Diagnostic and severity scores for Cockayne syndrome.

BACKGROUND: Cockayne syndrome is a progressive multisystem genetic disorder linked to defective DNA repair and transcription. This rare condition encompasses a very wide spectrum of clinical severity levels ranging from severe prenatal onset to mild adult-onset subtypes. The rarity, complexity and variability of the disease make early diagnosis and severity assessment difficult. Based on similar approaches in other neurodegenerative disorders, we propose to validate diagnostic and severity scores for Cockayne syndrome. METHODS: Clinical, imaging and genetic data were retrospectively collected

Long-Term Effect of Keratinized Tissue Width on Peri-implant Health Status Indices: An Updated Systematic Review and Meta-analysis.

PURPOSE: The keratinized tissue (KT) around dental implants may be useful to ease plaque control. This meta-analysis aimed to compare peri-implant health parameters based on KT width (≥ 2 mm vs < 2 mm). MATERIALS AND METHODS: A highly sensitive search of five databases was performed up to February 1, 2020. The quality of eligible studies was assessed using a modified version of the Newcastle-Ottawa checklist. A random-effects model was used to pool standardized mean differences with 95% confidence intervals. To discover sources of heterogeneity, subgroup analysis and meta-regression were used

Dental Fluorosis according to Birth Cohort and Fluoride Markers in an Endemic Region of Colombia.

OBJECTIVES: To analyze changes in the dental fluorosis (DF) incidence according to a birth cohort and explore current exposure to DF in a case series. METHODS: Repeated cross-sectional study of two periods: 2015 and 2018. Two standardized examiners registered DF using the Thylstrup-Fejerskov index in permanent teeth of children aged 7-18 years. Period and birth cohort frequencies were estimated by a generalized linear model, binomial family, and logarithmic link function. Period estimates are presented as prevalence ratios (PR) and birth cohort estimates as cumulative incidence ratios (RR); 9

Biologic and pathologic aspects of osteocytes in the setting of medication-related osteonecrosis of the jaw (MRONJ).

Medication-related osteonecrosis of the jaw (MRONJ) is a potentially severe, debilitating condition affecting patients with cancer and patients with osteoporosis who have been treated with powerful antiresorptives (pARs) or angiogenesis inhibitors (AgIs). Oral risk factors associated with the development of MRONJ include tooth extraction and inflammatory dental disease (e.g., periodontitis, periapical infection). In bone tissues, osteocytes play a bidirectional role in which they not only act as the "receiver" of systemic signals from blood vessels, such as hormones and drugs, or local signals

Awareness of Malocclusion Is Closely Associated with Allergic Rhinitis, Asthma, and Arrhythmia in Late Adolescents.

BACKGROUND: Oral infectious diseases are typified by dental caries and periodontal diseases and are known to be associated with various systemic diseases. However, clear associations between occlusal disorders and systemic diseases have not yet been established. We investigated the association between an awareness of malocclusion and common diseases in late adolescence. METHODS: We retrospectively reviewed the data of mandatory medical questionnaires that are a legal requirement of the freshman medical checkup. We collected the data of all the students who completed the questionnaires between

The role of macrophages in osseointegration of dental implants: An experimental study in vivo.

To investigate the role of macrophages in the osseointegration of dental implants through induced macrophage reduction in a murine model. Fifty-four Sprague-Dawley rats with bilateral maxillary first molars replaced by titanium implants were randomly assigned into three groups. For the test group, macrophages were depleted by tail-vein injection of clodronate liposome (20 mg/kg) 3 days before implantation and reinjection every 3 days until the sacrifice of the rats (10 mg/kg). Animals treated with Phosphate Buffer saline (PBS) alone or empty liposome were included as controls. Samples containe

The impact of oro-facial pain conditions on oral health-related quality of life: A systematic review.

Pain in the oro-facial region is one of the most common reasons for patients to seek dental treatment. Oral health-related quality of life (OHRQoL) can be affected not only by pain, but also by other oral disorders. Four main dimensions, Oral Function, Orofacial Pain, Orofacial Appearance and Psychosocial Impact, have been suggested to cover different areas of OHRQoL. The aim of this systematic review was to map the impact of oro-facial pain conditions on the Orofacial Pain dimension of OHRQoL (PROSPERO registration: CRD42017064033). Studies were included if they reported Oral Health Impact Pr

Association between parity and tooth loss among northern Nigerian Hausa women.

BACKGROUND: Female reproduction is associated with physiological, metabolic, and nutritional demands that can negatively affect health and are possibly cumulative when parity is high. While it is probable that maternal oral health is similarly affected, available evidence is based on fairly low parity populations and likely confounders affecting oral health status were not considered. AIM: To determine the relationship between parity and tooth loss in a population with many high parity women. Contributions of age, reproductive and socioeconomic parameters, and oral health practices were consi

Biomechanical performance of an immature maxillary central incisor after revitalization: a finite element analysis.

AIM: To investigate the stress distribution in an immature maxillary incisor and the same tooth after simulated revitalization with deposition of tubular dentine or cementum by finite element analysis (FEA). METHODOLOGY: A finite element model of a maxillary central incisor was developed on the basis of a µCT scan. The tooth was segmented in two parts: a part that represented a tooth in an immature state and an apical part that represented the tissue formed after revitalization. The apical part was given the mechanical properties of dentine or cementum. The immature tooth and the same tooth r

WNT3A accelerates delayed alveolar bone repair in ovariectomized mice.

UNLABELLED: Our goal was to evaluate alveolar bone healing in OVX mice, and to assess the functional utility of a WNT-based treatment to accelerate healing in mice with an osteoporotic-like bony phenotype. INTRODUCTION: Is osteoporosis a risk factor for dental procedures? This relatively simple question is exceedingly difficult to answer in a clinical setting, for two reasons. First, as an age-related disease, osteoporosis is frequently accompanied by age-related co-morbidities that can contribute to slower tissue repair. Second, the intervals at which alveolar bone repair are assessed in a c

Sustained and repeated mouth opening leads to development of painful temporomandibular disorders involving macrophage/microglia activation in mice.

Temporomandibular disorder (TMD) is a set of heterogeneous musculoskeletal conditions involving the temporomandibular joint (TMJ) and/or the masticatory muscles. Up to 33% of the population has had at least 1 symptom of TMD with 5% to 10% of them requiring treatment. Common symptoms include limited jaw movement, joint sound, and pain in the orofacial area. Once TMD becomes chronic, it can be debilitating with comorbidities that greatly reduce one's overall quality of life. However, the underlying mechanism of TMD is unclear because of the multicausative nature of the disease. Here, we report a

Deletion of epithelial cell-specific Cdc42 leads to enamel hypermaturation in a conditional knockout mouse model.

Recent evidence suggests that GTPases Rho family plays an important role in tooth development; however, the role of Cdc42 in tooth development remains unclear. We aimed to investigate the function of Cdc42 in tooth development and amelogenesis. We generated an epithelial cell-specific K5-Cdc42 knockout (KO) mouse to evaluate post-eruption dental phenotypes using a K5-Cre driver line. This model overcomes the previously reported perinatal lethality. Tooth phenotypes were analyzed by micro X-ray, micro-computed tomography (CT), scanning electron microscopy (SEM), energy dispersive X-ray spectros

Transplantation of human dental pulp stem cells ameliorates brain damage following acute cerebral ischemia.

AIMS: Numerous experimental studies have shown that cellular therapy, including human dental pulp stem cells (DPSCs), is an attractive strategy for ischemic brain injury. Herein, we examined the effects of intravenous DPSC administration after transient middle cerebral artery occlusion in rats. METHODS: Male Sprague-Dawley rats received a transient 90 min middle cerebral artery occlusion. DPSCs (1 × 106 cells) or vehicle were administered via the femoral vein at 0 h or 3 h after ischemia-reperfusion. PKH26, a red fluorescent cell linker, was used to track the transplanted cells in the brain.

Dentinal mineralization is not limited in the mineralization front but occurs along with the entire odontoblast process.

The mineralization-front theory is historically rooted in mineralization research fields for many decades. This theory is widely used to describe mineralization events in both osteogenesis and dentinogenesis. However, this model does not provide enough evidence to explain how minerals are propagated from the pulp-end dentin to dentin-enamel junction (DEJ). To address this issue, we modified the current research approaches by a) extending the mineral deposition windows of time from minutes to hours, instead of limiting the mineralization assay on days and weeks only; b) switching a regular fluo

Oral health-related quality of life in patients with disorders of nutrition.

BACKGROUND: Researchers demonstrated a correlation between nutrition and health-related quality of life (HRQoL). Little is known about the correlation between disorders of nutrition (DON) and Oral HRQoL (OHRQoL). OBJECTIVES: (a) To measure OHRQoL in DON patients compared to patients attending nutritional counselling for other reasons (eg, systemic diseases or general dietary counselling) (NCOR) and to healthy controls attending routine dental screenings. (b) To analyse associations between OHRQoL and demographics, health-related behaviours and nutritional practices. We hypothesised that OHRQo

The effect of adjusting the pH of local anaesthetics in dentistry: a systematic review and meta-analysis.

The acidic nature of commercial local anaesthetics (LAs) can cause pain during infiltration and delay the onset of anaesthesia. It is suggested that adjusting the pH of anaesthetic agents could minimize these effects. This systematic review aimed to evaluate the efficacy of buffered LAs in reducing infiltration pain and onset time during dental procedures. MEDLINE, Embase, Scopus and Scielo databases were searched up to April 2017. Randomized controlled trials comparing buffered and unbuffered LAs for intraoral injections were included. Risk of bias was assessed using the Cochrane Collaboratio
