Mesona chinensis Benth polysaccharides protect against oxidative stress and immunosuppression in cyclophosphamide-treated mice via MAPKs signal transduction pathways.

In this study, the immune responses of Mesona chinensis Benth polysaccharides (MP) in vitro and in vivo were investigated. Results showed that MP presented immunomodulatory activities on macrophages and T lymphocytes in vitro. Compared with the cyclophosphamide (Cy)-induced immunosuppressive mice, the body weights, spleen indexes (3.45 to 4.91) and thymus indexes (0.78 to 1.04) of the mice treated with MP were increased, and the peripheral blood levels were recovered. MP treatment also increased superoxide dismutase, glutathione peroxidase and catalase activities, and reduced malondialdehyde l

Overview of gene therapy in spinal muscular atrophy and Duchenne muscular dystrophy.

Both 5q-linked spinal muscular atrophy (SMA) and Duchenne muscular dystrophy (DMD) are fatal monogenic neuromuscular disorders caused by loss-of-function mutations. SMA is an autosomal recessive disorder affecting motor neurons that is typically caused by homozygous whole-gene deletions of SMN1. DMD is an X-linked recessive muscle disease most often due to exon deletions, but also duplications and smaller sized variants within the DMD gene. Gene replacement therapy offers the opportunity to correct the underlying genetic defect by the introduction of a functional gene. We review the transforma

Targeted deubiquitination rescues distinct trafficking-deficient ion channelopathies.

Impaired protein stability or trafficking underlies diverse ion channelopathies and represents an unexploited unifying principle for developing common treatments for otherwise dissimilar diseases. Ubiquitination limits ion channel surface density, but targeting this pathway for the purposes of basic study or therapy is challenging because of its prevalent role in proteostasis. We developed engineered deubiquitinases (enDUBs) that enable selective ubiquitin chain removal from target proteins to rescue the functional expression of disparate mutant ion channels that underlie long QT syndrome (LQT

Multi-functional MnO2-doped Fe3O4 nanoparticles as an artificial enzyme for the colorimetric detection of bacteria.

The authors describe a rapid enzyme-free colorimetric assay for food- and water-borne pathogens by using the multi-functional MnO2-doped magnetic ferrite nanoparticles (MCDs-MnO2 NPs). These nanoparticles not only can recognize, absorb, and separate the analyte from the matrix efficiently but also can directly catalyze the oxidation of TMB into a blue colored product without H2O2. In the presence of target, the nanoparticles preferentially bind to the target surface and the interaction between nanoparticles and TMB can be blocked, providing turn-off sensing of bacteria. By combining the superi

Identification and Characterization of Edible Cricket Peptides on Hypertensive and Glycemic In Vitro Inhibition and Their Anti-Inflammatory Activity on RAW 264.7 Macrophage Cells.

Recent studies continue to demonstrate the potential of edible insects as a protein base to obtain bioactive peptides applicable for functional food development. This study aimed at identifying antihypertensive, anti-glycemic, and anti-inflammatory peptides derived from the in vitro gastrointestinal digests of cricket protein hydrolysates. After sequential fractionation, the protein digest subfraction containing the lowest molecular weight (<0.5 kDa), hydrophobic (C18) and cationic peptides (IEX) was found responsible for the most bioactivity. The cationic peptide fraction significantly reduce

Nano-reduction of starch from underutilised millets: Effect on structural, thermal, morphological and nutraceutical properties.

Starch nanoparticles from pearl (Pe) and proso (Pr) millets were characterised for morphological, thermal, rheological and nutraceutical properties which are important parameters to be considered for predicting applicable domain of nanoparticles in food and other industrial applications. In the present study after using collision ball milling to achieve the nano-reduction, dynamic light scattering (DLS) revealed the average hydrodynamic particle diameter of 636 nm and 417 nm for nano-reduced pearl (PeN) and proso (PrN) millet starches. Further the nano-particles produced were having greater st

Reaction-Based Ratiometric and Colorimetric Chemosensor for Bioimaging of Biosulfite in Live Cells, Zebrafish, and Food Samples.

In this work, a reaction-based ratiometric and colorimetric sensor was designed and synthesized for probing bisulfite (HSO3-) by coupling coumarin (CM) with barbituric (BA) moiety. Further tests have shown that CM-BA has high selectivity and sensitivity for the recognition of HSO3-, which can be applied for the detection of HSO3- in environmental and biological systems very effectively. The fluorescence intensity ratios (F462/F568) exhibited an outstanding HSO3--dependent response with ultrafast response time (within 20 s) and a lower detection limit (105 nM). Meanwhile, the color of the CM-BA

Effects of Cornelian Cherry on Atherosclerosis and Its Risk Factors.

Functional food represents an important alternative management of atherosclerosis, its risk factors, and clinical complications. Atherosclerosis is characterized by microinflammation, formation of atheromatous lipoprotein-rich plaques, and protrombogenic status. Cornelian cherry (Cornus mas L., CC) contains polyphenols influencing all three components of atherosclerosis. Its high antioxidant potential, verified in experimental studies, exhibited a pronounced decrease of inflammatory markers. CC treatment demonstrated a favourable effect on lipid spectrum (comparable with statins), decrease of

Preparation and antioxidant properties of crayfish (Procambarus clarkii) By-products protein hydrolysates and ultra filtration fractions.

Protein isolate from crayfish by-products (CBPI) were hydrolyzed using Alcalase, neutrase, pancreatin and bromelain. Hydrolysis by Alcalase had more remarkable digesting efficiency on crayfish by-products protein than that by the other enzymes. Therefore, protein hydrolysate from Alcalase digestion (CBPHa) was selected to be fractionated by ultrafiltration according to molecular weight into three fractions F1 (MW <1kDa), F2 (MW 1-3kDa) and F3 (MW 3-10kDa). The amino acid determination revealed that CBPI had essential amino acid (EAA) close to that required for human protein synthesis. In vitro

Protective effects of lycopene on kainic acid-induced seizures.

Lycopene (LCP) is a carotenoid that protects against many diseases by alleviating oxidative stress. However, the effect of LCP on epileptic seizures has not been examined well in previous studies. In the current work, we employed kainic acid (KA) to induce experimental epileptic seizures in mice, and investigated the function of LCP during this process. We found that the onset and extent of KA-induced seizures were alleviated in LCP-pretreated mice. Nissl staining of hippocampus showed that the granule cell dispersion lesion induced by KA was improved by the LCP treatment. Additionally, we ana

Peracetic acid: Structural elucidation for applications in wastewater treatment.

Peracetic acid (PAA) is an oxidizer widely used for the sterilization of equipment in hospitals, pharmaceutical, cosmetic and food industries and also for water and wastewater disinfection. Even with its increasing applications, there have been no previous theoretical studies that explain the experimental results based on its molecular behavior. In this context, this work used calculations based on the density functional theory (DFT) combined with experimental results to elucidate the decomposition mechanisms of PAA for predicting its stability and the possible products generated from its deco

The role of mast cells in pediatric gastrointestinal disease.

Mast cells are granulocytes derived from CD34+ pluripotent progenitor cells that demonstrate plasticity in their development, leaving the bone marrow and differentiating in the tissue where they ultimately reside. They are best known for their role in the allergic response, but also play a prominent immunoregulatory role in other processes, including immune tolerance, the innate immune response, angiogenesis, wound healing and tissue remodeling. Mast cells are found throughout the gastrointestinal tract; their metabolic products influence and regulate intestinal epithelial and endothelial func

First Metatarsophalangeal Joint Space Area Decreases Within 1 Month After Implantation of a Polyvinyl Alcohol Hydrogel Implant: A Retrospective Radiographic Case Series.

In 2016, the U.S. Food and Drug Administration approved the first and only polyvinyl alcohol hydrogel implant for the treatment of hallux rigidus. The implant functions as a bumper to maintain first metatarsophalangeal joint space to prevent contact of the phalangeal base with the first metatarsal head. Short-term and intermediate outcomes with this implant have reported positive outcomes with no radiographic outcomes of implant wear or subsidence. We performed a retrospective radiographic review of 27 consecutive patients who received the implant and measured preoperative and postoperative jo

Gastroparesis.

Gastroparesis is a disorder characterized by delayed gastric emptying of solid food in the absence of a mechanical obstruction of the stomach, resulting in the cardinal symptoms of early satiety, postprandial fullness, nausea, vomiting, belching and bloating. Gastroparesis is now recognized as part of a broader spectrum of gastric neuromuscular dysfunction that includes impaired gastric accommodation. The overlap between upper gastrointestinal symptoms makes the distinction between gastroparesis and other disorders, such as functional dyspepsia, challenging. Thus, a confirmed diagnosis of gast

Mouse models of cancer-induced cachexia: Hind limb muscle mass and evoked force as readouts.

The majority of patients with advanced cancer suffer from cachexia, a systemic wasting syndrome, which subsequently impacts the tolerance to anti-cancer treatments, response to therapy, quality of life, and eventually, survival. Despite a high unmet medical need, there is currently no specific remedy available for an effective treatment of cachexia and its sequelae. A key feature of cachexia is the inexorable loss of skeletal muscle mass, which constitutes a main contributor to body weight loss and progressive functional impairments. Therefore, it's crucial to identify early readouts to detect

Antitumor and immunomodulatory activities of total flavonoids extract from persimmon leaves in H22 liver tumor-bearing mice.

Persimmon (Diospyros kaki L.) leaves are commonly used in Asia as tea infusion and as an agent in traditional medicine. The present study aims to explore the antitumor and immunomodulatory effects of total flavonoids extract from persimmon leaves (PLF) in H22 liver tumor-bearing mice. We found that the PLF showed significant inhibition on the liver tumor growth in mice with a tumor inhibition rate of up to 49.35%. In contrast to the severe side effects of cyclophosphamide (CTX), the PLF exhibited anti-cachexia effect and showed no alternation in the body weight and food intake in mice. Moreove
