Molecular and Therapeutic Aspects of Hyperbaric Oxygen Therapy in Neurological Conditions.

In hyperbaric oxygen therapy (HBOT), the subject is placed in a chamber containing 100% oxygen gas at a pressure of more than one atmosphere absolute. This treatment is used to hasten tissue recovery and improve its physiological aspects, by providing an increased supply of oxygen to the damaged tissue. In this review, we discuss the consequences of hypoxia, as well as the molecular and physiological processes that occur in subjects exposed to HBOT. We discuss the efficacy of HBOT in treating neurological conditions and neurodevelopmental disorders in both humans and animal models. We summariz

Modeling succinate dehydrogenase loss disorders in C. elegans through effects on hypoxia-inducible factor.

Mitochondrial disorders arise from defects in nuclear genes encoding enzymes of oxidative metabolism. Mutations of metabolic enzymes in somatic tissues can cause cancers due to oncometabolite accumulation. Paraganglioma and pheochromocytoma are examples, whose etiology and therapy are complicated by the absence of representative cell lines or animal models. These tumors can be driven by loss of the tricarboxylic acid cycle enzyme succinate dehydrogenase. We exploit the relationship between succinate accumulation, hypoxic signaling, egg-laying behavior, and morphology in C. elegans to create ge

Pyogranulomatous pleuropneumonia caused by Nocardia asiatica in a dog coinfected with canine morbillivirus (canine distemper virus).

Nocardia asiatica causing pyogranulomatous pleuropneumonia is reported for the first time in a dog coinfected with canine morbillivirus (CM), diagnosed based on epidemiological, clinical, haematological, images, microbiological, histopathological, polymerase chain reaction and hsp65 gene sequencing findings. The immunosuppression of CM probably favoured the opportunistic behaviour of N. asiatica. Despite the therapeutic measures, the animal died, mainly due to respiratory distress. The association of methods to improve early diagnosis, therapy procedures and prognosis of canine nocardiosis is

Metabolomics: Strategies to Define the Role of Metabolism in Virus Infection and Pathogenesis.

Metabolomics is an analytical profiling technique for measuring and comparing large numbers of metabolites present in biological samples. Combining high-throughput analytical chemistry and multivariate data analysis, metabolomics offers a window on metabolic mechanisms. Because they intimately utilize and often rewire host metabolism, viruses are an excellent choice to study by metabolomics techniques. Studies of the effects of viruses on metabolism during replication in vitro and infection in animal models or human subjects have provided novel insights into these networks and provided new tar

Recent advances in regenerative medicine to treat enteric neuropathies: use of human cells.

As current options for treating most enteric neuropathies are either non-effective or associated with significant ongoing problems, cell therapy is a potential attractive possibility to treat congenital and acquired neuropathies. Studies using animal models have shown that following transplantation of enteric neural progenitors into the bowel of recipients, the transplanted cells migrate, proliferate, and generate neurons that are electrically active and receive synaptic inputs. Recent studies have transplanted human enteric neural progenitors into the mouse colon and shown engraftment. In thi

Recombinant alpha-1-microglobulin: a potential treatment for preeclampsia.

Preeclampsia is a serious pregnancy-specific condition, affecting 10 million women annually worldwide. No specific treatment is currently available. Recent studies have demonstrated abnormal production and accumulation of free fetal hemoglobin in the preeclamptic placenta, and identified subsequent leakage into the maternal circulation as an important factor in the development of preeclampsia. A recombinant version of alpha-1-microglobulin, an endogenous well-characterized heme and radical scavenger, has been developed. Intravenous administration of recombinant alpha-1-microglobulin in animal

Zika crisis in Brazil: challenges in research and development.

Infection with the Zika virus (ZIKV) usually causes a mild acute illness, but two major severe syndromes have been described during the epidemic in Brazil: microcephaly and the Guillain-Barré Syndrome. There is now much evidence to show that ZIKV can infect and damage neuronal cells in vitro. In experimental animals, ZIKV has significant neurotropism and can cause brain damage. At present, diagnosis is still a challenge in the field and there is no treatment available. Another major challenge is that one must devise therapies for pregnant women, at all stages of pregnancy. LISA: Surfactant administration in spontaneous breathing. Which evidence from the literature?

Recent human and animal studies demonstrated that surfactant can be delivered intratracheally without traditional intubation and bagging, but using a fine catheter inserted into the trachea of spontaneously breathing preterm infants on CPAP. This strategy, known as LISA (less invasive surfactant administration) or MIST (minimal invasive surfactant therapy), seems to reduce failure of non-invasive respiratory approach. Avoiding mechanical ventilation and manual inflation it is possible to reduce lung injury due to baro-volutrauma. Moreover leaving the infants supported by N-CPAP during the mane

Brain networks in Huntington disease.

Recent studies have focused on understanding the neural mechanisms underlying the emergence of clinical signs and symptoms in early stage Huntington disease (HD). Although cell-based assays have focused on cell autonomous effects of mutant huntingtin, animal HD models have revealed alterations in the function of neuronal networks, particularly those linking the cerebral cortex and striatum. These findings are complemented by metabolic imaging studies of disease progression in premanifest subjects. Quantifying metabolic progression at the systems level may identify network biomarkers to aid in

Tranexamic acid protects against rotenone-induced apoptosis in human neuroblastoma SH-SY5Y cells.

Rotenone is a pesticide that has been shown to induce the pathological symptoms of Parkinson's disease (PD) in animal models. In this study, we investigated the protective effects of tranexamic acid (TA) on rotenone-induced apoptosis in human dopaminergic SH-SY5Y cells. TA blocked the rotenone-induced phosphorylation of JNK and P38, the downregulation of BCL2 and the upregulation of BAX. Furthermore, TA not only decreased the rotenone-induced cleavage of caspase 9, PARP, and caspase 3, but also increased caspase 3 enzymatic activity. Our findings indicate that TA is able to protect neuronal ce

Cutaneous horn: a potentially malignant entity.

A cutaneous horn is a conical, dense, hyperkeratotic protrusion that often appears similar to the horn of an animal. It is a morphologic designation referring to an unusually cohesive keratinized material, not a true pathologic diagnosis. Cutaneous horns occur in association with, or as a response to, a wide variety of underlying benign, pre-malignant, and malignant cutaneous diseases. The most important common concern is distinguishing a hyperkeratotic actinic keratosis from a cutaneous squamous cell carcinoma. Keratoacanthoma is another cause, as illustrated herein as a projective cutaneous

Potential use of fluorocarbons in lung surfactant therapy.

Exogenous surfactant therapy based on animal lung extract preparations has been developed successfully for the treatment of neonatal respiratory distress syndrome. However, because of the inherent limitations of these natural preparations, the development of new synthetic surfactants is a major objective. We report here that a perfluorocarbon gas (perfluorooctyl bromide, gPFOB) inhibits the formation of the semi-crystalline domains that occur during compression of a Langmuir monolayer of dipalmitoyl phosphatidylcholine (DPPC), taken as a simplified model of lung surfactant. gPFOB also facilita

General principles for clinical use of repetitive transcranial magnetic stimulation (rTMS).

Repetitive transcranial magnetic stimulation (rTMS), a non-invasive technique allowing stimulating neurons in the cerebral cortex, is able to modify durably local as well as distant neuronal activity. Results obtained by stimulation of the primary motor cortex and measurements of induced muscle responses suggest that effects on cortical excitability depend on stimulation frequency and intensity, as well as of pulse-train duration. Such data, as well as results of animal studies have brought a physiological basis for the use of rTMS for treatment of various neurological and psychiatric disorder

Ménière's disease review 2005.

Ménière's disease (MD) is a complex, multifactorial disorder of the inner ear that is the most common cause of the syndrome of episodic vertigo combined with fluctuating hearing loss. In spite of a century of investigation, the etiology and pathophysiology of MD remain controversial and incompletely understood. Among the factors that have contributed to these controversies are the absence of (1) a validated clinical test, (2) an appropriate animal model, and (3) a specific treatment. Nonetheless, physicians are able to assist MD patients with a variety of tailored, symptom-specific medications

Vitamin D-dependent rickets type II in a cat.

A cat with clinical signs Indicating rickets was diagnosed as having a defect of vitamin D receptors. Clinical signs had been seen from four months of age. Treatment with calcium supplementation and various forms of vitamin D did not alter plasma calcium levels or reverse skeletal lesions of lateral antebrachial bowing, lumbar spinal lordosis and costochondral beading. Analgesics were effective for relieving skeletal pain during the bone growth phase and were withdrawn when the animal reached skeletal maturity. Therapy for hip osteoarthritis was given from five years of age until the cat was e

Oral disorders of exotic ungulates.

There are many species classified as ungulates that possess a large variety of dental formulae and dental adaptations. Despite the diversity found within the ungulate group, the list of dental abnormalities remains short. Regardless of the contributing factors, cases of aveolitis accompanied by osteomyelitis ("lumpy jaw") occur more frequently than some of the other dental problems that are encountered in these species (fractures of tusks/canines, malocclusion, eruption cysts). Commonly, treatment of "lumpy jaw" requires exodontia with additional antibiotic therapy using a drug that is appropr

A cytochemical assay for osteoclast cathepsin K activity.

Cathepsin K is a member of the papain superfamily of cysteine proteases and plays a pivotal role in osteoclast-mediated bone resorption. This enzyme is an excellent target for antiresorptive therapies for osteopenic disorders such as osteoporosis.(1) Although isolated inhibitor studies on purified enzymes is required to discover potent and selective inhibitors of cathepsin K, a quantitative cytochemical assay(2) for cathepsin K would allow inhibitors to be tested on actual osteoclasts within sections of bone. Furthermore cathepsin K activity could be used to identify and analyse osteoclasts at

[The neurobiological approaches to obsessive-compulsive disorder].

The lifetime prevalence rate of obsessive-compulsive disorder (OCD) is more than 2 percent of the population. Its contemporary pathophysiological models have been explored. As serotonin reuptake inhibitors and cognitive behavior therapy are both considered first-line treatments of OCD, the treatment interventions provide us with clues. In this review, the authors summarized that genetics, neuropathology in the cortico-striatal-thalamic-cortical (CTSC) circuits, the association between OCD and Tourette's syndrome, the possibility of autoimmune-mediated pathophysiology containing PANDAS, serolog

Pentoxifylline (PTX), methylxanthine derivative and phosphodiesterase inhibitor, has been in use for more than 20 years (with minimal side effects) for the potent hemorrheologic properties and was recently discovered to have influence on function of some immune cells and production of cytokines. Human and animal studies have shown that PTX therapy results in a variety of physiologic changes at the cellular level, which may be important in treating a diverse group of human afflictions. Immune modulation includes increased leukocyte deformability and chemotaxis, decreased endothelial leukocyte a

[Sporotrichosis in a horse].

A 9-year old male Arabian horse was referred to the Department of Large Animal Surgery of the University of Utrecht because of multiple nodules on the inner side of the right hind leg. The nodules seemed to follow a cutaneolymphatic pattern. Histopathology of a nodule showed a granulomatous inflammation with the presence of multinucleated giant cells. In PAS- and Grocott-stained sections, spheroid yeast-like organisms with some budding were found throughout the tissue. A preliminary diagnosis of sporotrichosis was made. A fresh nodule was cultured and the presence of Sporothrix c.f. schenckii
