Hepatitis C virus (HCV) readily sets up persistence after acute infection. Cellular immune responses are thought to play a major role in control of the virus. Failure of CD4+ T-cell responses in acute disease is associated with viral persistence but the dynamics of this are poorly understood. We aimed to assess such responses using a novel set of Class II tetrameric complexes (tetramers) to study helper T-cells ex vivo in acute disease. We analysed the HCV-specific CD4+ T-cell response in a patient with acute hepatitis c infection. We were able to track the virus-specific CD4+ T-cells directly

Uveitis in children are less frequent than in adults. Their prognosis is variable because it may be found as an isolated and idiophatic condition or in association with definite clinical entities. The associated noninfectious diseases with predominantly renal involvement are tubulointerstitial nephritis and uveitis syndrome (TINU syndrome), mesangial glomerulonephritis isolated or in association with Behçet's disease. A case of 14-years-old girl with intermediate uveitis (pars planitis) and mesangial glomerulonephritis is presented. The ocular symptoms was eye redness and ocular pain and she h

Certain cytokines, the prototype being the highly pleiotropic TNF, have many homeostatic physiological roles, are involved in innate immunity, and cause inflammation when in excess. These cytokines have long been accepted to have central roles in the pathogenesis of systemic or local non-cerebral disease states, whether acute or chronic, and whether or not caused by infectious agents. Over the last decade they have also been appreciated to be broadly important in brain physiology. As in other organs, excessive levels in brain are harmful, and its physiological complexity leads to corresponding

BACKGROUND: Immunoglobulin A nephropathy (IgAN) is a common immune-mediated kidney disease. Its pathogenesis involves dysregulated B cell activation, excessive production of galactose-deficient immunoglobulin A1 (Gd-IgA1), and formation of IgA immune complex, which collectively drive glomerular injury. Telitacicept, a fusion protein that blocks B cell activation pathways, may counteract these mechanisms, but observational data and reliable biomarkers of therapeutic response remain limited.
OBJECTIVE: This study aimed to evaluate the clinical efficacy and safety of telitacicept and to determin

Most medically important bacterial and viral pathogens gain entry into the body either via the skin or a mucosal surface. Vaccination provides a viable and cost-effective strategy for the prevention of such diseases and it has always been a principal aim with vaccinologists, to be able to promote simultaneously, protective immune responses both systemically and at mucosal surfaces. The paradigm that mucosal immunity is best stimulated by exposure to antigen via a mucosal route simply because inductive sites such as Peyer's patches and bronchial associated lymphoid tissues are located in the mu

Human papillomavirus (HPV) infection is a major driver of anogenital disease and virus-related carcinogenesis. Although most infections resolve spontaneously, persistent infection with high-risk genotypes may progress to high-grade squamous intraepithelial lesions (HSILs) and cancer, particularly in the setting of impaired immune surveillance. Unlike previous HPV-related reviews focused primarily on cervical disease, vaccination, or isolated immunosuppressed populations, this narrative review comparatively examines the clinical expression, colposcopic findings, screening strategies, and therap

Inflammatory bowel diseases (IBDs), encompassing Crohn's disease and ulcerative colitis, are complex and heterogeneous diseases characterized by a multifactorial etiology, therefore demanding a multimodal approach to disentangle the main pathophysiological components driving disease onset and progression. Adoption of a systems biology approach is increasingly advocated with the advent of multiomics profiling technologies, aiming to improve disease classification, to identify disease biomarkers, and to accelerate drug discovery for patients with IBD. However, clinical translation of multiomics-

We report on a 2-year-old girl with chronic bullous disease of childhood (CBDC). Concomitantly with a feverish gastrointestinal infection caused by Salmonella enteritidis, the child presented with lesions resembling impetigo contagiosa on the legs and face. Following antibiotic treatment with cephalosporins p.o. and flucloxacillin i.v. she developed typical symptoms of CBDC, i.e. tense blisters on erythematous skin in the perineal area, on the flexor aspects of the thighs and upper arms and on the face. Direct immunofluorescence revealed a linear IgA deposition along the basement membrane zone

In the absence of lupus, mesangial electron-dense deposits in membranous nephropathy are thought to be uncommon. In this study 18 renal biopsies of 16 cases seen over a 2-year period were evaluated by light and electron microscopy and immunofluorescence, directing particular attention to the mesangium. Lupus had been excluded in every instance by the usual serologic tests. In seven of the 18 biopsies, mesangial electron-dense deposits were found. Clinically, almost all of these patients were characterized by atypical features: five patients had underlying diseases which have been associated wi

Atherosclerosis (AS), a chronic inflammatory disease driven by lipid accumulation and immune dysregulation, predisposes to acute cardiovascular events via plaque rupture and thrombosis. The molecular mechanisms linking immune microenvironment imbalance to plaque instability remain incompletely defined. Utilizing the GSE100927 dataset (69 AS patients vs 35 controls), we identified differentially expressed genes (DEGs) and constructed co-expression networks via weighted gene co-expression network analysis. Hub genes were intersected with DEGs to determine core genes. Immune cell infiltration was

Sepsis is a life-threatening condition arising from a dysregulated host response to infection, characterized by a complex interplay between pro-inflammatory and anti-inflammatory immune mechanisms. B cells, that are key components of humoral immunity, are essential for antibody production, antigen presentation, and immune modulation. Sepsis is commonly associated with B cell lymphopenia, particularly in severe cases, as evidenced by a marked reduction in circulating B cells in both adult and pediatric patients. However, the mechanisms driving this depletion have not been completely understood,

Psoriasis is a common chronic dermatological disease, affecting approximately 1-3% of the global population, and is associated with numerous comorbidities, impaired quality of life, and reduced life expectancy compared with the general population. The pathogenesis of psoriasis is complex and multifactorial, involving genetic susceptibility, external environmental factors, and immune system dysregulation. Psoriasis is perceived as a systemic disorder associated with a systemic inflammatory condition. In psoriasis, a chronically activated immune response results in the expression of numerous pro

Advances in coronavirus disease 2019 management has led to decreases in disease burden; however, immunocompromised patients continue to experience prolonged or persistent infections. We investigated immunological factors associated with time to viral clearance of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infections in immunocompromised patients. By integrating clinical follow-up data, SARS-CoV-2-specific humoral and cellular response data, and single-cell RNA transcriptomic data, we uncovered differential immune responses between patients exhibiting short or long time-to-cle

Autoimmune diseases result from a breakdown of immune tolerance influenced by genetic and environmental factors. Regulatory T cells (Tregs) maintain immune homeostasis, while interferon-γ (IFNγ) has context-dependent proinflammatory and regulatory roles. In B10.S mice, mercury-induced autoimmunity (HgIA) emerges within approximately 4 weeks of Hg exposure and is marked by antinucleolar antibody (ANoA) production, polyclonal B-cell activation, and deposition of immune complexes in the kidney. We investigated whether Tregs attenuate HgIA and evaluated IFNγ's role in this regulation. Female WT an

Membranous nephropathy (MN) is an immune-mediated glomerular disease and the most common cause of nephrotic syndrome in adults. Classical paradigms concentrate on the binding of circulating autoantibodies (e.g., anti-PLA2R, anti-THSD7A) to podocytes, resulting in subepithelial immune deposits, complement activation, and podocyte damage. Nonetheless, mounting evidence suggests that lipid metabolism in podocytes is a crucial regulator of MN pathophysiology. Podocyte slit diaphragms are situated within specialized cholesterol-enriched lipid rafts that orchestrate essential structural and signalin

Chronic viral hepatitis caused by hepatitis B virus (HBV) and hepatitis C virus (HCV) remains a leading global public health burden, driving progressive liver fibrosis, cirrhosis, and hepatocellular carcinoma (HCC) through complex interactions between viral replication, host immune responses, and extracellular matrix (ECM) remodelling. Matrix metalloproteinases (MMPs) are a family of zinc-dependent endopeptidases that serve as central regulators of ECM homoeostasis and immune modulation in the liver. While physiological MMP activity is essential for tissue repair and immune surveillance, dysre

BACKGROUND: The aetiology of multiple sclerosis (MS) is complex and remains unclear. Increasing evidence suggests that environmental factors, including infectious agents, may contribute to disease initiation and progression. This review aims to summarize current evidence on the role of microorganisms in MS pathogenesis.
METHODS: A systematic literature search was conducted in PubMed, SCOPUS, and Web of Science. We enrolled studies published between 2020 and 2025. Eligible papers investigating associations between microbial agents and MS risk, activity, or progression were assessed for eligibi

However, the current state of knowledge of potential interactions among iodine-selenium-thyroid-immune system is not sufficient. The aim of the study was to evaluate iodine and selenium statuses and examine the relationship between them and the functioning of the thyroid and immune system in a group of women of reproductive age, without previously diagnosed disease. Methods: The study involved a group of 60 women aged 19-40 from southern Poland. The concentrations of iodine

BACKGROUND: Severe aplastic anemia (SAA) is an uncommon life-threatening disorder characterized by hypocellular bone marrow and pancytopenia. It is typically associated with immune-mediated mechanisms, requiring immunosuppressive therapy (IST) or hematopoietic stem cell transplantation (HSCT). Infections, especially invasive fungal infections such as mucormycosis and aspergillosis, constitute principal causes of morbidity and mortality in patients with SAA. Genetic predispositions, including perforin (PRF1) polymorphisms, may further complicate disease outcomes by impairing immune function.

Type 1 diabetes mellitus (T1DM) is a chronic autoimmune disorder characterized by the destruction of insulin-producing pancreatic beta cells, resulting in lifelong insulin dependence. While genetic susceptibility-particularly human leukocyte antigen (HLA) class II alleles-is a major risk factor, accumulating evidence implicates viral infections as potential environmental triggers in disease onset and progression. This narrative review synthesizes current findings on the role of viral pathogens in T1DM pathogenesis. Enteroviruses, especially Coxsackie B strains, are the most extensively studied
