Associations between childhood adversity and human papillomavirus vaccination in Australian women: a cross-sectional study.

BACKGROUND: Adversity in childhood can lead to greater adoption of risky health behaviours, but less use of preventive health care. The impact of childhood adversity on the uptake of the human papillomavirus (HPV) vaccination in a country with a schools-based vaccination program has not been explored. Our aim was to investigate the associations between adverse childhood factors and HPV vaccination in a cohort of Australian women born in 1989-95 from the Australian Longitudinal Study on Women's Health (aged 12-18 years in 2007 when the Australian schools-based program began). METHODS: Survey a

Evaluation of the impact of different notification methods for unvaccinated individuals in a ACWY meningococcal vaccination campaign.

INTRODUCTION: Invasive meningococcal disease (IMD) in Spain remains rare, yet rising trends in serogroups W and Y prior to 2019 prompted the national recommendation to replace adolescent meningococcal C vaccination with the quadrivalent meningococcal ACWY (MenACWY) vaccine and to implement a catch-up campaign for individuals born between 2001 and 2007. In the Region of Murcia, the COVID-19 pandemic delayed implementation until September 2021. After an initial mass-vaccination phase and a long period without active outreach, coverage remained suboptimal, prompting a multi-phase reminder strateg

Epigenetic plus cytokine therapy induces stem-like CD8+ T-cell reservoirs and intratumoral immune niches to overcome anti-PD-1 resistance.

BACKGROUND: Checkpoint blockade (CPB) has limited efficacy in patients with colorectal cancer (CRC), warranting improved therapeutic strategies capable of remodeling the tumor microenvironment. Stem-like TCF1+CD8+ T cells, cytotoxic CD8+ T cells, and lymphoid aggregates have been associated with CPB responsiveness, either individually or as coordinated immune hubs. METHODS: We evaluated the combination of αPD-1, the class I histone deacetylase inhibitor entinostat, and the IL-15 superagonist N-803 in CPB-resistant murine CRC and breast cancer models with varying antigen presentation deficienc

Clinical and body composition parameters as predictors of response to chemotherapy plus PD-1 inhibitor in gastric cancer.

BACKGROUND: Predicting the treatment efficacy of programmed cell death protein 1 (PD-1) inhibitors is crucial for guiding optimal treatment plans and preventing unnecessary complications for cancer patients. We aimed to develop a prediction model using clinical and body composition parameters to identify gastric cancer (GC) patients who would respond to chemotherapy plus PD-1 antibody. METHODS: Clinical data of GC patients treated with chemotherapy plus PD-1 antibody (immunotherapy cohort, n = 120) or chemotherapy alone (chemotherapy cohort, n = 82) following surgical resection were reviewed

Neoadjuvant chemotherapy with or without PD-1/PD-L1 inhibitors in resectable esophageal squamous cell carcinoma: a meta-analysis based on randomized controlled trials.

BACKGROUND: Neoadjuvant chemotherapy (NC) is a cornerstone in the management of resectable esophageal squamous cell carcinoma (ESCC). The integration of PD-1/PD-L1 inhibitors into NC (NIC) regimens has shown promise; however, its efficacy and safety remain uncertain. This meta-analysis aims to compare the potential risks and clinical benefits of NIC versus NC in patients with resectable ESCC based on randomized controlled trials (RCTs). METHODS: A thorough search of six databases was performed to identify RCTs evaluating NIC and NC in resectable ESCC. Key outcomes analyzed included the pathol

Increased PD-1 expression in livers associated with PD-1-antibody-induced hepatotoxicity.

Vanishing bile duct syndrome (VBDS) is a serious drug induced liver injury characterized by chronic cholestasis and loss of intrahepatic bile ducts. VBDS has been reported also following checkpoint inhibitor treatment. We compared CD3 + , CD4 + , CD8 + , CD20 + , CD57 + , PD-1 + and PD-L1 + lymphocyte infiltrates in liver biopsies of patients that encountered VBDS (n = 2) or hepatotoxicity (n = 3) after pembrolizumab (n = 4) or nivolumab (n = 1) treatment with samples from normal liver (n = 10), non-alcohol steatohepatitis (NASH, n = 10), primary biliary cholangitis (PBC, n = 10) or pembrolizu

Uridine depletion impairs CD8⁺ T cell antitumor activity through N-glycosylation.

Immune checkpoint blockade (ICB) faces limitations owing to high cost and restricted efficacy. This study identifies SNX17 as a key mediator of ICB resistance. Elevated SNX17 correlates with poor anti-PD-1 response in humans and mice. SNX17 deletion in tumor cells inhibits tumor growth via CD8+ T cell-dependent mechanisms. SNX17 reduces uridine in the tumor microenvironment (TME), suppressing IFN-γ and upregulating PD1 in CD8+ T cells. Exogenous uridine shows antitumor efficacy comparable to anti-PD-1/PD-L1 in low-SNX17 tumors and overcomes resistance in high-SNX17 models. Uridine enhances CD8

Depletion of myeloid-derived suppressor cells sensitizes murine multiple myeloma to PD-1 checkpoint inhibitors.

BACKGROUND: Cancer immunotherapy using immune checkpoint blockade (ICB) has revolutionized cancer treatment. However, patients with multiple myeloma (MM) rarely respond to ICB. Accumulating evidence indicates that the complicated tumor microenvironment (TME) significantly impacts the efficacy of ICB therapy. Therefore, investigating how TME components in MM influence ICB treatment is urgent. METHODS: We employed two well-established murine myeloma models, 5TGM1 and Vk*MYC, by intravenously injecting 5TGM1 or Vk*MYC cells into mice, respectively, to determine ICB therapeutic efficacy in MM. Impact of Exposure to Benzodiazepines on Adverse Effects and Efficacy of PD-1/PD-L1 Blockade in Patients With Non-Small Cell Lung Cancer.

BACKGROUND: The impact of concomitant medications on immune-related adverse events (irAEs) and immune checkpoint inhibitor (ICI) efficacy in non-small cell lung cancer (NSCLC) remains unclear. Benzodiazepine receptor agonists (BZRAs), commonly prescribed for anxiety and insomnia in cancer care, may influence antitumor immunity via γ-aminobutyric acid (GABA) signaling. Here, we retrospectively analyzed medical records of NSCLC patients treated with ICIs. METHODS: In an initial exploratory analysis, BZRA use was significantly associated with a lower incidence of irAEs, prompting further evaluat

Investigation of immune checkpoint molecules (CTLA-4, PD-1, PD-L1, Tim-3) expressions in preeclampsia: A comparative study of membranous and soluble forms.

Preeclampsia (PE) is characterized by immune dysfunction, including altered expression levels of multiple immune checkpoints (ICs), which are essential for inducing immune tolerance during pregnancy. While the pivotal role of ICs in PE is well-established, a limited understanding remains of the changes in their various forms, particularly in their membranous and secretory states. This study focused on exploring the probable role of ICs in the pathophysiology of PE via measuring the levels of their transmembrane and soluble forms. Initially, expression levels of transmembrane CTLA-4, PD-1, PD-L
