Subcutaneous delivery of icariin via a gelatin methacryloyl (GelMA) hydrogel sustained-release system improves ovarian function in reproductively aged mice.

Ovarian aging is characterized by a progressive decline in oocyte quality and quantity with age. Icariin (ICA), a flavonoid compound derived from Epimedium species, has demonstrated potential as an agent for ovarian restoration. In this study, a subcutaneous implantation system using gelatin methacryloyl (GelMA) hydrogel embedded with ICA was developed to restore ovarian function in aged female mice. Mice were assigned to receive subcutaneous implantation of GelMA alone (GelMA group), GelMA containing ICA (GelMA/ICA group), or a sham operation. Ovarian morphology, serum hormone levels, follicl

Genomic and Developmental Models to Predict Cognitive and Adaptive Outcomes in Autistic Children.

IMPORTANCE: Although early signs of autism are often observed between 18 and 36 months of age, there is considerable uncertainty regarding future development. Clinicians lack predictive tools to identify those who will later be diagnosed with co-occurring intellectual disability (ID). OBJECTIVE: To predict ID in children diagnosed with autism. DESIGN, SETTING, AND PARTICIPANTS: This prognostic study involved the development and validation of models integrating genetic variants and developmental milestones to predict ID. Models were trained, cross-validated, and tested for generalizability ac

Pyridoxine supply in human development.

Vitamin B(6) has an important role in the function of the human nervous system. Experimental data are not generally available on the role in human development, but significant conclusions may be made from studies of the effect of disorders of B(6) vitamer metabolism. Vitamin B(6) comprises seven compounds - pyridoxal, pyridoxine, pyridoxamine and their respective 5' phosphates. The common active form in human tissue is the 5'-phosphate form of pyridoxal (PLP) most of which is found in muscle bound to phosphorylase. Like many vitamins, B(6) can function both as a co-enzyme and as a chaperone. P

Structure and functional analysis of two insulin superfamily members in the Pacific white shrimp Litopenaeus vannamei.

The insulin superfamily constitutes a phylogenetically conserved hormonal system that critically regulates growth, metabolism, and reproduction in Metazoa. However, the functional diversification and the architecture of its regulatory networks of this superfamily remains poorly characterized in crustaceans. Here, we identify and characterize two little-studied insulin superfamily genes in the Pacific white shrimp Litopenaeus vannamei, gonadulin (LvGon) and insulin-related peptide (LvIRP). LvGon is predominantly expressed in neural tissues (brain and thoracic ganglia), while LvIRP is specifical

Signalling networks orchestrating the development of Drosophila indirect flight muscles.

Indirect flight muscles (IFMs) of Drosophila melanogaster are among the most structurally ordered and metabolically demanding tissues in the animal kingdom. Importantly, IFMs comprise two anatomically and mechanistically distinct muscle groups: the dorsal longitudinal muscles (DLMs) and the dorsoventral muscles (DVMs). Their ability to sustain high-frequency, stretch-activated oscillations depends not only on specialised sarcomeric architecture but on multilayered signalling networks that coordinate lineage specification, myoblast proliferation, fibre patterning, mechanotransduction, metabolic

Metformin attenuates valproic acid-induced developmental cardiotoxicity through activation of the AMPK/PGC-1α axis and restoration of mitochondrial and redox homeostasis in zebrafish embryos.

Prenatal valproic acid (VPA) exposure is strongly linked to developmental cardiotoxicity, yet no targeted pharmacological countermeasure exists. Converging mechanistic evidence indicates that VPA cardiotoxicity centers on AMPK/PGC-1α suppression, positioning the AMPK activator metformin (MET) as a rational candidate for in vivo protection. We examined MET's protective effects against VPA-induced developmental cardiotoxicity in zebrafish embryos. Embryos were randomly allocated to four groups (Control, VPA 0.1 mM, MET 10 mM, VPA + MET) immediately after fertilization and chronically exposed unt

OsPLT1 negatively regulates phosphorus deficiency tolerance by modulating nitric oxide-mediated cell wall phosphorus reutilization in rice.

BACKGROUND AND AIMS: Phosphorus (P) deficiency severely limits rice productivity, yet the role of root developmental regulators in P homeostasis remains unclear. Here, we show that the rice PLETHORA transcription factor OsPLT1 suppresses rice tolerance to P-deficient conditions. Under P-starved conditions, osplt1 mutants accumulated more biomass and exhibited higher soluble and total P concentrations, whereas OsPLT1-overexpressing lines were hypersensitive to P deficiency. KEY RESULTS: Mechanistically, osplt1 mutants displayed reduced P retention in root cell walls, particularly in the water-

Transient fetal hyperglycemia reprograms adipogenesis, lipogenesis, and insulin signaling in zebrafish.

Gestational diabetes mellitus (GDM) exposes the fetus to chronic hyperglycemia and increases the risk of later metabolic disease. The mechanisms by which early hyperglycemia reprograms lipid and glucose metabolism remain unclear, largely due to maternal and placental confounders in mammalian models. To overcome these limitations, we developed a zebrafish model that isolates fetal hyperglycemia and enables direct investigation of its developmental and long-term metabolic effects. To mimic late-onset GDM, zebrafish embryos were exposed at 4 days post-fertilization to glucose concentrations compa

Adverse outcome pathway-based analysis of vanillic acid-induced neurodevelopmental toxicity and cellular senescence in zebrafish.

Vanillic acid (VA), widely used in industrial and therapeutic applications, also presents potential ecological and biological risks. This study systematically investigated the developmental and neurotoxic effects of VA in zebrafish and explored its mechanistic pathways within an Adverse Outcome Pathway (AOP) framework. VA exposure resulted in concentration-dependent toxicity, with an LC50 of 638.3 μM. High VA concentrations induced developmental abnormalities, including malformations of the eyes, otic vesicles, craniofacial structures, and tails. Behavioral assessments revealed a significant r

Modelling Tetralogy of Fallot: insights and limitations of animal models.

Tetralogy of Fallot (TOF) is the commonest cyanotic congenital heart disease, arising from antero-cephalad deviation of the outlet septum during outflow tract (OFT) morphogenesis, producing a ventricular septal defect, overriding aorta, pulmonary stenosis, and right ventricular hypertrophy. Despite advances in surgical correction, the developmental origins of TOF remain incompletely understood, and the genetic architecture of the majority of non-syndromic cases is yet to be resolved. Experimental models have been central to progress in this field, yet each model may capture only part of the di

Maternal Western Diet Programmes Bile Acid Dysregulation and Hepatic Fibrosis in Fetal and Juvenile Macaques.

BACKGROUND AND AIMS: Maternal obesity increases the risk of the paediatric form of metabolic dysfunction-associated steatotic liver disease (MASLD), affecting up to 30% of youth, but the developmental origins remain poorly understood. METHODS: Using a Japanese macaque model, we investigated the impact of maternal Western-style diet (mWSD) or chow diet followed by postweaning WSD (pwWSD) or chow diet focusing on bile acid (BA) homeostasis and hepatic fibrosis in livers from third-trimester fetuses and 3-year-old juvenile offspring. RESULTS: Juveniles exposed to mWSD had increased hepatic coll

Reduced Availability of Essential Amino Acids Disrupts Differentiation of Anorexigenic POMC Neurons in the Fetal Rat Hypothalamus.

Intrauterine growth restriction (IUGR) is associated with an elevated risk of long-term metabolic disorders, including obesity and type 2 diabetes, aligning with the Developmental Origins of Health and Disease hypothesis. However, the mechanisms underlying metabolic programming remain elusive. This study investigates the impact of gestational protein restriction (PR) on fetal hypothalamic development, focusing on the formation of neuronal populations regulating appetite and energy balance. Using a rat isocaloric PR model (8% protein preconception, 4% during gestation), we examined hypothalamic

Q241R mutation of Braf causes neurological abnormalities in a mouse model of cardio-facio-cutaneous syndrome, independent of developmental malformations.

Constitutively active mutants of BRAF cause cardio-facio-cutaneous (CFC) syndrome, characterized by growth and developmental defects, cardiac malformations, facial features, cutaneous manifestations, and mental retardation. An animal model of human CFC syndrome, the systemic BrafQ241R/+ mutant mouse, has been reported to exhibit multiple CFC syndrome-like phenotypes. In this study, we analyzed the effects of Braf mutations on neural function, separately from their effects on developmental processes. To this end, we generated Braf mutant mice expressing BRAFQ241R specifically in mature excitato

Colchicine induces developmental defects and renal toxicity in zebrafish by upregulating the oxidative stress.

Colchicine is a type of alkaloid commonly used clinically for treating diseases such as gouty arthritis. Studies suggest that colchicine is toxic to the kidneys. However, the specific mechanisms are unclear. Herein, we used zebrafish to validate the renal toxicity of colchicine and investigate the underlying mechanisms. Zebrafish larvae at 3 days post-fertilization were exposed to various concentrations (15, 20, and 25 mg/L) of colchicine solution for 72 h. Results showed that colchicine caused death, slowed heart rate, slowed growth, increased interpupillary distance, caused periorbital and r

Toxicity profiling of phenazopyridine in zebrafish: An emerging pharmaceutical contaminant in the environment.

Phenazopyridine is an azo dye analgesic routinely used in the management of urinary tract infections. An overdose of this pharmaceutical agent causes methemoglobinemia and renal failure across all age groups. There is limited knowledge regarding the implications of early phenazopyridine exposure, whether via medicinal use or inadvertent contact, or its toxicity in specific organs in living organisms. Moreover, 40-50 % of this drug gets eliminated in its native form and hence it contributes to the pharmaceutical burden in the environment. In this study, we used zebrafish, a versatile model syst

Melatonin alleviates UV-induced embryonic toxicity via PARP1 expression in early mouse embryos.

Maintaining genome integrity is essential for preimplantation embryos, yet light poses a significant genotoxic threat. The application of assisted reproductive technologies (ART) increases embryo exposure to environmental factors, with ultraviolet (UV) light being particularly concerning. UV can cause various forms of DNA damage, such as sister chromatid exchange and DNA breakage in preimplantation embryos. Melatonin (MLT), a neuroendocrine hormone secreted by the pineal gland, positively influences various physiological processes, including regulating DNA damage responses. Here, we demonstrat

Expanded Phenotype of the Cln6nclf Mouse Model.

Neuronal ceroid lipofuscinoses (NCLs) are a group of autosomal recessive neurogenetic disorders caused by mutations in 14 different genes. CLN6 disease manifests as variant late-infantile NCL (vLINCL) or as an adult variant. In childhood, symptoms include speech delay, vision loss, cognitive and motor decline, seizures, and early death. An in-depth characterization of a naturally occurring Cln6 mutant mouse (Cln6nclf) is presented, with implications for translational research. The expanded phenotype provides data showing early death, vision loss, and motor deficits in male and female Cln6nclf
