Sex differences in fear memory consolidation via Tac2 signaling in mice.

Memory formation is key for brain functioning. Uncovering the memory mechanisms is helping us to better understand neural processes in health and disease. Moreover, more specific treatments for fear-related disorders such as posttraumatic stress disorder and phobias may help to decrease their negative impact on mental health. In this line, the Tachykinin 2 (Tac2) pathway in the central amygdala (CeA) has been shown to be sufficient and necessary for the modulation of fear memory consolidation. CeA-Tac2 antagonism and its pharmacogenetic temporal inhibition impair fear memory in male mice. Surp

Self-Face Activates the Dopamine Reward Pathway without Awareness.

The self-face advantage has been demonstrated not only at the supraliminal level, but also at the subliminal level. However, it remains unclear whether subliminal self-face processing involves the same neural networks as those for supraliminal self-face processing. Here, we show that the ventral tegmental area, a center of the dopamine reward pathway, exhibited greater activation to subliminal presentations of the self-face than those of the others' faces, whereas subliminal presentations of the others' faces induced activation in the amygdala, which generally responds to unfamiliar informatio

An antibody to RGMa promotes regeneration of cochlear synapses after noise exposure.

Auditory neuropathy is caused by the loss of afferent input to the brainstem via the components of the neural pathway comprising inner hair cells and the first order neurons of the spiral ganglion. Recent work has identified the synapse between cochlear primary afferent neurons and sensory hair cells as a particularly vulnerable component of this pathway. Loss of these synapses due to noise exposure or aging results in the pathology identified as hidden hearing loss, an initial stage of cochlear dysfunction that goes undetected in standard hearing tests. We show here that repulsive axonal guid

Auditory activity is diverse and widespread throughout the central brain of Drosophila.

Sensory pathways are typically studied by starting at receptor neurons and following postsynaptic neurons into the brain. However, this leads to a bias in analyses of activity toward the earliest layers of processing. Here, we present new methods for volumetric neural imaging with precise across-brain registration to characterize auditory activity throughout the entire central brain of Drosophila and make comparisons across trials, individuals and sexes. We discover that auditory activity is present in most central brain regions and in neurons responsive to other modalities. Auditory responses

Transcription of the SCL/TAL1 interrupting Locus (Stil) is required for cell proliferation in adult Zebrafish Retinas.

The human oncogene SCL/TAL1 interrupting locus (Stil) is highly conserved in vertebrate species. Previously, we identified a homolog of the Stil gene in zebrafish mutant (night blindness b, nbb), which showed neural defects in the retina (e.g. dopaminergic cell degeneration and/or lack of regeneration). In this research, we examined the roles of Stil in cell proliferation after degeneration in adult zebrafish retinas. We demonstrated that knockdown of Stil gene expression or inhibition of Sonic hedgehog (Shh) signaling transduction decreases the rate of cell proliferation. In contrast, activat

The fiber connections of the temporal lobe with emphasis on the rhesus monkey.

The temporal lobe has three functionally distinct areas which have minimal interconnections within the lobe, but extensive connections with other parts of the brain. The superior temporal gyrus contains primary and association auditory areas. Inferotemporal cortex is exclusively a visual association area. The temporal pole is involved in social behavior. In addition, the superior temporal sulcus functions as a sensory integration area. From a review of the literature, a schema is proposed for organizing the reciprocal connections of the three functional areas into a series of loops which follo

The immune regulation of complement system in intracerebral hemorrhage(ICH): the double-edged effects of mechanisms and therapeutic strategies.

The complement system plays a dual role in the immunopathophysiological process following intracerebral hemorrhage. Its excessive activation during the acute phase can exacerbate neuroinflammation, disrupt the blood-brain barrier integrity, recruit destructive immune cells, and directly induce cell lysis, thereby worsening secondary brain injury. However, during the subacute and chronic phases, the complement system shows repair potential, including promoting hematoma clearance via pathways such as C3b-CR3, regulating inflammation resolution, and possibly participating in neural remodeling. Tissue-specific mRNA m6A reprogramming unveils vitamin-driven post-transcriptional regulation in mice.

Vitamins are essential metabolic cofactors, yet their roles in epitranscriptomic regulation, particularly N 6-methyladenosine (m6A) modification, remain unclear. Here, we investigate the effects of various vitamins (VB2, VB6, and VB12) on the mRNA m6A epitranscriptome in multiple mouse tissues (brain, liver, and testis). Clustering analyses reveal closer similarity between the brain and testis m6A profiles, whereas the liver exhibits a unique pattern, reflecting tissue-specific regulatory dynamics. More than 90% of m6A sites are independent of mRNA abundance, highlighting the post-transcriptio

Stress-induced neuroinflammation and synaptic dysregulation: Linking HPA axis to glutamate and NMDA receptors.

Chronic stress is a major risk factor for psychiatric and neurological disorders, operating through interconnected molecular cascades that link neuroendocrine dysfunction to synaptic pathology. This review mechanistically examines how stress-induced hypothalamic-pituitary-adrenal (HPA) axis hyperactivation sustains glucocorticoid release, driving microglial activation and astrocytic reactivity toward pro-inflammatory phenotypes characterized by immunometabolic reprogramming. Key inflammatory mediators including tumor necrosis factor-alpha (TNF-α), interleukin-1 beta (IL-1β), and quinolinic aci

Integrated Genetic Networks and Epigenetic Regulation inTooth Development and Maturation.

Tooth development or odontogenesis is a complex morphogenetic process that requires tightly regulated interactions between the oral epithelium and mesenchyme of neural crest origin. In this narrative review, we compile existing knowledge regarding gene regulatory networks and epigenetic factors throughout tooth development from initiation to eruption. Signaling between the epithelium and mesenchyme is mediated by four conserved pathways-Wnt/β-catenin, bone morphogenetic protein (BMP), fibroblast growth factor (FGF), and Sonic hedgehog (Shh)-which operate iteratively and interact through extens

Comprehensive Transcriptomic Profiling of Nonatopic Prurigo Nodularis in Korean Patients Reveals Il-22-driven Epidermal Stress and Neuro-immuno-fibrotic Remodelling.

Prurigo nodularis (PN) is a chronic pruritic dermatosis with incompletely defined pathogenesis, and molecular data from East Asian populations are limited. We characterized the transcriptomic signatures of non-atopic PN in Korean patients to identify pathways linked to chronic itch and lesion persistence. RNA sequencing was performed on lesional and non-lesional skin from 17 PN patients and normal skin from 11 controls, followed by differential expression, functional enrichment, and correlation analyses. Lesional PN skin showed distinct transcriptional signatures with upregulation of Th22/IL-2

Migraine and auditory dysfunction: beyond comorbidity.

Migraine operates as a systemic disorder of multisensory sensitization. Within this spectrum, auditory manifestations, including sensorineural hearing loss, tinnitus, and hyperacusis, represent prevalent yet frequently overlooked otologic phenotypes. Epidemiological data show that 15%-49% of migraine patients present with comorbid auditory symptoms, and nearly two-thirds exhibit auditory electrophysiological abnormalities. However, optimal clinical management remains constrained by an incomplete understanding of pathogenic mechanisms, variable audiological assessment protocols, and dispersed t

Mapping the iceberg: from focal trauma to a spectrum of systemic complications in spinal cord injury.

Spinal cord injury (SCI) has traditionally been viewed as a focal trauma to the central nervous system, with research focusing primarily on the disruption of motor and sensory pathways. However, emerging evidence reveals that SCI rapidly induces systemic changes that extend far beyond the spinal cord. This review synthesizes current evidence to conceptualize SCI as a comprehensive systemic disorder, utilizing the "neuro-immune-metabolic network" as a central explanatory framework. Following the initial injury, the disruption of descending autonomic pathways contributes to widespread physiologi

Neuronal Actin Remodeling and Its Role in Higher Nervous Activity.

The dynamic interaction of memory and forgetting processes determines the formation, stability, and specificity of the engram. While the molecular genetic processes of learning and memory have been intensively studied, the mechanisms of active forgetting have only recently attracted the attention of neuroscientists. The emergence and disappearance of memory traces in the brain are regulated by specific signaling cascades that influence the morphological and functional properties of synaptic connections. Actin remodeling is known to be the basis of neuroplasticity. Cofilin normally acts as an a

Transcriptomic analysis reinforces the implication of spatacsin in neuroinflammation and neurodevelopment.

Hereditary spastic paraplegia (HSP) encompasses a group of rare genetic diseases primarily affecting motor neurons. Among these, spastic paraplegia type 11 (SPG11) represents a complex form of HSP caused by deleterious variants in the SPG11 gene, which encodes the spatacsin protein. Previous studies have described several potential roles for spatacsin, including its involvement in lysosome and autophagy mechanisms, neuronal and neurites development or mitochondria function. Despite these findings, the precise function of the spatacsin protein remains elusive. To elucidate its function, we cond

Stromal Pbrm1 mediates chromatin remodeling necessary for embryo implantation in the mouse uterus.

Early gestational loss occurs in approximately 20% of all clinically recognized human pregnancies and is an important cause of morbidity. Either embryonic or maternal defects can cause loss, but a functioning and receptive uterine endometrium is crucial for embryo implantation. We report that the switch/sucrose nonfermentable (SWI/SNF) remodeling complex containing polybromo-1 (PBRM1) and Brahma-related gene 1 (BRG1) is essential for implantation of the embryonic blastocyst on the wall of the uterus in mice. Although preimplantation development is unaffected, conditional ablation of Pbrm1 in u

Dihydrotestosterone reduces neuroinflammation in spinal cord injury through NF-κB and MAPK pathway.

Neuroinflammation induced by microglia following spinal cord injury (SCI) leads to secondary neurologic injury. Androgens including testosterone and dihydrotestosterone (DHT) show as endogenous neuroprotective factors against multiple neurologic diseases, while their therapeutic role in SCI-induced neuroinflammation and underlying mechanism remains elusive. In the study, we aimed to investigate the role of DHT against microglia-induced neuroinflammation in SCI and evaluate its protective treatment.
