An allosteric interleukin-1 receptor modulator mitigates inflammation and photoreceptor toxicity in a model of retinal degeneration.

BACKGROUND: Inflammation and particularly interleukin-1β (IL-1β), a pro-inflammatory cytokine highly secreted by activated immune cells during early AMD pathological events, contribute significantly to retinal neurodegeneration. Here, we identify specific cell types that generate IL-1β and harbor the IL-1 receptor (IL-1R) and pharmacologically validate IL-1β's contribution to neuro-retinal degeneration using the IL-1R allosteric modulator composed of the amino acid sequence rytvela (as well as the orthosteric antagonist, Kineret) in a model of blue light-induced retinal degeneration. METHODS:

Mitigating Surface Poisoning by Bidirectional Regulation for Efficient C1-C2 Molecule Electrooxidation.

The rational design of Pt-based electrocatalysts to alleviate CO intermediates poisoning during electrooxidation of C1-C2 fuels (methanol, ethanol, formic acid) presents a grand challenge. Current detoxification strategies predominantly focus on enhancing OH* formation to accelerate CO* oxidation. However, the periodic voltage/current oscillations induced by accumulated OH species are often overlooked. Such accumulation of secondary poisoning intermediates inevitably leads to a high overpotential and poor operational stability. Herein, guided by machine learning screening, we resolve this dile

Microbiota-Targeted Chitooligosaccharides Intervention Restores Glucose Homeostasis After Islet Cell Transplantation in Rapamycin-Treated Mice.

Islet cell transplantation (ICT) is an effective treatment for diabetes mellitus, but postoperative islet function recovery and inflammation are closely linked to immunosuppressants. Using multi-omics and fecal microbiota transplantation (FMT) in human microbiota-associated (HMA) mice, this study explored rapamycin-induced gut dysbiosis and its impacts on islet function and inflammation post-ICT. ICT significantly altered the gut microbiota of type 2 diabetes mellitus (T2DM) patients, and FMT from these patients to antibiotic-treated mice recapitulated metabolic disorders in the mice. These di

A Cerebral Organoid Model of Familial Alzheimer's Disease Using Amyloid Precursor Protein Mutation, Val669Leu (APPSeoul).

Various animal and cellular Alzheimer's disease (AD) models harboring familial AD (fAD) mutations have been developed and widely used for AD research. In this study, we established an AD cerebral organoid (CO) model using a novel Val669Leu (APPSeoul) mutation in the APP gene. We generated a human embryonic stem cell (hESC) line overexpressing APPSeoul, referred to as the fAD-S hESC line. Using this line, we produced COs and confirmed robust AD-associated pathologies, including amyloid-β (Aβ) accumulation and tau phosphorylation. In addition, increased expression of β-secretase was observed in

Homogenous catalysis and heterogenous recycle: Cellulose-supported acetylacetonate ion pair catalyst for cycloaddition of carbon dioxide to epoxide.

Designing catalysts that operate homogeneously during reactions while allowing heterogeneous recovery remains a significant challenge in carbon dioxide utilization. We developed a bifunctional cellulose-supported organocatalyst composed of an acetylacetonate anion and a protonated organic base for the cycloaddition of CO₂ with epoxide (CCE) reactions. 3-Acetylacetonate group was regioselectively anchored to C-6 position on cellulose backbone, then converted to acetylacetonate carbanion ion pair catalyst. An optimal cellulose-supported ion pair catalyst containing acetylacetonate carbanion (aca

Hemiparetic koneramine NNP ligand comprising strategic hard and soft donors: transition metal complexes, CS2 activation and therapeutic potential.

A novel koneramine-derived tridentate NNP ligand has been designed and synthesized, and its coordination chemistry explored with hard and soft metal centres, including Zn(II), Pd(II), Ru(II), Cu(I), Cu(II), and Ni(II). The resulting complexes were characterized by spectroscopic, structural, and electrochemical methods, revealing the ligand's electronic flexibility and its ability to stabilize copper in multiple oxidation states (Cu(I)/Cu(II)) via selective donor interactions and adaptable coordination geometries. A ruthenium hydride complex, [Ru(NNP)(H)Cl(CO)], exhibits efficient hydride trans

Therapeutic Potential of Carbon Monoxide (CO) and Hydrogen Sulfide (H2S) in Hemolytic and Hemorrhagic Vascular Disorders-Interaction between the Heme Oxygenase and H2S-Producing Systems.

Over the past decades, substantial work has established that hemoglobin oxidation and heme release play a pivotal role in hemolytic/hemorrhagic disorders. Recent reports have shown that oxidized hemoglobins, globin-derived peptides, and heme trigger diverse biological responses, such as toll-like receptor 4 activation with inflammatory response, reprogramming of cellular metabolism, differentiation, stress, and even death. Here, we discuss these cellular responses with particular focus on their mechanisms that are linked to the pathological consequences of hemorrhage and hemolysis. In recent y

Interleukin 34 Serves as a Novel Molecular Adjuvant against Nocardia Seriolae Infection in Largemouth Bass (Micropterus Salmoides).

DNA vaccines have been widely employed in controlling viral and bacterial infections in mammals and teleost fish. Co-injection of molecular adjuvants, including chemokines, cytokines, and immune co-stimulatory molecules, is one of the potential strategies used to improve DNA vaccine efficacy. In mammals and teleost fish, interleukin-34 (IL-34) had been described as a multifunctional cytokine and its immunological role had been confirmed; however, the adjuvant capacity of IL-34 remains to be elucidated. In this study, IL-34 was identified in largemouth bass. A recombinant plasmid of IL-34 (pcIL

Prevention of Bacterial Contamination of a Silica Matrix Containing Entrapped β-Galactosidase through the Action of Covalently Bound Lysozymes.

β-galactosidase was successfully encapsulated within an amino-functionalised silica matrix using a "fish-in-net" approach and molecular imprinting technique followed by covalent binding of lysozyme via a glutaraldehyde-based method. Transmission electron microscopy (TEM), X-ray diffraction (XRD), scanning electron microscopy (SEM), and Fourier transform infrared (FTIR) spectroscopy were used to characterise the silica matrix hosting the two enzymes. Both encapsulated β-galactosidase and bound lysozyme exhibited high enzymatic activities and outstanding operational stability in model reactions.

Blast injury leads to chronic shifts in protein expression of distinct astrocyte populations.

Blast traumatic brain injury results in chronic pathology, especially for those receiving repetitive injuries. To evaluate cellular changes induced by these pressure waves, we studied post-mortem prefrontal cortex of military personnel with a history of multiple blast exposures and military controls with no battlefield experience. Chronic increases in IBA1 (microglia) and GFAP (glial fibrillary acidic protein; astrocyte) immunoreactivity occurred in injured brains and also confirmed that GFAP-expressing astrocytes altered predominantly at interface regions of the brain: around blood vessels, t

Effects of chlorpromazine on growth and development of Dictyostelium discoideum.

Chlorpromazine (5 x 10(-3) M) administered to Dictyostelium discoideum cells inhibited its growth and morphogenesis. Cells treated with chlorpromazine were found to have distorted morphology. At lower doses of chlorpromazine the development was delayed. Early developmental events such as cell streaming, cell aggregations, development of EDTA stable cell contacts, cAMP-chemotaxis etc, were inhibited. Chlorpromazine was also found to inhibit spore formation. Culturing D. discoideum cells on chlorpromazine agar, in supernatant taken from the chlorpromazine treated cells, or co-culturing of chlorp

Gene context drift identifies drug targets to mitigate cancer treatment resistance.

Cancer treatment often fails because combinations of different therapies evoke complex resistance mechanisms that are hard to predict. We introduce REsistance through COntext DRift (RECODR): a computational pipeline that combines co-expression graph networks of single-cell RNA sequencing profiles with a graph-embedding approach to measure changes in gene co-expression context during cancer treatment. RECODR is based on the idea that gene co-expression context, rather than expression level alone, reveals important information about treatment resistance. Analysis of tumors treated in preclinical

Atomically Dispersed Ni-N-C Catalysts for Electrochemical CO2 Reduction.

The atomic dispersion of nickel in Ni-N-C catalysts is key for the selective generation of carbon monoxide through the electrochemical carbon dioxide reduction reaction (CO2RR). Herein, the study reports a highly selective, atomically dispersed Ni1.0%-N-C catalyst with reduced Ni loading compared to previous reports. Extensive materials characterization fails to detect Ni crystalline phases, reveals the highest concentration of atomically dispersed Ni metal, and confirms the presence of the proposed Ni-Nx active site at this reduced loading. The catalyst shows excellent activity and selectivit

Co-upgrading of ethanol-assisted depolymerized lignin: A new biological lignin valorization approach for the production of protocatechuic acid and polyhydroxyalkanoic acid.

This study presents a promising biological co-upgrading of ethanol-assisted depolymerized lignin (EDL) into protocatechuic acid (PCA) and polyhydroxyalkanoic acid (PHA) without any separation process. A depolymerized alkali lignin containing various G-lignin-type monomers at a concentration of 77 mg/mL was used for co-upgrading. An engineered Pseudomonas putida KT2440 strain was constructed by knocking out the protocatechuate 3, 4-dioxygenase, expression of the formaldehyde utilization pathway, and the expression of aldehyde dehydrogenase to enhance the efficiency of the ethanol utilization pa

A 3D culture model of innervated human skeletal muscle enables studies of the adult neuromuscular junction.

Two-dimensional (2D) human skeletal muscle fiber cultures are ill-equipped to support the contractile properties of maturing muscle fibers. This limits their application to the study of adult human neuromuscular junction (NMJ) development, a process requiring maturation of muscle fibers in the presence of motor neuron endplates. Here we describe a three-dimensional (3D) co-culture method whereby human muscle progenitors mixed with human pluripotent stem cell-derived motor neurons self-organize to form functional NMJ connections. Functional connectivity between motor neuron endplates and muscle

Studies on Effects of Certain Quinones: II. Photosynthetic Incorporation of CO(2) by Chlorella.

The effects of various quinone herbicides and fungicides on the photosynthetic (14)CO(2) fixation and the incorporation of (14)C among the products of photosynthesis in Chlorella pyrenoidosa was investigated. Addition of 30 mum 2,3-dichloro-1,4-naphthoquinone (dichlone), 2-amino-3-chloro-1,4-naphthoquinone (06K-quinone), or 2,3,5,6-tetrachloro-1,4-benzoquinone (chloranil) inhibited CO(2) fixation, whereas 1,4-benzoquinone had no effect. Treatment with 3 mum or higher concentrations of dichlone, 06K-quinone or 1,4-benzoquinone also produced marked changes in the pattern of (14)C distribution. A

Cell models for studying myasthenia gravis.

Understanding the pathophysiology of Myasthenia Gravis (MG) and developing effective treatments requires using cell models that replicate key features of the disease, particularly those involved in the autoimmune response and neuromuscular dysfunction. This chapter reviews the various cell-based models used in MG research and those with potential for preclinical MG studies, including muscle cells and co-culture models to form neuromuscular junctions (NMJ). We discuss the strengths and limitations of these models, further outline methods for characterizing these, and provide an outlook on the f

Ex Vivo Model of Spontaneous Neuroretinal Degeneration for Evaluating Stem Cells' Paracrine Properties.

Ex vivo neuroretina cultures closely resemble in vivo conditions, retaining the complex neuroretina cells dynamics, connections, and functionality, under controlled conditions. Therefore, these models have allowed advancing in the knowledge of retinal physiology and pathobiology over the years. Furthermore, the ex vivo neuroretina models represent an adequate tool for evaluating stem cell therapies over neuroretinal degeneration processes. Here, we describe a physically separated co-culture of neuroretina explants with stem cells to evaluate the effect of stem cells paracrine properties on spon
