Pattern of core and surface expression in liver tissue reflects state of specific immune response in hepatitis B.

A series of 67 liver biopsies (20 kidney transplant recipients and 47 outpatients with hepatitis) was investigated for the presence of hepatitis B antigen core (HBc) and surface (HBs) components by immunofluorescence and electron microscopy. The variable appearance of the core in liver cell nuclei and of the surface in the cytoplasm allowed the recognition of expression patterns which, together with histologic parameters, could be integrated into four reaction types of diagnostic and prognostic implications: Type I (Elimination Type). No components or only occasional expression of HBc; histolo

IL-2Ralpha-Directed monoclonal antibodies provide effective therapy in a murine model of adult T-cell leukemia by a mechanism other than blockade of IL-2/IL-2Ralpha interaction.

Adult T-cell leukemia (ATL) develops in a small proportion of human T-cell lymphotrophic virus-I infected individuals. The leukemia consists of an overabundance of activated T cells, which are characterized by the expression of CD25, or IL-2Ralpha, on their cell surface. Presently, there is not an accepted curative therapy for ATL. We developed an in vivo model of ATL in non-obese diabetic/severe combined immunodeficient (NOD/ SCID) mice by introducing cells from an ATL patient (MET-1) into the mice. The leukemic cells proliferated in these mice that lack functional T, B, and natural killer (N

Epidemiological insights into bovine viral diarrhea virus and bovine rotavirus in diarrheic Yaks on the Qinghai-Tibet Plateau.

Diarrhea is common in yak calves and causes substantial economic losses to the yak industry on the Qinghai-Tibet Plateau. Bovine viral diarrhea virus (BVDV) and bovine rotavirus (BRV) are important enteric pathogens in cattle, but their epidemiology and potential association in yaks remain unclear. In this study, 1,623 fecal samples were collected from yaks across different locations, age groups, seasons, and breeds to investigate BVDV, BRV, and BVDV/BRV dual positivity. Viral positivity was detected using PCR amplification, and associated factors were analyzed using chi-square tests and multi

Neurological Manifestations in People Living with Human Immunodeficiency Virus (PLHIV) in India: A Scoping Review and the Need for a National HIV-Neurology Special Interest Group.

BACKGROUND AND OBJECTIVES: Despite a low national prevalence of 0.20%, India's absolute burden of 2.544 million people living with human immunodeficiency virus (HIV) presents clinicians with varied neurological complications. While combination antiretroviral therapy (ART) has extended survival, the neuro-HIV profile in India, characterized by the predominance of HIV-1 clade C, remains distinct from that reported in Western cohorts. This scoping review aimed to identify patterns of neurological manifestations in India across the pre-ART, ART, and "treat-all" eras and to support the establishmen

Computational screening of bacterial and fungal natural compounds targeting monkeypox virus DNA polymerase (F8) holoenzyme.

The Orthopoxvirus known as the monkeypox virus (MPXV) has resurfaced as a global public health concern and is becoming increasingly difficult to control due to growing resistance to tecovirimat and the limitations of current antiviral treatments. This study focused on natural product-based inhibitors targeting the viral DNA polymerase holoenzyme (PDB ID: 8HG1), an essential replication complex mechanistically distinct from conventional therapies. A comprehensive computer-aided drug design workflow was used to screen 36,395 natural compounds through molecular docking, followed by pharmacokineti

HSV-1 infections of the prefrontal cortex: regional vulnerability, cellular mechanisms, and neurocognitive consequences.

Herpes simplex encephalitis (HSE), caused by herpes simplex virus type 1 (HSV-1), is the most common cause of sporadic viral encephalitis and remains associated with substantial mortality and long-term neurological morbidity despite effective antiviral therapy. Although HSE is classically characterized by involvement of the medial temporal lobes and limbic system, recent clinical, preclinical and neuroimaging studies highlight involvement of prefrontal cortical regions, including the medial prefrontal cortex (mPFC) and orbitofrontal cortex (OFC). The functional consequences of HSV-1 associated

Designing a multi-epitope vaccine against La Crosse virus using immunoinformatics approach.

La Crosse virus (LACV) is a mosquito-borne Orthobunyavirus responsible for pediatric encephalitis, with no currently approved vaccine or targeted antiviral therapy. This study aimed to design a multi-epitope subunit vaccine targeting the conserved nucleoprotein of LACV using an integrated immunoinformatics approach. The nucleoprotein exhibited strong antigenicity and favorable physicochemical properties, including stability and hydrophilicity. Highly conserved CTL, HTL, and B-cell epitopes were initially predicted, from which high-affinity, non-allergenic, and non-toxic epitopes were selected.

Structural characterization of HCoV-229E and H163A main protease (Mpro) interactions with the inhibitor narlaprevir.

Narlaprevir, originally developed as a clinical-stage inhibitor of the hepatitis C virus (HCV) NS3/4A protease, has recently demonstrated potent inhibitory activity against the SARS-CoV-2 main protease (Mpro), with its co-crystal structure already resolved. To elucidate the mechanistic basis of its cross-genus inhibition and drug resistance profiles, we determined the crystal structures of narlaprevir in complex with HCoV-229E Mpro and the SARS-CoV-2 Mpro(H163A) mutant at resolutions of 2.01 Å and 2.14 Å, respectively. By integrating these crystallographic data with molecular dynamics (MD) sim

Interactions between Tick-Borne Encephalitis Virus Nonstructural Protein 1 and Blood-Brain Barrier Tight Junction Proteins: Potential Clues to Strain-Specific Neuropathogenicity.

Tick-borne encephalitis virus (TBEV) invades the central nervous system (CNS) through strain-specific mechanisms that remain poorly understood. For mosquito-borne orthoflaviviruses such as dengue and yellow fever viruses, the nonstructural protein 1 (NS1) has been shown to disrupt endothelial barrier integrity by targeting tight junction proteins (TJPs), facilitating viral neuroinvasion. However, comparable mechanisms in TBEVs remain largely unexplored. Here, we investigated the potential interaction of NS1 from high (Hypr)- and low (Vs)-pathogenic TBEV strains with different blood-brain barri

Evaluating Vaginal Microbiome Effects on Antiretroviral Exposure in the Female Genital Tract Using Pharmacometric and Machine Learning Approaches.

Antiretroviral pre-exposure prophylaxis (PrEP) is less effective in women than expected, partly because drug exposure within the female genital tract is variable and may be influenced by the vaginal microbiome. We developed an integrated pharmacometrics and machine learning framework to evaluate associations between vaginal microbiome composition and cervical antiretroviral exposure in women with human immunodeficiency virus (HIV) receiving tenofovir (TFV)-, lamivudine (3TC)-, or emtricitabine (FTC)-containing regimens. Pharmacokinetic and microbiome data were obtained from two single-center s

ZBP1 and IFI16: novel innate sensors and inflammation regulation under infection stress.

The innate immune system is the first line of defense against infection, using pattern recognition receptors to sense pathogens and abnormal host-derived nucleic acids. Among these, Z-DNA-binding protein 1 (ZBP1) and interferon gamma-inducible protein 16 (IFI16) are two important nuclear and cytosolic nucleic acid sensors attracting recent attention. These sensors play key roles in detecting abnormal nucleic acids from viruses, bacteria, and damaged host cells. They trigger downstream innate immune responses to regulate inflammation. Despite much progress, key questions remain about how they a

Genomic Surveillance of Dengue Virus During the 2024 Outbreak in a Northeastern Brazilian State.

Dengue remains a major public health challenge in tropical regions, where viral genetic diversity, vector ecology and human mobility contribute to recurrent outbreaks. In 2024, Brazil experienced its largest recorded dengue epidemic, with intense transmission across the Northeast region. This study characterizes the epidemiological, genomic and phylogeographic features of dengue virus (DENV) circulation during the 2024 outbreak in Alagoas, northeastern Brazil. Of 6102 suspected cases analysed by molecular diagnostics, 1607 (26.3%) were confirmed as DENV infections. Epidemiological analyses rev

Targeted Removal of HCV E2 N2 N-Glycan Is Associated with Improved Immune Responses in Mice.

Hepatitis C virus (HCV) still lacks a licensed vaccine. The envelope glycoprotein E2 is a key neutralizing target, but its dense N-glycan shield can hinder epitope exposure. In this study, we revisit E2 glycan editing and examine whether single-site deletion preserves antigen integrity while improving immune responses in mice under a DNA immunization setting. Using a secreted E2 ectodomain (sE2384-661), we generated five N to D mutants at conserved sites (N1, N2, N4, N6, and N11) and evaluated them in a unified DNA immunization model with identical CpG content and delivery conditions across gr

Perfluorooctanoic acid disrupts lysine metabolism and autophagy to promote white spot syndrome virus infection in shrimp.

Perfluorooctanoic acid (PFOA), a globally distributed per- and polyfluoroalkyl substance (PFAS), is increasingly recognized for its immunotoxic and metabolic effects in aquatic organisms; however, its role in viral disease susceptibility remains poorly understood. Using Pacific white shrimp (Penaeus vannamei) and white spot syndrome virus (WSSV) as a model system, we investigated the effects of chronic PFOA exposure on antiviral immunity and viral transmission. Chronic exposure to environmentally relevant concentrations of PFOA resulted in significant accumulation of PFOA in the hepatopancreas

Chemogenetic Activation of Neurons Promotes Axonal Regeneration and Ameliorates Neurological Deficits in Mice with Intracerebral Hemorrhage by Modulating Mitochondrial Function.

Intracerebral hemorrhage (ICH) is a devastating neurological condition characterized by high morbidity and mortality, with limited treatment options for promoting neurological recovery. Enhancing cortical excitability has emerged as a potential strategy for promoting neurological recovery. However, the role of neuronal activation in mitochondrial regulation and axonal regeneration after ICH remains unclear. A chemogenetic approach using adeno-associated virus (AAV)-human M3 muscarinic designer receptor exclusively activated by designer drugs (hM3Dq) was employed to selectively activate cortica

An oral rotavirus-vectored vaccine confers protection against Clostridium perfringens and rotavirus.

The segmented genome of live-attenuated rotavirus (RV) vaccines provides an ideal platform for developing multivalent vaccines against enteric pathogens. Here, we engineered the commercial human RV vaccine strain LLR as a vector to deliver Clostridium perfringens alpha-toxin (CPA), a critical virulence factor associated with gastroenteritis. A screen of four 2A peptide sequences identified that porcine teschovirus-1 2A (P2A) preceded by an N-terminal GSG spacer enabled optimal cleavage and antigen expression. In mice, the resulting recombinant virus elicited potent systemic antibody responses

Estimating the effect of self-protection on transmission dynamics of SARS-CoV-2 in Germany in 2021: a modelling study.

BACKGROUND: During the COVID-19 pandemic, German states implemented non-pharmaceutical interventions, while individuals also adopted self-initiated protective behavior. Most epidemiological studies tend to focus on one of these aspects, but in reality, both factors influence transmission dynamics simultaneously. In this study, we investigate the effect of self-protection and NPIs on the transmission dynamics of SARS-CoV-2 in Germany during 2021 and identify the corresponding model parameters based on publicly available data. METHODS: We present a unified mathematical model that integrates both

Targeting the lethal weakness of SFTSV: Latest advances in antiviral strategies and immune prevention.

In recent years, severe fever with thrombocytopenia syndrome (SFTS), caused by the SFTS virus (SFTSV), has a fatality rate ranging from 12% to 50%, but there are currently no approved therapies to effectively treat the infection. Host cell receptors CCR2 and LRP1 have been identified as key entry points for viral invasion, and viral non-structural proteins evade the host innate immune response through multiple mechanisms. Several clinical studies have demonstrated that inflammatory markers (such as, IL-6, TNF-α, etc.), lipid metabolites, and ferritin are reliable prognostic indicators, aiding
