Discovery of Pyrazole-Fusaric Acid Derivatives as the Potential Herbicidal Lead Compounds Targeting Both Transketolase and Protoporphyrinogen Oxidase.

Transketolase (TKL, EC 2.2.1.1) was confirmed as a potential new target for herbicides. In this study, a series of pyrazole-fusaric acid derivatives 9a-9o were designed and synthesized via structural optimization of compound 7r. Among them, compound 9f exhibited good inhibition on Digitaria sanguinalis and Amaranthus retroflexus (inhibition >95%) even at 10 mg/L via the small cup method. It exhibited excellent inhibition against D. sanguinalis and A. retroflexus (inhibition >90%, 150 g ai/ha) via the pot bioassay superior to the positive control. Significantly, it showed good crop safety to ma

Deciphering the role of human peroxiredoxins for combating lead and zinc mediated cellular toxicity using a yeast model.

Elevated levels of heavy metals in cells lead to an imbalance of cellular redox homeostasis which is also seen in zinc and lead ion mediated toxicity. Human peroxiredoxins (hPrxI and hPrxII) are central regulators of redox balance; however, their role in mitigating heavy metal-induced stress remains unclear. Previously, we showed that the Saccharomyces cerevisiae model lacking TSA1 and TSA2 (tsa1tsa2Δ) showed growth defects in the presence of redox stressors that can be rescued by expressing hPrxI and hPrxII. In the present study, we utilized the tsa1tsa2Δ strain to understand the role of huma

Effect of Sm and Ni co-doping on optical and electrical properties of lead magnesate niobate titanate ceramics.

The ability to manipulate the bandgap while maintaining strong ferroelectricity in perovskite oxides offers numerous functional advantages, including multimodal energy harvesting/sensing, current modulation, dual-source actuation, electric-free poling, and enhanced bulk photovoltaic effect. However, bandgap engineering of Pb(Mg1/3Nb2/3)O3-PbTiO3 (PMN-PT) - renowned for its outstanding piezoelectric properties - via Ni2+ doping remains unexplored, despite its prior success in alkaline niobates. This study provides a comprehensive characterization of the structure-property relationships in Sm/Ni

Integrating machine learning and deep learning with multiple molecular fingerprints for topoisomerase I inhibitor screening and lead identification.

Topoisomerase I (TOP1) is a crucial anticancer target, but the development of traditional TOP1 inhibitors suffers from long research cycles, high costs, and low success rates. Existing artificial intelligence (AI)-driven studies lack systematic comparisons of molecular fingerprints and algorithms, as well as user-friendly predictive application tools. To address these gaps, this study retrieved TOP1 inhibitor activity data from the ChEMBL database, integrated five types of molecular fingerprints (AtomPairs, MACCS, Morgan, PharmacoPFP, and RDKitDes), and constructed and compared classical machi

Laser vs rotational transvenous lead extraction: A systematic review and meta-analysis of procedural safety and efficacy outcomes.

BACKGROUND: The rise in cardiac implantable electronic devices implantation has resulted in a concomitant rise in transvenous lead extractions (TLEs). Advanced extraction tools such as the excimer laser and mechanical rotational sheaths have improved acute procedural success when manual extraction fails. However, it is unclear how the laser sheath compares with the rotational sheath in safety and efficacy outcomes. OBJECTIVE: We aimed to compare the safety and efficacy of laser and rotational TLEs using a meta-analysis. METHODS: A systematic literature search was performed for studies involv

Prenatal lead exposure: associations with growth and anthropometry in early childhood in a UK observational birth cohort study.

BACKGROUND: Lead is a neurotoxic metal that crosses the placenta freely. It has adverse effects on a range of birth outcomes. The few studies reporting on the associations of prenatal exposure to lead and child growth have had conflicting results. This study aimed to examine the effect of prenatal exposure to lead on children's growth from 4 to 61 months of age. METHODS: Pregnant women were enrolled in the UK Avon Longitudinal Study of Parents and Children (ALSPAC). Whole blood samples for pregnancies with a live birth were analysed for lead (n = 4140). A 10% subsample of the offspring cohort

Caffeic Acid Mitigates Behavioral and Biochemical Alterations in Lead-induced Neurotoxicity in Rats With Possible Involvement of TFEB.

Lead (Pb) neurotoxicity is characterized by persistent cognitive and motor impairments that arise from converging disturbances in mitochondrial function, oxidative balance, and neuroinflammatory signaling. Increasing evidence suggests that these pathological outcomes are closely linked to disruption of Transcription Factor EB (TFEB), a key regulator of autophagy-lysosomal pathways and mitochondrial quality control. Impaired TFEB function can promote the accumulation of dysfunctional mitochondria and perpetuate oxidative and inflammatory cascades, thereby exacerbating Pb-induced neurodegenerati

Design, synthesis, and Lead optimization of novel Quinazoline-based FLT3 inhibitors with potent anti-acute myelogenous leukemia activity.

FLT3 mutations, including internal tandem duplications (ITD) and tyrosine kinase domain (TKD) variants, are key drivers of acute myeloid leukemia (AML) and represent attractive therapeutic targets. Guided by a scaffold-hopping strategy based on G-749 (denfivontinib), a series of quinazoline-based derivatives was designed and synthesized to explore structure-activity relationships (SAR). Among them, compound W4 showed the most promising profile, exhibiting potent antiproliferative activity against MV4-11 and MOLM-13 cells and strong inhibition of FLT3-ITD (IC50 = 16.0 nM) and FLT3-D835Y (IC50 =

Disc degeneration did not lead to an increased incidence of adjacent vertebral fractures in wedge-type OVCF patients.

BACKGROUND: Stress concentration is a primary pathogenesis of adjacent vertebral fracture (AVF) following percutaneous vertebroplasty (PVP) for osteoporotic vertebral compression fractures (OVCF). Disc degeneration (DD) is highly prevalent in the elderly, including OVCF patients. As a key biomechanical function of the intervertebral disc (IVD) is to ensure uniform load distribution, the progression of DD may impair this capacity, leading to localized stress concentrations. This study aims to investigate the impact of DD on AVF risk and elucidate its underlying biomechanical mechanisms. METHOD

Accuracy, Repeatability, and Reproducibility of a Radiographic Technique to Assess Spinal Cord Stimulation Lead Position: A Validation Study.

Spinal cord stimulation with implantable leads is a valuable therapy used to treat a variety of chronic pain conditions. However, lead migration is a common complication causing loss of efficacy. Previous reports have characterized lead migration using radiographs, but methods are not consistent and lack rigorous validation. The purpose of this study was to develop a technique to perform radiographic measurements of the position of epidural spinal cord leads within the lumbosacral spinal canal and establish its accuracy, repeatability, and reproducibility. Computed tomography scans were acquir

Do Uniform Fasting Instructions Lead to Reproducible Stomach Volumes in Patients Being Treated With Radiation to the Pancreas?

PURPOSE: This retrospective study determines if uniform fasting protocols successfully limit stomach volume variation and proximity to the planning target volume throughout radiation treatment. METHODS AND MATERIALS: A total of 74 patients with cancer were instructed to fast for 2 hours before receiving stereotactic body radiation therapy to their pancreas. The entire stomach and the stomach near the planning target volume (stomach_3cm) were contoured at simulation and on each treatment image. Patients were grouped into 3 clusters based on their SD in stomach volume across treatments. The sim

Defects in tissue-resident macrophages lead to smaller eardrums and abnormal neurovascular networks with increased middle-ear infection.

The tympanic membrane (TM), or eardrum, is essential for hearing. Macrophages, the primary innate immune cells, are densely distributed in the eardrum after birth, especially near blood vessels and nerve fibers. During postnatal development, neonatal tissue-resident macrophages (TRMs) gradually polarize, and their population declines. These dynamic changes closely parallel the maturation of neurovascular networks. What are the precursors of the early 'wave' of TRMs? Are they critical for postnatal TM development? Using fate mapping, single-cell RNA sequencing, and macrophage depletion, this st

Delayed Partial Protrusion of the Lead After Left Bundle Branch Pacing: The Importance of Programming-A Case Report.

Delayed lead perforation into the left ventricular (LV) cavity after left bundle branch area pacing (LBBAP) is less common than in the acute setting and could be associated with severe adverse events. We present the case of a young patient with symptomatic second-degree atrioventricular block who developed partial protrusion of the LBBAP lead into the LV cavity after discharge. While adequate conduction system capture was still seen with bipolar pacing, this potentially serious complication was self-diagnosed by the patient due to the initial ventricular pacing programming in a unipolar settin

Dual-Lead Spinal Cord Stimulation for Refractory Diabetic Small Fiber Neuropathy With Erythromelalgia-Like Features: A Case Report.

BACKGROUND: Small fiber neuropathy is a painful diabetes complication that may present with burning pain, dysesthesia, and autonomic features such as swelling, color changes, and hyperhidrosis. Some cases overlap with erythromelalgia, a rare condition marked by episodic erythema, temperature sensitivity, and vasomotor instability. Spinal cord stimulation has shown promise for refractory neuropathic pain, but evidence for its use for small fiber neuropathy and erythromelalgia is limited. CASE REPORT: A 32-year-old man with longstanding Type 1 diabetes reported more than a decade of progressive

Wastewater-based surveillance for seasonal respiratory viruses provides a one-week lead to clinical cases and hospital admissions.

Wastewater-based surveillance (WBS) is a complementary tool for infectious disease surveillance, providing independent data that is inclusive of entire populations. We evaluated the role of WBS in tracking seasonal respiratory viral infections caused by influenza A (IAV) and B (IBV), and respiratory syncytial virus (RSV) - pathogens responsible for significant morbidity and mortality and imposing substantial burdens on healthcare systems. Longitudinal surveillance was performed 1-3 times weekly from January 2022-June 2024, encompassing multiple respiratory virus seasons, across the three munic

Discovery of Arenarine D and its active derivatives as a novel lead targeting DYRK1B/MYH9 in hepatic fibrosis.

Hepatic fibrosis is characterized by excessive deposition of extracellular matrix (ECM) components, which can potentially progress to cirrhosis and hepatocellular carcinoma. In this study, a natural product, Arenarine D, was isolated from the endophytic fungus Pallidocercospora crystallina of Ajuga decumbens Thunb. Mechanistic studies established DYRK1B and MYH9 as potential targets of Arenarine D. Through systematic structural optimization, we obtained a series of derivatives of Arenarine D. Among them, the active derivatives 8a, 5b, 11a, and 11b demonstrated enhanced anti-fibrotic activity t

Impact of soil lead on children's urinary levels and parent-reported learning disabilities in areas of diffuse contamination.

Children's lead exposure has declined with the phasing out of leaded gasoline, while second order Pb sources, such as contaminated soils, have become more prevalent. Lead remains a pollutant of concern in contaminated urban and post-industrial areas, where legacy diffuse soil contamination levels are deemed acceptable with current soil management policies. However, the contribution of soil Pb, particularly its bioaccessible fraction, to children's internal Pb dose remains insufficiently characterized in these settings. This study investigates chronic Pb exposure in 172 children (6-11 years) li

Septal thickness to QRS voltage ratio in Lead I/aVR: A simple tool for estimating cardiac amyloid burden.

BACKGROUND: Cardiac amyloidosis (CA) is an infiltrative cardiomyopathy associated with adverse outcomes. Quantification of myocardial amyloid burden is essential for risk stratification. Although cardiac magnetic resonance (CMR)-derived extracellular volume (ECV) is the reference standard for non-invasive assessment, its use is limited by cost and availability. Therefore, simple and widely accessible tools to estimate amyloid burden are needed. This study aimed to evaluate whether a novel integrated echocardiographic-electrocardiographic (echo-ECG) index, based on the ratio of maximum septal t
