Advancements in intratumoral therapies for liver tumors.

Liver tumors, including hepatocellular carcinoma (HCC), intrahepatic cholangiocarcinoma (iCCA), and liver-dominant metastases, remain associated with high mortality despite advances in systemic therapy. Intratumoral therapies have emerged as a promising strategy to achieve high local drug concentrations, modulate the tumor microenvironment, and enhance systemic anti-tumor immunity while limiting systemic toxicity. Although intratumoral approaches have demonstrated clinical success in melanoma, their translation to liver malignancies presents unique biological, immunologic, and technical challe

Folate receptor-α targeted therapies in ovarian cancer: recent advances and emerging therapeutic strategies.

Epithelial ovarian cancer (EOC) remains a biologically heterogeneous and therapeutically difficult malignancy because most advanced-stage tumors initially respond to platinum-taxane chemotherapy but later recur with progressively reduced platinum sensitivity. Folate receptor-alpha (FRα/FOLR1) has become a clinically relevant precision-oncology target because it is enriched in high-grade serous ovarian carcinoma, is relatively shielded from circulating drugs in many polarized normal epithelia, and undergoes receptor-mediated internalization after ligand or antibody binding. This review critical

Quantitative Assessment of Additive Effects of Mirabegron and Solifenacin in Overactive Bladder: Re-analysis of SYNERGY Study.

OBJECTIVE: To quantitatively determine whether the clinical benefit of mirabegron plus solifenacin for overactive bladder (OAB) reflects pharmacological synergy or independent drug action. METHODS: We conducted a post hoc quantitative reanalysis of the SYNERGY study comparing solifenacin monotherapy, mirabegron monotherapy, and their combinations (S5+M25, S5+M50). A copula-based independent drug action (IDA) framework with a Gaussian copula was applied to model correlated response propensity between monotherapies. Co-primary endpoints were zero urgency urinary incontinence (Zero UI) and norma

Daily patterns of opioid and stimulant use and associated risk-behavior outcomes: A cohort study.

BACKGROUND: People who use opioid and stimulant drugs have high overdose rates. Little is known about day-to-day variation in patterns of use of these drugs and their association with other drug use behaviors. METHODS: Participants with past-two-week opioid or stimulant use completed daily cellphone surveys over a two-week period. We classified each day's opioid-stimulant combination into five mutually exclusive groups. We estimated associations of each group with prevalence of 7 same-day outcomes -alcohol, cigarette, THC, and tranquilizer use, injecting drugs, using xylazine, and using drugs

Dual-targeted protein-coated PLGA nanoparticles for pancreatic cancer therapy: a novel approach using esculetin and curcumin.

The study aimed to investigate the effects of dual-targeted poly-lactic-co-glycolic acid (PLGA) nanoparticles (NP) containing esculetin (ESC) and curcumin (CURC), coated with membrane and cytosolic proteins, isolated from PID-PC (pancreatic islet-derived precursor cells) and MIA PaCa-2 cells, on pancreatic cancer. MIA PaCa-2 cells' viability treated with different combinations of the above-mentioned molecules and proteins was investigated in vitro. The sizes of all formed nanoparticles were determined by the "dynamic light scattering" method. The drug release profile of drug-containing PLGA-NP

P329G-engager: a universal mix & match antibody-based adaptor platform for cancer immunotherapy.

Targeting various combinations of tumor antigens and immune cell receptors is of increasing importance in antibody-based cancer immunotherapy. Here, we present a novel modular P329G-engager platform that enables rapid combination of primary tumor-targeting and secondary immune effector antibodies. The platform utilizes two antibodies, each selected from: 1) a set of tumor-targeting adaptor antibodies, bearing P329G mutations in the Fc region, and 2) a set of P329G-targeting (bispecific) cell engagers, including innate and T cell engagers, costimulators and immunocytokines. Specifically, upon d

D1 dopamine / mu opioid receptor interactions in operant conditioning assays of pain-depressed responding and drug-induced rate suppression, and a conditioned place preference procedure: assessment of therapeutic index in male Sprague Dawley rats.

RATIONALE AND OBJECTIVES: In vivo receptor interactions vary as a function of behavioral endpoint, with key differences between reflexive and non-reflexive measures that assess the motivational aspects of pain and pain relief. There have been no assessments of D1 dopamine agonist / mu opioid receptor (MOR) agonist interactions in non-reflexive behavioral measures of pain. We examined the hypothesis that D1/MOR mixtures show enhanced effectiveness in blocking pain depressed behaviors while showing decreased side effects such as sedation and drug reward. METHODS: SKF82958 and methadone were use

Glucagon-like peptide 1 agonists are potentially useful drugs for treating metabolic dysfunction-associated steatotic liver disease.

In this editorial, we comment on Yin et al's recently published Letter to the editor. In particular, we focus on the potential use of glucagon-like peptide 1 receptor agonists (GLP-1RAs) alone, but even more so in combination therapy, as one of the most promising therapies in metabolic dysfunction-associated steatotic liver disease (MASLD), the new definition of an old condition, non-alcoholic fatty liver disease, which aims to better define the spectrum of steatotic pathology. It is well known that GLP-1RAs, having shown outstanding performance in fat loss, weight loss, and improvement of ins

Belantamab mafodotin, pomalidomide and dexamethasone in refractory multiple myeloma: a phase 1/2 trial.

Due to evolving treatment standards for newly diagnosed multiple myeloma, many patients will be triple-class exposed after initial relapses and have poor survival. Novel therapies and combinations are therefore required to improve outcomes. B cell maturation antigen (BCMA)-targeted biologics have emerged as an important new area of therapeutics for relapsed multiple myeloma. The two-part ALGONQUIN trial evaluated various doses and schedules of the anti-BCMA antibody-drug conjugate belantamab mafodotin plus pomalidomide and dexamethasone for patients who are lenalidomide refractory and proteoso

Efficacy of aspergillomarasmine A/meropenem combinations with and without avibactam against bacterial strains producing multiple β-lactamases.

The effectiveness of β-lactam antibiotics is increasingly threatened by resistant bacteria that harbor hydrolytic β-lactamase enzymes. Depending on the class of β-lactamase present, β-lactam hydrolysis can occur through one of two general molecular mechanisms. Metallo-β-lactamases (MBLs) require active site Zn2+ ions, whereas serine-β-lactamases (SBLs) deploy a catalytic serine residue. The result in both cases is drug inactivation via the opening of the β-lactam warhead of the antibiotic. MBLs confer resistance to most β-lactams and are non-susceptible to SBL inhibitors, including recently ap

In vitro, in vivo metabolism and quantification of the novel synthetic opioid N-piperidinyl etonitazene (etonitazepipne).

OBJECTIVES: N-piperidinyl etonitazene (etonitazepipne) is a newly synthesized opioid related to the 2-benzylbenzimidazole analog class. Etonitazepipne has been formally notified and placed under intensive monitoring in Europe in January 2022. Nitazenes have high affinity at µ-opioid receptor (MOR). Etonitazepipne, specifically shows a EC50 of 2.49 nM, suggesting about 50 times higher potency combined with higher efficacy compared to morphine. Antinociceptive potency l ('hot plate test' with rats) was 192-fold greater than that of morphine. METHODS: Here we report on a post-mortem case involvi

Cutaneous Atypical Mycobacterial Infections: A Brief Review.

Nontuberculous mycobacterial (NTM) infections are increasingly recognized, particularly in tropical regions and are often found in immunocompetent individuals. These infections are emerging as significant health concerns, especially pulmonary NTM, which is reported more frequently and is known to be associated with hospital environments. While pulmonary NTM infections are on the rise, partly due to drug resistance and possible patient-to-patient transmission, there is no current evidence indicating an increase in cutaneous NTM infections. The clinical manifestations of NTM infections, except f

Prdx5 in the Regulation of Tuberous Sclerosis Complex Mutation-Induced Signaling Mechanisms.

(1) Background: Tuberous sclerosis complex (TSC) mutations directly affect mTORC activity and, as a result, protein synthesis. In several cancer types, TSC mutation is part of the driver mutation panel. TSC mutations have been associated with mitochondrial dysfunction, tolerance to reactive oxygen species due to increased thioredoxin reductase (TrxR) enzyme activity, tolerance to endoplasmic reticulum (ER) stress, and apoptosis. The FDA-approved drug rapamycin is frequently used in clinical applications to inhibit protein synthesis in cancers. Recently, TrxR inhibitor auranofin has also been i

Secondary Acute Myeloid Leukemia in Myelodysplastic Syndrome Patients Aged Over 60 Years.

In myelodysplastic syndrome (MDS), neoplastic cells originate in hematopoietic stem cells of the bone marrow, causing dysplasia in multiple cell lines. This may ultimately lead to cytopenia and anemia. MDS generally occurs in patients aged over 60 years, and if left unchecked, it can lead to secondary acute myeloid leukemia (AML), which has a worse prognosis than de novo AML. Hence, it is important to find methods to treat and manage MDS and prevent secondary AML. This review tries to point out the best methods to find out the best possible treatment for MDS, which can lead to its remission or

Functional Testing to Characterize and Stratify PI3K Inhibitor Responses in Chronic Lymphocytic Leukemia.

PURPOSE: PI3K inhibitors (PI3Ki) are approved for relapsed chronic lymphocytic leukemia (CLL). Although patients may show an initial response to these therapies, development of treatment intolerance or resistance remain clinical challenges. To overcome these, prediction of individual treatment responses based on actionable biomarkers is needed. Here, we characterized the activity and cellular effects of 10 PI3Ki and investigated whether functional analyses can identify treatment vulnerabilities in PI3Ki-refractory/intolerant CLL and stratify responders to PI3Ki. EXPERIMENTAL DESIGN: Periphera

Personalized drug testing in human pheochromocytoma/paraganglioma primary cultures.

Aggressive pheochromocytomas and paragangliomas (PPGLs) are difficult to treat, and molecular targeting is being increasingly considered, but with variable results. This study investigates established and novel molecular-targeted drugs and chemotherapeutic agents for the treatment of PPGLs in human primary cultures and murine cell line spheroids. In PPGLs from 33 patients, including 7 metastatic PPGLs, we identified germline or somatic driver mutations in 79% of cases, allowing us to assess potential differences in drug responsivity between pseudohypoxia-associated cluster 1-related (n = 10) a

ROADMAPS: An Online Database of Response Data, Dosing Regimens, and Toxicities of Approved Oncology Drugs as Single Agents to Guide Preclinical In Vivo Studies.

UNLABELLED: Preclinical studies provide valuable data in the early development of novel drugs for patients with cancer. Many cancer treatment regimens now utilize multiple agents with different targets to delay the emergence of drug-resistant tumor cells, and experimental agents are often evaluated in combination with FDA-approved drugs. The Biological Testing Branch (BTB) of the U.S. NCI has evaluated more than 70 FDA-approved oncology drugs to date in human xenograft models. Here, we report the first release of a publicly available, downloadable spreadsheet, ROADMAPS (Responses to Oncology A
