Development of In Vitro Potency Assays for Gene Therapy Products Targeting cGMP-Related Retinopathies.

The emergence of efficient viral vectors derived from adeno-associated viruses (AAV) has led many groups to develop gene therapies for inherited retinal diseases. To evaluate the potency of new gene therapy vectors in a preclinical context, it is common to use animal models, such as gene-deficient or mutant animal models of a given human disease, and then assess transgene expression and resulting restoration of vision with functional or behavioral assays. While such animal models are invaluable to the preclinical testing process, they cannot be readily used as batch release tests during manufa

Prime editing of a pathogenic Scn1a allele ameliorates seizure phenotypes in a GEFS+ mouse model.

Generalized epilepsy with febrile seizures plus (GEFS+) is an inherited epileptic disorder predominantly linked to autosomal-dominant, loss-of-function mutations in the sodium voltage-gated channel α subunit 1 (SCN1A) gene, which encodes the α subunit of the neuronal voltage-gated sodium ion channel type 1 (NaV1.1). Reduced NaV1.1 function in γ-aminobutyric acid (GABA)-ergic interneurons impairs inhibitory signaling and leads to neuronal hyperexcitability. Clinically, GEFS+ is characterized by a spectrum of seizure types, often beginning with febrile seizures in early childhood and progressing

Evolution in metazoans of the TRPM channel family involves multiple gains and losses of genes and domains.

Transient receptor potential (TRP) ion channels of the melastatin family (TRPM) have eight members in mammals with a broad spectrum of functions. We investigated the evolution of this complex gene family across metazoans. The characteristic aminoterminal melastatin domain and the carboxyterminal NUDT9 homology domain with similarity to ADP-ribose pyrophosphatase were added to the common ancestor of TRPM and its sister channel TRPS. Gene duplications before the origin of bilaterians resulted in four TRPM genes: α, β, βlike, and γ. The two latter were discovered in this study. All four and TRPS

AAV-mediated long-term TBX18 expression causes cardiac fibrosis and fails to induce pacemaker activity in rodents.

Gene therapy-based biological pacemakers have been proposed as an alternative to their hardware-based counterparts. In this context, short-term ectopic expression of the T-box transcription factor 18 (TBX18) in the ventricle has been reported to generate potent, short-term pacemaker function in various animal models. Here, we investigated the impact of adeno-associated virus-mediated (AAV-mediated), long-term expression of TBX18 and compared the outcomes with those of the pacemaker ion channel hyperpolarization-activated cyclic nucleotide-gated potassium and sodium channel 2 (Hcn2). Our findin

Propylene/Ethylene Separation via a Pressure-Induced Flexible Ultramicroporous Metal-Organic Framework.

Efficient separation of propylene/ethylene mixtures remains a formidable challenge because of the very similar physicochemical properties of the two olefins, particularly in methanol-to-olefins product streams. Herein, we report a pressure-responsive flexible ultramicroporous metal-organic framework, Co-PPA, constructed from pipemidic acid ligands and Co(II) ions, featuring pearl-strand-like one-dimensional channels and reversible guest-induced gate opening. At 298 K and 0.1 bar, Co-PPA exhibits a high C3H6 uptake of 25.21 cm3 g-1, whereas the uptake of C2H4 is only 0.26 cm3 g-1, affording an

Microfluidics-Enabled Nanoparticle for Multiscale Synergetic Osteoarthritis Therapy.

Osteoarthritis (OA) is a complex degenerative joint disease perpetuated by a vicious cycle of biomechanical, biochemical, and chondrocyte homeostasis imbalances, ultimately leading to progressive cartilage degradation. A simultaneous approach to biomechanical disturbances, chondrocyte metabolic imbalances, and maintaining cartilage homeostasis simultaneously holds promise in tackling this challenge. Herein, we report a microfluidics-enabled nanoparticle, termed ZCMC, formed by grafting a magnesium-modified chondroitin sulfate polymer layer onto a zein core preloaded with cannabidiol (CBD), for

A rare ORAI1 missense variant associates with risk of vascular diseases in White British adults.

BACKGROUND: Pathological remodelling of native vascular smooth muscle cells (VSMC) within the arterial wall is a key contributor to vascular disease. A driver of this remodelling is platelet-derived growth factor BB (PDGF-BB) and its signalling via activation of the store-operated calcium ion channel, ORAI1. Here, we investigated if there are associations of ORAI1 polymorphisms with human cardiovascular disease. METHODS AND RESULTS: We conducted candidate gene association analysis and revealed that a missense ORAI1 variant (rs3741596, S218G) associates with an increased risk of hospital-diagn

Enhanced calcium activity and transcriptomic alterations in iPSC-derived neurons from BAFME patients with repeat expansions.

Benign adult familial myoclonus epilepsy (BAFME) is caused by intronic TTTCA and TTTTA repeat expansions in SAMD12 and other genes; the neuronal basis of cortical hyperexcitability, however, remains unclear. We generated induced pluripotent stem cell (iPSC)-derived glutamatergic and GABAergic neurons from three BAFME1 patients and examined functional and transcriptomic phenotypes. Patient-derived neurons retained the pathogenic repeat expansions and showed a tendency toward upstream intronic RNA accumulation. Calcium imaging revealed increased spontaneous Ca2 + transient frequency in both neur

Hemp Flower Oil and Occupational Pain Management in Farming Communities: A Systematic Review.

Musculoskeletal pain is a major concern for agricultural workers worldwide. However, access to safe and non-opioid pain relief measures is limited in resource-scarce settings. Hemp flower oil, a rich source of cannabinoids and terpenes, has been recognised as a promising adjunct pain therapy due to its analgesic and anti-inflammatory activities through the activation of CB2 receptors, TRP channels, cytokine downregulation and antioxidant mechanisms. This study is a systematic scoping review with narrative synthesis conducted in accordance with PRISMA 2020 guidelines which aims at addressing th

Polymorphic ventricular tachycardia with mutation in KCNJ2: case report.

Polymorphic ventricular tachycardia (VT), particularly in the absence of structural heart disease, has a strong genetic foundation primarily rooted in mutations affecting cardiac ion channels and associated regulatory proteins. Here, we report two patients with polymorphic VT harboring potassium channel subfamily J member 2 (KCNJ2) gene mutations. The first case is a 13-year-old girl presenting with periodic paralysis, polymorphic VT, and a prolonged QT interval. She was sequentially treated with metoprolol, verapamil, amiodarone, moricizine, and mexiletine, along with three radiofrequency cat

Protein sorting and proteostasis mechanisms in CFTR-related exocrine pancreas dysfunction: A systematic narrative review.

The pancreas consists of exocrine and endocrine compartments. In the exocrine pancreas, cystic fibrosis transmembrane conductance regulator (CFTR) functions mainly in ductal epithelial cells as a chloride and bicarbonate channel. Its activity depends on proper protein folding, trafficking, and localization to the apical membrane. This systematic narrative review aims to synthesize the available evidence on the role of protein sorting machinery in CFTR channelopathies and its contribution to exocrine pancreatic dysfunction. A thorough search was conducted using PRISMA criteria on PubMed, Wiley

Avicularin and Senegenin Suppress TRPV3-Associated TSLP Expression in Human Keratinocytes.

Transient receptor potential vanilloid channel 3 (TRPV3) is a keratinocyte-expressed ion channel that regulates calcium signaling and thymic stromal lymphopoietin (TSLP) production, contributing to pruritic inflammatory skin diseases. Although avicularin (AVC), a flavonoid from Polygonum aviculare L., and senegenin (SNG), the bioactive aglycone of Polygala tenuifolia Willd., possess anti-inflammatory properties, their effects on TRPV3-associated signaling remain unknown. A natural product library was screened for nitric oxide (NO) production in carvacrol-stimulated human keratinocytes. Followi

Establishment of High Channel-Count Packaging in Active Implantable Medical Devices for Neuroprosthesis.

Neuroprostheses have contributed extensively to humankind's capacity to treat disease and injury. Hearing loss is routinely overcome; pain is controlled; and myriad neurological, sensory, motor, and psychological disorders are managed through interventions involving neuromodulation. Fabrication of active implantable medical devices (AIMDs) often relies on highly skilled operators, leading to high costs, reliability challenges, and limitations to further device miniaturization and manufacturing output. Extending AIMD benefits to other disorders may also require a substantial increase in the num

Scorpion venom peptides: Novel therapeutic approaches for inflammatory and hepatic disorders.

Chronic hepatic disorders, such as metabolic dysfunction associated steatohepatitis (MASH), alcohol associated liver disease (ALD), and viral hepatitis (Hepatitis B virus [HBV]/Hepatitis C virus [HCV]), are primarily driven by persistent immune-mediated inflammation and hepatic stellate cell activation leading to fibrosis, yet conventional therapies lack tissue and molecular specificity. Scorpion venom peptides, refined through evolutionary selection, provide highly potent, target specific scaffolds capable of modulating intrahepatic inflammatory networks. Recent in vivo preclinical studies in

Interaction of CFTR Modulators with Mammalian Membrane Mimetics: The Role of Cholesterol.

Lumacaftor and Ivacaftor are two FDA-approved medications currently used to treat cystic fibrosis (CF), a genetic disease caused by mutations in the cystic fibrosis transmembrane conductance regulator (CFTR), a chloride ion channel located in epithelial cell membranes; however, the detailed mechanism(s) of their action remains to be elucidated. Both drugs, termed modulators, bind CFTR at a protein-lipid interface, yet Lumacaftor acts at the endoplasmic reticulum (ER), while Ivacaftor acts at the plasma membrane (PM). A major difference among biological membranes is their level of cholesterol (

Effects of Blocking Multiple Sources of Calcium in Hippocampus During Spatial Learning and Memory Using a Rapid Acquisition Variant of the Morris Water Task.

Long-term potentiation (LTP) is proposed to be the molecular mechanism underlying learning and memory in the brain. A key event for LTP is the influx of calcium into post-synaptic neurons via multiple ion channel control systems. One such system involves N-methyl-D-aspartate receptors (NMDARs), which were originally believed to be essential for LTP and new learning. Recent studies have demonstrated that hippocampal NMDARs are critical for learning new spatial information in a novel environment; however, when pre-training occurs prior to new spatial learning, these receptors are not needed. Metabolite-centric perspectives on QT prolongation: Molecular mechanisms and clinical implications beyond the parent drug.

Prolongation of the heart-rate-corrected QT interval is a widely accepted surrogate for delayed ventricular repolarization and torsade de pointes risk. Although initial drug-safety efforts targeted parent compounds, accumulating evidence shows that circulating and tissue-accumulated metabolites are often the primary drivers of heart-rate-corrected QT interval change. This review synthesizes mechanistic, translational, and clinical findings to explain how metabolites prolong QT through several routes: direct human ether-à-go-go-related gene and other ion-channel inhibition, modulation of sodium

Optimization strategy for modeling sick sinus syndrome in rats: Balancing effect and animal care.

OBJECTIVE: This study aimed to validate the feasibility of establishing a sick sinus syndrome (SSS) rat model by injecting 3 % sodium hydroxide (NaOH) into the jugular vein at a rate of 0.01 mL/s, and to assess reductions in animal mortality and vascular injury. RESULTS: Compared with the conventional 10 % NaOH method, the modified 3 % NaOH approach yielded a significantly higher modeling success rate (78 % vs. 44 %), substantially lower mortality (15 % vs. 48 %), and reduced jugular vein injury. Electrophysiological evaluations demonstrated a time-dependent decrease in post-modeling heart ra

The proteostasis interactomes of trafficking-deficient variants of the voltage-gated potassium channel KV11.1 associated with long QT syndrome.

The voltage-gated potassium ion channel KV11.1 plays a critical role in cardiac repolarization. Genetic variants that render Kv11.1 dysfunctional cause long QT syndrome (LQTS), which is associated with fatal arrhythmias. Approximately 90% of LQTS-associated variants cause intracellular protein transport (trafficking) dysfunction, which pharmacological chaperones like E-4031 can rescue. Protein folding and trafficking decisions are regulated by chaperones, protein quality control factors, and trafficking machinery comprising the cellular proteostasis network. Here, we test whether trafficking d

Intravenous administration of recombinant Phα1β: Antinociceptive properties and morphine tolerance reversal in a cancer-associated pain model.

Cancer-related pain is considered one of the most prevalent symptoms for those affected by cancer, significantly influencing quality of life and treatment outcomes. Morphine is currently employed for analgesic treatment in this case, however, chronic use of this opioid is limited by the development of analgesic tolerance and adverse effects, such as digestive and neurological disorders. Alternative therapies, such as ion channel blockade, are explored. The toxin Phα1β has demonstrated efficacy in blocking calcium channels, making it a potential candidate for alleviating cancer-related pain. Th
