The neuronal P2X7R controversy: Revisiting evidence, methods, and unresolved questions.

The P2X7 receptor (P2X7R) is an ATP-gated ion channel belonging to the purinergic ligand-gated P2X receptor family. In the central nervous system (CNS), activation of this receptor has been proposed to play a key role in the pathogenesis of various neurodegenerative disorders. Its expression has been clearly demonstrated in microglia, where it regulates numerous cellular processes, including cell activation, cytokine release, and calcium signaling. Recent data show convincing arguments for the presence of P2X7R also in other cell types of the nervous system, such as astrocytes, oligodendrocyte

Neuronal Activity Suppresses NEUROD-Dependent Transcription and Genome Organization.

Neuronal activity induces gene expression programs associated with plasticity in the brain, but it also transiently suppresses gene expression through poorly understood mechanisms. Here, we report that neuronal activity downregulates NEUROD-dependent gene expression in cerebellar granule neurons in adult mice. Using NEUROD1/2 double conditional knock-out mice of both sexes, we show that NEUROD binds and activates its target gene enhancers and establishes a three-dimensional genome architecture that facilitates transcription when neuronal activity is low. Moreover, NEUROD antagonizes activity-d

Disulfidptosis in neurodegenerative diseases: From redox imbalance to neuronal dysfunction.

Disulfidptosis is a recently identified form of regulated cell death driven by disulfide stress and cytoskeletal collapse under conditions of impaired reducing capacity. Neurodegenerative diseases (NDs), including Parkinson's disease, Alzheimer's disease, and amyotrophic lateral sclerosis, are characterized by oxidative stress, mitochondrial dysfunction, metabolic impairment, protein aggregation, and cytoskeletal instability-features that may provide a permissive intracellular context for disulfidptosis. However, its occurrence and pathological relevance in these disorders remain incompletely

A Case of Neuronal Intranuclear Inclusion Disease Mimicking Acute Stroke.

Neuronal intranuclear inclusion disease (NIID) is a rare neurodegenerative disorder with diverse clinical manifestations, including encephalitis-like, stroke-like, and seizure episodes. We report a female patient who experienced recurrent stroke-like attacks with cerebrovascular changes resembling an embolism. At the ages of 65 and 71, she exhibited transient stenosis and hypoperfusion of the left middle cerebral artery, followed by hyperperfusion. Initial imaging lacked characteristic diffusion-weighted imaging (DWI) abnormalities, resulting in a misdiagnosis of infarction. The diagnosis of N

Glioblastoma With a Primitive Neuronal Component: A Case Report and Review of the Literature.

Glioblastoma (GBM) with a primitive neuronal component (PNC) is an uncommon GBM subtype. We report a rare case of GBM with PNC originating in the left frontal lobe and review the relevant literature. A 60-year-old woman with no previous medical history presented with a generalized seizure. MRI revealed a lesion with ring enhancement in the left frontal lobe, suggestive of a high-grade glioma. The patient underwent gross total tumor resection. Intraoperative pathological examination suggested a high-grade glioma; therefore, photodynamic therapy was applied to the resection cavity. Histopatholog

The working lives of neuronal 5-HT2A receptors from the outside in.

5-HT2A receptors have been shown to play critical roles in regulating neuronal signalling networks and brain plasticity. They have become important targets for treatment of resistant forms of mental health disorders such as depression, anxiety and post-traumatic stress disorder. Like many G protein-coupled receptors (GPCRs), signalling downstream of 5-HT2A depends on where the receptor is in the cell. Events and signalling pathways modulated by the cell surface pool of receptors may be distinct from those regulated by an internal pool of receptors. In this review, we parse out how the cell org

Disrupted astrocyte-neuron metabolic coupling in amyotrophic lateral sclerosis.

Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease characterised by progressive loss of motor neurons. In addition to neurodegeneration, ALS is increasingly recognised as a disorder associated with widespread metabolic dysfunction, including hypermetabolism, weight loss, and dyslipidaemia, all of which correlate with disease progression and survival. Astrocytes play a central role in maintaining metabolic homeostasis in the central nervous system by supporting neuronal energy demands, regulating glutamate levels, buffering oxidative stress, and maintaining lipid balance.

Neuronal detection of social actions directs collective escape behaviour.

Animals in groups obtain information from social partners to engage in adaptive behaviour1-3. Social information transmission is observed in fish schools4-6, bird flocks7,8 and human groups9,10, but the neural representation of such socially acquired information is poorly understood3,11,12. Here we show that, in the schooling glassfish Danionella cerebrum13-15, collective escape from danger can be mediated by an individual's visual perception of other escaping animals. To understand the neural basis of socially transmitted escape behaviour, we imaged neural activity from adult glassfish viewin

Spatial Organization of Morpho-Electric Subtypes of Pyramidal Neuron in the Subiculum.

The subiculum is a main output structure of the hippocampus, transmitting information from the CA1 to the entorhinal cortex in a spatially structured manner. Prior studies revealed a high electrophysiological and molecular heterogeneity of subicular pyramidal neurons (PYNs) with evidence for further spatial subdivisions of the subiculum. In this study we focused on the cellular organization of the proximal to mid-distal part of the subiculum, designated as subregion 2 (Sub2) and performed a comprehensive electrophysiological and morphological characterization of PYNs by whole-cell patch-clamp

Perinatal exposure to metal mixtures disrupts neuronal function and behavior.

BACKGROUND: Environmental exposure to heavy metals such as lead (Pb), arsenic (As), hexavalent chromium [Cr(VI)], and cadmium (Cd), (PACC), is linked to neurodevelopmental disorders. These metals often co-occur in contaminated environments, but their combined effects on brain development remain poorly understood. OBJECTIVE: To test the hypothesis that perinatal exposure to a mixture of environmentally relevant levels of Pb, As, Cd, and Cr(VI), causes developmental defects in cognition, behavior, and neuronal function. METHODS: Female C57BL/6J mice were exposed to either a single metal or the

Capillary Density and Neuronal Homeostasis in Human Primary Visual Cortex.

BACKGROUND: Hypoxia is a critical determinant of neuronal function and vulnerability in the brain. While neuronal density has been extensively characterized across cortical regions, the spatial relationship between neurons and the microvasculature represents an additional structural constraint on oxygen diffusion and metabolic support. In particular, the distance between neurons and nearby capillaries influences local oxygen availability required to sustain neuronal activity and maintain ionic homeostasis. METHODS: In this study, we quantified neuron-capillary spatial relationships across cor

Biomimetic ferroelectric-semiconductor transistor enables neuronal multisensory integration.

Human brain seamlessly integrates multisensory stimuli to synthesize complementary information for enhanced perceptions, depending on neural principles of superadditivity, inverse effectiveness, and temporal congruency. Replicating multisensory integration in artificial intelligences has remained challenging due to the inefficiency of algorithmic fusions and the absence of hardware-native mechanisms. Here, we demonstrate biomimetic audiovisual integration at the device level of Bi2O2Se ferroelectric-semiconductor field-effect transistors (FeS-FETs) through multiphysics coupling. Our FeS-FETs s

Memristive Motion-Streak Neuron for Spatiotemporal Multiple Object Detection.

Conventional artificial vision systems process dynamic scenes inefficiently by reconstructing motion from discrete frames, which requires post-processing. In contrast, real-world environments containing multiple moving objects demand sensor-level discrimination. This work presents a memristive motion-streak neuron that performs spatiotemporal encoding by integrating an Al/InGaZnO/Al optomemristor with an Ag/HfO2/Pt dynamic memristor, whose relaxation dynamics provide temporal memory. In this system, the presence time of moving objects is detected by decay of the output current, allowing motion

Increased firing rates monotonically expand neuronal coding bandwidth.

Sensory information is often processed by populations of neurons that vary with respect to their levels of spiking activity. It appears plausible that neurons that fire more often are capable of following faster changes in the input signal than those with lower firing rates. In this study, we test this intuitive assumption by systematically investigating how the spiking activity of a neuron affects information transmission. To this end, we employ the coherence function as a spectral measure of information transmission and study the relation between the width of the coherence function, which we

Halting neuron-fibroblast crosstalk disarms tumor and augments immunotherapy.

Innervation propels tumor progression through intricate mechanisms. Recently, Zhang et al. demonstrated that sensory neurons interact with cancer-associated fibroblasts via the calcitonin gene-related peptide (CGRP)-receptor activity-modifying protein 1 (RAMP1) axis to mediate immune exclusion in triple-negative breast cancer. Halting the neuron-fibroblast crosstalk via rimegepant holds promise for reorganizing the extracellular matrix and synergizing with cancer immunotherapy.

Optimized genetic tracers for viral mediated neuronal projection mapping.

Mapping fine axonal projections with high sensitivity remains a major challenge in neuroanatomy. Here, we developed Codon Optimized Membrane Embedded Tracers (COMET), a family of genetically encoded, membrane-anchored fluorescent proteins optimized for adeno-associated virus (AAV) delivery. COMET tracers—based on codon-optimized SuperFolder GFP (gCOMET) and TdTomato (rCOMET) fused to an H-RAS CaaX prenylation motif—enable robust plasma membrane targeting and enhance fluorescent signal localization to fine-caliber axons. In vivo, COMET tracers significantly outperformed conventional soluble flu

MitoPG: An engineered biocompatible nanocarrier for drug delivery to neuronal mitochondria.

Mitochondrial dysfunction is a key contributor to the pathogenesis of major neurodegenerative diseases such as Parkinson's disease. Targeted drug delivery to neuronal mitochondria is often limited by the inherent toxicity and inefficiency of drug carriers. Here we report the mitochondrial targeting ability of MitoPG, a novel nanocarrier based on dendritic polyglycerol (PG) conjugated with triphenylphosphonium (TPP+). Among a library of MitoPGs studied, MitoPG3 demonstrated superior mitochondrial localization and minimal cytotoxicity, without adverse effects on mitochondrial functions. Notably,

Neuronal autoantibodies in neurodegenerative dementia: From evidence to clinical framework.

Neuronal autoantibodies, including against neuronal surface receptors or intracellular antigens, are established pathogenic mediators and therapeutic targets in autoimmune encephalitis (AIE) and paraneoplastic neurological syndromes (PNS). These antibodies are now increasingly reported in patients with clinically diagnosed neurodegenerative dementia, prompting re-evaluation of a neurodegenerative etiology. Within neurodegenerative trajectories, whether such antibodies act as pathogenic drivers, non-causal markers, or immune modulators remains unresolved, and heterogeneous cohorts, assays, and

Neuronal Activity Regulating the Dauer Entry Decision in Caenorhabditis elegans.

The life cycle of the model nematode Caenorhabditis elegans involves a choice between two alternative developmental trajectories. Hermaphroditic larvae can either become reproductive adults or, under conditions of crowding or low food availability, enter a long-term, stress-resistant diapause known as the dauer stage. Chemical signals from a secreted larval pheromone promote the dauer trajectory in a concentration-dependent manner, and their influence can be antagonized by increased availability of a microbial food source. The decision is known to be under neuronal control, involving both sens

AKT is necessary for neuronal hypertrophy and epilepsy caused by Pten knockout.

Phosphatase and tensin homolog on chromosome 10 (PTEN) is a key negative regulator of the AKT/mTOR signaling pathway. Mutations in PTEN are highly implicated in Autism Spectrum Disorder (ASD), epilepsy, congenital hydrocephaly, and macrocephaly. While the conditional genetic knockout of Pten in murine neurons results in hypertrophy, increased migration, excitatory synaptogenesis, hyperexcitability, and epileptiform activity, the specific downstream signalling mediators of these pathologies remain to be fully elucidated. Using retroviral-mediated genetic manipulation of individual neurons withi
