LPGAT1 controls MEGDEL syndrome by coupling phosphatidylglycerol remodeling with mitochondrial transport.

Phosphatidylglycerol (PG) is a mitochondrial phospholipid required for mitochondrial cristae structure and cardiolipin synthesis. PG must be remodeled to its mature form at the endoplasmic reticulum (ER) after mitochondrial biosynthesis to achieve its biological functions. Defective PG remodeling causes MEGDEL (non-alcohol fatty liver disease and 3-methylglutaconic aciduria with deafness, encephalopathy, and Leigh-like) syndrome through poorly defined mechanisms. Here, we identify LPGAT1, an acyltransferase that catalyzes PG remodeling, as a candidate gene for MEGDEL syndrome. We show that PG

Genetic Factors Associated with Response to Vitamin E Treatment in NAFLD.

Non-alcoholic fatty liver disease (NAFLD) is becoming the predominant liver disease worldwide, and vitamin E has been clinically shown to improve histological parameters in a subset of patients. In this narrative review, we investigate whether genetic factors may help to explain why some patients show histological improvements upon high-dose alpha-tocopherol (αT) treatment while others do not. In summary, we identified two factors that are associated with treatment response, including genetic variations in haptoglobin as well as fatty acid desaturase 1/2 (FADS1/FADS2). Other genetic variants s

Adlay (Coix lacryma-jobi L.) Polyphenol Improves Hepatic Glucose and Lipid Homeostasis through Regulating Intestinal Flora via AMPK Pathway.

SCOPE: Non-alcoholic fatty liver disease (NAFLD) is a type of metabolic syndrome characterized of abnormal lipid deposition in the liver. Adlay polyphenol (AP), an effective component extracted from Coix lacryma-jobi L., has been reported that it can be used as a dietary supplement to prevent NAFLD. In this study, the mechanism and action of AP on lipid metabolism and regulation of intestinal flora are investigated. METHODS AND RESULTS: AP significantly decreases the lipid accumulation in free fatty acid-treated HepG2 cells. Western blot results indicate that AP improves lipid metabolism via

Rapid Onset of Weight Gain and Liver Dysfunction Successfully Treated With Nutrition and Exercise.

Physical inactivity is one of the causes of most metabolic syndromes. The incidence of metabolic syndrome is expected to increase in the near future because of the reduced opportunities for exercise caused by COVID-19. Non-alcoholic fatty liver disease (NAFLD) is currently the most common cause of chronic liver disease. Changes in diet and lifestyle have led to a dramatic increase in the prevalence of NAFLD in the world. NAFLD is characterized by excessive triglyceride (TG) accumulation in the hepatocytes due to both increased inflow of free fatty acids and de novo hepatic lipogenesis. Thus fa

Liver Cirrhosis, Etiology and Clinical Characteristics Disparities Among Minority Population.

Liver cirrhosis (LC) is a common disease with varied primary causes and ethnic disparities. Clinical characteristics and outcomes of Arab Bedouin (AB) and Jewish patients with LC were retrospective collected and compared. We included 1048 patients, 95 (9%) Arab Bedouin and 953 (91%) Jewish patients. The incidence of cirrhosis was much lower among AB. Age at diagnosis was 47 ± 18 years among Bedouins compared to 61 ± 13 years (p < 0.001) among Jews. The most frequent causes of cirrhosis among Bedouin patients were fatty liver 21.1%, cryptogenic 20%, hepatitis B 17.9% and autoimmune hepatitis 15

Analysis of the intricate effects of polyunsaturated fatty acids and polyphenols on inflammatory pathways in health and disease.

Prevention and treatment of non-communicable diseases (NCDs), including cardiovascular disease, diabetes, obesity, cancer, Alzheimer's and Parkinson's disease, arthritis, non-alcoholic fatty liver disease and various infectious diseases; lately most notably COVID-19 have been in the front line of research worldwide. Although targeting different organs, these pathologies have common biochemical impairments - redox disparity and, prominently, dysregulation of the inflammatory pathways. Research data have shown that diet components like polyphenols, poly-unsaturated fatty acids (PUFAs), fibres as

Patterns of hepatocyte injury in man.

Three patterns of hepatocyte injury in man, direct, immunological, and cholestatic, are described. The characteristics of the direct pattern are predominantly mitochondrial damage, central (zone 3) necrosis, and, usually, fatty change. It can be subdivided into the alcohol type (also seen with obesity, in diabetes, as a reaction to perhexiline, in Wilson's disease, and in Indian childhood cirrhosis) and the Reye's syndrome type (also seen with tetracycline toxicity, fatty liver of pregnancy, and cytotoxic drugs). Reactive drug metabolites, metal poisoning, and anoxia are also associated with t

EXPERIMENTAL LIVER NECROSIS. V. THE FATS AND LIPOIDS.

1. Changes which occur in the fat content of the liver of dogs receiving haemotoxic serum bear no relation to the degree of necrosis produced by this serum. 2. An increase in water content of the tissue seldom occurs, but where present is due to the nitrogenous autolysis rather than to the deposition of fat. 3. The appearance of fat in the cell is not associated with a decomposition of the proteid component of the compound fats, but rather to a simple splitting off of the fatty radical. This is shown by the slight variations occurring in the percentage nitrogen of the fat-free substance. 4. Th

Investigation of the relationship between skin-emitted volatile fatty acids and skin surface acidity in healthy participants-a pilot study.

Volatile organic compounds (VOCs) emitted from human skin are of great interest in general in research fields including disease diagnostics and comprise various compound classes including acids, alcohols, ketones and aldehydes. The objective of this research is to investigate the volatile fatty acid (VFA) emission as recovered from healthy participant skin VOC samples and to characterise its association with skin surface acidity. VOC sampling was performed via headspace-solid phase microextraction with analysis via gas chromatography-mass spectrometry. Several VFAs were recovered from particip

Asymmetric heptamethine quinocyanines with large Stokes shifts for high-fidelity imaging of mitochondrial viscosity and NAFLD.

Mitochondrial viscosity is a key parameter reflecting the functional status of mitochondria, and its dysregulation is linked to various diseases. In this study, we designed and synthesized two asymmetric heptamethine quinocyanine dyes (QCy7-1 and QCy7-2). QCy7-1 exhibits viscosity-activated fluorescence (λem = 780 nm; Stokes shift = 239 nm) and a 20.8-fold brightness increase in glycerol versus PBS. Probe QCy7-1 enables real-time monitoring of mitochondrial viscosity changes induced by nystatin, monensin, lipopolysaccharide, and starvation, and in vivo imaging of non-alcoholic fatty liver dise

An Unexpected Cause of Severe Steatohepatitis in a Young Male.

A 24-year-old male with a body mass index (BMI) of 37.4 kg/m2 was referred to our hospital following the incidental discovery of abnormal liver echogenicity on a routine physical examination. The patient denied any history of alcohol consumption or illicit drug use and reported no family history of liver or metabolic diseases. Abdominal ultrasonography performed in December 2025 revealed heterogeneous liver echotexture with diffuse nodular hyperechoic areas, initially suggestive of nodular fatty infiltration or cirrhosis, and was accompanied by mild splenomegaly. Contrast-enhanced abdominal co

PKM2: A novel helmsman of lipid metabolism.

PKM2 is an isoform of pyruvate kinase, which has low catalytic activity and is abundantly present in rapidly proliferating cells. Numerous studies have demonstrated the important role of PKM2 in diseases such as cancer, non-alcoholic fatty liver disease, atherosclerosis and other abnormalities of lipid metabolism. In this paper, we review the roles played by PKM2 in different pathways of lipid metabolism, and suggest that PKM2 may be a potential therapeutic target in disorders of lipid metabolism. In addition, this review introduces common PKM2 intervention modalities, aiming to provide new po

The role of the nervous system in liver diseases.

The nervous system significantly participates in maintaining homeostasis, and modulating repair and regeneration processes in the liver. Moreover, the nervous system also plays an important role in the processes associated with the development and progression of liver disease, and can either potentiate or inhibit these processes. The aim of this review is to describe the mechanisms and pathways through which the nervous system influences the development and progression of liver diseases, such as alcohol-associated liver disease, nonalcoholic fatty liver disease, cholestatic liver disease, hepa

The role of ferroptosis in metabolic diseases.

The annual incidence of metabolic diseases such as diabetes, non-alcoholic fatty liver disease (NAFLD), osteoporosis, and atherosclerosis (AS) is increasing, resulting in a heavy burden on human health and the social economy. Ferroptosis is a novel form of programmed cell death driven by iron-dependent lipid peroxidation, which was discovered in recent years. Emerging evidence has suggested that ferroptosis contributes to the development of metabolic diseases. Here, we summarize the mechanisms and molecular signaling pathways involved in ferroptosis. Then we discuss the role of ferroptosis in

The role of hepatocyte nuclear factor 4α (HNF4α) in tumorigenesis.

Hepatocyte Nuclear Factor 4 Alpha (HNF4α) is a master transcription factor mainly expressed in the liver, kidney, intestine and endocrine pancreas. It regulates multiple target genes involved in embryonic development and metabolism. HNF4α-related diseases include non-alcoholic fatty liver disease (NAFLD), obesity, hypertension, hyperlipidemia, metabolic syndrome and diabetes mellitus. Recently, HNF4α has been emerging as a key player in a variety of cancers. In this review, we summarized the role and mechanism of HNF4α in different types of cancers, especially in liver and colorectal cancer, a

Friend or foe? The roles of inulin-type fructans.

Inulin-type fructans (ITFs) as functional fructans and soluble dietary fiber are a mixture of inulin, oligofructose and fructooligosaccharide with β configuration. They are modified by gut microbiota at the end of ileum, subsequently, improve digestive system, metabolic syndrome, immune system and inflammatory diseases, and prevent against infection and cancer. However, it has been reported that inadequate consumption of ITFs aggravates the development of non-alcoholic fatty liver disease, results in gastrointestinal symptoms, liver cancer and intestinal inflammation. Therefore, this review su

The influence of probiotics on bile acids in diseases and aging.

Recent evidence indicates the use of probiotics in the prevention and treatment of diseases. Probiotics are capable of changing the gut microbiota composition and bile acid synthesis to elicit health benefits such as cholesterol-lowering, weight reduction, and improving insulin sensitivity. The aging population is prone to develop diseases because of their decreased physiological and biological systems. Probiotics are one of the promising supplements that may potentially counteract these detrimental effects. This review will discuss the influence of probiotics on bile acids in different popula

Letter to the Editor: Comments on "Obesity associated with a novel mitochondrial tRNACys m.5802A>G mutation in a Chinese family".

In a recent article Wang et al. reported about a 9-year-old Chinese male with obesity, insulin resistance, acanthosis nigricans, hyperuricemia, and non-alcoholic fatty liver disease (NALFD) being attributed to the variant m.5802A>G in tRNA(Cys) (Bioscience Reports (2020) 40(1), BSR20192153). Pathogenicity of the variant was substantiated by documentation of perturbed stability and mobility of the tRNA(Cys). The interesting study has a number of shortcomings, which do not allow drawing conclusions as provided before they are solved. Obesity is multifactorial and many differential causes of mtDN
