Magnesium Phosphate Composite Hydrogel Dressing Promotes Diabetic Wound Repair Through Anti-Inflammatory, Antioxidant, Angiogenic, and Metabolic Regulatory Mechanisms.

Chronic non-healing diabetic wounds pose a significant clinical challenge, attributed to a pathogenic microenvironment characterized by persistent inflammation, excessive accumulation of reactive oxygen species, impaired angiogenesis, and mitochondrial dysfunction. Herein, we reported a novel multifunctional hydrogel dressing aimed at promoting diabetic wound healing through the synergistic action of anti-inflammatory, antioxidant, angiogenic, and metabolic regulatory mechanisms. First, nicotinamide mononucleotide (NMN)-templated magnesium phosphate nanoparticles (NMP NPs) were synthesized thr

Composite Hydrogel With Ultra-High Mechanical Anisotropy and Biotissue-Like Softness by Confining Electrostatic and Entropic Repulsion Between Cofacially Aligned Nanosheets.

Owing to their biotissue-like nature, mechanically anisotropic hydrogels have attracted extensive attention. Recent studies have achieved high mechanical anisotropy by densely assembling oriented polymers and/or nanofillers into stiff networks. However, this strategy involves soft-direction hardening to afford intrinsically hard hydrogels, limiting their applications, particularly in biomedical fields. Here, we developed the first hydrogel that simultaneously achieves excellent mechanical anisotropy and a low elastic modulus in the soft direction, using magnetically oriented, negatively charge

A mechanically tunable photo-cured hydrogel system based on methacrylated collagen-keratin inter-penetrating networks for rapid and enhanced wound healing.

Conventional hydrogel wound dressings have non-tunable mechanical properties and serve only as passive barriers, resulting in slow wound healing. Advanced hydrogel dressings that can stimulate wound contraction and tissue deformation can potentially accelerate the healing of chronic wounds characterized by long healing times and high infection risk. In this study, we developed injectable photo-cured methacrylated collagen-keratin interpenetrating network (ColMA-K@IPN) hydrogels for rapid enhanced wound healing. Gastrointestinal In Situ Self-Assembled Gastrodia Elata Polysaccharide Hydrogel Enables Parkinson's Disease Therapy via Gut-Brain Axis Modulation.

Parkinson's disease (PD) is closely associated with abnormal α-synuclein propagation along the gut-brain axis and progressive dopaminergic neuronal loss. Traditional oral preparations suffer from weak gastrointestinal resistance, rapid degradation and poor gut-brain axis regulation. Here, we constructed a gastrodia elata polysaccharide-functionalized dual-network GEPH hydrogel via thiol-maleimide click crosslinking. GEPH possesses favorable biocompatibility, regular porous microstructure and tailored rheological properties suitable for gastrointestinal delivery, showing outstanding erosion res

Synergistic role of quercetin in sodium alginate/κ-carrageenan hydrogel microspheres for the protection and controlled release of the novel antimicrobial peptide hz-01.

To address the poor stability and high susceptibility to degradation of antimicrobial peptides, a pH-responsive hydrogel microsphere system was developed based on sodium alginate (SA) and κ-carrageenan (Car), with quercetin (Que) for intestinal delivery of chemically synthesized free hz-01 peptide. The optimized formulation containing 0.45% Car exhibited the highest encapsulation efficiency (77%), corresponding to an absolute increase of 13 percentage points compared with the system without Que. The microspheres remained stable under simulated gastric conditions but enabled controlled intestin

Construction of a chitosan/γ-poly(glutamic acid) composite hydrogel crosslinked by ZIF-8 for enhanced α-glucosidase immobilization and inhibitor screening.

Herein, a composite hydrogel based on two natural macromolecules-chitosan (CS) and γ-poly(glutamic acid) (γ-PGA)-was developed, crosslinked by zeolitic imidazolate framework-8 (ZIF-8) nanoparticles, which serve as reactive crosslinking hubs to covalently connect the biopolymer chains for efficient immobilization of α-glucosidase (α-Glu). A carboxyl-terminated poly(ethylene glycol)-poly(glutamic acid) copolymer was first synthesized and its activated carboxyl groups underwent amidation with amino groups of CS to rapidly form a three-dimensional PC/ZIF-8 hydrogel. The effects of ZIF-8 content an

Curcumin-loaded copper/iron bimetallic nanoparticle-incorporated hydrogel scaffold: a sequential microenvironment reprogramming platform for accelerated chronic wound healing.

Diabetic wound management represents a substantial clinical challenge owing to the deteriorative tissue microenvironment including the excessive reactive oxygen species (ROS), persistent inflammation, and potential bacterial infection. To address these issues, herein, a hybrid hydrogel scaffold (SF@NP-Cur) capable of sequentially reprogramming the wound microenvironment was developed through microfluidic 3D printing technique for infected chronic wound healing. Such scaffold incorporates curcumin-loaded copper/iron bimetallic nanoparticles (NP-Cur), which confers not only the oxidase- and pero

Core-shell hydrogel beads co-encapsulating kunitz-type trypsin inhibitor: A synergistic strategy for enhanced stability of oral active peptides.

This study repurposed Kunitz-type trypsin inhibitor (KTI) as a synergistic component for oral peptide co-delivery to overcome the burst release and enzymatic degradation of liraglutide. A pH-responsive chitosan (CS)‑sodium alginate (ALG) core-shell hydrogel co-delivery system co-encapsulating KTI and liraglutide, named ACKL (ALG-CS-KTI-liraglutide), was accordingly constructed. The optimized microspheres, with a 3% sodium tripolyphosphate (TPP)-crosslinked core and 0.8% ALG shell, exhibited uniform morphology, a distinct core-shell structure, and excellent mechanical properties. Autologous Neural Stem Cell Encapsulation in CT04-Integrated Fibrin Hydrogel Enhances Tissue Regeneration After Spinal Cord Injury in Monkeys.

Spinal cord injury (SCI) is a major traumatic insult that leads to significant motor and sensory impairments. Following SCI, an inhibitory microenvironment develops, accompanied by substantial neuronal loss. A single therapeutic approach may not achieve substantial functional recovery. Therefore, strategies aimed at addressing the inhibitory environment and enhancing cellular engraftment are crucial for optimal recovery. In this study, we investigated the efficacy of combining an injectable fibrin hydrogel with subventricular zone-derived neural stem cells (SVZ-NSCs) and CT04, a RhoA inhibitor

Hyaluronic acid-based reactive oxygen species responsive nanocomposite hydrogel for sequential drug delivery and effective prevention of postoperative abdominal adhesions.

Postoperative abdominal adhesion (PAA) affects over 90% of abdominal surgery patients and remains a huge clinical challenge. Sustained inflammation with excessive reactive oxygen species (ROS) production and subsequent peritoneal mesothelial cells (PMCs)-mesenchymal transition (MMT) are key events driving PAA formation. Herein, we engineer a ROS-responsive nanocomposite hydrogel (P/H/PBLA@PFD) for ROS-triggered sequential delivery of antioxidant/anti-inflammatory drugs and MMT inhibitors for effective PAA prevention. P/H/PBLA@PFD hydrogel is fabricated via Schiff base crosslinking between alde

Amphotericin B-cholesterol Co-encapsulated Polymeric Nanoparticles in Protein Hydrogel-based Injectable Depot System for Long-acting Drug Delivery.

Amphotericin B exhibits dose-dependent nephrotoxicity and hepatotoxicity due to its non-selective interaction with mammalian cholesterol. The aim of the present study was to develop an Amphotericin B-cholesterol co-encapsulated albumin hydrogel-based injectable depot formulation to achieve prolonged plasma drug concentrations with reduced toxicity. Amphotericin B-loaded polymeric nanoparticles (Amphotericin B-PNs) were prepared by the nanoprecipitation method. Cholesterol co-encapsulated nanoformulation has been able to keep Amphotericin B in a less aggregated state (aggregation ratio -1.40),

Post-modified biosynthesis strategy to achieve a heme-mimicking bacterial cellulose hydrogel with synergistic antibacterial functions for infected skin defect therapy.

The emergence of global multi-drug-resistant (MDR) bacteria has occurred due to the excessive abuse of antibiotics. Multiple antibacterial technologies have been explored to prevent MDR bacteria. Here, we developed a post-modified biosynthesis strategy to achieve a heme-mimicking bacterial cellulose hydrogel integrating a quaternary ammonium compound (i.e., [VPIM]Br) with a heme-mimicking gallium porphyrin (i.e., Ga-CHP) with synergistic antibacterial functions for infected skin defect therapy. The obtained hydrogel exhibited high in vitro antibacterial activity (99.9%) against both Escherichi

3D-Printed Ordered Porous CSMA/GelMA Hydrogel Scaffolds Enhance ADSC Reparative Function With FAK-YAP-Related Mechanotransductive Changes.

Chronic diabetic wounds persist in a hostile microenvironment that compromises the survival and reparative activity of transplanted stem cells. Here, we developed a structurally defined 3D-printed ordered porous chitosan methacrylate/gelatin methacryloyl (CSMA/GelMA) hydrogel scaffold platform to test whether topological order acts as a structurally relevant biomaterial cue for adipose-derived mesenchymal stem cell (ADSC) regulation. To enrich for topology-dependent effects, ordered and disordered scaffolds were fabricated from the same photocrosslinkable chitosan methacrylate/gelatin methacry

Layered double hydroxide-nanoengineered injectable hydrogel based on oxidised pectin/carboxymethyl chitosan for sustained enoxaparin release in deep vein thrombosis.

Deep vein thrombosis prophylaxis with enoxaparin (ENO) requires frequent subcutaneous injections, posing treatment burden and compliance challenges. However, the sustained delivery of ENO remains challenging due to its highly anionic nature and rapid diffusion from conventional delivery matrices. Here, we developed an injectable hydrogel incorporating ENO-loaded layered double hydroxide (LDH) nanocarriers within an oxidised pectin/carboxymethyl chitosan (OP/CMCs) matrix to enable prolonged release. The hydrogel forms via Schiff base chemistry, yielding rapid gelation (∼7 min), self-healing pro

Mg-integrated octopus-inspired hydrogel dressing enables autonomous adhesion and wound closure for enhanced healing via sequential microenvironment regulation.

Diabetic wounds are characterized by persistent infection, excessive oxidative stress, and impaired healing. Conventional dressings often lack autonomous adhesion and wound-closure capabilities, and fail to balance essential antimicrobial and anti-inflammatory properties. Inspired by the sucker morphology and thermo-responsive behavior of the octopus, this study developed a Three-Dimensional (3D)-printed, photosensitive/thermosensitive dual-responsive poly(N-isopropylacrylamide) (PNIPAM)/gelatin hydrogel dressing. The hydrogel was functionalized with mesoporous MgO nanoparticles coated with se

Ferroptosis-driven in situ vaccine-like antitumor effects: NIR-triggered IFBM hydrogel synergizes with sorafenib to unleash systemic antitumor immunity.

The immunosuppressive tumor microenvironment and immune evasion mechanisms inhibit the development of effective antitumor responses and enduring immune memory, thereby diminishing the efficacy of cancer immunotherapy. In situ vaccines (ISVs) have been developed to overcome these challenges by inducing localized immunogenic cell death (ICD) and activating systemic immunity via dendritic cells (DCs) and CD8+ T cells. In this study, we engineered ferrous sulfide-bovine serum albumin-cell membrane nanoclusters to trigger ferroptosis, which were co-encapsulated with the photothermal agent indocyani
