Segmentation and the development of the vertebrate nervous system.

1. Recent experiments on the development of neural segmentation in chick embryos are reviewed. 2. Segmentation of the spinal peripheral nerves is governed by a subdivision of the somite-derived sclerotome into anterior and posterior halves. Migrating neural crest cells and outgrowing motor axons are confined to the anterior sclerotome as a result, in part, of inhibitory interactions with posterior sclerotome cells. 3. The sclerotomal distribution of certain molecules known to influence growing nerve cells in vitro, namely laminin, fibronectin, N-CAM, N-Cadherin and J1/tenascin/cytotactin, sugg

Are sugars involved in the binding of rhodopsin-membranes by the retinal pigment epithelium?

The binding by the retinal pigment epithelium (RPE) cells of the embryonic chick, in culture, of recombinant membranes composed of phosphatidylcholine and bovine rhodopsin (rhodopsin-liposomes [RL]) was investigated to explore the influence that the carbohydrate groups of rhodopsin might have on this process. The reaction was quantitated by measuring the amount of RL associated with the cells using a radioimmunoassay for rhodopsin. The process was saturated at about 8 microM RL (calculated as rhodopsin-equivalents), and a two- to threefold greater rate and extent of binding was observed at 37

Case report: Ulocladium spp. (mold) catheter-related peritoneal dialysis-associated peritonitis.

Peritoneal dialysis-associated peritonitis (PD peritonitis) is a serious complication of PD, with fungal cases carrying high morbidity and mortality. We report a 65-year-old female with PD peritonitis caused by Ulocladium, a filamentous fungus not previously implicated in the literature. She was asymptomatic at presentation but noticed black spots in her PD catheter lumen; effluent analysis confirmed peritonitis. Initial cultures grew nonspecific mold, prompting treatment initiation with intravenous liposomal amphotericin B and PD catheter removal. Final cultures identified Ulocladium spp., se

Subclavian mycotic aneurysm caused by Aspergillus flavus in a child with acute lymphoblastic leukemia: a case report.

Invasive aspergillosis is a severe opportunistic infection in immunocompromised children, particularly those receiving chemotherapy for hematologic malignancies. We report the case of a 2-year-old girl with acute lymphoblastic leukemia who developed massive hemoptysis during consolidation chemotherapy. Thoracic computed tomography revealed a saccular pseudoaneurysm of the proximal left subclavian artery. Surgical resection and autologous vein graft replacement were performed, and Aspergillus flavus DNA was detected in the resected tissue using Aspergillus-specific polymerase chain reaction. Th

Secondary Hemophagocytic lymphohistiocytosis in an immunocompetent adult: a case report distinguishing sarcoidosis from histoplasmosis.

Distinguishing between histoplasmosis and sarcoidosis can be challenging as both can present with non-specific clinical findings. We present a 73-year-old immunocompetent Caucasian male who was recently diagnosed with sarcoidosis via left upper lobe biopsy who presented with lethargy and poor oral intake. The patient was found to have fever, splenomegaly, pancytopenia, hypofibrinogenemia, and elevated inflammatory markers. After bone marrow biopsy, he was diagnosed with disseminated histoplasmosis and secondary hemophagocytic lymphohistiocytosis (HLH). The patient was treated with liposomal am

Aspergillus Spondylodiscitis in an Immunocompetent Patient With Recurrent Aspergillus Endocarditis; A Clinical Case Report.

INTRODUCTION: Fungal endocarditis is a very uncommon and deadly illness that causes inflammation in the heart's lining, including the valves. Aspergillus endocarditis is the second most common cause of prosthetic endocarditis, especially the aortic valve, after Candida spp. Aspergillus endocarditis can occur on native and prosthetic valves, even in immunocompetent hosts. CASE REPORT: In this article, we describe a case of recurrent aortic-valve Aspergillus endocarditis occurring in a Caucasian man without previously known immunocompromised status with multiple brain septic emboli and spondylo

AI-guided design of a CXCR4-targeted core-shell nanocarrier for co-delivery of berberine/paclitaxel in cancer therapy.

An integrated three-step artificial-intelligence (AI) workflow was developed to accelerate the design of a CXCR4-targeted, dual-drug nanocarrier for colorectal cancer therapy. The workflow combined AI-based drug synergy prediction, peptide ligand discovery, and formulation optimization to create a core-shell nanocarrier consisting of a mesoporous silica core coated with a liposomal shell, co-delivering berberine (BBR) and paclitaxel (PTX). The nanocarrier exhibited efficient drug loading, sustained release, and selective uptake by CXCR4-positive cancer cells. In vitro, it synergistically inhib

Pulmonary Mucormycosis with Endobronchial Involvement Complicated by Pleural Effusion.

BACKGROUND: Pulmonary mucormycosis is an infection caused by a fungus with a severe and often fatal course, with a mortality rate exceeding 50%. It can spread through blood or lymphatic fluids or by inhalation of sporangiospores. Immunocompromised patients are especially susceptible to this high mortality rate. Histopathology is used to identify nonseptate hyphae in tissue, and culture is used to confirm the diagnosis. Treatment usually involves a combination of aggressive surgery and medication. CASE REPORT: A patient with diabetes mellitus received corticosteroid treatment for facial paraly

An 80-year-old man with a rare disease manifestation of disseminated cryptococcosis.

We present an extraordinary manifestation of disseminated cryptococcosis in a man who developed a triad of acute panuveitis, autoimmune hemolytic anemia, and pulmonary cryptococcal infection. The patient exhibited progressive clinical deterioration with mediastinal lymphadenopathy, fever and severe hemolysis. Despite initial negative cryptococcal antigen testing, histopathological examination of a pulmonary nodule demonstrated yeast forms consistent with cryptococcal infection. The diagnosis was confirmed through molecular diagnostics and isolation of Cryptococcus neoformans from vitreous humo

Focus on Trabectedin in Ovarian Cancer: What Do We Still Need to Know?

In the era of single and combination maintenance therapies as well as platinum and Poly (ADP-ribose) polymerase inhibitors (PARPi) resistance, the choice of subsequent treatments following first-line platinum-based chemotherapy in recurrent ovarian cancer (ROC) patients has become increasingly complex. Within the ovarian cancer treatment algorithm, particularly in the emerging context of PARPi resistance, the role of trabectedin, in combination with pegylated liposomal doxorubicin (PLD) still preserves its significance. This paper offers valuable insights into the multifaceted role and mechani

In vitro assessment of nanomedicines' propensity to cause palmar-plantar erythrodysesthesia: A Doxil vs. doxorubicin case study.

Palmar-plantar erythrodysesthesia (PPE), also known as hand and foot syndrome, is a condition characterized by inflammation-mediated damage to the skin on the palms and soles of the hands and feet. PPE limits the successful therapeutic applications of anticancer drugs. However, identifying this toxicity during preclinical studies is challenging due to the lack of accurate in vitro and in vivo animal-based models. Therefore, there is a need for reliable models that would allow the detection of this toxicity early during the drug development process. Herein, we describe the use of an in vitro sk

Nanotechnology-assisted combination drug delivery: a progressive approach for the treatment of acute myeloid leukemia.

Acute myeloid leukemia (AML), a heterogeneous hematopoietic cancer prevalent in adults, has been a leading cause of leukemia-associated deaths for decades. Despite advancements in understanding its pathology and pharmacological targets, therapeutic strategies have seen minimal change. The standard treatment, combining cytarabine and anthracycline, has persisted, accompanied by challenges such as pharmacokinetic issues and non-specific drug delivery, leading to severe side effects. Nanotechnology offers a promising solution through combination drug delivery. FDA-approved CPX351 (VYXEOS™) a lipo

Recurrent Mucor indicus central venous catheter infection in a five year old child on long term parenteral nutrition for short gut syndrome: could gut translocation be responsible?

A five year old girl with life-long TPN dependence for short gut syndrome presented with two episodes of non-fatal Mucor indicus central line associated blood stream infection (CLABSI). Each episode occurred fifteen months apart, without any evidence of ongoing mould infection whilst off antifungal therapy in the intervening time period. Both episodes were treated with removal of the infected central venous catheter (CVC) and 6 weeks of intravenous liposomal amphotericin B and/or posaconazole, with good clinical, microbiological, and radiological response. The possibility of gut translocation

Challenges in the treatment of cutaneous leishmaniasis caused by L. braziliensis in four travelers: a case series.

We describe a group of four travelers returning to the United States and Canada who acquired Leishmania braziliensis infection in the Peruvian Amazon. Pentavalent antimonials are the preferred treatment option for cutaneous leishmaniasis (CL) in most endemic countries in Central and South America. However, we initially treated our patients with liposomal amphotericin B (LAB) and miltefosine since these are the only two available Food and Drug Administration approved drugs in the United States. Refractory disease was common as three of the four travelers required repeated courses of miltefosine

Rhinocladiella mackenziei cerebral phaeohyphomycosis with a definitive outcome.

A right-handed man in his early 60s with hypertension controlled by a single prescription medication presented with left-sided heaviness and intermittent right occipital headache. Initial diagnostic workup was unremarkable. CT revealed an enhancing lesion located in the right parietal lobe, with mild mass effect on the right occipital horn, indicative of a brain abscess. The patient was initially treated with a course of empirical antibiotics, including ceftriaxone, vancomycin, metronidazole and dexamethasone. Case Report: Extensive Facial Cutaneous Leishmaniasis in a Neonate.

Cutaneous leishmaniasis (CL) is a skin infection caused by various species of the Leishmania parasite and is spread by the bite of an infected female sandfly. In southern Israel, CL caused by Leishmania major is endemic. Cutaneous leishmaniasis is considered a self-limiting disease, characterized by progressive, long-lasting nodulo-ulcerative skin lesions, which usually resolve in several months to years, and leads to scarring, cosmetic disfigurement, and future stigmatization. Although CL is a common disease among children, reports of CL in children younger than 1 year are rare. We present a
