Because of their small size, bacterial cells have long kept details about their inner workings a secret. We are starting to decipher their mechanistic secrets, in no small part due to the development of single-molecule and super-resolution fluorescence imaging, the subject of the 2014 Nobel Prize in Chemistry. These new methods have yielded a surge of discoveries about the subcellular organization and dynamics inside microbes. One example is an increased understanding of the virulence pathway within the cholera-causing microbe, Vibrio cholerae. Here, expression of the cholera toxin is regulate

Chlamydiae are obligate intracellular bacterial parasites that infect a wide range of metazoan hosts. Some Chlamydia species are important causes of chronic inflammatory diseases of the ocular, genital and respiratory tracts in humans. Genes located in a variable region of chlamydial genomes termed the plasticity zone are known to be key determinants of pathogenic diversity. The plasticity zone protein CT153, present only in select species, contains a membrane attack complex/perforin (MACPF) domain, which may mediate chlamydial interactions with the host cell. CT153 is present throughout the C

The objective of the study was to find out if human amniotic membrane could be used for corrective surgery after trauma to the orbital wall. Because of its proposed antiadhesive qualities, it seemed to be potentially suitable. We studied 8 men (mean age 37 (range 19-74) years) who had deficient ocular movement after fractures of the orbital floor. Five of them had already been operated on. Inclusion criteria were trauma dating back more than 4 months and a soft tissue stricture in the orbital floor diagnosed by magnetic resonance imaging. Patients were treated secondarily with lysis of adhesio

Bromosulfalein is an organic anion dye used in the study of a variety of membrane carriers expressed in animal tissues and involved in transport of drugs and metabolites. The spectrophotometric assay of electrogenic bromosulfalein transport in membrane vesicles, isolated from various mammalian organs or tissues, enables to specifically measure the transport activity of bilitranslocase (TCDB 2.A.65.1.1). The latter is a bilirubin- and flavonoid-specific transporter expressed in rat liver, the organ where its function has been best characterized. The spectrophotometric assay of electrogenic brom

Acquired resistance to antibiotics and other chemotherapeutic agents is a major problem in the practice of neurology and other branches of medicine. There are several mechanisms by which drug resistance is acquired. Multidrug transporters are important glycoproteins located in the cell membrane that actively transport small lipophilic molecules from one side of the cell membrane to the other, most often from the inside to the outside of a cell. They have important protective role yet may prove inconvenient in chemotherapy. In epilepsy and other disorders this mechanism augments the elimination

Effluents from recirculating aquaculture systems (RAS) contain high concentrations of nitrogen and phosphorous wastes and thus often require proper treatment to prevent potential detrimental impacts on receiving water bodies. The purpose of this study was to evaluate the feasibility of membrane filtration coupled with chemical precipitation as a pretreatment step with emphasis on phosphorus removal from RAS effluents. Chemical precipitation tests were conducted by adding magnesium chloride and alum at different chemical concentrations and pH values, respectively. Crossflow, flat-sheet membrane

PURPOSE: The tyrosine kinase receptor Tie2 and its ligands, the angiopoietins (Angs), play important roles in vascular integrity and neovascularization, modulating vascular endothelial growth factor (VEGF) activity. To elucidate the potential role of Angs and the Tie2 system in retinopathy of prematurity (ROP), we have investigated the expression of Angs, Tie2 and VEGF within fibroproliferative membranes in ROP.
METHODS: Fibroproliferative membranes were obtained from 38 cases with stage 5 ROP at the time of vitrectomy. Membranes were fixed in formalin and embedded in paraffin. Each specimen

Five consecutive patients underwent epikeratoplasty for keratoconus. Postoperatively, four patients had poor visual acuity (average, 20/200) secondary to folds in Descemet's membrane and interface scarring. Two underwent penetrating keratoplasty eight months later. Histopathologic examination of the host corneas and the overlying lenticules disclosed epithelial irregularity and subepithelial fibrosis. The host corneas showed folds in Descemet's membrane and focal posterior stromal fibrosis. Electron microscopy disclosed breaks in Bowman's membrane with irregular collagen, posterior aggregates

Biopolymeric nanofibers are crucial in tissue engineering, providing structural support and cell attachment. Electrospun membranes have shown potential in treating various wounds, including vascular injuries that require rapid hemostasis. In this study, a collagen (Coll)/chitosan (CTS) nanofiber membrane was fabricated via electrospinning for the topical delivery of tranexamic acid (TXA), an antifibrinolytic and first-aid agent. A key methodological innovation was the use of a non-cytotoxic PBS (1×):ethanol solvent system, ensuring biocompatibility of the final material. Collagen was extracted

Retinal fibrosis is a blinding complication of proliferative diabetic retinopathy (PDR), proliferative vitreoretinopathy (PVR), age-related macular degeneration (AMD) and idiopathic epiretinal membrane (iERM), yet effective therapies remain lacking. The purpose of this study was to investigate the role of BMP2 and BMP4 (BMP2/4)-activin receptor-like kinase (ALK) signaling in retinal fibrosis. BMP2 was detected in the vitreous of PDR, PVR, and rhegmatogenous retinal detachment (RRD) samples at concentration of 70, 180, and 85 pg/mL respectively, whereas it was present only at trace levels or wa

The plasma membrane (PM) is characterized by asymmetric bilayer organization in terms of both the chemistry of lipid headgroups and the physical properties of lipid acyl chains. Sphingomyelin (SM), a major outer-leaflet lipid, has long been considered a key partner of cholesterol (Chol) in the formation of ordered membrane domains. However, mammalian cells contain multiple SM species with distinct N-acyl chains, most predominantly C16:0, C24:0, and C24:1, whose structural differences can change membrane packing, Chol distribution, and interleaflet communication. Evidence from model membranes s

Transmembrane domains of immune complexes transmit precise signals across lipid bilayers. Probing their interactions has the potential to yield mechanistic insights relevant to therapeutic design. However, our capability to generate molecules that bind lipid-embedded sites is limited. Here, we created synthetic polypeptides targeting a prominent mediator of inflammatory signaling directly within membranes, Toll-like receptor 4 (TLR4), and clarify structural principles of its cross-membrane signaling mechanism. Doing so, we test emerging design principles for computationally encoding protein in

Increasing temperature fluctuations threaten crop productivity worldwide, emphasizing the need for a deeper understanding of plant adaptation to such extremes. Lipids are fundamental biological molecules that furnish structural, metabolic, and regulatory roles in plant growth and development, and responses to environmental stresses. The potential of lipids as key targets for crop improvement under changing climates is emerging. This systematic review and meta-analysis presents comprehensive syntheses of current knowledge on plant lipidome responses to heat and cold stresses. The analysis revea

OBJECTIVES: Pre-vascularization of the collagen membranes with autologous platelet concentrates is a standard procedure in oral and maxillofacial surgery. This study analyzed the possible interaction of an acellular collagen membrane of porcine origin (NM) with platelet-rich fibrin (PRF) regarding its rehydration protocol with differences in pH values and effect on angiogenesis.
MATERIALS AND METHODS: NM was analyzed alone and combined with solid PRF by plotting or co-culturing with injectable PRF. Different media (venous blood, buffer solution with a fixed pH value of 7, saline solution, and

Interactions between glycan-binding proteins (GBPs) and glycosphingolipids (GSLs) present in cell membranes are implicated in a wide range of biological processes. However, studying GSL binding is hindered by the paucity of purified GSLs and the weak affinities typical of monovalent GBP-GSL interactions. Native mass spectrometry (nMS) performed using soluble model membranes is a promising approach for the discovery of GBP ligands, but the detection of weak interactions remains challenging. The present work introduces MEmbrane ANchor-assisted nMS (MEAN-nMS) for the detection of low-affinity GBP

α-Synuclein (αS) is a presynaptic protein that binds to cell membranes and is linked to Parkinson's disease (PD). Binding of αS to membranes is a likely first step in the molecular pathophysiology of PD. The αS molecule can adopt multiple conformations, being largely disordered in water, adopting a β-sheet conformation when present in amyloid fibrils, and forming a dynamic multiplicity of α-helical conformations when bound to lipid bilayers and related membrane-mimetic surfaces. Multiscale molecular dynamics simulations in conjunction with nuclear magnetic resonance (NMR) and cross-linking mas

Cannabidiol (CBD) is the primary nonpsychotropic phytocannabinoid found in Cannabis sativa, which has been proposed to be therapeutic against many conditions, including muscle spasms. Among its putative targets are voltage-gated sodium channels (Navs), which have been implicated in many conditions. We investigated the effects of CBD on Nav1.4, the skeletal muscle Nav subtype. We explored direct effects, involving physical block of the Nav pore, as well as indirect effects, involving modulation of membrane elasticity that contributes to Nav inhibition. MD simulations revealed CBD's localization

Patients with end-stage renal disease are prone to developing a complication of hyperhomocysteinemia, manifesting as an elevation of the homocysteine (Hcy) concentration in human plasma. However, Hcy as a protein-bound toxin is barely removed by conventional hemodialysis membranes. Here, we report a novel hemodialysis membrane by preparing a bioactive coating of pyridoxal 5'-phosphate (PLP) and adding biocompatible hyperbranched polyglycerol (HPG) brushes to achieve Hcy removal. The dip-applied PLP coating, a coenzyme with a role in Hcy metabolism, dramatically promoted a decrease in the Hcy c

BACKGROUND: Defective complement inhibition can lead to the formation of membrane attack complexes (MAC; C5b-9) on the plasma membranes of vascular endothelial cells, resulting in injury that drives the progression of thrombotic microangiopathy (TMA), a key pathology in kidney disease.
OBJECTIVE/METHODS: We examined the response of human endothelial cells to complement-mediated damage using blood outgrowth endothelial cells (BOECs) derived from healthy donors. BOECs were sensitized to complement factors present in normal human serum to induce the formation of C5b-9 on their plasma membranes.
