Curcumin as a natural therapeutic agent for osteoarthritis: molecular mechanisms, preclinical evidence, clinical applications, and future perspectives.

Osteoarthritis is a prevalent and progressive musculoskeletal disorder characterized by a complex interplay of mechanical stress, low-grade inflammation, oxidative stress, extracellular matrix degradation, chondrocyte dysfunction, and subchondral bone remodeling. Current pharmacological and surgical approaches primarily provide symptomatic relief and have limited capacity to modify the underlying structural progression of the disease. Curcumin, a major bioactive polyphenol derived from Curcuma longa, has attracted considerable interest as a potential therapeutic candidate because of its abilit

Exosome-mediated modulation of osteogenesis under inflammatory conditions: a systematic review of in vitro studies on human periodontal ligament stem cells.

Exosomes have emerged as promising cell-free therapeutic agents for tissue regeneration. However, chronic inflammation, a hallmark of periodontal disease, may impair their osteogenic potential. This systematic review evaluated how inflammatory conditions influence the ability of exosomes to modulate osteogenesis in human periodontal ligament stem cells (hPDLSC) in vitro. Electronic databases and grey literature were searched up to February 2026, and study selection followed a two-phase screening process. Risk of bias was assessed using the Risk of Bias in Dental Preclinical Research Tool (RobD

Spatial transcriptomics identifies dysregulated programs across neural and non-neural tissues in spinal muscular atrophy.

Spinal muscular atrophy (SMA) is caused by insufficient levels of the survival motor neuron (SMN) protein and clinically manifests as profound weakness due to motor neuron degeneration. While recent evidence suggests it is a multisystem disorder, the pathological programs in neural and peripheral tissues are poorly understood. We applied spatial transcriptomics to cross-sections from the entire lumbar region of pre-symptomatic SMA and control mice to define early, tissue-wide transcriptional consequences of SMN deficiency. This approach enabled spatially preserved analysis of spinal cord, dors

Immune microenvironment of infantile hemangioma: cellular dynamics, molecular networks, and emerging immunomodulatory strategies.

Infantile hemangioma (IH), the most common vascular tumor of infancy, is characterized by a paradoxical natural history of rapid proliferation followed by spontaneous but often incomplete involution. Although dysregulated angiogenic signaling has long been considered central to IH pathobiology, an angiogenesis-only model is insufficient to explain its phase-dependent behavior, marked clinical heterogeneity, syndromic associations, or variable response to therapy. Emerging evidence instead supports the view of IH as a dynamically remodeled immune-vascular lesion driven by signaling crosstalk am

Differences in Reproductive Performance Between Shiqi and Dabao Pigeons: Insights from Multi-Omics Analysis.

Background/Objectives: Shiqi and Dabao pigeons are two Chinese meat-pigeon breeds with distinct breeding backgrounds and production-related phenotypes, but whether these differences extend to ovarian physiology and molecular profiles remains unclear. This study compared ovarian physiological traits and multi-omics profiles between the breeds. Methods: Seven-month-old reproductively mature females were studied. Flock-level laying and fertilization records were obtained from 456 breeding pairs per breed and recorded every 2 days. Serum reproductive hormones were measured in 10 females per breed.

Multi-trait and Gene-Based Analyses Identify Genetic Variants Associated with Spontaneous Coronary Artery Dissection.

BACKGROUND AND AIMS: Spontaneous coronary artery dissection (SCAD) is a non-atherosclerotic cause of acute myocardial infarction (MI) that predominantly affects young women. As an under-recognized cause of MI, large genome-wide association studies (GWAS) remain challenging. We aimed to leverage SCAD shared genetic basis with related vascular diseases to uncover genetically determined biological mechanisms. METHODS: Summary statistics for SCAD GWAS (1,917 cases, 9,293 controls) was harmonised with seven related vascular traits: fibromuscular dysplasia, intracranial aneurysm, cervical artery di

Cellular and Molecular Changes Underpinning the Formation of Intimal Hyperplasia in a Model of Static Human Saphenous Vein Ex Vivo Culture.

OBJECTIVE: Intimal hyperplasia (IH) significantly limits the long term patency of saphenous vein grafts following bypass surgery, with no widely accepted human models that fully capture its complex pathogenesis. Although animal models, primarily murine systems, have provided mechanistic insights into IH, limitations persist in translating these findings to human pathophysiology. This study aimed to evaluate the translational value of a static ex vivo culture model using human saphenous vein segments to study IH. METHODS: Human saphenous vein segments obtained from patients who underwent lower

Patient-Derived 3D-Bioprinted Models of Pancreatic Cancer: Toward Personalized Therapy and Overcoming Tumor Microenvironment Challenges.

Pancreatic ductal adenocarcinoma (PDAC) is one of the most aggressive and treatment-resistant solid tumors, mainly due to its complex tumor microenvironment (TME). Characterized by dense desmoplastic stroma, immune suppression, and metabolic rewiring, the TME impairs drug delivery and eventually leads to therapeutic failure. Conventional models, such as two-dimensional (2D) cultures and mouse models, fail to recapitulate the cellular and mechanical intricacies of PDAC, limiting their translational relevance. Three-dimensional (3D) models have emerged as promising tools to better simulate tumor

Platelet-derived growth factor signaling in porcine testicular tubular somatic cells.

Mechanisms governing somatic cell interactions in the testis are not well defined. The platelet-derived growth factor (PDGF) pathway mediates epithelial-mesenchymal interactions and is involved in testicular morphogenesis in rodents. However, its roles in the testis of higher mammals remain largely unknown. Here, we investigated how PDGF signaling inhibition affects immature (1-week-old) porcine testicular tubular somatic cells (TTSCs), including cell-cell communication and morphogenesis. From scRNA-seq data, we established the PDGF pathway signatures in crosstalk between testicular cells, ide

A phase I/IIa trial of PXS-5505, a novel pan-lysyl oxidase inhibitor, in advanced myelofibrosis.

Myelofibrosis is a progressive disease characterized by accumulation of extracellular matrix in the bone marrow (BM), which impairs normal hematopoiesis. While Janus kinase inhibitors reduce spleen volume and provide symptomatic improvement, they do not resolve BM fibrosis and may cause or exacerbate anemia and thrombocytopenia. An antifibrotic therapy capable of normalizing the BM microenvironment and function remains a significant gap in the current treatment landscape. Myelofibrosis is associated with elevated expression of lysyl oxidases, enzymes responsible for maturation of the most abun

Neutrophil extracellular trap inhibition revitalizes PDAC immunotherapy responsiveness via reduced fibrosis and TCF1+CD8+ progenitor T-cell expansion.

Pancreatic ductal adenocarcinoma (PDAC) is an aggressive disease with poor survival. The immunosuppressive tumor microenvironment (TME) drives resistance to therapy, including immunotherapy. This may be, in part, mediated by the formation of neutrophil extracellular traps (NETs), formed when neutrophils release their intracellular contents. NETs are elevated in PDAC and are associated with nearly every stage of tumor progression. We investigated the influence of NETs on the PDAC TME and immunotherapy response. An orthotopic PDAC model was utilized in C57BJ6 or PAD4-/- mice receiving one of the

Molecular insights in HIV-associated cardiac dysfunction.

BACKGROUND: Antiretroviral therapy (ART) in human immunodeficiency virus (HIV) markedly improved life expectancy in persons living with HIV (PLWH), but cardiac disease has emerged as the leading cause of death in this population. The prevalence of impaired diastolic function and heart failure with preserved ejection fraction (HFpEF) is higher among PLWH on ART. However, the specific role that HIV itself, and/or ART may have on the cardiomyocyte phenotype is unknown. We investigated modified molecular pathways in cardiomyocytes after exposure to serum from the same patients who were initially A

Filter-aided expansion proteomics for the spatial analysis of single cells and organelles in FFPE tissue samples.

Filter-aided expansion proteomics (FAXP) is a spatial proteomics approach designed for high-resolution analysis of formalin-fixed, paraffin-embedded (FFPE) tissues. Here we describe the integration of hydrogel-based tissue expansion with mass spectrometry, enabling isotropic expansion and robust protein retention while preserving spatial features. The FAXP workflow consists of several sequential steps, including tissue section dewaxing, in situ protein anchoring, hydrogel embedding, homogenization, staining, isotropic expansion, microdissection and filter-aided in-gel digestion to maximize pep

Tadalafil protects against renal fibrosis after ischemia-reperfusion injury by inhibiting Akt/NF-κB signaling pathway.

Renal fibrosis is a common feature of chronic kidney diseases, characterized by a complex and dynamic process involving the infiltration of inflammatory cells, abnormal formation and deposition of the extracellular matrix, and damage to the kidney's intrinsic cells. Renal fibrosis can lead to end-stage renal disease, but developing antifibrotic drugs remains a significant challenge. Phosphodiesterase 5 inhibitors have anti-apoptotic and antioxidant properties, making them promising candidates for treating ischemia-reperfusion injuries. However, no phosphodiesterase 5 inhibitors are currently a
