Influence of carboxymethyl chitosan chain length on its interaction with vitamin C and curcumin: molecular docking and molecular dynamic approach.

CONTEXT: The molecular understanding of the interaction between carboxymethyl chitosan (CMCs) with the encapsulated drug (vitamin C and curcumin) has not been clearly understood yet. This study seeks to ascertain how the polymer length affects the molecular weight of CMCs as a matrix by using molecular dynamics and molecular docking. We have used pentamer (N-CMCs-5), decamer (N-CMCs-10), and pentadecamer(N-CMCs-15). Molecular docking and dynamic simulations showcase diverse interaction strengths, revealing medium-to-weak hydrogen bonds and hydrophobic interactions within the N-CMCs…vitamin C a

New Approach for Targeting Small-Molecule Candidates for Intrinsically Disordered Proteins.

Intrinsically disordered proteins (IDPs), such as the Alzheimer's-associated tau protein, pose challenges for conventional drug discovery. This study applied the Informational Spectrum Method for Small Molecules (ISM-SM), a computational technique utilizing electron-ion interaction potentials (EIIPs), to identify potential tau modulators. Characteristic interaction frequencies derived from known ligands and conserved mammalian tau sequences were used to screen DrugBank and the COCONUT natural product database. The screening identified approved drugs previously reported to indirectly influence

Identification of medication-microbiome interactions that affect gut infection.

Most people in the USA manage their health by taking at least one prescription drug, and drugs classified as non-antibiotics can adversely affect the gut microbiome and disrupt intestinal homeostasis1,2. Here we identify medications that are associated with an increased risk of gastrointestinal infections across a population cohort of more than one million individuals monitored over 15 years. Notably, the cardiac glycoside digoxin and other drugs identified in this epidemiological study are sufficient to alter the composition of the microbiome and the risk of infection with Salmonella enterica

LncRNA H19 overexpression induces bortezomib resistance in multiple myeloma by targeting MCL-1 via miR-29b-3p.

Radiotherapy, chemotherapy, autologous/allogeneic stem cell transplantation, and targeted drug therapy are currently available therapeutic options for multiple myeloma (MM), but the clinical outcome remains unsatisfactory owing to frequent occurrence of drug resistance. Anti apoptosis is one of the main mechanisms to mediate drug resistance. Studies have shown that MCL-1 plays a key role in the growth of cancer cells "escaping" drug attacks, but the underlying mechanism remains unclear. Our previous study demonstrated that lncRNA H19 was highly expressed in the serum of MM patients. Bioinforma

Peptides and Drug Delivery.

Peptides have been used as drugs to treat various health conditions, and they are also being developed as diagnostic agents. Due to their receptor selectivity, peptides have recently been utilized for drug delivery to target drug molecules to specific types of cells (i.e. cancer cells, immune cells) to lower the side effects of the drugs. In this case, the drug is conjugated to the carrier peptide for directing the drug to the target cells (e.g. cancer cells) with higher expression of a specific receptor that recognizes the carrier peptide. As a result, the drug is directed to the target disea

Methods for quantifying intrafollicular drug delivery: a critical appraisal.

IMPORTANCE OF THE FIELD: In recent years there has been increasing awareness that the hair follicles and their associated pilosebaceous structures may act as significant permeation pathways and/or reservoirs for topically applied drugs. This has implications in terms of dermatological therapy for acne, hirsutism, alopecias or certain skin cancers as well as systemic drug delivery. As the processes modulating follicular drug penetration are poorly understood at present, there is an emergent need for methodologies that can quantify follicular drug penetration and deposition. So far, a review art

Quantum mechanics-guided parameterization enhances molecular dynamics accuracy for flavonoid-membrane biophysical properties.

Understanding how small molecule parameterization influences the outcomes of molecular dynamics (MD) simulations is crucial for accurately modeling drug-membrane interactions. This study examines the impact of refined force-field parameterization on the ability of MD simulations to reproduce in vitro experimental biophysical outcomes for three flavonoids (baicalein, chrysin, and luteolin) interacting with model lipid membranes composed of dioleoylphosphocholine and dipalmitoylphosphocholine. We compared an analogy-based approach using CHARMM General Force Field (CGenFF ver. 4.0) with a quantum

Amantadine-induced reorganization of model SARS-CoV-2 lipid envelopes.

Amantadine attracted renewed interest during the COVID-19 pandemic because of its known antiviral activity against influenza A. In this work, we investigated how amantadine affects a simplified model of the SARS-CoV-2 lipid envelope composed of DOPC : DMPS : PI (50 : 35 : 15), as well as monolayers of the individual lipids: DOPC, DMPS and PI. Langmuir experiments showed that the effect of the drug depends on the lipid type. In DOPC and PI monolayers, amantadine increased the area per molecule, suggesting its incorporation into more fluid layers. In contrast, in more compact DMPS monolayers, st

Mechanochemical Activation of Olanzapine in Mixed Solid Dispersions: Impact of Excipients on Release and Permeation Rates.

Background: The key parameters determining the bioavailability of an active pharmaceutical ingredient are its solubility/dissolution rate in physiological fluids and permeability across biological membranes. Highly accurate in vitro prediction of bioavailability is a key issue that typically arises during the development of new drug formulations and the improvement of existing ones. Objectives: The objective of the present work is to study the dissolution/release and permeation of olanzapine (OLZ) from two- and three-component solid dispersions (SDs) with sulfobutylether-β-cyclodextrin (SBE-β-

Molecular dynamics-driven optimization of triterpenoid, amidinium, and flavonoid inhibitors targeting dengue NS2B-NS3 protease.

Dengue virus remains one of the most significant worldwide threats and has yet to yield to effective antiviral therapies. The viral NS2B-NS3 protease is a critical factor for dengue virus infection and is a validated target of therapeutic interest. In this work, we present a computational drug-design and similarity-based prioritization approach to evaluate triterpenoid, amidinium, and flavonoid inhibitors with their structural analogues. We have examined the binding modes, conformational stability, and energetic profiles of the molecules through molecular docking and explicit-solvent molecular

Prediction of bile salt export pump inhibition using biomimetic chromatographic descriptors: Integrating membrane affinity and protein binding.

BACKGROUND AND PURPOSE: Bile salt export pump (BSEP) inhibition is a key mechanistic driver of cholestatic drug-induced liver injury, yet current prediction approaches rely mainly on in vitro potency and calculated physicochemical descriptors that do not adequately reflect local exposure at the canalicular membrane. This study aimed to evaluate whether biomimetic chromatographic descriptors could improve predictions of BSEP inhibition by incorporating mechanistically relevant information on membrane affinity and protein binding. EXPERIMENTAL APPROACH: A dataset of 62 structurally diverse comp

Mechanistic insights into P-glycoprotein-driven transport of anti-cancer PARP inhibitors.

P-glycoprotein (Pgp), a membrane-bound transporter protein, plays significant roles in the development of multidrug resistance (MDR) and in drug disposition. Understanding the interactions between Pgp and its substrates is therefore crucial for optimal drug design and ultimately clinical success. In this study, we conducted a comparative analysis of the four FDA-approved anti-cancer PARPi and their interactions with Pgp to elucidate the mechanisms driving their Pgp-mediated transport. Through an assessment of PARPi-induced effect on Pgp-coupled ATP hydrolysis, we found that olaparib and talazo
