In vivo selection of genetically modified erythroid cells using a jak2-based cell growth switch.

Cell-based therapies have potential widespread applications in clinical medicine, and methods for controlling the fate of transplanted cells are needed. We have previously described a means for directing the growth of genetically modified cells in vivo using a derivative of the thrombopoietin receptor, mpl, that is reversibly activated by a drug called a chemical inducer of dimerization (CID). Since Jak2 participates in signaling from a number of different cytokine receptors (including mpl), we tested whether direct activation of the JH1 domain of Jak2 would broaden the repertoire of hematopoi

Evidence of a possible epigenetic inactivation mechanism operating on a region of mouse chromosome 19 in gamma-radiation-induced thymic lymphomas.

Loss of heterozygosity (LOH) analysis, performed in 68 gamma-radiation-induced primary thymic lymphomas of F1 hybrid mice, provided evidence of significant LOH on chromosome 19 in a region defined by the D19Mit106 (22 cM) and D19Mit100 (27 cM) markers (Thymic Lymphoma Suppressor Region 8, TLSR8). Cd95 and Pten, two genes mapped at this region, were inactivated in a vast majority of these tumors (85.3% for Cd95 and 61.8% for Pten). Moreover, altered expression of Cd95 and Pten occurred concomitantly in 34 of 68 (50%) thymic lymphomas suggesting a coordinated mechanism of inactivation of these g

Rho kinase cascade activation in circulating leukocytes in patients with diabetes mellitus type 2.

BACKGROUND: The intracellular ROCK signaling pathway is an important modulator of blood pressure and of cardiovascular and renal remodeling when Rho-kinase activity is increased. Besides, in preclinical models of diabetes, ROCK activation has also a role in abnormal glucose metabolism as well as in subsequent vascular and myocardial dysfunction. In humans, there are a few data assessing ROCK activation in patients with type 2 diabetes mellitus (T2D) and no studies assessing upstream/downstream components of the ROCK pathway. We assessed here levels of ROCK activation and some of the RhoA/ROCK

Modulation of 5-fluorouracil activation of toll-like/MyD88/NF-κB/MAPK pathway by Saccharomyces boulardii CNCM I-745 probiotic.

BACKGROUND AND AIM: Chemotherapy drugs that act via Toll-like receptors (TLRs) can exacerbate mucosal injury through the production of cytokines. Intestinal mucositis can activate TLR2 and TLR4, resulting in the activation of NF-κB. Intestinal mucositis characterized by intense inflammation is the main side effect associated with 5-fluorouracil (5-FU) treatment. Saccharomyces boulardii CNCM I-745 (S.b) is a probiotic yeast used in the treatment of gastrointestinal disorders. The main objective of the study was to evaluate the effect of S.b treatment on the Toll-like/MyD88/NF-κB/MAPK pathway ac

PKD1 induces p21(waf1) and regulation of the cell cycle via direct activation of the JAK-STAT signaling pathway in a process requiring PKD2.

Autosomal dominant polycystic kidney disease is characterized by cyst formation in the kidney and other organs and results from mutations of PKD1 or PKD2. Previous studies suggest that their gene products have an important role in growth regulation. We now show that expression of polycystin-1 activates the JAK-STAT pathway, thereby upregulating p21(waf1) and inducing cell cycle arrest in G0/G1. This process requires polycystin-2, a channel protein, as an essential cofactor. Mutations that disrupt polycystin-1/2 binding prevent activation of the pathway. Mouse embryos lacking Pkd1 have defectiv

A Derivative of Plumbagin Targets the JAK2/STAT3 Pathway to Inhibit the Progression of Oral Squamous Cell Carcinoma.

Signal transducer and activator of transcription 3 (STAT3) drives multiple hallmarks of tumorigenesis, making it a validated therapeutic target for diverse cancers. The 1,4-naphthoquinones, such as plumbagin (PL), exhibit anticancer activity via inhibiting STAT3 phosphorylation and dimerization, but their severe cytotoxicity precludes clinical translation. Here, we screened 23 PL derivatives for STAT3 inhibitory activity and normal cell cytotoxicity, identifying III-1a as the optimal candidate with superior STAT3 inhibition and reduced toxicity compared to PL. Molecular docking and cellular th

Melatonin Promotes Neurogenesis via the JAK2/STAT3 Pathway in Hypoxic-Ischemic Neonatal Rats.

Neonatal hypoxic-ischemic brain damage (HIBD) is a leading cause of neurological dysfunction and long-term disability in newborns. Lactate accumulation and metabolic disturbances after brain injury inhibit neurogenesis, while the restorative capacity of endogenous neural stem cells (NSCs) is essential for neural reconstruction. Melatonin (Mel) alleviates neonatal brain injury, but its effects on NSCs proliferation and migration remain unclear, and the visualization methods for dynamic monitoring of metabolic changes are inadequate. In this study, a neonatal rat model of HIBD was established, a

Effects of Type I Interferon Signaling and JAK Kinase Inhibition in a Mouse ACL Reconstruction Model.

Improving anterior cruciate ligament reconstruction (ACL-R) healing remains a clinical challenge. Macrophages play a crucial role in musculoskeletal tissue injury repair. However, the mechanism by which macrophages modulate the healing after ACL-R remains incompletely characterized. Macrophages with an interferon gene expression signature gradually accumulate at the bone-to-tendon interface during healing. This study investigated the effects of interferon signaling on ACL-R healing using type I interferon receptor 1 (IFNAR1)-deficient mice and an FDA-approved JAK inhibitor baricitinib. Sixteen

Dysregulated JAK2 signaling delays fracture healing by enhancing STAT3/NF-κB signaling and inhibiting BMP2-mediated osteogenesis.

AIMS: Fracture nonunion is increasingly recognized in metabolic disorders such as obesity and type 2 diabetes (T2D), yet the molecular mechanisms underlying impaired fracture healing remain incompletely understood. We investigated how dysregulated JAK2 signaling affects fracture repair under metabolic stress and conditions of sustained JAK2 activation. MATERIALS AND METHODS: We integrated bioinformatic analyses with multiple mouse models of obesity, T2D, and JAK2 overexpression. Osteogenic differentiation, callus maturation, trabecular microarchitecture, and mechanical properties were evaluat

PTPRO represses breast cancer lung metastasis by inhibiting the JAK2-YAP axis.

Lung metastasis is the primary cause of breast cancer-related mortality. Protein tyrosine phosphatases such as PTPRO are important in cancer progression. However, the role and underlying mechanisms of PTPRO in breast cancer lung metastasis are largely unknown. The function of PTPRO in breast cancer metastasis was examined in mice with ptpro deficiency driven by the PyMT promoter. The regulatory role of PTPRO in JAK2-YAP activation was tested in cell-based knockdown, overexpression and catalytic-dead mutation assays. Bioinformatics analyses and assays of human cancer specimens and mouse tumour

Telocyte-derived exosomes promote angiogenesis and alleviate acute respiratory distress syndrome via JAK/STAT-miR-221-E2F2 axis.

Acute respiratory distress syndrome (ARDS) is characterized by severe respiratory failure and significant inflammation, leading to vascular and epithelial cell damage. The absence of effective pharmacologic treatments underscores the need for novel therapeutic approaches. Telocytes (TCs), a newly identified type of interstitial cells, have shown potential in tissue repair and angiogenesis, particularly through the release of exosomal microRNAs (miRNAs). Exosomes were isolated from LPS (lipopolysaccharide)-stimulated TCs and characterized using western blotting and nanoparticle tracking analysi
