Proteome-wide Mendelian randomization identifies causal links between blood proteins and severe COVID-19.

In November 2021, the COVID-19 pandemic death toll surpassed five million individuals. We applied Mendelian randomization including >3,000 blood proteins as exposures to identify potential biomarkers that may indicate risk for hospitalization or need for respiratory support or death due to COVID-19, respectively. After multiple testing correction, using genetic instruments and under the assumptions of Mendelian Randomization, our results were consistent with higher blood levels of five proteins GCNT4, CD207, RAB14, C1GALT1C1, and ABO being causally associated with an increased risk of hospital

Estimating causal effects of atherogenic lipid-related traits on COVID-19 susceptibility and severity using a two-sample Mendelian randomization approach.

BACKGROUND: As the number of COVID-19 deaths continues to rise worldwide, the identification of risk factors for the disease is an urgent issue, and it remains controversial whether atherogenic lipid-related traits including serum apolipoprotein B, low-density lipoprotein (LDL)-cholesterol, and triglyceride levels, are risk factors. The aim of this study was to estimate causal effects of lipid-related traits on COVID-19 risk in the European population using a two-sample Mendelian randomization (MR) approach. METHODS: We used summary statistics from a genome-wide association study (GWAS) that

Association and mediation pathways of maternal hyperglycaemia and liability to gestational diabetes with neonatal outcomes: A two-sample Mendelian randomization study.

AIMS: Maternal hyperglycemia is linked to adverse neonatal outcomes. However, current evidence was insufficient for mechanistic pathways. We aim to use two-sample Mendelian randomization (MR) to obtain a comprehensive understanding of the causal association and mediation pathways. MATERIALS AND METHODS: Genetic variants of fasting glucose (FG), insulin sensitivity index (ISI), glycated haemoglobin (HbA1c), gestational diabetes mellitus (GDM) and type 2 diabetes (T2D) were used as instruments (N = 50 404-898 130). The associations with offspring birthweight, gestational duration, spontaneous p

Colon importance in short bowel syndrome.

BACKGROUND: Aging can be understood as a consequence of the declining force of natural selection with age. Consistent with this, the antagonistic pleiotropy theory of aging proposes that aging arises from trade-offs that favor early growth and reproduction. However, evidence supporting antagonistic pleiotropy in humans remains limited. METHODS: Mendelian randomization (MR) was applied to investigate the associations between the ages of menarche or first childbirth and age-related outcomes and diseases. Ingenuity Pathway Analysis was employed to explore gene-related aspects associated with sig

Use of Bioinformatics and Machine Learning to Identify Circadian Rhythm Disruption- and Endoplasmic Reticulum Stress-Associated Biomarkers in Nonalcoholic Fatty Liver Disease.

Nonalcoholic fatty liver disease (NAFLD) is a common chronic liver disorder closely linked to circadian rhythm disruption (CRD) and endoplasmic reticulum stress (ERS). We aimed to identify CRD- and ERS-associated biomarkers in NAFLD samples and elucidate their roles in shaping disease pathogenesis. We first performed Mendelian randomization (MR) to identify circadian rhythm-related genes (CRRGs) with putative causal effects on NAFLD. Differential expression analysis was conducted via GSE126848 (discovery dataset) to identify differentially expressed genes (DEGs) between NAFLD and controls. Wei

Association of vitamin D status with arterial blood pressure and hypertension risk: a mendelian randomisation study.

BACKGROUND: Low plasma 25-hydroxyvitamin D (25[OH]D) concentration is associated with high arterial blood pressure and hypertension risk, but whether this association is causal is unknown. We used a mendelian randomisation approach to test whether 25(OH)D concentration is causally associated with blood pressure and hypertension risk. METHODS: In this mendelian randomisation study, we generated an allele score (25[OH]D synthesis score) based on variants of genes that affect 25(OH)D synthesis or substrate availability (CYP2R1 and DHCR7), which we used as a proxy for 25(OH)D concentration. We me

Role of TRAIL in mediating the effect of lipidome on breast cancer: a Mendelian randomization study.

BACKGROUND: Breast cancer (BC) is the most common malignancy in women and remains a major cause of cancer-related death. Growing observational and multi-omics evidence suggests that dysregulated lipid metabolism and alterations in circulating lipidomes are involved in BC development, yet their causal relationships and underlying mechanisms remain unclear. This study aimed to elucidate the causal effect of lipidomes on BC, as well as its possible mechanism of action, and to investigate the mediating effect of tumor necrosis factor (TNF)-related apoptosis-inducing ligand (TRAIL) on the risk of B

CR1(+) tumor-associated macrophages orchestrate an immunosuppressive niche in hepatocellular carcinoma: a genetic and multi-omics dissection.

BACKGROUND: Hepatocellular carcinoma (HCC) remains a major global health burden and a leading cause of cancer-related mortality. Advanced disease is characterized by a profoundly immunosuppressive tumor microenvironment (TME) and limited durable responses to therapy. However, the upstream genetic determinants that drive tumor-associated macrophage (TAM) dysfunction in HCC remain poorly defined. Using an integrative genetic and multi-omics framework, we investigated complement receptor 1 (CR1) as a candidate regulator of this immunosuppressive niche. METHODS: We combined Mendelian randomizatio

Integrative profiling of lactylation reveals prognostic biomarkers and an immunosuppressive niche in acute myeloid leukemia.

BACKGROUND: The overall survival rate of acute myeloid leukemia (AML) remains less than 30%. Metabolic reprogramming of leukemia cells, such as the Warburg effect, enables them to adapt to the microenvironment and thereby develop. Elucidating the landscape of lactate regulation in AML helps clarify the pathogenesis from the perspective of metabolic reprogramming and identify possibilities for optimizing current treatment modalities. METHODS: RNA and single-cell sequencing data for AML were obtained from the Gene Expression Omnibus (GEO) and The Cancer Genome Atlas (TCGA) databases. Seurat, li

Obesity, White Matter Hyperintensities, and Cognitive Decline: Sex-Specific DTI-ALPS Index Mediation.

BACKGROUND: Obesity is a risk factor for white matter hyperintensities (WMH) and cognitive decline, but underlying mechanisms regarding sex differences, obesity metrics, and the glymphatic system remain unclear. METHODS: Using data from the UK Biobank, we performed sex-stratified analyses. Our approach integrated obesity trajectory modeling (over a mean 9-year period), genome-wide association studies, and 2-sample Mendelian randomization. We examined associations of body mass index, waist-hip ratio, and body fat percentage with glymphatic system function assessed via the diffusion tensor imag

Erucic acid aggravates cardiac fibrosis after myocardial infarction by CD36-mediated metabolic reprogramming with impaired mitochondrial aerobic oxidation.

BACKGROUND: Post-myocardial infarction (MI) cardiac fibrosis is a key driver of heart failure, with dysregulated cardiac metabolism playing a central role. The specific impact of circulating fatty acid metabolites on mitochondrial function and fibrotic remodeling remains unclear. Erucic acid, a very-long-chain fatty acid found in certain edible oils, has a historical association with cardiac lipidosis, yet its causal role and mechanism in post-MI fibrosis are unknown. METHODS: We employed an integrative, two-stage strategy. First, a hypothesis-free two-sample Mendelian Randomization (MR) anal

Assessing causal relationships between iron level, disorders of iron metabolism, and hepatocellular carcinoma: a Mendelian randomisation study : The first author.

BACKGROUND: Emerging evidence indicates that elevated systemic iron levels and dysregulated iron metabolism are closely associated with the pathogenesis of hepatocellular carcinoma (HCC). Observational studies have linked iron overload (e.g., hereditary hemochromatosis) to an increased risk of HCC, potentially mediated through iron-induced oxidative stress, DNA damage, and chronic inflammatory responses. However, the causal relationship between specific iron-related factors-such as organ-specific iron accumulation, systemic iron metabolism disorders, or exogenous iron exposure (dietary/drug ir
