Functional and structural impact of pirfenidone on the alterations of cardiac disease and diabetes mellitus.

A synthetic compound, termed pirfenidone (PFD), is considered promising for the treatment of cardiac disease. It leads to beneficial effects in animal models of diabetes mellitus (DM); as well as in heart attack, atrial fibrillation, muscular dystrophy, and diabetic cardiomyopathy (DC). The latter is a result of alterations linked to metabolic syndrome as they promote cardiac hypertrophy, fibrosis and contractile dysfunction. Although reduced level of fibrosis and stiffness represent an essential step in the mechanism of PFD action, a wide range of functional effects might also contribute to t

Stabilizers of neuronal and mitochondrial calcium cycling as a strategy for developing a medicine for Alzheimer's disease.

For the last two decades, most efforts on new drug development to treat Alzheimer's disease have been focused to inhibit the synthesis of amyloid beta (Aβ), to prevent Aβ deposition, or to clear up Aβ plaques from the brain of Alzheimer's disease (AD) patients. Other pathogenic mechanisms such as the hyperphosphorylation of the microtubular tau protein (that forms neurofibrillary tangles) have also been addressed as, for instance, with inhibitors of the enzyme glycogen synthase-3 kinase beta (GSK3β). However, in spite of their proven efficacy in animal models of AD, all these compounds have so

The role of the calcium and the voltage clocks in sinoatrial node dysfunction.

Recent evidence indicates that the voltage clock (cyclic activation and deactivation of membrane ion channels) and Ca(2+) clocks (rhythmic spontaneous sarcoplasmic reticulum Ca(2+) release) jointly regulate sinoatrial node (SAN) automaticity. However, the relative importance of the voltage clock and Ca(2+) clock for pacemaking was not revealed in sick sinus syndrome. Previously, we mapped the intracellular calcium (Ca(i)) and membrane potentials of the normal intact SAN simultaneously using optical mapping in Langendorff-perfused canine right atrium. We demonstrated that the sinus rate increas

New concepts in atrial fibrillation: neural mechanisms and calcium dynamics.

Atrial fibrillation (AF) is a complex arrhythmia with multiple possible mechanisms. It requires a trigger for initiation and a favorable substrate for maintenance. Pulmonary vein myocardial sleeves have the potential to generate spontaneous activity, and this arrhythmogenic activity is surfaced by modulation of intracellular calcium dynamics. Direct autonomic nerve recordings in canine models show that simultaneous sympathovagal discharges are the most common triggers of paroxysmal atrial tachycardia and paroxysmal AF. Autonomic modulation as a potential therapeutic strategy has been targeted

Impact of Ca2+ flux inhibitors on acrosome reaction of hamster spermatozoa.

Ca(2+) plays a prominent role in the regulation of critical functions of spermatozoa, such as capacitation, acrosome reaction (AR), and fertilization. While there is consensus that Ca(2+) is essential, researchers have reported conflicting results as to what happens to Ca(2+) flux across the sperm membrane during capacitation and AR. The purpose of the present study was to further delineate the function of Ca(2+) channels and their role in sperm capacitation and AR. Epididymides were obtained from healthy adult male hamsters. Spermatozoa were washed with modified Tyrode medium supplemented wit

Contributions of extracellular and intracellular Ca2+ to regulation of sperm motility: Release of intracellular stores can hyperactivate CatSper1 and CatSper2 null sperm.

In order to fertilize, mammalian sperm must hyperactivate. Hyperactivation is triggered by increased flagellar Ca(2+), which switches flagellar beating from a symmetrical to an asymmetrical pattern by increasing bending to one side. Thimerosal, which releases Ca(2+) from internal stores, induced hyperactivation in mouse sperm within seconds, even when extracellular Ca(2+) was buffered with BAPTA to approximately 30 nM. In sperm from CatSper1 or CatSper2 null mice, which lack functional flagellar alkaline-activated calcium currents, 50 microM thimerosal raised the flagellar bend amplitudes from

Calcium regulation and muscle disease.

Changes in intracellular Ca2+-concentration play an important role in the excitation-contraction-relaxation cycle of skeletal muscle. In this review we describe various inheritable muscle diseases to highlight the role of Ca2+-regulatory mechanisms. Upon excitation the ryanodine receptor releases Ca2+ in the cytosol. During and after contraction the sarcoplasmic reticulum (SR) Ca2+ATPase (SERCA) pumps Ca2+ back in the SR resulting in relaxation. An abnormal change in the intracellular Ca2+-concentration results in defective muscle contraction and/or relaxation, which is the cause of various mu

Management of disturbed calcium metabolism in uraemic patients: 1. Use of vitamin D metabolites.

Chronic renal failure is characterized by diminished synthesis of, and resistance to, the active vitamin D metabolite 1,25-dihydroxy-vitamin D3 (1,25(OH)2D3, calcitriol). Calcitriol results from the biotransformation of the precursor 25-hydroxy-vitamin D3 (25(OH)D3) to 1,25(OH)2D3. 25(OH)D3 is synthesized in the liver, and 1alpha-hydroxylase, the rate-limiting enzyme for its biotransformation into the most active metabolite, 1,25(OH)2D3, is located in the kidney. The regulation of 1alpha-hydroxylase in renal failure is not well known. Recent work indicates that, in contrast to previous opinion

[Severe neonatal hyperparathyroidism in a family with familial hypocalciuric hypercalcemia].

Familial hypocalciuric hypocalcaemia (FHH) is a rare disorder, inherited in an autosomal dominant manner. It has earlier been believed that neonatal severe hyperparathyroidism (NSHPT) is the homozygous form of FHH, but in this case story we show that it is not always like that. We describe a girl who presents with a calcium metabolic disorder from birth. Genetic examination of the girl and her family shows a single abnormal allele in the calcium ion sensitive receptor. We discuss why some heterozygotic inactivating calcium receptor mutations cause NSHPT, while the majority of other mutations o

Effects of the removal of extracellular Ca2+ on [Ca2+]i responses to FCCP and acetate in carotid body glomus cells of adult rabbits.

The effects of the removal of extracellular Ca2+ on the responses of cytosolic concentrations of Ca2+ ([Ca2+]i) to acidic stimuli, a protonophore carbonyl cyanide p-trifluoromethoxyphenylhydrazone (FCCP) and an organic acid acetate, were examined in clusters of cultured carotid body glomus cells of adult rabbits using fura-2 microfluorometry. Application of FCCP (1 microM) induced an increase in [Ca2+]i (mean +/- S.E.M., 108 +/- 14%). After withdrawal of the protonophore the increased [Ca2+]i returned slowly to a resting level. The [Ca2+]i response was attenuated by an inorganic Ca2+ channel a

Releasable pools and the kinetics of exocytosis in adrenal chromaffin cells.

Using high resolution capacitance (Cm) measurement techniques, we mapped the kinetics of exocytosis evoked by brief (5-100 ms) depolarizations that activate voltage-dependent Ca2+ channels in rat adrenal chromaffin cells. After correcting the Cm signal for contributions from Na+ channel-gating charge movements, the initial kinetics of exocytosis were consistent with depolarization-triggered release occurring initially from an immediately releasable pool of only approximately 17 secretory vesicles. In contrast, repetitive application of longer depolarizations evoked release from a distinct, lar

Electrical resonance of isolated hair cells does not account for acoustic tuning in the free-standing region of the alligator lizard's cochlea.

The cochlea of the alligator lizard is divided into two morphologically and physiologically distinct regions. In the "tectorial region," hair bundles of hair cells are draped by a tectorial membrane, whereas in the "free-standing region," hair bundles are said to be free-standing because there are no overlying tectorial structures. The acoustic tuning of the free-standing region depends at least in part on mechanical resonances of the hair bundles. In the turtle cochlea, in contrast, acoustic tuning depends in large part upon the electrical properties of the hair cells. We have investigated th

[Ca2+ channels and the abnormal electrical activity of demyelinated nerve].

Peripheral nerve demyelination was produced in adult rats by placing loosely-constrictive ligatures around the common sciatic nerve. The postoperative behavior of these rats indicated that hyperalgesia, allodynia and possible spontaneous pain were produced. In the meantime, abnormal spontaneous afferent activities (ectopic firings)originating from the demyelinated region were recorded. Evidence showed that the application of Ca2+ and Ca2+ channel blockers modulated the abnormal activity of the injured nerve. Ca2+ facilitation was dependent on its concentration (in the range of 10-20 mol/L), wh

Effect of calcium inhibitors and calcium mobilizers on doxorubicin accumulation in rat myocardial tissue.

The effect of four calcium-modulating agents on myocardial doxorubicin (DXR) accumulation was studied: the calcium-inhibitors verapamil (0.1 microM), trifluoperazine (5 microM) and flunarizine (0.05 microM) and the calcium-mobilizer taurine (50 mM). Myocardial cells of isolated Langendorff rat hearts receiving the drugs showed a significantly enhanced 14C-DXR uptake. These observations are in contrast with the hypothesis that DXR accumulation processes may be related to a decrease in calcium influx and suggest that a combined therapy with DXR and calcium-modulating agents might enhance the dru

[Possibilities in the future application of calcium antagonists. Is the prevention of blood vessel calcinosis possible?].

One of the most important effects of the new pharmacological family of Ca-antagonistic drugs is the prevention of deleterious myocardial Ca overload which is the decisive etiological factor in the production of cardiac necroses, as for instance b-adrenergic overstimulation. However, at an advanced age, also in the human arterial walls, cytotoxic degrees of Ca overload are reached, which probably play an important role in the pathogenesis of arteriosclerotic lesions. It is interesting that severe diabetics and heavy smokers exhibit a much faster progression of age-dependent arterial calcinosis

Involvement of TRPV2 and SOCE in calcium influx disorder in DMD primary human myotubes with a specific contribution of α1-syntrophin and PLC/PKC in SOCE regulation.

Calcium homeostasis is critical for several vital functions in excitable and nonexcitable cells and has been shown to be impaired in many pathologies including Duchenne muscular dystrophy (DMD). Various studies using murine models showed the implication of calcium entry in the dystrophic phenotype. However, alteration of store-operated calcium entry (SOCE) and transient receptor potential vanilloid 2 (TRPV2)-dependant cation entry has not been investigated yet in human skeletal muscle cells. We pharmacologically characterized basal and store-operated cation entries in primary cultures of myotu

Bovine CACNA1A gene and comparative analysis of the CAG repeats associated to human spinocerebellar ataxia type-6.

A small expansion of a CAG repeat domain in exon 47 of the human CACNA1A gene, which codes for the pore-forming alpha1A subunit of P/Q-type Ca2+ channels, causes spinocerebellar ataxia type-6. Only the human alpha1A protein has been demonstrated to contain the poly(Q) tract, although this locus has also recently been detected in ape genomes. To our knowledge, no further information has been published on other mammal species. Here, we have cloned the full-length alpha1A subunit in a non-primate species, the cow. The results have made it possible to explore the exon organization of the bovine CA

K+ Channels in Membrane Potential Dynamics and Migration of Neutrophil Granulocytes.

Neutrophil granulocytes exhibit dynamic changes in membrane potential upon stimulation with the complement component C5a. It is not well-defined which ion channels are responsible for membrane potential dynamics in neutrophils, although ion channel signaling has been linked to neutrophil behavior. Here, we investigated the contribution of K+ channels that are important players in different immune cells: KCa3.1, Kir2.1, and KV1.3. We studied their roles in membrane potential regulation, chemotaxis, and LTB4 secretion using live-cell imaging, analytical methods, and mathematical modeling. We ide
