The new SCCmec type methicillin-resistant Staphylococcus aureus carried CRISPR-cas system isolated from a pig in China.

Methicillin-resistant Staphylococcus aureus (MRSA) lineages circulate globally in healthcare, community, and livestock-associated (LA) settings. Nine MRSA isolates were recovered from swine in China, all exhibiting resistance to ampicillin and multidrug resistance phenotypes. Among eight ST9-t899 isolates, SCCmec type XII(9C2) predominated. However, we identified a novel staphylococcal cassette chromosome mec (SCCmec) type, designated XIII (9A), in an LA-MRSA strain (LS45). Structural analysis revealed SCCmec XIII(9A) comprises a CRISPR-Cas system (cas10-csm2-csm3-csm4-csm5-csm6). Functional a

Human neutrophil α-defensin HNP1 interacts with bacterial cell wall components to promote biofilm formation by Staphylococcus aureus.

Staphylococcus aureus is a major opportunistic pathogen whose ability to form biofilms significantly enhances its persistence and antibiotic resistance. We previously found that human neutrophil peptide 1 (HNP1), a member of the α-defensin family, promotes biofilm formation in Acinetobacter baumannii. Here, we demonstrate that HNP1 enhances biofilm formation across a range of Gram-positive and Gram-negative bacteria, with particularly strong effects in S. aureus, which increases its tolerance to multiple antibiotics. Mechanistically, HNP1 promotes bacterial adhesion and self-aggregation, facil

Artesunate Perturbs GTP Binding of the Conserved GTPase Obg Thereby Alleviating Antibiotic Resistance in Methicillin-Resistant Staphylococcus aureus.

Methicillin-resistant Staphylococcus aureus (MRSA) is an important nosocomial pathogen that causes various secondary infections among hospital-associated patients. The pathogen is challenging to treat due to its resistance to a wide spectrum of antibiotics, including the last-resort antibiotic vancomycin and newly developed drugs, such as linezolid and daptomycin. While the invention of entirely new drugs to combat MRSA infection seems almost impossible, potentiating the efficacy of conventional antibiotics is critical. Our article explores the novel application of the antimalarial drug artesu

Methicillin-resistant Staphylococcus aureus in Saudi Arabia: genomic evidence of recent clonal expansion and plasmid-driven resistance dissemination.

OBJECTIVES: Staphylococcus aureus is a leading cause of hospital-acquired infections worldwide. Over recent decades, methicillin-resistant Staphylococcus aureus (MRSA), which is resistant to multiple antimicrobials, has emerged as a significant pathogenic strain in both hospital and community settings. The rapid emergence and dissemination of MRSA clones are driven by a dynamic and evolving population, spreading swiftly across regions on epidemiological time scales. Despite the vast geographical expanse and diverse demographics of the Kingdom of Saudi Arabia and the broader West Asia region, t

Synergistic antibacterial activity and prevention of drug resistance of daptomycin combined with fosfomycin against methicillin-resistant Staphylococcus aureus.

The combination of daptomycin and fosfomycin is attracting attention due to the rising resistance observed in methicillin-resistant Staphylococcus aureus (MRSA). Most studies indicate that this combination exhibits synergistic or additive antimicrobial activity against MRSA. However, its capacity to prevent resistance remains uncertain. In this study, we first investigated the antibacterial effect of daptomycin and fosfomycin on MRSA, followed by predicting MRSA resistance under the influence of different drugs using a resistance mutation selection window. Subsequently, we conducted preliminar

2-Aminothiophene derivatives reduce resistance to fluoroquinolones in Staphylococcus aureus strains which overexpress NorA and MepA efflux pumps.

In response to the growing threat of antibiotic resistance, this study aimed to identify novel compounds capable of modulating and/or even restoring antibiotic efficacy by inhibiting bacterial efflux pumps. Thirteen 2-aminothiophene (2-AT) derivatives were synthesized and tested against Staphylococcus aureus strains overexpressing NorA and MepA pumps, which confer resistance to fluoroquinolones. Although the 2-ATs displayed little inherent antibacterial activity, several-particularly compounds P4, P7, and P8-significantly potentiated the effects of norfloxacin, ciprofloxacin, and ethidium brom

Treatment outcomes in methicillin-resistant Staphylococcus aureus bacteraemia patients receiving vancomycin: a retrospective study at a tertiary hospital.

BACKGROUND: Methicillin-resistant Staphylococcus aureus (MRSA) bacteraemia remains a major clinical challenge with high mortality, despite guideline-recommended vancomycin therapy. Although a ratio of the area under the curve to the minimum inhibitory concentration (AUC/MIC) ≥400 mg·hour/L is advocated for efficacy, the association between pharmacokinetic/pharmacodynamic (PK/PD) target attainment and clinical outcomes remains uncertain. OBJECTIVE: To determine the in-hospital mortality rate among patients with MRSA bacteraemia receiving vancomycin. The secondary objectives were to (1) evaluat

False positive results obtained with latex agglutination identification of Staphylococcus aureus caused by Macrococcus caseolyticus and Mammaliicoccus fleurettii.

Reliable and rapid identification of Staphylococcus aureus constitutes a critical issue in food safety. A total of 180 meat samples were analysed, and 30 isolates that phenotypically matched S. aureus were obtained. All 30 isolates generated positive results in agglutination with the SlidexStaph test. The results of agglutination were verified with the matrix-assisted laser desorption ionisation time-of-flight mass spectrometry (MALDI-TOF MS) method, S. aureus-specific PCR and sequencing of the 16 S rRNA genes. Among 30 isolates positive in the SlidexStaph test, 17 were finally identified as S

Bacterial cell wall-specific nanomedicine for the elimination of Staphylococcus aureus and Pseudomonas aeruginosa through electron-mechanical intervention.

Personalized synergistic antibacterial agents against diverse bacterial strains are receiving increasing attention in combating antimicrobial resistance. However, the current research has been struggling to strike a balance between strain specificity and broad-spectrum bactericidal activity. Here, we propose a bacterial cell wall-specific antibacterial strategy based on an in situ engineered nanocomposite consisting of carbon substrate and decorated TiOx dots, termed TiOx@C. The fiber-like carbon substrate of TiOx@C is able to penetrate the bacterial membrane of Pseudomonas aeruginosa (P. aeru

Synergistic antimicrobial effects of polyphenolic Chinese medicine active compounds-loaded photothermal nanoparticles against multidrug-resistant Staphylococcus aureus infections.

BACKGROUND: Given the global decline in fertility rates, ensuring the healthy development of infants has become an urgent priority. Staphylococcus aureus (S. aureus) is the leading cause of lactation mastitis, a condition that often results in premature breastfeeding cessation due to the absence of effective and safe preventive measures, posing risks to infant growth and development. While antibiotics remain the primary treatment option, their overuse contributes to the emergence of antimicrobial resistance. PURPOSE: This study aimed to develop a green, safe, and effective strategy for eradic

Matrine alleviates Staphylococcus aureus-induced acute lung injury in mice by inhibiting MLKL and NLRP3-mediated inflammatory activity.

Acute lung injury (ALI) is a serious clinical condition with high incidence and mortality. The inflammatory response induced by gram-positive bacteria, especially Staphylococcus aureus (S. aureus), is a key factor contributing to ALI progression and other infectious diseases. Matrine, known for its diverse biological and pharmacological properties, has not been fully explored for its potential to prevent or treat S. aureus-induced ALI. Our study demonstrated that matrine exerts a protective effect against lung injury in mice infected with S. aureus. Specifically, matrine reduced pulmonary edem
