typically, MPM exhibits high molecular heterogeneity, providing opportunities for exploiting therapeutic vulnerabilities in well-defined subgroups of patients. Nevertheless, these options have been insufficiently explored. METHODS & RESULTS: We found that inactivation of Protein Phosphatase 2A (PP2A) is a recurrent event in MPM, affecting the cancer cell phenotype and chemotherapy response. Spe

Current methods and emerging strategies for the detection of foodborne bacterial protein toxins.

Foodborne bacterial pathogens are a major source of disease and mortality worldwide and have a significant impact on the global economy. Many of these pathogens produce lethal toxins that may persist in various food matrices even in the absence of viable microorganisms, making direct toxin detection essential for food safety surveillance. This review focuses on five major foodborne bacterial exotoxins selected based on acute clinical severity, structural and mechanistic diversity across toxin classes, a large volume of published data on detection platforms relative to other foodborne protein t
