[Mycotic infection in immunosuppressed patients. An anatomopathologic study].

Mycotic infections are a frequent and often severe complication in the immunosuppressed patient. A review of autopsy findings in 54 cases with gross, histologic and mycologic studies was undertaken among immunocompromised patients after chemotherapy or allogenic bone marrow transplantation: fungal infections were either localized especially in lungs and gastrointestinal tract, or disseminated. Fungi were various: principally Candida and Aspergillus, but also Fusarium, Torulopsis and Trichosporon. In acquired immunodeficiency syndrome (11 autopsy cases), mycotic infections appeared different. O

[The use of prophylactic antibiotics in oral and maxillofacial surgery].

Cultures of postoperative wounds after head and neck surgery reveal a variety of oropharyngeal anaerobic and aerobic flora, aerobic gram negative rods and fungi. Control clinical trials have shown that preoperative antibacterial prophylaxis reduces the incidence of postoperative wound infections. When an antibiotic is used prophylactically, it must be active against the micro-organisms known to cause the infection which the patient is supposed to be protected from. Antibacterial prophylaxis should begin just before the operation beginning, earlier is unnecessary and beginning later is less eff

Ultrastructural study of microbiologic colonization of urinary catheters.

We examined ten urinary catheters, associated with catheter-related urinary tract infections, by scanning and transmission electron microscopy to study the morphology of bacterial adherence. We confirmed that the bacteria associated with catheter-associated urinary tract infections grow in glycocalyx-enclosed microcolonies in a biofilm on the catheter surface. The bacterial populations demonstrated a heterogeneity that was not evident from the culture results, and it was demonstrated that only a small proportion of the microorganisms, including fungi, identified morphologically by scanning or

Bacteriology of chronic otitis media affecting children living in Rio de Janeiro.

A bacteriologic analysis was performed on the purulent exudates obtained from the middle ears of 45 children living in Rio de Janeiro with CSOM and spontaneous perforations of the ear drum. Anaerobic cultures showed anaerobic bacteria in association with aerobic or facultatively anaerobic bacteria, or both, in 30 (66.7%) specimens. Facultative anaerobic microorganisms occurred in 42 (93.3%) of the specimens analyzed, strict aerobes in 17 (37.8%), capnophilic in 6 (13.3%), and fungi in 4 (8.9%). The rate of different species of bacteria isolated per sample was 4.4:1.87 for anaerobic and 1.84 fo

Necrotizing fasciitis: a clinical, microbiologic, and histopathologic study of 14 patients.

We studied the clinical, microbiologic, and histopathologic findings from 14 patients with necrotizing fasciitis; also tissue obtained at autopsy was available from six patients. In all cases, material cultured was positive for multiple organisms, including Clostridia and fungi; organisms were identified by histochemical staining of tissue sections in 12 of 14 cases. The histologic pattern comprised edema, necrosis, and inflammation of skin, subcutaneous fat, and fascial tissue. Hyalin necrosis of sweat glands was observed in five patients. Thrombosis of vessels at all levels was a prominent f

Subacute disseminated mucormycosis in a diabetic male.

Mucormycosis is an opportunistic invasive infection caused by fungi of the order Mucorales. Rhizopus, Absidia, and Mucor are the most commonly encountered genera. Disease is characterized by vascular invasion, thrombosis, and tissue necrosis. Rhinocerebral disease is the most common manifestation but pulmonary, cutaneous, gastrointestinal, and widely disseminated forms have been reported. Pulmonary and disseminated disease are usually seen in neutropenic patients with leukemia or lymphoma. Both present as fever and unexplained pulmonary infiltrates unresponsive to antibacterials and corticoste

Stopping antibiotic therapy in neutropenic patients.

Infection is the major cause of death in neutropenic patients. Because of the lack of acute inflammatory cells, the usual signs of infection are often absent in these patients. Therefore, unexplained fever in a neutropenic patient requires prompt initiation of antibiotic therapy. Many physicians advocate continuing antibiotic therapy until neutropenia resolves. However, prolonged treatment with broad-spectrum antibiotics increases the risks of drug toxicity and superinfection with resistant bacteria and fungi. Based on a critical review of the literature and a large personal clinical experienc

Carcinoid tumor of the gastrointestinal tract with metastases to the brain.

A 56-year-old women with carcinoid tumor of the gastrointestinal tract, which was metastatic to the mesenteric lymph nodes, was seen four months after bowel resection with new neurologic findings. Results of an initial examination suggested infection with mycobacteria, or fungi, but the condition of the patient deteriorated, and she died after antituberculous and antifungal therapy was begun. She never suffered from carcinoid syndrome, and 5-hydroxyindoleacetic acid and 5-hydroxytryptamine were not detected in blood samples. A postmortem examination disclosed widespread metastases of carcinoid

Microbial eczema of the feet.

Twenty-seven patients were investigated for eczema of the feet which mainly affected the medial interdigital spaces and the dorsal areas of the medial toes. The lesions tended to spread to the dorsum of the foot, rather than the plantar surface as seen in tinea pedis, which typically affects the lateral interdigital spaces. In all patients S. aureus and/or haemolytic streptococci were cultured, whereas scrapings for fungi were negative. All responded well to treatment aimed at eradicating bacteria and dermatitis, but relapses occurred in 33% of the cases. We believe this clinical entity is due

Progress in antimycotic chemotherapy 1945-1980.

Before 1950 no reliable or safe therapy existed for systemic and invasive mycoses, and only traditional and empirical topical preparations were available for dermatomycoses. Two distinct eras of rapid progress in antifungal therapy followed: first, in the 1950's came the introduction of the polyenes, nystatin and pimaricin for cutaneous, vaginal and intestinal candidiasis, and amphotericin B for the treatment of severe systemic mycoses. The second phase saw the successful introduction and clinical use of 5-fluorocytosine and several imidazole derivatives some twenty years later, at a time when

Factors influencing the production of cellulases by Sporotrichum thermophile.

Cellulase production and growth of a strain of Sporotrichum thermophile were studied by using a mineral salts medium supplemented with yeast extract and insoluble cellulose. The effects of cultural conditions, such as pH, nitrogen source, substrate concentration, and temperature, were examined. Maximum production of C1 and CX cellulases occurred at 45 C in 2 to 4 days, in the presence of 1% Solka/Floc as substrate, when NaNO3 or urea used as sources of nitrogen. Under these conditions, cellulolytic activity of culture filtrates appeared to be similar to that reported for Trichoderma viride gro

YlAaf1-YlAaf2, a bipartite SANT domain-containing complex of transcriptional activator, promotes filamentous growth in the dimorphic yeast Yarrowia lipolytica.

The yeast-to-filament transition is important for dimorphic fungi to adapt to different environmental conditions. Transcriptional activators that activate downstream target genes in response to environmental stimulations are particularly important for the control of this morphological transition. However, only a limited number of key activators of filamentation have been identified. Here, we report YlAaf1 and YlAaf2, a novel pair of SANT domain-containing transcription factors that promote filamentation in the dimorphic yeast Yarrowia lipolytica. We show that both deletion and co-overexpressio

The Effect of the Extraction Method on the Content of Bioactive Compounds and the Biological Activity of Nigella sativa Extracts.

Ensuring food safety and consumer health are crucial elements of sustainable food safety management, requiring the use of substances that inhibit the growth of undesirable microorganisms at various stages of production. The negative impact of many of these on human health and the environment has led to increased interest in alternative solutions, such as plant extracts. The aim of this study was to determine the biological activity of extracts obtained from Nigella sativa seeds using various methods, including Soxhlet and ultrasound-assisted extraction (UAE) using hexane and methanol and super

Characterization of the forehead skin microbiome in the early phase of acne.

BACKGROUND: The skin microbiota is known to be imbalanced in acne vulgaris, but the changes occurring during the early stages of acne onset remain poorly described. OBJECTIVES: To characterize the skin microbiome of subclinical stages of acne in adults and adolescents. METHODS: The composition and diversity of the microbiota from non-lesional skin on the forehead of subjects with mild-to-moderate acne were compared to the ones from non-acne subjects. Analyses of skin swab samples were performed using high-throughput sequencing of the V1-V3 regions of the bacterial 16S ribosomal RNA gene, the

Scedosporium/Lomentospora Species Induce the Production of Siderophores by Pseudomonas aeruginosa in a Cystic Fibrosis Mimic Environment.

Over the last years, the interkingdom microbial interactions concerning bacteria and fungi cohabiting and/or responsible for human pathologies have been investigated. In this context, the Gram-negative bacterium Pseudomonas aeruginosa and fungal species belonging to the Scedosporium/Lomentospora genera are widespread, multidrug-resistant, emergent, opportunistic pathogens that are usually co-isolated in patients with cystic fibrosis. The available literature reports that P. aeruginosa can inhibit the in vitro growth of Scedosporium/Lomentospora species; however, the complex mechanisms behind t

The fungus Clonostachys epichloë alters the influence of the Epichloë endophyte on seed germination and the biomass of Puccinellia distans grass.

The fungal grass endophyte Epichloë typhina (Pers.) Tul. & C. Tul. (Ascomycota: Clavicipitaceae) grows intercellulary in aerial plant parts and reproduces asexually by invading host seeds. In this phase, it enhances seed production and germination, which accelerates its vertical spread. This relationship may be distorted by other seed-born fungi, whose spread is not so directly dependent on the success of the grass. Recently, the fungus Clonostachys epichloë Schroers has been observed on Puccinellia distans (Jacq.) Parl seeds originating from grass clumps infested with stromata, sexual structu

Serine/Threonine Phosphatase Calcineurin Orchestrates the Intrinsic Resistance to Micafungin in the Human-Pathogenic Fungus Mucor circinelloides.

Procedures such as solid-organ transplants and cancer treatments can leave many patients in an immunocompromised state. This leads to their increased susceptibility to opportunistic diseases such as fungal infections. Mucormycosis infections are continually emerging and pose a serious threat to immunocompromised patients. Recently there has been a sharp increase in mucormycosis cases as a secondary infection in patients battling severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infections. Mucorales fungi are notorious for presenting resistance to most antifungal drugs. The absence

Value of Acid-Fast Bacilli and Fungal Cultures in Foot and Ankle Surgery in a US Hospital System.

BACKGROUND: Acid-fast bacilli(AFB) and fungi are generally slow-growing, difficult to culture, and rarely the cause of infection. The goal of this study was to evaluate the value of routinely obtaining AFB and fungal cultures in foot and ankle surgery at a US hospital. METHODS: A retrospective review was conducted to determine the number of positive AFB and fungal cultures out of the total number of foot and ankle samples tested. Between 2014 and 2019, patients who underwent surgery for a foot and ankle infection for soft tissue infection, septic arthritis, infectious postoperative complicati

A Rare Case of Pulmonary and Gastrointestinal Mucormycosis Due to Rhizopus spp. in a Child with Chronic Granulomatous Disease.

Mucormycosis is a rare but serious fungal infection caused by a mold family known as the Mucorales. These fungi exist throughout the environment, especially in the soil, leaves, compost piles, or decaying woods. Humans contract mucormycosis by coming in contact with the spores from fungus either by inhalation or through cuts on the skin. The population at risk for this life-threatening infection includes diabetes mellitus patients, cancer patients, premature infants, burn patients, and immunocompromised patients. The fungi that most commonly cause mucormycosis are the Rhizopus species, and the

SERS-PLSR Analysis of Vaginal Microflora: Towards the Spectral Library of Microorganisms.

The accurate identification of microorganisms belonging to vaginal microflora is crucial for establishing which microorganisms are responsible for microbial shifting from beneficial symbiotic to pathogenic bacteria and understanding pathogenesis leading to vaginosis and vaginal infections. In this study, we involved the surface-enhanced Raman spectroscopy (SERS) technique to compile the spectral signatures of the most significant microorganisms being part of the natural vaginal microbiota and some vaginal pathogens. Obtained data will supply our still developing spectral SERS database of micro
