Pressor responses from noncardioselective beta-blockers.

This study reviews more than fifty papers dealing with pressor responses from noncardioselective beta-blockers. It is concluded that the responses are usually mild. They occur mainly in situations of increased sympathetic activity. Therefore some patients seem to be at risk, eg, patients with unstable diabetes type 1, sportsmen performing isometric exercise, and heavy smokers. In orthostatic hypotension, noncardioselective beta-blockers may be beneficial. Cardiac output tends, however, to decrease, and patients with orthostatic hypotension will probably not benefit from this effect.

[The possibility of predicting the development of bronchial asthma].

A total of 235 patients with bronchial asthma and preasthma and their 85 relatives of the 1st degree of kinship were investigated. A detailed clinicofunctional and immunological investigation including a study of histocompatibility antigens, revealed the relationships between bronchial asthma development and antigens A2, B13, B21, B35 carriage. The authors presented a complex of methods (provocative acetylcholine tests, anaprilin test, determination of HLA antigens) permitting the prediction of this disease in patients with an aggravated hereditary and allergological history at the preclinical

Abundance in the embryonic brainstem of adrenaline during the absence of detectable tyrosine hydroxylase activity.

The activities of the catecholamine synthetic enzymes tyrosine hydroxylase and phenylethanolamine N-methyltransferase, and the concentrations of the catecholamines and their respective metabolites, have been measured in the dorsal and ventral halves of the brainstem at various ages in the embryonic and adult rat. The activity of phenylethanolamine N-methyltransferase in both parts of the brainstem at day 14 of gestation is at or greater than adult levels and thereafter displays relatively small variations during ontogeny. Tyrosine hydroxylase activity, in contrast, is undetectable at day 14 an

Depletion of adrenal catecholamines alters the amnestic effect of amygdala stimulation.

Amygdala stimulation produces a robust retrograde amnesia which appears to be related to peripheral hormonal responses to stress. This present study investigated the effects of electrical stimulation of the amygdala delivered immediately after inhibitory and active avoidance training to rats depleted of adrenal catecholamines or sham adrenal operated rats. Comparable findings were obtained with the two training tasks: amygdala stimulation impaired retention of sham adrenal operated rats but it enhanced retention of rats depleted of adrenal catecholamines. These findings suggest that adrenal ca

Neuropharmacological antagonism of the beta-carboline-induced "anxiety" response in rhesus monkeys.

A behavioral and physiological syndrome of stress-related responses was reported in primates following treatment with the benzodiazepine receptor antagonist, beta-carboline-3-carboxylic acid ethyl ester (beta-CCE). The behavioral and physiological effects of beta-CCE are similar to those observed during stressful or "anxiety"-related conditions characterized in rhesus monkeys under natural conditions. Pharmacological agents which are known to antagonize anxiety responses in other paradigms were tested for their ability to antagonize the actions of beta-CCE. Diazepam (1 mg/kg) completely blocke

Vasoactivity of adrenaline in regions covering the canine gastrointestinal tract.

Little is known about the action adrenaline has on vascular areas of functionally different parts of the stomach and intestine. Therefore the vasoactivity of adrenaline (1-1024 ng/kg i.v.) was studied by electromagnetic flow measurements in 8 vascular beds covering the gastrointestinal tract. Adrenaline induced 1) a vasodilation in the lesser curvature and antrum of the stomach; 2) a vasoconstriction in the greater curvature of the stomach; 3) a vasoconstriction followed by a vasodilation in the duodenum and jejunum; 4) a vasoconstriction in the ileum, caecum/colon and colon/rectum. A case of glycogen storage disease type I associated with an incomplete type of Fanconi syndrome; the protective role of lysosomal alpha 1,4-glucosidase and insulin deficiency against hypoglycemia.

A 59-year-old Japanese farmer with asymptomatic fasting hypoglycemia and with exaggerated hypoglycemic episodes induced by insulin and oral hypoglycemic agent administered for his postprandial hyperglycemia was diagnosed as glycogen storage disease type I. This diagnosis was suggested by unresponsiveness of blood glucose level to glucagon and confirmed by 13% normal level of glucose 6-phosphatase activity in liver biopsy specimen and by the presence of PAS positive amylase digestable glycogen in liver specimen. This case was associated with an incomplete type of Fanconi syndrome characterized

Adrenaline causes hypokalaemia in man by beta 2 adrenoceptor stimulation.

Increased circulating adrenaline produces systemic hypokalaemia by the stimulation of a membrane bound Na/K ATPase. In man, this enzyme appears to be linked to an adrenoceptor of the beta-subtype. We have further studied the subtype of beta-adrenoceptor involved by infusing adrenaline intravenously in normal volunteers after pretreatment with either a selective beta 2 antagonist (ICI 118551) or placebo. During the adrenaline infusion the serum potassium fell from 4.08 +/- 0.21 to 3.32 +/- 0.25 mmol/l (P less than 0.002). This adrenaline induced hypokalaemia was completely blocked by ICI 118551

Effects of verapamil on lipolysis due to dibutyryl cyclic AMP.

The effects of verapamil, a calcium antagonist, on lipolysis in isolated rat adipocytes were studied. Verapamil (100 microM) potentiated lipolysis due to dibutyryl cyclic AMP (Bt2cAMP) at submaximal concentrations, with or without extracellular Ca2+. Lipolysis due to 0.5 mM-Bt2cAMP was potentiated by verapamil in a dose-dependent manner up to 200 microM, whereas at concentrations higher than 100 microM the stimulatory effect of verapamil was progressively diminished with or without extracellular Ca2+. Verapamil showed only an inhibitory effect on lipolysis due to adrenaline (0.1-10 microM) or

Neural and humoral mechanisms involved in blood pressure variability.

In order to study blood pressure variability we have measured blood pressure, heart rate, plasma noradrenaline and adrenaline concentration and plasma renin activity during sleep and during the waking process in 20 subjects with borderline hypertension. The responses to a number of standardized tests were also measured. These were reading, mental arithmetic, change in posture, physical exercise and the response to intravenously injected phenylephrine and noradrenaline. The sensitivity of the baroreflex for heart rate control was also determined from the relationship between heart period (R-R i

Prior thiazide diuretic treatment increases adrenaline-induced hypokalaemia.

Hypokalaemia is a common finding in acutely ill patients and may be related in part to increased sympathoadrenal activity. In an investigation to determine whether pretreatment with thiazide diuretics causes the serum potassium to fall to an even lower level during increased sympathoadrenal activity, adrenaline was infused into healthy subjects after pretreatment for 7 days with either bendrofluazide (5 mg) or placebo. Thiazide diuretic pretreatment had no effect on the adrenaline-induced changes in blood pressure and heart rate. However, not only was the baseline serum potassium lower after b

Hormonal activation of type-L hormone-sensitive lipase measured in defatted heart powders.

Adrenaline, 3-isobutyl-1-methylxanthine (MIX) and dibutyryl cyclic AMP (Bt2 cyclic AMP) stimulated type-L hormone-sensitive lipase (HSL) activity when measurements were made on defatted rat heart powders. These lipolytic agents stimulated the activity of this enzyme in a time- and dose-dependent manner. This activation was reversible, because removal of adrenaline from the perfusate was accompanied by the return of type-L HSL activity to control levels. We have reported [Palmer, Caruso & Oscai (1981) Biochem. J. 198, 159-166] that perfusion with low levels of adrenaline, MIX or Bt2 cyclic AMP

Time course of release of catecholamine and other granular contents from perifused adrenal chromaffin cells of guinea-pig.

Experiments were carried out to investigate the time course of the release of catecholamine, dopamine-beta-hydroxylase (DBH) and adenine nucleotides from isolated chromaffin cells of guinea-pig adrenal gland. When the isolated chromaffin cells were incubated with medium containing acetylcholine (ACh) (0.1 mM), veratridine (0.1 mM) or scorpion (Leiurus quinquestriatus) venom, (10 micrograms/ml.), catecholamine was released into the medium. Catecholamine secretion induced by veratridine or scorpion venom was inhibited by tetrodotoxin (1 microM) but not by atropine (0.1 mM) plus hexamethonium (0.

Peripheral catecholamine release by alpha-latrotoxin in the rat.

Intraarterial injection of alpha-latrotoxin (alpha LTx), the major toxin of the black widow spider (Latrodectus mactans tredecimguttatus) venom, into carotid catheterized rats, induced prompt and marked rises in plasma adrenaline and noradrenaline concentrations, indicating that the toxin stimulates catecholamine release from both the adrenal medulla and sympathetic nerve terminals. Pretreatment with the ganglionic blocker chlorisondamine greatly reduced the plasma adrenaline response to alpha LTx but had almost no effect on the noradrenaline response, indicating that alpha LTx-stimulation of

Possible role of lipoprotein lipase in the regulation of endogenous triacylglycerols in the rat heart.

1. Adrenaline has a biphasic effect on intracellular lipoprotein lipase activity and on endogenous triacylglycerol content in heparin-perfused heart. 2. A high concentration of adrenaline (1 microM in the perfusion buffer) activated endogenous lipoprotein lipase activity and, at the same time, decreased intracellular triacylglycerol stores. 3. In contrast, a low concentration (0.005 microM-adrenaline) inhibited intracellular lipoprotein lipase activity. Under these conditions, cardiac triacylglycerol content was elevated above control values. 4. Perfusing the heart with high and low concentrat

Cardiovascular, hormonal and metabolic responses to graded exercise in juvenile diabetics with and without autonomic neuropathy.

Thirteen juvenile diabetics were studied in order to determine if decreased beat-to-beat variation during deep respiration, indicating abnormal autonomic nerve function, imply that cardiovascular, hormonal and metabolic responses are impaired. Patients with decreased beat-to-beat variation had to be more heavily stressed during exercise to reach a certain heart rate or catecholamine level. The relation between other metabolic and hormonal response is discussed.

[Different cardiovascular and hormonal consequences of A-V fistula in uremic patients (author's transl)].

The Authors studied 11 uremic patients for whom it was necessary to make an arterio-venous (A-V) fistula for dialysis purposes. Before and after creation of the fistula, the following parameters were determined: arterial pressure (PA) heart rate (HR), peripheral vascular resistances (PVR), renin, adrenalin and nor-adrenalin blood levels. The patients were divided in two groups according to the increase of the systolic output (SO) and o the cardiac index (CI) after the fistula (1st group of 5 patients: less than 30% increase of these values as compared to those before the fistula; 2nd group: mo

Changes in the adenohypophysis and genital tract of cryptorchid rats: some morphological and pharmacological aspects.

It has been confirmed that cryptorchidism may affect the control mechanism of gonadotrophin secretion. This has been based on morphological aspects of the adenohypophysis of cryptorchid rats which have shown an increase in basophilic and vacuolated basophilic cells, an increase known to be related to the production of gonadotrophin. The morphological alterations observed in the testes, prostates, seminal vesicles and ductus deferentes of these cryptorchid animals, the quiescence and the responses of their seminal vesicles and ductus deferentes to sympathomimetic and parasympathomimetic drugs i
