[Neuronal dysplasia of the colon. A case report].

A clinical case of Neuronal Intestinal Dysplasia (NID) is reported. This is an important pathology which is part of congenital alterations of bowel's innervation. This disease consists of two forms (A and B) which occur during different ages causing problems in intestinal motility with consequent various symptoms. Actually NID is diagnosed easier than in the past because histopathological techniques are very refined and permit to distinguish NID from cases once classified as Hirschsprung's disease. Histology can show an increase of Ach and Ldh activity in cholinergic fibers of lamina propria a

Mechanism of interleukin-1β induced-increase in mouse intestinal permeability in vivo.

UNLABELLED: Interleukin-1β (IL-1β) has been shown to play an essential role in mediating intestinal inflammation of Crohn's disease and other inflammatory conditions of the gut. Previous studies from our laboratory have shown that IL-1β causes an increase in intestinal tight-junction permeability in Caco-2 monolayers in vitro. However, the IL-1β effect on the intestinal epithelial barrier in vivo remains unclear. AIMS: the major aims of this study were to examine the effect of IL-1β on mouse intestinal epithelial barrier in vivo and to delineate the mechanisms involved using an in vivo model

Reply to comment on "Ultrasonographic scores for ileal Crohn's disease assessment: better, worse or the same as contrast‑enhanced ultrasound?

We read the comments by Nylund K et al. regarding our paper "Ultrasonographic scores for ileal Crohn's disease assessment: Better, worse or the same as contrast‑enhanced ultrasound?". Intestinal ultrasound has become one of the most valuable developments in the past decade, a non-invasive, well-tolerated exam, with an easy repeatability, and absence of sedation, ionizing radiation, or preparation. Particularly for inflammatory bowel disease, where there is a lack of agreement of patient's symptoms with disease activity, in an era where the paradigm of mucosal healing is changing to transmural

Melatonin and inflammatory bowel disease: From basic mechanisms to clinical application.

Inflammatory bowel disease is a chronic inflammatory disease and has periods of recurrence and remission. Improper immune responses to gut flora bacteria, along with genetic susceptibility, appear to be involved in causing this complex disease. It seems dysbiosis and oxidative stress may also be involved in IBD pathogenesis. A significant number of clinical studies have shown an interesting association between sleep disturbances and IBD. Studies in animal models have also shown that sleep deprivation has a significant effect on the pathogenesis of IBD and can aggravate inflammation. These inte

Appraisal of gut lavage in the study of intestinal humoral immunity.

Direct investigation of intestinal humoral immunity requires collection of intestinal secretions or mucosal biopsy specimens, or both. A non-invasive technique of gut lavage, with a polyethyleneglycol electrolyte lavage solution as a means of collecting intestinal secretions for immunoglobulin and antibody studies, was evaluated. Fifty patients were studied--25 immunologically normal patients or volunteers, 15 patients with untreated coeliac disease, and 10 patients with active Crohn's disease. Protease inhibitors were added promptly to samples to prevent proteolysis of immunoglobulin content.

The Gut-Lung Axis, Epigenetics and Respiratory Disease.

The first and second phases of the human microbiome project provided a view of mucosal surfaces and the skin of humans that mapped an abundant and complex ecosystem (microbiota) that is composed of bacteria (bacteriobiota), fungi (mycobiota), viruses (virobiota), enteric phages, archaea, protists, and helminths. Intestinal dysbiosis describes an adverse shift in microbial homeostasis in the gut that enhances intestinal epithelial permeability, translocating toxins that may lead to endotoxemia. Numerous intestinal and extra-intestinal illnesses have been linked to gut dysbiosis, including infla

From in vitro development to accessible luminal interface of neonatal bovine-derived intestinal organoids.

BACKGROUND: Intestinal organoids provide physiologically relevant in vitro models that bridge the gap between conventional cell culture and animal studies. Although these systems have been developed for adult cattle, their use in neonatal calves-who are particularly vulnerable to enteric disease-has not been well established. Neonatal diarrhea remains a major health concern in modern agriculture, yet age-appropriate models for studying its pathogenesis are lacking. Given that host-pathogen interactions vary with developmental stage, there is a need for culture systems that reflect the distinct

Impaired intestinal vitamin B1 (thiamin) uptake in thiamin transporter-2-deficient mice.

BACKGROUND & AIMS: Intestinal thiamin uptake process is vital for maintaining normal body homeostasis of the vitamin; in vitro studies suggest that both thiamin transporter-1 (THTR-1) and -2 (THTR-2) are involved. Mutations in THTR-1 cause thiamin-responsive megaloblastic anemia, a tissue-specific disease associated with diabetes mellitus, megaloblastic anemia, and sensorineural deafness. However, in patients with thiamin-responsive megaloblastic anemia, plasma thiamin levels are within normal range, indicating that THTR-2 (or another carrier) could provide sufficient intestinal thiamin absorp

Interleukin-17 Weakens the NAFLD/NASH Process by Facilitating Intestinal Barrier Restoration Depending on the Gut Microbiota.

Interleukin-17 (IL-17) is associated with nonalcoholic fatty liver disease (NAFLD) and gut microbiota, and how IL-17 mediates the NAFLD/nonalcoholic steatohepatitis (NASH) process depending on the gut microbiota is unclear. We found that T helper 17 (TH17) cells were decreased in the small intestine in a methionine choline-deficient (MCD) diet-induced NASH model. IL-17-deficient (Il17-/-) mice showed alterations in intestinal microbiota, including the inhibition of probiotic growth and the overgrowth of certain pathogenic bacteria, and were prone to higher endotoxemia levels and more severe ga

High-sugar and carrageenan-containing diets synergistically impair the intestinal mucosal barrier through the gut microbiota: in vitro and in vivo studies.

Increased consumption of high-sugar processed foods is accompanied by a higher daily intake of food additives like carrageenan, which is strongly correlated with the development of intestinal diseases. While previous studies indicate long-term consumption of high-sugar and carrageenan-containing foods impairs colonic barrier, the underlying mechanisms remain unclear. In this study, combined in vitro and in vivo models were employed and the mediating role of gut microbiota was elucidated. In animal studies, high-sugar and carrageenan-containing diets decreased antimicrobial substances expressio

Spatial metabolomics reveals the therapeutic mechanisms of Shengjiang Xiexin decoction in modulating gut-liver axis fatty acid metabolism for the treatment of Clostridioides difficile infection.

ETHNOPHARMACOLOGICAL RELEVANCE: Shengjiang Xiexin Decoction (SXD) is a classic formula documented in the Treatise on Cold Damage and Miscellaneous Diseases. It has long been used to treat infectious gastrointestinal diseases characterized by diarrhea, dysentery, and abdominal distension. Its core pathogenesis involves spleen-stomach disharmony and damp-heat accumulation, consistent with clinical features of bacterial or viral enteritis. Although SXD effectively relieves gastrointestinal symptoms, its active components and mechanisms remain unclear, especially regarding spatial regulation of gu

Drug repurposing DMSO from an old solvent to a new candidate for the treatment of colitis and sepsis in male mice.

Normal microvascular function is critical for tissue perfusion, and intestinal vascular dysfunction is a major pathological feature of colitis and sepsis. Although dimethyl sulfoxide (DMSO) as a versatile solvent exerts anti-inflammatory and vasoactive properties in conduit vessels, its pharmacological effects on arterioles have not been explored. This study repurposed it as a potential therapeutic agent for these two gut-derived inflammatory diseases, aiming to elucidate its direct effects and underlying mechanisms on intestinal resistance arterioles under both physiological and pathological

ΔFleQ of Aeromonas hydrophila generated as a live attenuated vaccine in common carp (Cyprinus carpio).

Aeromonas hydrophila is a pathogen responsible for motile Aeromonas septicemia (MAS) in fish and an opportunistic pathogen that poses a significant threat to aquaculture and human health. Live attenuated vaccines are considered to be an effective and environmentally friendly method for the prevention and control of bacterial infectious diseases in aquaculture. The pathogenic mechanisms of bacterial flagella include assisting bacterial movement and attachment to the host's intestinal tract, as well as promoting biofilm formation. FleQ is a key gene that regulates flagellar assembly in A. hydrop

FGF1ΔHBS ameliorates DSS-induced ulcerative colitis by reducing neutrophil recruitment through the MAPK pathway.

BACKGROUND AND PURPOSE: Inflammatory bowel diseases (IBDs) constitute chronic inflammatory disease of the gastrointestinal tract, with escalating global prevalence. There is a pressing demand for safe and effective treatments for IBDs. Fibroblast growth factor 1 (FGF1) variant FGF1ΔHBS, characterised by reduced mitogenic capacity, has shown promising therapeutic potential in various inflammatory conditions, including obesity and diabetic nephropathy. Hence, exploring the therapeutic impact of FGF1ΔHBS on colitis is warranted. EXPERIMENTAL APPROACH: The protective role of FGF1ΔHBS was evaluate

Generally-healthy individuals with aberrant bowel movement frequencies show enrichment for microbially-derived blood metabolites associated with reduced kidney function.

Bowel movement frequency (BMF) has been linked to changes in the composition of the human gut microbiome and to many chronic conditions, like metabolic disorders, neurodegenerative diseases, chronic kidney disease (CKD), and other intestinal pathologies like irritable bowel syndrome and inflammatory bowel disease. Lower BMF (constipation) can lead to compromised intestinal barrier integrity and a switch from saccharolytic to proteolytic fermentation within the microbiota, giving rise to microbially-derived toxins that may make their way into circulation and cause damage to organ systems. Prevalence of Amoebiasis and Associated Complications in India: A Systematic Review.

PURPOSE: Intestinal protozoan parasites are responsible for diarrheal diseases causing significant morbidity and mortality. Entamoeba histolytica is the principle protozoan parasite associated with diarrheal infections. The infection is often associated with inaccessibility to clean drinking water and poor sanitary conditions in low- and middle-income countries including India. A comprehensive systematic review was performed to evaluate a reliable nationwide estimate for prevalence and geographic distribution of amoebiasis in India and the complications associated with it. METHODS: We used th

A netrin domain-containing protein secreted by the human hookworm Necator americanus protects against CD4 T cell transfer colitis.

The symbiotic relationships shared between humans and their gastrointestinal parasites present opportunities to discover novel therapies for inflammatory diseases. A prime example of this phenomenon is the interaction of humans and roundworms such as the hookworm, Necator americanus. Epidemiological observations, animal studies and clinical trials using experimental human hookworm infection show that hookworms can suppress inflammation in a safe and well-tolerated way, and that the key to their immunomodulatory properties lies within their secreted proteome. Herein we describe the identificati
