Overexpression of the mdr1 gene in blast cells from patients with acute myelocytic leukemia is associated with decreased anthracycline accumulation that can be restored by cyclosporin-A.

Typical multi-drug resistance (MDR) in human and animal cell lines is caused by overactivity of a unidirectional drug efflux pump. This pump is composed of a 170-kDa transmembrane glycoprotein (P-glycoprotein) that is encoded by the so-called mdr1 gene. The functionally relevant characteristic of MDR cells is a defect in drug accumulation that can be restored by agents which inhibit the P-glycoprotein pump. The purpose of our study was to find out whether P-glycoprotein inhibitors could increase the daunorubicin (DNR) accumulation in acute myelocytic leukemia (AML) cells, overexpressing the md

A mutation in the gene for type III procollagen (COL3A1) in a family with aortic aneurysms.

Experiments were carried out to test the hypothesis that familial aortic aneurysms, either thoracic or abdominal, are caused by mutations in the gene for type III procollagen (COL3A1) similar to mutations in the same gene that have been shown to cause rupture of aorta and other disastrous consequences in the rare genetic disorder known as Ehlers-Danlos syndrome type IV. A family was identified through a 37-yr-old female captain in the United States Air Force who was scrutinized only because many of her direct blood relatives had died of ruptured aortic aneurysms. The woman was heterozygous for

Genetic regulation of early survival and cyst number after peroral Toxoplasma gondii infection of A x B/B x A recombinant inbred and B10 congenic mice.

Genetics of two traits, survival and brain cyst number after peroral Toxoplasma gondii infection, were studied by using recombinant inbred strains of mice derived from resistant A/J (A) and susceptible C57BL/6J (B) progenitors, F1 progeny of crosses between A/J and C57BL/6J mice, and congenic mice (B10 background). Analysis of strain distribution pattern of survival of A x B/B x A recombinant mice indicated that survival is regulated by a minimum of five genes. One of these genes appears to be linked to the H-2 complex and another is related to an as yet unmapped gene controlling resistance to

Selective gene amplification in mammalian cells after exposure to 60Co gamma rays, 241Am alpha particles, or uv light.

Simian Virus 40 wild type (SV40)-transformed Chinese hamster embryo cells (Co631) contain about five viral copies integrated per cell genome. These SV40 sequences were used as endogenous indicator genes to study the response of mammalian cells to radiation at the gene level. An increase in copy number was detected by dispersed cell blotting and Southern analysis in combination with specific DNA hybridization. All types of radiation tested induce a 15- to 25-fold amplification of SV40 sequences without producing intact virus. The amplification is dose dependent and increases with time after irr

Time-Dependent Effects of Cisplatin on Autophagy Gene Expression in Bladder Cancer Cells.

Background/Objectives: Bladder cancer treatment is frequently hindered by chemoresistance to agents such as cisplatin, a process in which autophagy is hypothesized to play a cytoprotective role. This study aimed to investigate the time-dependent transcriptional dynamics of autophagy-related genes in response to cisplatin in bladder cancer cell lines to better elucidate the molecular underpinnings of this resistance. Methods: Two human bladder cancer cell lines, T24 and 5637, were exposed to varying concentrations of cisplatin. Cell viability and half-maximal inhibitory concentration (IC50) val

Diabetes-associated MYT1 and ST18 genes regulate human beta cell insulin secretion and survival via other diabetes risk genes.

AIMS/HYPOTHESIS: Genetic and environmental factors work together to cause islet beta cell failure, leading to type 2 diabetes. How these factors are integrated to regulate beta cells remains largely unclear. Based on our previous findings that the family of myelin transcription factors (MYT TFs; including MYT1, MYT1L and ST18) prevents mouse beta cell failure by repressing the overactivation of stress response, their regulation by obesity-related nutrition signals in human beta cells and their association with type 2 diabetes, we postulate that these factors prevent human beta cell failure und

Cross-Host Adaptation of Campylobacter jejuni Is Shaped by Chromosomal Backgrounds and Mobile Gene Acquisition, with Human-Associated Traits Emerging Under Limited Mutational Diversification.

Campylobacter jejuni is a major zoonotic pathogen that circulates among birds, livestock, humans, and environmental reservoirs, yet the genomic mechanisms that enable persistence and transmission across divergent hosts remain incompletely understood. Here, we sequenced 61 C. jejuni isolates recovered from multiple host-associated sources in Shenzhen, China, from 2016 to 2023, and analyzed them together with 312 dereplicated publicly available high-quality reference genomes. Phylogenomic analyses resolved three major clades, including one avian-restricted clade and two clades showing frequent c

Molecular Evolution and Expression Analysis of the ADH Gene Family in Apple Bud Mutants.

Alcohol dehydrogenase (ADH) catalyzes the reduction of aldehydes to alcohols, key precursor substrates for volatile ester biosynthesis, which determines the characteristic aroma of apple fruit. However, a comprehensive genome-wide investigation of the ADH gene family in apple has been lacking. In this study, we systematically identified ADH genes in the apple genome using integrated bioinformatics approaches, including phylogenetic analysis, synteny evaluation, promoter cis-element prediction, codon usage bias assessment, and protein interaction network modeling. Expression patterns were exami

Knockdown of Arabidopsis RUS4 impacts pollen coat formation by affecting flavonoid and lipid metabolism.

The DUF647 family member RUS4 contributes to pollen coat formation in Arabidopsis by modulating transcript levels of key genes for flavonoid and lipid metabolism. RUS4 encodes a member of the broadly conserved DUF647 domain-containing protein family that is distributed across eukaryotes. We have previously shown that artificial microRNA knockdown Arabidopsis RUS4 lines (known as amiR-RUS4) exhibit reduced male fertility due to reduced pollen viability, decreased pollen germination rate, and impaired pollen tube growth. However, amiR-RUS4 plants exhibit a previously uncharacterized phenotype in

Multi-Tissue Transcriptomic Analysis Reveals Tissue-Specific Thermal Responses and Sex-Biased Functional Differentiation in Reproductive Tissues of the Brown Frog (Rana dybowskii).

The brown frog (Rana dybowskii) is an important cold-temperate amphibian species in northeastern China, with considerable ecological and resource value. Because its survival, metabolism, and reproduction are highly sensitive to environmental temperature, elucidating the molecular mechanism's underlying temperature response is of great significance for understanding environmental adaptation and reproductive regulation in amphibians. In this study, R. dybowskii was used as the experimental model, and a multi-tissue transcriptomic analysis was conducted on the brain, liver, spleen, ovary, oviduct

Temporal transcriptomic and lipidomic analysis reveals multi-omics dynamic profiles of B. napus seed germination.

BACKGROUND: Lipids represent the major storage reserve in Brassica napus seeds. During germination, lipid mobilization delivers indispensable energy to support seedling establishment, thereby profoundly influencing germination vigor and subsequent seedling growth capacity. Although lipid mobilization is fundamentally required for successful rapeseed germination, the temporal coordination between transcriptional reprogramming and lipid metabolic conversion remains largely elusive, and the underlying molecular regulatory network remains to be systematically deciphered. RESULTS: We conducted an

Multi-strain probiotic suppression of Staphylococcus aureus virulence: Growth kinetics, enterotoxin production, and sea/seb gene expression under Lactobacillus casei, Lactobacillus plantarum, Bifidobacterium bifidum and consortia co-culture conditions.

BACKGROUND: Staphylococcus aureus causes staphylococcal food poisoning (SFP), mediated through heat-stable enterotoxins encoded by virulence genes sea and seb. Probiotic biocontrol represents a promising upstream intervention, yet simultaneous mechanistic evidence spanning growth inhibition, toxin suppression, and virulence gene transcriptional downregulation remains limited. OBJECTIVES: To evaluate the capacity of Lacticaseibacillus casei (formerly Lactobacillus casei), Lactiplantibacillus plantarum (formerly Lactobacillus plantarum), Bifidobacterium bifidum, and their defined consortia to s

Single-cell transcriptomics reveals that RNA pseudouridylation-related gene NHP2 promotes the progression of uveal melanoma and validated in vivo and in vitro.

Uveal melanoma (UM) stands as the predominant primary intraocular malignancy encountered among adults. There is still limited research on the function of RNA pseudouridylation-associated genes in UM. This study identified RNA modification-related genes that play crucial role in UM by single-cell RNA sequencing analysis. We validated their expression in UM tissues and cell lines using immunohistochemistry and Western blotting experiments. Gene knockdown and overexpression experiments were conducted in UM cell lines, and their effects on UM proliferation, migration, invasion and apoptosis were v

Secondhand Smoke Exposure Timing Triggers Distinct Placental Responses in Mouse Pregnancy.

Secondhand smoke (SHS), found in about 57.6% of global public areas as a widespread environmental hazard, has been associated with negative effects during pregnancy, such as preeclampsia (PE) and intrauterine growth restriction (IUGR). Our research investigated the impact of SHS on placental issues in a C57BL/6 model that simulates PE and IUGR in mice. We administered SHS to pregnant mice through a nose-only delivery method, beginning either on embryonic day 12.5 (prior to spiral artery (SA) invasion; labeled SHS-6D) or day 14.5 (following SA invasion; labeled SHS-4D), continuing up to E18.5.
